RESUMO

Creativity, a high-order cognitive ability, has received wide attention from researchers and educators who are dedicated to promoting its development throughout one's lifespan. Currently, creativity is commonly assessed with divergent thinking tasks, such as the Alternative Uses Task. Recent advancements in neuroimaging techniques have enabled the identification of brain markers for high-order cognitive abilities. One such brain structure of interest in this regard is the hippocampus, which has been found to play an important role in generating creative thoughts in adulthood. However, such role of the hippocampus in childhood is not clear. Thus, this study aimed to investigate the associations between creativity, as measured by divergent thinking, and both the volume of the hippocampus and its resting-state functional connectivity in 116 children aged 8-12 years. The results indicate significant relations between divergent thinking and the volume of the hippocampal head and the hippocampal tail, as well as the volume of a subfield comprising cornu ammonis 2-4 and dentate gyrus within the hippocampal body. Additionally, divergent thinking was significantly related to the differences between the anterior and the posterior hippocampus in their functional connectivity to other brain regions during rest. These results suggest that these two subregions may collaborate with different brain regions to support diverse cognitive processes involved in the generation of creative thoughts. In summary, these findings indicate that divergent thinking is significantly related to the structural and functional characteristics of the hippocampus, offering potential insights into the brain markers for creativity during the developmental stage.

BACKGROUND: Early iron deficiency (ID) is a common risk factor for poorer neurodevelopment, limiting children's potential and contributing to global burden. However, it is unclear how early ID alters the substrate of brain functions supporting high-order cognitive abilities and whether the timing of early ID matters in terms of long-term brain development. This study aimed to examine the effects of ID during fetal or early postnatal periods on brain activities supporting proactive and reactive cognitive control in pre-adolescent children. METHODS: Participants were part of a longitudinal cohort enrolled at birth in southeastern China between December 2008 and November 2011. Between July 2019 and October 2021, 115 children aged 8-11 years were invited to participate in this neuroimaging study. Final analyses included 71 children: 20 with fetal ID, 24 with ID at 9 months (postnatal ID), and 27 iron-sufficient at birth and 9 months. Participants performed a computer-based behavioral task in a Magnetic Resonance Imaging scanner to measure proactive and reactive cognitive control. Outcome measures included accuracy, reaction times, and brain activity. Linear mixed modeling and the 3dlme command in Analysis of Functional NeuroImages (AFNI) were separately used to analyze behavioral performance and neuroimaging data. RESULTS: Faster responses in proactive vs. reactive conditions indicated that all groups could use proactive or reactive cognitive control according to contextual demands. However, the fetal ID group was lower in general accuracy than the other 2 groups. Per the demands of cues and targets, the iron-sufficient group showed greater activation of wide brain regions in proactive vs. reactive conditions. In contrast, such condition differences were reversed in the postnatal ID group. Condition differences in brain activation, shown in postnatal ID and iron-sufficient groups, were not found in the fetal ID group. This group specifically showed greater activation of brain regions in the reward pathway in proactive vs. reactive conditions. CONCLUSIONS: Early ID was associated with altered brain functions supporting proactive and reactive cognitive control in childhood. Alterations differed between fetal and postnatal ID groups. The findings imply that iron supplement alone is insufficient to prevent persisting brain alterations associated with early ID. Intervention strategies in addition to the iron supplement should consider ID timing.

Traditional oxidase-like (OXD) nanozymes rely primarily on O2-mediated superoxide anion (O2·-) process for catalytic oxidation and organophosphorus (Ops) detection. While during the actual detection process, the concentration of O2 is inconstant that can be easily changed with the external environment, distorting detection results. Herein, highly-oxidizing Au@MnO2-X nanozymes with core-shell nanostructure are designed which trigger substantial electron transfer from inner Au core to outer ultrathin MnO2-X layer. According to experimental and theoretical calculations, the core-shell nanostructure and ultrathin MnO2-X of Au@MnO2-X result in the large surface defects, high oxygen vacancies and MnIII ratios. The specially structured Au@MnO2-X nanozymes are therefore highly-oxidizing and the catalytic oxidation can be completed merely through electrons transferring instead of the O2-mediated O2·- process. Based on this, an oxygen independent and ultrasensitive nanozyme-based sensor is established using homogeneous electrochemistry (HEC), its Ops is detected at a LOD of 0.039 ng mL-1. Combined with the UV-vis spectrum of 3,3',5,5'-tetramethylbenzidine (TMB), the linear discriminant analysis of five Ops i.e., Ethion, Omethoate, Diazinon, Chlorpyrifos methyl and Dipterex has achieved superior discrimination results. Therefore, HEC based on strong oxidizing nanozymes provide a new avenue for the development of high-performance electrochemical sensors and demonstrate potential applicability to pesticide residue determination in real samples.

Physical activity is critical for maintaining cognitive and brain health. Previous studies have indicated that the effect of physical activity on cognitive and brain function varies between individuals. The present study aimed to examine whether mind wandering modulated the relations between physical activity and resting-state hippocampal functional connectivity. A total of 99 healthy adults participated in neuroimaging data collection as well as reported their physical activity in the past week and their propensity to mind wandering during typical activities. The results indicated that mind wandering was negatively related to the resting-state functional connectivity between hippocampus and right inferior occipital gyrus. Additionally, for participants with higher level of mind wandering, physical activity was negatively related to hippocampal connectivity at left precuneus and right precentral gyrus. In contrast, such relations were positive at right medial frontal gyrus and bilateral precentral gyrus for participants with lower level of mind wandering. Altogether, these findings indicated that the relations between physical activity and hippocampal functional connectivity vary as a function of mind wandering level, suggesting that individual differences are important to consider when we aim to maintain or improve cognitive and brain health through increasing physical activity.

BACKGROUND: Talar cartilage injury is a kind of disease that causes long-term and chronic pain of ankle joint. Autologous osteochondral transplantation has been viewed as an alternative choice for treating these lesions, but donor-site morbidity has limited its application. This study aimed to analyze the efficacy of iliac bone autografting for Hepple V osteochondral lesions of the talus. METHODS: This retrospective study included 32 patients surgically treated for Hepple V osteochondral lesions of the talus from January 2015 to January 2020. All patients underwent open surgery. Ipsilateral iliac bone grafts were taken and filled with talar cartilage injury area. The improvement of postoperative ankle pain was evaluated by Visual Analogue Scale (VAS), and the improvement of ankle function was evaluated by the American Orthopaedic Foot & Ankle Society (AOFAS). During the postoperative follow-up, X-ray examination of the front and side of the ankle joint and CT of the ankle joint were performed to evaluate the bone cartilage healing in the graft area. RESULTS: Thirty-two patients (32 ankles) (100%) returned for clinical and radiologic follow-up at an average of 28 (range 24-36) months postoperatively. At 3 months postoperatively and at the last follow-up, the AOFAS scores were (80.4 ± 3.6) and (89.2 ± 6.4), respectively, which were significantly improved compared with the preoperative score (49.7 ± 8.1), and the difference was statistically significant (P < 0.05). The VAS scores were (2.1 ± 0.9) and (1.5 ± 0.8), respectively, which were significantly better than the preoperative score (6.2 ± 1.7), and the difference was statistically significant (P < 0.05). Re-examination of the front and side of the ankle joint X-rays and CT showed that the bone healing at the osteotomy of medial malleolus and osteochondral transplantation area. All patients had no pain at the donor site. No complications occurred in 32 patients at the last follow-up. CONCLUSIONS: With iliac bone autografting for Hepple V osteochondral lesions of the talus can effectively relieve ankle joint pain and significantly improved ankle function. Exercise fatigue can exert deleterious effects on the body. This study evaluated the effects and mechanisms by which Lonicera caerulea berry polyphenols extract (LCBP) improved the treadmill endurance of mice. Comparison was performed between the effects at 25°C and low temperatures (-5°C). Energy storage, product metabolism, and other biochemical indices were determined using vitamin C (VC) as a positive control. Co-immunoprecipitation was performed to detect the interaction between different proteins. Dietary supplementation with LCBP significantly prolonged the exhaustion time during treadmill exercise by 20.4% (25 °C) and 27.4% (-5 °C). LCBP significantly regulated the expression of antioxidant and inflammatory proteins, Bcl-2 /Bax apoptosis proteins, and the PKCα -NOx2 / Nox4 pathway proteins, and activated the expression of AMPK-PGC1α -NRF1-TFAM proteins in skeletal muscle mitochondria. The gene and protein expression of miRNA-133a/IGF-1/PI3K/Akt/mTOR in skeletal muscle cells was also activated. Molecular docking confirmed that the main components of LCBP such as cyanidin-3-glucoside, catechin, and chlorogenic acid, have strong binding affinity toward AMPKα. LCBP alleviates exercise fatigue in mice by reducing oxidative stress, inflammation, and apoptosis of skeletal muscle cells, enhances mitochondrial biosynthesis and cell proliferation, reduces fatigue, and enhances performance. These effects are also significant in a low-temperature environment (Graphical Abstract). Consequently, these results provide novel insights into the anti- fatigue roles of LCBP in exercise fatigue.

HE is a natural extract with strong antioxidant capacity. Drosophila melanogaster was used to explore HE could delay aging in this study. We detected that 3 mg/ml HE could increase stress tolerance (heat, cold, starvation, oxidative stress), reduce intestinal dysfunction, and prolong the lifespan of D. melanogaster. Network pharmacology analysis showed HE could act through the PI3K-Akt pathway. Meanwhile, HE intervention inhibited the gene expression of InR, PI3K, and Akt-1, and further increased the gene expression of Atg1, Atg5, Atg8a, and Atg8b. Furthermore, HE inhibited the unnatural propagation of ISCs and increased the number of lysosomes. Supplement with HE may be an effective intervention for aging D. melanogaster. PRACTICAL APPLICATIONS: In recent years, diseases that come with aging have seriously affected people's healthy life. Hawthorn is a kind of nutrient-rich substance that is rich in flavonoids and thus has many potential biological and pharmacological functions. Our results showed that HE has good antioxidant properties and can maintain intestinal homeostasis, which provides a good theoretical basis for the development and research using HE as an effective natural antioxidant for the elderly.

The medical field has gradually become intelligent, and information and the research of intelligent medical diagnosis information have received extensive attention in the medical field. In response to this situation, this paper proposes a Hadoop-based medical big data processing system. The system first realized the ETL module based on Sqoop and the transmission function of unstructured data and then realized the distributed storage management function based on HDFS. Finally, a MapReduce algorithm with variable key values is proposed in the data statistical analysis module. Through simulation experiments on the system modules and each algorithm, the results show that because the traditional nondistributed big data acquisition module transmits the same scale of data, it consumes more than 3200 s and the transmission time exceeds 3000 s. The time consumption of smart medical care under the 6G protocol is 150 s, the transmission time is 146 s, and the time is reduced to 1/10 of the original. The research of intelligent medical diagnosis information based on big data has good rationality and feasibility.

Physical activity may improve cognitive control and episodic memory. Cognitive control could exert positive or negative influences on episodic memory. This study aimed to test whether physical activity modulated the effect of proactive and reactive control on episodic memory. Participants reported their physical activity in the past week, encoded episodic memory incidentally in proactive and reactive conditions, and subsequently retrieved their memories of items and sources. Subsequent item memory was better when items were encoded in proactive vs. reactive condition. Smaller condition difference in subsequent item memory was related to better cognitive control ability. Cognitive control completely mediated the relation between physical activity and the condition difference in subsequent item memory. Additionally, condition difference in subsequent source memory was negatively related to cognitive control. After controlling for cognitive control, greater physical activity was positively related to the difference in subsequent source memory between proactive and reactive conditions. Altogether, the findings suggested that physical activity modulated the effect of proactive and reactive control on subsequent item memory through improving cognitive control ability, but it was independent of cognitive control for subsequent source memory.

The increasing emergence of multi-drug resistant Escherichia coli (E. coli) has become a global concern, primarily due to the limitation of antimicrobial treatment options. Phage therapy has been considered as a promising alternative for treating infections caused by multi-drug resistant E. coli. However, the application of phages as a promising antimicrobial agent is limited by their narrow host range and specificity. In this research, a recombinant T4-like phage, named WGqlae, has been obtained by changing the receptor specificity determinant region of gene 37, using a homologous recombination platform of T4-like phages established by our laboratory previously. The engineered phage WGqlae can lyse four additional hosts, comparing to its parental phages WG01 and QL01. WGqlae showed similar characteristics, including thermo and pH stability, optimal multiplicity of infection and one-step growth curve, to the donor phage QL01. In addition, sequencing results showed that gene 37 of recombinant phage WGqlae had genetically stable even after 20 generations. In planktonic test, phage WGqlae had significant antimicrobial effects on E. coli DE192 and DE205B. The optical density at 600 nm (OD600) of E. coli in phage WGqlae treating group was significantly lower than that of the control group (P < 0.01). Besides, phage WGqlae demonstrated an obvious inhibitory effect on the biofilm formation and the clearance of mature biofilms. Our study suggested that engineered phages may be promising candidates for future phage therapy applications against pathogenic E. coli in planktonic and biofilm forms.

Escherichia coli (E. coli) strains, from the gut of animals and humans, harbor wide range of drug resistance genes. A comparative study is conducted on the intestinal E. coli from fecal samples of healthy chicken from China and Sudan in order to monitor the antimicrobial sensitivity pattern. A number of 250 E. coli isolates from chicken farms, including 120 from China and 130 from Sudan, were isolated and identified. All isolates were subjected to susceptibility tests against 10 antibiotics and the distribution of antibiotic resistant genes was confirmed by PCR amplification, involving genes such as ampC, tetA, pKD13, acrA, ermA, ermB, ermC, tetB, mphA, aadA14, aadA1, aac3-1, and aac3- III. Many isolates were found to exhibit resistance against more than one antibiotic. However, the Chinese isolates showed more antibiotics resistance and resistance genes compared to the Sudanese isolates. For better understanding of the multidrug resistance factors, we conducted whole genome analyses of E. coli D107 isolated from China, which revealed that the genome possesses multiple resistance genes including tetracycline, erythromycin, and kanamycin. Furthermore, E. coli D4 isolate from Sudan was more sensitive to antibiotics such as erythromycin, tetracycline, and gentamicin. After analysis by RAST and MAUVE, the two strains showed 89% average nucleotide identity. However, the genomes mostly differed at the number of antibiotics-related genes, as the genome of D107 revealed a considerable number of antibiotics resistance genes such as ermA and mphD which were found to be absent in D4 genome. These outcomes provided confirmation that the poultry farms environment in different countries (China and Sudan) may serve as a potential reservoir of antimicrobial resistance genes and also indicated the evolutionary differences of strains in terms of resistant genes expression.
