RESUMO

A Gram-stain-variable, strictly anaerobic, rod-shaped, catalase-negative and endospore-forming bacterial strain, designated MJC39T, was isolated from grass silage preserved in Hokkaido, Japan. Growth occurred at 20-42 °C, pH 5.0-7.0 and NaCl concentrations up to 2 % (w/v). The isolated strain MJC39T produced butyric acid in peptone yeast extract medium with glucose. The DNA G+C content of strain MJC39T was 34.4±0.2 mol%. The major cellular fatty acids (>10 %) were C14 : 0, C16 : 0 and summed feature 3 (including C16 : 1ω7c/C16 : 1ω6c). No respiratory quinones were detected. The polar lipids of strain MJC39T were phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, one unidentified lipid, one unidentified aminolipid, two unidentified glycolipids, one unidentified phospholipid, one unidentified aminoglycolipid and one unidentified phosphoaminoglycolipid. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain MJC39T was a member of the genus Clostridium and is closely related to Clostridium tyrobutyricum JCM 11008T (95.8 % similarity) and Clostridium algifaecis MB9-7T (95.5 % similarity). Based on the genotypic, phenotypic and chemotaxonomic characteristics, strain MJC39T represents a novel species of the genus Clostridium, for which the name Clostridium pabulibutyricum sp. nov. is proposed. The type strain is MJC39T (=JCM 31506T=DSM 103944T).

OBJECTIVE: This study aimed to evaluate whether the methane (CH4) to carbon dioxide (CO2) ratio (CH4/CO2) and methane-related traits obtained by the sniffer method can be used as indicators for genetic selection of Holstein cows with lower CH4 emissions. METHODS: The sniffer method was used to simultaneously measure the concentrations of CH4 and CO2 during milking in each milking box of the automatic milking system to obtain CH4/CO2. Methane-related traits, which included CH4 emissions, CH4 per energy-corrected milk, methane conversion factor (MCF), and residual CH4, were calculated. First, we investigated the impact of the model with and without body weight (BW) on the lactation stage and parity for predicting methane-related traits using a first on-farm dataset (Farm 1; 400 records for 74 Holstein cows). Second, we estimated the genetic parameters for CH4/CO2 and methane-related traits using a second on-farm dataset (Farm 2; 520 records for 182 Holstein cows). Third, we compared the repeatability and environmental effects on these traits in both farm datasets. RESULTS: The data from Farm 1 revealed that MCF can be reliably evaluated during the lactation stage and parity, even when BW is excluded from the model. Farm 2 data revealed low heritability and moderate repeatability for CH4/CO2 (0.12 and 0.46, respectively) and MCF (0.13 and 0.38, respectively). In addition, the estimated genetic correlation of milk yield with CH4/CO2 was low (0.07) and that with MCF was moderate (-0.53). The on-farm data indicated that CH4/CO2 and MCF could be evaluated consistently during the lactation stage and parity with moderate repeatability on both farms. CONCLUSION: This study demonstrated the on-farm applicability of the sniffer method for selecting cows with low CH4 emissions.

This study proposed a method for measuring the methane (CH4 )/carbon dioxide (CO2 ) ratio from individual beef cattle under on-farm conditions and estimated the variance components of the CH4 /CO2 ratio. Gas measurements were conducted using 166 Japanese Black cattle group-housed in pens equipped with individual feed bins. The gas containing the animal's breath was measured individually after concentrate feeding by covering the feed bin with a sheet with sampling inlets. Measurements were performed six times (three consecutive days, twice daily) per individual. Most of the sampled gas contained more than 1000 ppm of the mean background-corrected CO2 , suggesting that the method proposed in this study successfully collected sufficient breath concentration to accurately measure the CH4 /CO2 ratio. The between-animal variance accounted for 31.7% of the total variance in the CH4 /CO2 ratio. The results showed that the gas collection method proposed in this study could be a useful tool for measuring the CH4 /CO2 ratio under on-farm conditions. The variance component obtained from this study will help to establish protocols for generating data for genetic evaluation and performing dietary experiments with sufficient statistical power.

This study investigated the physiological characteristics and carcass performance associated with residual methane emissions (RME), and the effects of bull differences on CH4-related traits in Japanese Black cattle. Enteric methane (CH4) emissions from 156 Japanese Black cattle (111 heifers and 45 steers) were measured during early fattening using the sniffer method. Various physiological parameters were investigated to clarify the physiological traits between the high, middle, and low RME groups. CH4-related traits were examined to determine whether bull differences affected progeny CH4 emissions. Ruminal butyrate and NH3 concentrations were significantly higher in the high-RME group than in the low-RME group, whereas the propionate content was significantly higher in the low-RME group. Blood urea nitrogen, ß-hydroxybutyric acid, and insulin concentrations were significantly higher, and blood amino acids were lower in the high-RME group than in the other groups. No significant differences were observed in the carcass traits and beef fat composition between RME groups. CH4-related traits were significantly different among bull herds. Our results show that CH4-related traits are heritable, wherein bull differences affect progeny CH4 production capability, and that the above-mentioned rumen fermentations and blood metabolites could be used to evaluate enteric methanogenesis in Japanese Black cattle.

In Japan, Japanese Black cattle, known for their exceptional meat quality owing to their abundant intramuscular fat, undergo a unique three-stage feeding system with varying concentrate ratios. There is limited research on physiological and rumen microbial changes in Japanese Black cattle during these stages. Therefore, this study aimed to examine Japanese Black steers in these three stages: early (T1, 12-14 months), middle (T2, 15-22 months), and late (T3, 23-30 months). The rumen bacteria of 21 cattle per phase was analyzed using 16S rRNA gene sequencing. Rumen bacterial diversity was significantly higher in T1, with a distinct distribution, than in T2 and T3. Specific phyla and genera were exclusive to each stage, reflecting the shifts in feed composition. Certain genera dominated each stage: T1 had Flexilinea, Streptococcus, Butyrivibrio, Selenomonas, and Kandleria; T2 had Bifidobacterium, Shuttleworthia, and Sharpea; and T3 had Acetitomaculum, Mycoplasma, Atopobium, and Howardella. Correlation analysis revealed significant associations between certain microbial populations and physiological parameters. These findings indicate that changes in energy content and feed composition are associated with physiological and ruminal alterations. This study may guide strategies to improve rumen health and productivity in Japanese Black cattle by modifying diets to specific fattening stages.

The objective of this study was to develop models for predicting the amount of feces and urine excreted by Japanese Black fattening steer using a dataset of 119 digestion trials for a total of 46 animals. Correlation analysis was used to analyze the relationships between feces and urine excretion and feed intake, feed digestibility, and nitrogen balance. Multiple regression analysis was conducted to develop models for predicting the amount of feces and urine excreted using the explanatory variables selected from various animal and dietary parameters based on P-value (<0.10) and variance inflation factor (<3.0). Resultingly, dry matter intake was a primary predictor of feces excreted. The prediction equation for the amount of feces excretion as a function of body weight, dry matter intake, and calculated total digestible nutrients fits the data well (adjusted coefficient of determination [adj R2 ] = 0.519, root mean square error = 1.57). Furthermore, the nitrogen content in the urine was the primary predictor of the urine excretion amount. Thus, the prediction equation for the amount of urine excreted using the nitrogen content in urine yielded a highly accurate model (adj R2 = 0.813, root mean square error = 4.12).

This study aimed to develop a prediction equation for methane (CH4 ) emissions from fattening cattle based on the CH4 /carbon dioxide (CO2 ) ratio and validate the predictive ability of the developed equation. The prediction equation was developed using the CH4 /CO2 ratio combined with oxygen consumption and respiratory quotient estimations that were theoretically calculated from the relation between gas emissions and energy metabolism. To validate the prediction equation, gas measurements in the headboxes were conducted using eight Japanese Black steers. The predictive ability of the developed equation was compared with that of two previously reported equations. As a result, the developed and reported equations had significant (P < 0.01) linear relationships between the observed and predicted CH4 emissions. Notably, only the developed equation had a significant (P < 0.01) linear relationship between the observed and predicted CH4 emissions when expressed per unit of dry matter intake. The results suggest that the developed prediction equation has a higher predictive ability than previously reported equations, particularly in evaluating the efficiency of CH4 emissions. Although further validation is required, the equation developed in this study can be a valuable tool for on-farm estimations of individual CH4 emissions from fattening cattle.

This study aimed to determine the physiological features and rumen microbial composition associated with the non-glucogenic-to-glucogenic short-chain fatty acids ratio (NGR). Holstein cows were housed in a free-stall barn with an automatic milking system and fed a partially mixed ration. Physiological and microbial analyses were performed on 66 datasets collected from 66 cows (50-250 days in milk). NGR was positively correlated with ruminal pH, relative abundances of protozoa and fungi, methane conversion factor, methane intensity, plasma lipids, parity, and milk fat, and negatively correlated with total short-chain fatty acids. To highlight the differences in bacterial and archaeal compositions between NGRs, low-NGR cows (N = 22) were compared with medium-NGR (N = 22) and high-NGR (N = 22) cows. The low-NGR group was characterized by a lower abundance of Methanobrevibacter and a higher abundance of operational taxonomic units belonging to the lactate-producing, such as Intestinibaculum, Kandleria, and Dialister, and the succinate-producing Prevotella. Our findings indicate that NGR affects the methane conversion factor, methane intensity, and blood and milk compositions. Low NGR is associated with a higher abundance of lactate- and succinate-producing bacteria and lower abundances of protozoa, fungi, and Methanobrevibacter.

Angiopoietin-like protein 3 (ANGPTL3) is expressed predominantly in the liver and plays a major role in regulating the circulating triglyceride and lipoprotein fraction concentrations by inhibiting lipoprotein lipase (LPL) activity. Given these physiological roles, ANGPTL3 may play an important role in metabolic changes related to fat accumulation during the fattening period in Japanese Black. This study aimed to reveal the physiological roles of hepatic ANGPTL3 in Japanese Black steers (Bos taurus) during the fattening period and investigate the regulatory effects of hepatic ANGPTL3. To investigate the gene expression and protein localization of ANGPTL3, 18 tissue samples were collected from tree male Holstein bull calves aged 7 wk. Biopsied liver tissues and blood samples were collected from 21 Japanese Black steers during the early (T1; 13 mo of age), middle (T2; 20 mo), and late fattening phases (T3; 28 mo). Relative mRNA expression, blood metabolite concentrations, hormone concentrations, growth, and carcass traits were analyzed. To identify the regulatory factors of hepatic ANGPTL3, primary bovine hepatocytes collected by two Holstein calves aged 7 wk were incubated with insulin, palmitate, oleate, propionate, acetate, or beta-hydroxybutyric acid (BHBA). The ANGPTL3 gene was most highly expressed in the liver, with minor expression in the renal cortex, lungs, reticulum, and jejunum in Holstein bull calves. In Japanese Black steers, relative ANGPTL3 mRNA expressions were less as fattening progressed, and blood triglyceride, total cholesterol, and nonesterified fatty acid (NEFA) concentrations increased. Relative ANGPTL8 and Liver X receptor alpha (LXRα) mRNA expressions decreased in late and middle fattening phases, respectively. Furthermore, relative ANGTPL3 mRNA expression was positively correlated with ANGPTL8 (r = 0.650; P < 0.01) and ANGPTL4 (r = 0.540; P < 0.05) in T3 and T1, respectively, and LXRα showed no correlation with ANGPTL3. Relative ANGTPL3 mRNA expression was negatively correlated with total cholesterol (r = -0.434; P < 0.05) and triglyceride (r = -0.645; P < 0.01) concentrations in T3 and T1, respectively; There was no significant correlation between ANGTPL3 and carcass traits. Relative ANGTPL3 mRNA expression in cultured bovine hepatocytes was downregulated in oleate treatment. Together, these findings suggest that ANGPTL3 downregulation in late fattening phases is associated with the changes in lipid metabolism.


The role of angiopoietin-like protein 3 (ANGPTL3) in various animal species under different physiological conditions remains largely unknown. We evaluated the physiological roles of hepatic ANGPTL3 in Japanese Black steers (Bos taurus) during the fattening period and investigated the expressional regulation of ANGPTL3 in bovine hepatocytes. Relative ANGPTL3 mRNA expression decreased late in the fattening phases. Relative ANGPTL3 mRNA expression was positively correlated with ANGPTL4 and ANGPTL8 and was negatively correlated with blood triglyceride concentrations in early fattening phases. Relative ANGPTL3 mRNA expression in cultured bovine hepatocytes was downregulated in oleate treatment. Fatty acids may influence ANGPTL3 expression in cultured bovine hepatocytes through possible regulatory factors. Our findings suggest that the physiological roles of ANGPTL3 are associated with the changes of lipid metabolism during the fattening period, and the ANGPTL family seem to be associated with blood lipid metabolites.

The purpose of this study was to investigate the carbon dioxide (CO2 ) concentration in the sampled gas to avoid low concentration of breath while measuring the methane (CH4 )/CO2 ratio using the sniffer method. This study also assessed the effect of selective elimination by applying the threshold of CO2 concentration to the CH4 /CO2 ratio. The gas measurement in the automatic milking system was conducted with 26 multiparous Holstein cows using an electric fan to manipulate the CO2 concentration in the sampled gas. Four different thresholds of the background-corrected CO2 concentrations (0, 0.025, 0.05, and 0.1%) were applied to every 1-s value of the individual gas measurement. Subsequently, three different upper limits of the proportion of eliminated values (none, 0.5, and 0.33) were applied to the individual records per milking. The results showed that the sampled gas must contain more than 0.1% of the corrected CO2 concentration to enable accurate calculation of the CH4 /CO2 ratio. It is recommended that at least half of the values in the data be larger than the threshold of the corrected CO2 concentration for unbiased measurement of the CH4 /CO2 ratio.

We investigated the physiological changes during the fattening period and production characteristics in Japanese Black steers bred and raised using the typical feeding system in Japan. Here, 21 Japanese Black steers aged 12 months were used, with experimental period divided into early (12-14 months of age), middle (15-22 months), and late fattening phases (23-30 months). The liver transcriptome, blood metabolites, hormones, and rumen fermentation characteristics were analyzed. Blood triglyceride and non-esterified fatty acid concentrations increased, whereas blood ketone levels decreased, with fattening phases. Blood insulin increased with fattening phases and was positively correlated with carcass weight and marbling in late fattening phases. Rumen fermentation characteristics showed high propionate levels and low butyrate levels in late fattening phases, likely due to increased energy intake. Genes related to glucose metabolism, such as SESN3, INSR, LEPR, and FOXO3, were down-regulated in late fattening phases. Genes related to lipid metabolism, such as FABP4, were up-regulated, whereas FADS1 and FADS2 were down-regulated. These findings suggest that the physiological changes resulted from changes in the energy content and composition of diets. Liver metabolism changed with changes in fat metabolism. Insulin was strongly associated with physiological changes and productivity in Japanese Black cattle.

The effects of long-term feeding of wood kraft pulp (KP) silage containing sweet-potato shochu distillery by-product (SDP) on feed intake, feed digestibility, rumen fermentation, and growth performance of Japanese Black steers were investigated during the early fattening period. Ten Japanese Black steers (9.8 ± 0.6 months of age) were used in this study. Five steers (KP group) were fed KP silage as a replacement for 10% timothy hay (dry matter bases), in contrast to the other five (control group). KP silage consisted of 92.9% KP and 7.1% SDP (dry matter bases). The experiment was conducted for 18 weeks. No significant differences were observed in terms of feed intake, feed digestibility, or daily body weight gain between the groups. In addition, diurnal changes in the rumen pH and ruminal lipopolysaccharide activity did not differ between the groups. However, the plasma concentration of aspartate transaminase in the KP group was slightly lower (P = 0.078) than that in the control group. Thus, our study suggested that feeding KP silage does not reduce feed intake or affect the rumen fermentation or growth performance of Japanese Black fattening steer.

In this study, using enteric methane emissions, we investigated the metabolic characteristics of Japanese Black cattle. Their methane emissions were measured at early (age 13 months), middle (20 months), and late fattening phases (28 months). Cattle with the highest and lowest methane emissions were selected based on the residual methane emission values, and their liver transcriptome, blood metabolites, hormones, and rumen fermentation characteristics were analyzed. Blood ß-hydroxybutyric acid and insulin levels were high, whereas blood amino acid levels were low in cattle with high methane emissions. Further, propionate and butyrate levels differed depending on the enteric methane emissions. Hepatic genes, such as SERPINI2, SLC7A5, ATP6, and RRAD, which were related to amino acid transport and glucose metabolism, were upregulated or downregulated during the late fattening phase. The above mentioned metabolites and liver transcriptomes could be used to evaluate enteric methanogenesis in Japanese Black cattle.

The effect of dietary vitamin K3 (VK3) on ruminant animals is not fully investigated. The aim of this study was to examine the effects of dietary VK3 on lactation performance, rumen characteristics, and VK1 and menaquinone (MK, or VK2) dynamics in the rumen, plasma, and milk of dairy cows. Eight Holstein dairy cows in late lactation periods were used in two crossover trials including a control (nontreatment) and a 50 or 200 mg/day (d) VK3 supplementation group. After 14 days, plasma, ruminal fluid, and milk were sampled and their VK1 and MKs contents were measured using fluorescence-high-performance liquid chromatography (HPLC). Milk production was unchanged after feeding 50 mg/day VK3 but marginally decreased after feeding 200 mg/day VK3. The molar ratio of propionate in ruminal fluid was significantly increased on feeding 200 mg/day VK3. Additionally, MK-4 concentrations significantly increased in both plasma and milk after VK3 feeding (50 and 200 mg/day). In ruminal fluid, MK-4 concentrations increased after 200 mg/day VK3 feeding. These results suggest that VK3 may be a good source of MK-4, the biologically active form of VK, in Holstein dairy cows during their late lactation periods. This study provides a basis for understanding the physiological role of VK in dairy cows.

This study was to examine the effects of dietary vitamin K (VK) 3 supplementation on immune-related substances in milk, oxidative stress indices in plasma and VK1, and menaquinone 4 (MK-4) in plasma and milk in periparturient dairy cows. Forty healthy perinatal Holstein-Friesian dairy cows were used in this study. Twenty-one animals were randomly selected and categorized into the VK3 supplemented (50 mg/day/head as VK3) group; the remaining 19 were categorized into the control group. On day 3 after calving, blood and milk were sampled, and their chemical components were determined. The VK3 supplemented group had significantly higher menaquinone 4 levels in plasma and milk on day 3 postpartum than the control group. In addition, there was a significant increase in the immunoglobulin G (IgG) level in milk. VK3 may be absorbed from the gastrointestinal tract and converted to MK-4, the biologically active form of VK, in the mammary gland and other tissues. It was thought that the increase in MK-4 level in plasma and milk induced an increase in the concentration of IgG in milk. VK3 supplementation to periparturient dairy cows may contribute to the production of colostrum with high concentrations of IgG and MK-4.

In this study, we evaluated the partial replacement of roughage with wood kraft pulp (KP) on rumen fermentation and productivity of dairy cows. Eighteen cows were divided into control and KP groups. The KP group started adaptation to KP 3 weeks before calving; after calving, they were fed a total mixed ration for 12 weeks, wherein 18% Timothy hay was replaced with KP. The dry matter intake, body weight, and milk yield and composition were similar in the control and KP groups. The average daily rumen pH was higher with KP feeding, and the average daily ruminal temperature remained lower at 16 days after calving (P < 0.05). The concentration of volatile fatty acids remained unaltered, the molar proportion of acetic acid decreased, and the molar proportion of propionic acid increased, indicating a low acetic acid:propionic acid ratio (P < 0.05). Lipopolysaccharide activity in the rumen fluid was higher in the KP group (P < 0.05); however, the rumen microbiota were unaffected. The digestibility of dry matter and neutral detergent fiber increased 12 weeks after calving, whereas excretion of urinary nitrogen decreased (P < 0.05). Partial replacement of roughage with KP did not suppress rumen fermentation and maintained postpartum productivity.

The aim of this study was to develop prediction equations for methane (CH4 ) emissions from lactating cows using the CH4 /carbon dioxide (CO2 ) ratio in the breath measured in the automatic milking system (AMS) and to evaluate the predicted values and factors affecting the CH4 /CO2 ratio. The model development was conducted using a dataset determined in respiration chambers or head boxes (n = 121). Then, gas measurements in the AMS as well as in the head box were carried out with six lactating cows fed one of three different levels of neutral detergent fiber (NDF) content, following a 3 × 3 Latin square experimental design. The obtained equation that is suitable for practical use on farms to predict CH4 was CH4 (L/day) = -507 + 0.536 live weight (kg) + 8.76 energy-corrected milk (kg/day) + 5,029 CH4 /CO2 (adjusted R2 = 0.83; root mean square error = 40.8 L/day). Results showed that the predicted values correlated positively with the observed values, the determined CH4 /CO2 ratio increased with increasing dietary NDF content, and the detected eructation rate was in the normal range. On the other hand, the CH4 /CO2 ratio was affected by the time interval between measurement and last eating before the measurement.

We investigated the effects of wood kraft pulp (KP) feeding on feed digestibility and rumen fermentation of Japanese Black (JB) steer in the middle fattening stage. The feeding experiment was carried out a replicated 3 × 3 Latin square design using six JB steers (16.2 ± 0.8 months of age). Steers were fed rice straw and three concentrated feeds: commercial formula feed (control), formula feed containing 7.5% KP on a dry matter (DM) basis (7.5% KP), and formula feed containing 15% KP on a DM basis (15% KP). Feed digestibility and rumen fermentation parameters were recorded for 3 days during the measurement period, after an adaptation phase of 11 days. There was no significant difference in DM intake per day among diets. A slightly higher digestibility of neutral detergent fiber was observed in steers fed 15% KP, compared with those fed other diets (P = 0.059). The butyric acid ratio in rumen fluid was higher in steers fed KP diets than in steers fed control (P = 0.083). The average ruminal pH was unaffected by KP diets. This study suggests that replacing 15% of concentrated feed with KP has little effect on feed digestibility and rumen fermentation in JB fattening steer.

The objective of this study was to develop a prediction equation for methane-related traits in beef cattle and evaluate this equation using datasets with different cattle breeds and roughage rates. Enteric methane emission (CH4 , l/day) was measured using open-circuit respiration chambers. Dry matter intake (DMI, kg/day), body weight (BW, kg), daily gain (DG, kg), total digestible nutrients (TDN, %DMI), and roughage rate (Rrate, %) were used as independent variables, and methane-related traits-CH4 , CH4 per DMI (CH4 /DMI, l/kg), and methane conversion factor (MCF, %)-were used as dependent variables. The best-fit equations to predict methane-related traits using a total of 76 records were CH4  = -676.7 + 0.04194 × BW + 29.88 × DMI + 7.883 × TDN + 4.367 × Rrate, CH4 /DMI = -52.24 - 1.193 × 10-3  × BW - 5.905 × DG + 1.077 × TDN + 0.5008 × Rrate, and MCF = -11.43 - 5.308 × 10-4  × BW - 1.223 × DG + 0.2336 × TDN + 0.1157 × Rrate. The predictive ability of the developed equations differed between roughage rates but not between breeds. For CH4 , the predictive ability of the developed equations was better compared with previously reported equations in the low roughage rate dataset, but not in the high roughage rate dataset. Our results suggest that the developed equations of methane-related traits can be applied in beef cattle fed with low roughage diets.

The objectives of this study were to estimate genetic parameters and to perform a genome-wide association study (GWAS) for predicted methane-related traits in Japanese Black steers. The methane production and yield traits were predicted using on-farm measurable traits, such as dry matter intake and average daily gain. A total of 4,578 Japanese Black steers, which were progenies of 362 sires genotyped with imputed 551,995 single nucleotide polymorphisms (SNPs), had phenotypes of predicted methane-related traits during the total fattening period (52 weeks). For the estimation of genetic parameters, the estimated heritabilities were moderate (ranged from 0.57 to 0.60). In addition, the estimated genetic correlations of methane production traits with most of carcass traits and feed-efficiency traits were unfavorable, but those of methane yield traits were favorable or low. For the GWAS, no genome-wide significant SNP was detected, but a total of four quantitative trait locus (QTL) regions that explained more than 5.0% of genetic variance were localized on the genome, and some candidate genes associated with growth and feed-efficiency traits were located on the regions. Our results suggest that the predicted methane-related traits are heritable and some QTL regions for the traits are localized on the genome in Japanese Black steers.
