RESUMO

Excessive intake of estrogen poses significant health risks to the human body; hence, there is a necessity to develop rapid detection methods to monitor its levels of addition. Gold nanoparticles (AuNPs), commonly utilized as colorimetric signal labels, find extensive application in lateral flow immunoassay (LFIA). However, the detection sensitivity of traditional AuNPs-LFIA is typically constrained by low molar extinction coefficients and reliance on a single signal. Herein, in this work, unique spark-type AuCuPt nanoflowers modified with tannic acid (AuCuPt@TA) were precisely designed by reasonable layer-by-layer element composition and green modification. The obtained AuCuPt displays robust broadband absorption spanning the visible to near-infrared spectrum, showcasing a notable molar extinction coefficient of 2.38 × 1012 M-1 cm-1 and a photothermal conversion efficiency of 48.5%. Based on this, selecting estriol (E3) as a model analyte, colorimetric/photothermal dual-signal LFIA (CLFIA and PLFIA) was developed. Limits of detection (LOD) of the CLFIA and PLFIA were achieved at 0.033 ng mL-1 and 0.021 ng mL-1, respectively, which represent a 9.3- and 14.6-fold improvement compared to the visual LOD of AuNPs-LFIA. Moreover, the application feasibility of the immunoassay was further evaluated in the milk and pork with satisfactory recoveries ranging from 86.21% to 117.91%. Thus, this work has enhanced the performance of LFIA for E3 detection and exhibited enormous potential for other sensing platform construction.

RESUMO

The multiple-readout capability of multimodal detection enhances the flexibility, reliability, and accuracy of lateral flow immunoassay (LFIA). The conjugation of two different metal-organic frameworks (MOFs) as a new-generation composite material offers extraordinary opportunities for developing multimodal LFIA. It is anticipated to compensate limitations of traditional single colorimetric signal LFIA and improve the analysis performance. Herein, an ultra-bright fluorescent AIE-MOF was proposed and coupled with an in-situ growth of Prussian blue (PB) nanoparticles strategy to obtain a novel multimodal signal tracer (AIE-MOF@PB). Thereafter, it was successfully applied to develop the multimodal LFIA platform for the detection of nitrofurazone metabolites. The synergy of AIE-MOF and PB endows AIE-MOF@PB with superb water dispersibility, robust fluorescence emission, brilliant colorimetric signal, marvelous photothermal conversion, and enhanced antibody coupling efficiency, all of which facilitate a highly sensitive triple-readout LFIA platform. The detection sensitivity improved by at least 5-fold compared with the colloidal gold-based LFIA. This work not only inspires the rational design of aggregation-induced emission luminogens (AIEgen)-based complex materials but also highlights the promising potential in flexible point-of-care applications.

Conventional lateral flow immunoassay (LFIA) usually suffers from poor antimatrix interference, unsatisfactory sensitivity, and lack of quantitative ability for target analyte detection in food matrices. In response to these limits, here, multifunctional nanomaterial ZnFe2O4 nanoparticles (ZFOs) were developed and integrated into LFIA for powerful magnetic separation/enrichment and colorimetric/photothermal target sensing. Under optimum conditions, the detection for clenbuterol (CL) with magnetic enrichment achieves 9-fold higher sensitivity compared to that without enrichment and 162-fold higher sensitivity compared to that based on traditional colloidal golds. Attributing the improved performances of ZFOs, CL can be detected at ultralow levels in pork and milk with 10 min of immunoreaction time. The vLODs were 0.01 µg kg-1 for two modes, and the cutoff values of CL were about 5 and 3 µg kg-1, respectively. More importantly, the enrichment ZFO-mediated LFIA (ZE-LFIA) exhibits a similar limit of detection (LOD) in both buffer solution and food matrix, demonstrating a universal resistance to the food matrix. The multitudinous performance merits of this ZE-LFIA with high sensitivity, matrix tolerance, accuracy, and specificity have ensured a broad application potential for target detection of clenbuterol and can serve as an experience for other veterinary drug residues' detection.

BACKGROUND: To explore the relationship between the degree of renal fibrosis in patients with immunoglobulin A nephropathy (IgAN) and their levels of Apelin-12, Average Optical Density of angiotensin II type 1 receptor (AODAT1R), and angiotensinogen (AGT). METHODS: A total of 156 patients with IgAN diagnosed by renal biopsy in our hospital were selected and divided into a T0 group (54 cases), T1 group (49 cases) and T2 group (53 cases). The levels of Apelin-12, AT1R, and AGT were compared among the three groups, and the relationship between the above three indicators and degree of renal fibrosis was analyzed among patients with IgAN. RESULTS: The AODAT1R and AGT level in the T2 group and T1 groups were significantly higher than those of the T0 group, and the Apelin-12 level of patients in the T2 group and T1 groups were significantly lower than that in T0 group. Significances of the same trend were observed among all the above indicators between the T2 group and T1 group. ROC curves showed that when the cutoff value of Apelin-12 was 2.36 µg/L, the area under curve (AUC), sensitivity, and specificity of T0-T1T2 were 0.889, 92.00%, and 88.00%, respectively. When the cut-off value of AODAT1R was 0.065, the AUC, sensitivity, and specificity were 0.706, 76.00%, and 76.00%, respectively, and when the cut-off value of AGT was 47.26 ng/mL, the AUC, sensitivity, and specificity were 0.899, 84.00%, and 88.00%, respectively. When the cutoff value of Apelin-12 was 0.92 µg/L, the AUC, sensitivity, and specificity of T0T1-T2 were 0.819, 84.62%, and 87.50%, respectively, and when the cutoff value of AODAT1R was 0.079, the AUC, sensitivity, and specificity were 0.699, 76.92%, and 79.17%, respectively. When the cut-off value of AGT was 92.96 ng/mL, the AUC, sensitivity, and specificity were 0.893, 84.62%, and 91.67%, respectively. CONCLUSIONS: Apelin-12 decreased with disease progression, while AT1R and AGT increased. The changes of levels of Apelin-12, AT1R, and AGT have certain significance in judging the degree of renal fibrosis in patients with IgA nephropathy, and the change of level of AGT has the highest correlation with the degree of renal fibrosis.

Taking the 4 hm2 fixed sampling plot in the Beigou forest farm of Weichang County in Hebei Province as test object, and by adopting the parameters (point pattern distribution, mingling, and size differentiation), this paper analyzed the community structure and distribution pattern of a natural secondary forest in the farm. There were eleven populations in the arbor layer of the forest, among which, Populus davidiana and Betula platyphylla had the obvious advantage in population density and basal area, being the dominant and constructive species of the arbor layer. Spatially, these two species all presented cluster modes remarkably, and competed each other greatly. The main accompanying species Larix principis-rupprechti and Acer truncatum also presented cluster modes, but the density and volume were significantly lower than the two dominant species', not able to compete with the dominate species. Affected by the low mingling of dominant species, the average mingling of the whole stand was only 0.40, while the mingling of accompanying species generally presented moderate or high. The mean size differentiation of the whole stand was 0.49, and P. davidiana, B. platyphylla, L. principis-rupprechti, and Quercus mongolica were of dominance or sub-dominance in the spatial structural units, while the other accompanying species had no obvious dominance.

To investigate the relations between morbidity of hemolytic diseases of newborn and ABO antibodies (HDN) in sera of Han and Yao nationality, pregnant women were examined before and after delivery. Antibodies screen, direct antiglobulin test, free antibodies and elution test were all performed. The results indicated that immunologic ABO antibodies of Han people were found in 673 cases out of 1,471 Han pregnant women, and was also found in 28 cases out of 65 Yao pregnant women, and there was no significant difference of incidences between Han and Yao nationality. 31 cases of HDN were found out of 288 newborn in Han and 3 cases of HDN were found out of 25 newborn in Yao, and there was no significant difference between Han and Yao nationalities. The characteristics of HDN in Han nationality were as same as in Yao nationality. In conclusion, the morbidity of HDN in Han and Yao nationalities of Shaoguan area did not showed essential difference, the immunologic ABO antibodies and its titration test, especially, elution test are important for prognosis of HDN.
