RESUMO

A fluorescent probe Y((1,1'-([1,1'-biphenyl]-4,4'-diylbis(3-(2-hydroxyphenyl)-4,5-dihydro-1H-pyrazole-5,1-diyl)) bis(ethan-1-one))) was designed and synthesized, which could be used to Cu2+ and Fe3+ sensors. Through the study of optical properties, the probe Y shows good selectivity and sensitivity to Cu2+ and Fe3+ in aqueous tetrahydrofuran solution [10.0 mM HEPES, pH 7.4, THF-H2O = 9:1(v/v)] with has excellent anti-interference performance, and its detection limits were 0.931 uΜ for Cu2+ and 0.401uΜ for Fe3+. The coordination mechanism of probe Y with Cu2+ and Fe3+ was speculated and verified at DFT level and HRNM. By Hela cytotoxicity and imaging tests, probe Y not only has good biocompatibility, but also can be used for sensing Cu2+ in cells.

An amino-functionalized terphenyl-tetracarboxylic acid, 2'-amino-[1,1':4',1″-terphenyl]-3,3″,5,5″-tetracarboxylic acid (H4tpta), was used as an adaptable linker to synthesize, under hydrothermal conditions, eight coordination polymers (CPs). The obtained products were formulated as [Co(µ6-H2tpta)]n (1), [Co(µ3-H2tpta)(2,2'-bipy)]n (2), [M3(µ6-Htpta)2(2,2'-bipy)2]n (M = Mn (3), Cd (4)), [Ni2(µ4-tpta)(phen)2(H2O)4]n (5), [Zn2(µ6-tpta)(phen)2]n (6), {[Zn2(µ6-tpta)(µ-4,4'-bipy)]·H2O}n (7), and [Zn2(µ6-tpta)(µ-H2biim)(H2O)2]n (8), wherein 2,2'-bipyridine (2,2'-bipy), 4,4'-bipyridine (4,4'-bipy), 1,10-phenanthroline (phen), or 2,2'-biimidazole (H2biim) are present as additional stabilizing ligands. The structural types of 1-8 vary from one-dimensional (1D) (2, 5) and two-dimensional (2D) (3, 4, 6) CPs to three-dimensional (3D) metal-organic frameworks (MOFs) (1, 7, and 8) with a diversity of topologies. The products 1-8 were investigated as catalysts in the Knoevenagel condensation involving aldehydes and active methylene derivatives (malononitrile, ethyl cyanoacetate, or tert-butyl cyanoacetate), leading to high condensation product yields (up to 99%) under optimized conditions. Various reaction conditions, substrate scope, and catalyst recycling were investigated. This work broadens the application of H4tpta as a versatile tetracarboxylate linker for the generation of diverse CPs/MOFs.

Widely utilized in the chemical industry and agriculture, hydrazine is easily absorbed by living things and can cause physical harm when in touch for an extended period of time. As a result, a novel cinnamaldehyde chalcone C5 was produced by Friedel Crafts process and aldol condensation reaction. Triphenylamine was used as the raw material for hydrazine determination in both reactions. Chalcone C5 exhibits significant AIE behavior in a mixed mixture of ethanol and water in addition to having great selectivity and a low detection limit (0.119 nm) for hydrazine. The solvent effect test revealed a linear relationship between the Stokes shift of C5 in the solvent and the rise in solvent orientation polarization. It is important to note that C5 is not harmful to MCF-7 cells, mouse kidney cells, or pig kidney cells. Furthermore, research on cell imaging has demonstrated that probe C5 may be utilized to image the fluorescence of hydrazine in active MCF-7 cells.

A pyridine-tricarboxylic acid, 5-(3',5'-dicarboxylphenyl)nicotinic acid (H3dpna), was employed as a adjustable block to assemble a series of coordination polymers under hydrothermal conditions. The seven new coordination polymers were formulated as [Co(µ3-Hdpna)(µ-dpey)]n·nH2O (1), [Zn4.5(µ6-dpna)3(phen)3]n (2), [Co1.5(µ6-dpna)(2,2'-bipy)]n (3), [Zn1.5(µ6-dpna)(2,2'-bipy)]n (4), [Co3(µ3-dpna)2(4,4'-bipy)2(H2O)8]n·2nH2O (5),[Co(bpb)2(H2O)4]n[Co2(µ3-dpna)2(H2O)4]n·3nH2O (6), and [Mn1.5(µ6-dpna)(µ-dpea)]n (7), wherein 1,2-di(4-pyridyl)ethylene (dpey), 1,10-phenanthroline (phen), 2,2'-bipyridine(2,2'-bipy),4,4'-bipyridine(4,4'-bipy),1,4-bis(pyrid-4-yl)benzene (bpb), and 1,2-di(4-pyridyl)ethane (dpea) were employed as auxiliary ligands. The structural variation of polymers 1-7 spans the range from a 2D sheet (1-4, 6, and 7) to a 3D metal-organic framework (MOF, 5). Polymers 1-7 were investigated as heterogeneous catalysts in the Knoevenagel condensation reaction, leading to high condensation product yields (up to 100%) under optimized conditions. Various reaction conditions, substrate scope, and catalyst recycling were also researched. This work broadens the application of H3dpna as a versatile tricarboxylate block for the fabrication of functional coordination polymers.

Two triphenylamine chalcone derivatives 1 and 2 were synthesized through the Vilsmeier-Haack reaction and Claisen-Schmidt condensation reaction. Through ultraviolet absorption spectroscopy and fluorescence emission spectroscopy experiments, it was confirmed that these two compounds exhibited good aggregation-induced emission (AIE) behavior in ethanol/water mixtures. The solvent effect test showed with the increase of the orientation polarizability of the solvent, the Stokes shift in the solvent of compound 1 and compound 2 shows a linear change trend. Through solid state fluorescence test and universal density function theory (DFT), the existence of π-π stacking interaction in the solid state of the compound has been studied, resulting in weak fluorescence emission. pH has no effect on the fluorescence intensity of the aggregate state of excited state intramolecular proton transfer (ESIPT) molecules in an acidic environment, but greatly weakens its fluorescence intensity in an alkaline environment. Cyclic voltammetry (CV) test shows that compound 1 was more prone to oxidation reaction than compound 2. The results of thermal stability test show that the thermal stability of compound 1 was better than that of compound 2, indicating that triphenylamine chalcone derivatives can improve the thermal stability of compounds by increasing the number of branches.

Firstly, a novel pyrazole-pyrazoline fluorescent probe was developed and synthesized. The probe can be used to determine Fe3+ ions in a series of cations in tetrahydrofuran aqueous solution with high selectivity and high sensitivity. After the addition of iron ions, the fluorescence intensity is significantly reduced, Its structure was characterized by 1H NMR, 13C NMR and HR-ESI-MS. UV absorption spectra and Fluorescence spectroscopy were used to study the selective recognition of probe M on metal ions. The probe M can selectivity and sensitivity to distinguish the target ion from other ions through different fluorescence phenomena. In addition, the binding modes of M with Fe3+ were proved to be 1:1 stoichiometry in the complexes by Job's plot, IR results. The combination of probe M and iron ions is 1:1, and the detection limit is 3.9 × 10-10 M. The binding mode and sensing mechanism of M with Fe3+ was verified by theoretical calculations using Gaussian 09 based on B3LYP/6-31G(d) basis.

Herein, we have presented a facile and efficient method of ring-opening nucleophilic fluorination of aziridines, affording highly regio-selective ß-fluorinated amines. Firstly, the example of ring-opening hydrofluorination of azetidines was reported. Then, the Olah's reagent also provided a promising method for the construction of enantioenriched ß-fluoro-α-amino acid derivatives, which could be used for the preparation of peptide-based bioactive molecules.

RESUMO

A tandem reaction consisting of a copper(I)-catalyzed cycloetherification and a hydrogen-bond-induced inverse-electron-demand oxa-Diels-Alder cycloaddition was performed from chiral propargyl alcohol, generating several kinds of optically pure [5, 6] spiroketals in excellent stereoselectivities and yields. The investigation on mechanism found that the cyclization prompted by a hydrogen bond not only improved the efficiency but also determined the diastereoselectivity.

RESUMO

A novel formal asymmetric synthesis of (-)-triptonide and (-)-triptolide, featuring a new alternative access to their known key intermediate 4, has been achieved through two synthetic routes in 9 steps with 13.6% total yield and 10 steps with 18.5% overall yield, respectively. This synthesis is scalable and hence has high potential for application to further synthetic elaboration and biologic investigation on such natural products.

Kidney fibrosis is an inevitable result of various chronic kidney diseases (CKDs) and significantly contributes to end-stage renal failure. Currently, there is no specific treatment available for renal fibrosis. ELA13 (amino acid sequence: RRCMPLHSRVPFP) is a conserved region of ELABELA in all vertebrates; however, its biological activity has been very little studied. In the present study, we evaluated the therapeutic effect of ELA13 on transforming growth factor-ß1 (TGF-ß1)-treated NRK-52E cells and unilateral ureteral occlusion (UUO) mice. Our results demonstrated that ELA13 could improve renal function by reducing creatinine and urea nitrogen content in serum, and reduce the expression of fibrosis biomarkers confirmed by Masson staining, immunohistochemistry, real-time polymerase chain reaction (RT-PCR), and western blot. Inflammation biomarkers were increased after UUO and decreased by administration of ELA13. Furthermore, we found that the levels of essential molecules in the mothers against decapentaplegic (Smad) and extracellular signal-regulated kinase (ERK) pathways were reduced by ELA13 treatment in vivo and in vitro. In conclusion, ELA13 protected against kidney fibrosis through inhibiting the Smad and ERK signaling pathways and could thus be a promising candidate for anti-renal fibrosis treatment.

RESUMO

The first enantioselective total synthesis of (+)-coriandrone A and B, two bioactive natural products, has been achieved in 10 steps and 11 steps starting from commercially available methyl 2-hydroxy-4-methoxybenzoate. Key reactions include a Claison rearrangement, a Shi-type epoxidation-cyclization sequence and ortho-metallation of t-butylbenzamides with (S)-(-)-propylene oxide reaction.

In this paper, four novel hydrazine fluorescent probes X1-X4 with bis-chalcone structure were designed and synthesized. Through the measurement of its optical properties, it is found that it can quickly identify hydrazine, high sensitivity, low detection limit, and good anti-interference ability. The recognition of hydrazine by probes X1-X4 is not affected in the pH range of 4-10, X2 has the highest sensitivity, and the detection limit is as low as 0.336 × 10-7 M. Through Gaussian quantization calculation of probe molecules and their reaction products with hydrazine, it is speculated that the recognition mechanism is the closure of intramolecular charge transfer effect. In addition, the cytotoxicity and imaging of HeLa cells were tested, which showed that probes X1-X4 could be used to detect hydrazine in cells.

A methoxy-functionalized isophthalic acid, 5-methoxy isophthalic acid (H2mia), was used a versatile linker for assembling six new metal(ii) compounds under hydrothermal conditions. The obtained products were [Cu2(µ2-mia)2(phen)2(H2O)2]·2H2O (1), [Mn(µ3-mia)(phen)]n (2), [Co(µ2-mia)(2,2'-bipy)(H2O)]n·nH2O (3), [Co(µ3-mia)(µ2-4,4'-bipy)]n·nH2O (4), [Co(µ3-mia)(py)2]n (5), and [Cd(µ2-mia)(py)(H2O)2]n·nH2O (6), where phen(1,10-phenanthroline), 2,2'-bipy(2,2'-bipyridine), 4,4'-bipy(4,4'-bipyridine) or py(pyridine) were incorporated as auxiliary ligands. The crystal structures of 1-6 range from 0D (1) and 1D (2, 3, 5, 6) CPs to a 2D network (4) with a variety of topological types. The catalytic behavior of 1-6 was studied in the cyanosilylation reaction between trimethylsilyl cyanide and aldehydes, resulting in up to 99% yields of products under optimized conditions. Various reaction parameters as well as catalyst recycling and substrate scope were investigated. This study widens the use of H2mia as a versatile dicarboxylate linker for assembling a diversity of functional metal-organic architectures with remarkable structural features and catalytic properties.

RESUMO

A new catalytic application of hypoiodite reagents generated in situ from iodide ions is found, which succeeded in the synthesis of bisbenzannelated spiroketal cores for the first time. Fluoride was proven to be obligatory for this spiroketalization, which is the first fluoride-promoted oxidative cycloetherification to aromatic spiroketals.

UCHL3 (Ubiquitin carboxyl-terminal hydrolase L3), a member of deubiquitinating enzymes, has been implicated in various cancers. However, the role of UCHL3 in esophageal squamous cell carcinoma (ESCC) remains unknown. In the current study, we aimed to investigate the role of UCHL3 in ESCC growth and migration, and whether UCHL3 could modulate CRY2 methylation through FOXM1. The expression of UCHL3 and CRY2 in ESCC tissues was assessed using qRT-PCR, western blotting and immunohistochemistry (IHC). Cell viability was determined by CCK-8 and colony formation assays. Hoechst 33342 and flow cytometry were used to detect cell apoptosis. Transwell assay was performed to investigate cell migration and invasion. In vivo animal model was used to assess cell tumorigenesis. Methylation-Specific PCR (MSP) was applied to detect CRY2 methylation in the promoter region. The results showed that UCHL3 expression was elevated in ESCC tissues and cells, while CRY2 expression was decreased. UCHL3 silencing inhibited cell viability, invasion, migration and induced cell apoptosis in vitro, repressed tumor growth in vivo, and increased CRY2 expression and decreased FOXM1 expression. In addition, UCHL3 knockdown decreased CRY2 methylation through downregulating FOXM1, leading to an increase in the expression of CRY2. Moreover, CRY2 silencing abolished UCHL3 deficiency-mediated inhibition in cell growth and migration. In summary, this study reveals that knockdown of UCHL3 inhibits ESCC growth and migration by reducing CRY2 methylation through downregulation of FOXM1 expression.

Hypervalent iodine catalyst-catalyzed nucleophilic fluorination of unsaturated amides using BF3·Et2O as the fluorine source and activating reagent was reported. Various 5-fluoro-2-oxazoline derivatives were synthesized in good to excellent yields (up to 95% isolated yield) within 10 min. The process was efficient and metal-free under mild conditions. A mechanism involving a fluorination/1,2-aryl migration/cyclization cascade was proposed on the basis of previous work and experimental results.

The origin of the nematic order remains unclear due to the strong coupling between orbital, spin and lattice degrees of freedom in iron-based superconductors. Although the driving force of hole-doped BeFe2As2is still controversial, the nematic fluctuation of electron-doped compounds is generally believed to be spin fluctuation driven. Here, we present a comprehensive study of the nematic phase transition in Ba(Fe0.962Cu0.038)2As2single crystal by using Mössbauer spectroscopy. The electric field gradient and its in-plane asymmetry on Fe nucleus, which are directly determined by the occupation of individualt2gorbital, are sensitive to the local nematicity of Fe ions. The nematic phase transition happens atTnem≈ 73.8 K in the compound while the band splitting betweendxz/dyzorbitals begins far aboveTnemand reaches 18.8 meV at 30 K. The temperature evolution of the hyperfine parameters proves the existence of electron-phonon interaction and non-Fermi-liquid behaviour nearTnem. However, the spin-lattice relaxation signal is only evident belowTnem. These observations show that the role of orbital degrees of freedom is more active in driving nematicity than in Co- or Ni-doped BaFe2As2compounds, and can be attributed to enhanced electronic localization caused by Cu doping.

Fluorination using chiral catalytic methods could result in a direct access to asymmetric fluorine chemistry. However, challenges in catalytic asymmetric fluorinations, especially the longstanding stereochemical challenges existed in BF3·Et2O-based fluorinations, have not yet been addressed. Here we report the catalytic asymmetric nucleophilic fluorination using BF3·Et2O as the fluorine reagent in the presence of chiral iodine catalyst. Various chiral fluorinated oxazine products were obtained with good to excellent enantioselectivities (up to >99% ee) and diastereoselectivities (up to >20:1 dr). Control experiments (the desired fluoro-oxazines could not be obtained when Py·HF or Et3N·3HF were employed as the fluorine source) indicated that BF3·Et2O acted not only as a fluorine reagent but also as the activating reagent for activation of iodosylbenzene.

Incorporating photochromic chromophores into polymer composites provides the possibility of a reversible photoswitch of the intrinsic properties of these materials. In this paper we report a route to attach azobenzene (AZO) moiety covalently to graphene oxide (GO) to create chromophore/graphene oxide (AZO-GO) hybrid, in which GO is both part of the chromophore and the template. Due to the high grafting density of AZO moiety and the low mass of the novel structure, the hybrid is a potential solar thermal storage material with high energy density of about 240 Wh·kg-1. It is found that C-H···π interaction between the cis-AZO chromophores and the aromatic rings of the substrate induces collective electronic modifications of GO at critical percentage of cis-isomers and reduce the thermal barrier of π-π* transition of the chromophores directly, which results in two sections of first-order reactions during the photoisomerization of trans- to cis-hybrid and also thermally stabilizes the cis-hybrid. Our findings demonstrate that high-performance AZO-GO hybrid can be manipulated by optimizing intermolecular nonbonding interactions.

OBJECTIVES: Radiotherapy for the management of keloids was first introduced in 1906, yet 107 years later optimal protocol has not yet been established. Most studies have been conducted using x-ray, ß-ray, or γ-ray. However, for high linear energy transfer radiation, clinical data are scarce. The aim of this study was to examine the efficacy and safety of postoperative carbon ion radiotherapy for keloids. MATERIAL AND METHODS: Case records of 16 patients with 20 keloids, who were given postoperative carbon ion radiotherapy with 16GyE/8 fractions in the therapy terminal at the Heavy Ion Research Facility in Lanzhou at the Institute of Modern Physics, Chinese Academy of Sciences, Lanzhou, China, were retrospectively reviewed. RESULTS: In a mean follow-up period of 29.7 months (range 24.3-35.3 months), overall survival and 95% success rates were achieved. No grade 3 or higher toxicity and complication occurred, and none of the 16 patients presented with local or systemic malignancy during the follow-up period. CONCLUSION: Here, the first evidence of postoperative carbon ion radiotherapy for keloids is provided. Surgical excision and immediate postoperative carbon ion radiotherapy is well tolerated and should be considered as a potential curative treatment modality of keloids in certain cases.
