RESUMO

Arabidopsis small GTPase RabF1 (ARA6) functions in endosomal vesicle transport and may play a crucial role in recycling and degradation of molecules, thus involved in stress responses. Here we have reported that complementary overexpression lines RabF1OE (overexpression), GTPase mutants RabF1Q93L (constitutively active) and RabF1S47N (dominant negative) lines show longer root growth than wild-type, rabF1 knockout and N-myristoylation deletion (Δ1-29, N-terminus) complementary overexpression mutant plants under salt induced stress, which indicates that N-myristoylation of RabF1 is indispensable for salt tolerance. Moreover, RabF1 is highly expressed during senescence and RabF1OE lines were more tolerant of dark-induced senescence (DIS) than wild-type and rabF1.

Grain size and shape are important components determining rice grain yield, and they are controlled by quantitative trait loci (QTLs). Here, we report the cloning and functional characterization of a major grain length QTL, qGL3, which encodes a putative protein phosphatase with Kelch-like repeat domain (OsPPKL1). We found a rare allele qgl3 that leads to a long grain phenotype by an aspartate-to-glutamate transition in a conserved AVLDT motif of the second Kelch domain in OsPPKL1. The rice genome has other two OsPPKL1 homologs, OsPPKL2 and OsPPKL3. Transgenic studies showed that OsPPKL1 and OsPPKL3 function as negative regulators of grain length, whereas OsPPKL2 as a positive regulator. The Kelch domains are essential for the OsPPKL1 biological function. Field trials showed that the application of the qgl3 allele could significantly increase grain yield in both inbred and hybrid rice varieties, due to its favorable effect on grain length, filling, and weight.

BACKGROUND: Rice blast caused by Magnaporthe oryzae is the most devastating disease in rice production. Compared with seedling blast, panicle blast is considered to be more destructive, which can occur without being preceded by severe seedling blast. However, panicle blast resistance research is rarely reported. RESULTS: Bodao, a japonica landrace from Taihu Lake region, showed a high level of panicle blast resistance. In this study, a mapping population of 212 recombination inbreeding lines (RILs) was developed from a cross of Bodao and the susceptible cultivar Suyunuo, and the RILs were evaluated for panicle blast resistance in three trials. Two quantitative trait loci (QTLs) qPb11-1 and qPb6-1 for panicle-blast resistance were identified, including a major QTL qPb11-1 (Pb-bd1) on chromosome 11 of Bodao explaining from 55.31% to 71.68% of the phenotype variance, and a minor QTL qPb6-1 on chromosome 6 of Suyunuo explaining from 3.54% to 6.98% of the phenotype variance. With the various segregation populations, Pb-bd1 was fine mapped in a 40.6 Kb region flanked by markers BS83 and BS98, and six candidate genes were identified within this region, including one gene encoding NAC domain-containing protein, one gene encoding unknown expression proteins, two genes encoding nucleotide binding site-leucine rich repeat (NBS-LRR) type disease resistance proteins, and two genes encoding von Willebrand factor type A (VWA) domain containing proteins. For application in rice breeding, three introgression lines of Pb-bd1with significantly enhanced panicle blast resistance were developed by using molecular assisted method (MAS) from the commercial variety Nanjing46 (NJ46). CONCLUSION: Two QTLs, qPb11-1(Pb-bd1) and qPb6-1 conferring panicle blast resistance, were identified from japonica landrace Bodao and Suyunuo.qPb11-1(Pb-bd1) was fine mapped in a 40.6 Kb region flanked by marker BS83 and BS98. Three introgression lines of Pb-bd1with significantly enhanced panicle blast resistance were developed by MAS method from the commercial variety NJ46. It indicated that Pb-bd1 would be useful gene source in panicle blast resistance breeding.

Since nonlinear optical materials used in the ultrafast all-optical switching is an important part for the modern optical technology, cuprates have been widely investigated for their specific Cu-O chain structure and intriguing optical properties. We present a new preparation method of microcrystalline Sr2CuO3 glass films on glass substrates combining spin-coating and co-sintering techniques. Then, the as-prepared samples were polished for different times to obtain microcrystalline Sr2CuO3 glass films with varying thickness. The influence of polishing time on the structure, the valence state and the nonlinear optical response were discussed, respectively. The purity of the Sr2CuO3 phase, surface morphology and the chemical composites of these synthesized glass films were given with scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS), X-ray powder diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). Importantly, optical absorption spectroscopy and Z-scan technique were used to measure linear absorption and third-order optical nonlinearity of the films. The experiments showed that third-order nonlinear susceptibility of the 140 min polished film sample with a thickness of 18 µm was up to 1.23 × 10-12 esu, indicating its potential application in the nonlinear field.

Chloroplasts maintain their lipid balance through a tight interplay with the endoplasmic reticulum (ER). The outer envelope membrane of chloroplasts contains a large proportion of the phospholipid phosphatidylcholine (PC), which is synthesized in the ER and also a possible precursor for thylakoid galactolipids. The mechanism for PC transport from the ER to chloroplasts is not known. Using isolated chloroplasts and liposomes containing radiolabeled PC we investigated non-vesicular transport of PC in vitro. PC uptake in chloroplasts was time and temperature dependent, but nucleotide independent. Increased radius of liposomes stimulated PC uptake, and protease treatment of the chloroplasts impaired PC uptake. This implies that the chloroplast outer envelopes contains an exposed proteinaceous machinery for the uptake of PC from closely apposed membranes.
