RESUMO

Developing highly active oxygen evolution reaction (OER) catalysts in acidic conditions is a pressing demand for proton-exchange membrane water electrolysis. Manipulating proton character at the electrified interface, as the crux of all proton-coupled electrochemical reactions, is highly desirable but elusive. Herein we present a promising protocol, which reconstructs a connected hydrogen-bond network between the catalyst-electrolyte interface by coupling hydrophilic units to boost acidic OER activity. Modelling on N-doped-carbon-layer clothed Mn-doped-Co3O4 (Mn-Co3O4@CN), we unravel that the hydrogen-bond interaction between CN units and H2O molecule not only drags the free water to enrich the surface of Mn-Co3O4 but also serves as a channel to promote the dehydrogenation process. Meanwhile, the modulated local charge of the Co sites from CN units/Mn dopant lowers the OER barrier. Therefore, Mn-Co3O4@CN surpasses RuO2 at high current density (100 mA cm-2 @ ~538 mV).

RESUMO

This work demonstrates the efficient tuning of incoherent and coherent coupling between emitters embedded in an epsilon-near-zero (ENZ) waveguide coated with a multilayer graphene. As a result, a tunable two-qubit quantum phase gate based on the ENZ waveguide is realized at the cutoff frequency. Furthermore, due to the vanishingly small permittivity of the ENZ waveguide, all incoherent coupling between any two identical emitters located in the central area of the slit approaches a maximum, enabling near-ideal bipartite and multipartite entanglement. The coherent coupling between emitters is much larger at an operating frequency far from the ENZ resonance frequency than at the cutoff frequency, and the coherent coupling and resulting energy transfer efficiency can also be effectively tuned by the Fermi level of graphene. These results demonstrate an efficiently tunable electro-optical platform for quantum devices.

RESUMO

Hydrophilicity is a requisite attribute for the 2D cell culture substrate's surface, facilitating cell adhesion and spreading. Conventional poly(dimethylsiloxane) (PDMS) microfluidic chips necessitate protein coatings to enhance hydrophilicity; however, this approach is afflicted by issues of transient efficacy, interference with cell analysis, and high costs. This paper presents a protein-free microfluidic chip, termed a "microfluidic Petri dish-chip (MPD-chip)", integrating PDMS as the cover and a tissue culture-treated (TC-treated) Petri dish as the substrate. Microstructures are hot-embossed onto the Petri dish substrate using a silicon mold. This meticulous replication process serves to establish stable flow field dynamics within the chip. A simplified method for irreversible bonding, utilizing plasma activation and silylation, is proposed for affixing the PDMS cover onto the microstructured Petri dish substrate. The prepared composite chip exhibits remarkable tightness, boasting a notable bond strength of 2825 kPa. Furthermore, the composite microfluidic chip demonstrates the capability to withstand flow velocities of at least 200 µL/min, effectively meeting the required injection standards for both cell suspension and culture medium. SH-SY5Y and HeLa cells are cultured dynamically in the MPD-chip and control groups. Outcomes encompassing normalized cell density, cell adhesion area, and cell viability metrics unequivocally highlight the superiority of the MPD-chip in facilitating long-term two-dimensional (2D) cell cultures.

RESUMO

The redox homeostasis in living cells is greatly crucial for maintaining the redox biological function, whereas accurate and dynamic detection of intracellular redox states still remains challenging. Herein, a reversible surface-enhanced Raman scattering (SERS) nanosensor based on covalent organic frameworks (COFs) was prepared to dynamically monitor the redox processes in living cells. The nanosensor was fabricated by modifying the redox-responsive Raman reporter molecule, 2-Mercaptobenzoquione (2-MBQ), on the surface of gold nanoparticles (AuNPs), followed by the in situ coating of COFs shell. 2-MBQ molecules can repeatedly and quickly undergo reduction and oxidation when successively treated with ascorbic acid (AA) and hypochlorite (ClO-) (as models of reductive and oxidative species, respectively), which resulted in the reciprocating changes of SERS spectra at 900 cm-1. The construction of the COFs shell provided the nanosensor with great stability and anti-interference capability, thus reliably visualizing the dynamics of intracellular redox species like AA and ClO- by SERS nanosensor. Taken together, the proposed SERS strategy opens up the prospects to investigate the signal transduction pathways and pathological processes related with redox dynamics.

RESUMO

Precisely modulating the spatial charge migration/separation constitutes the central issue in dictating the solar conversion efficiency of photoelectrochemical (PEC) cells, whereas it still remains a grand challenge. Here, we conceptually demonstrate the construction of hierarchically ordered metal oxide (MO)/transition-metal chalcogenide quantum dots (TMC QDs) multilayered heterostructured photoanodes, that is, MO/[TMC QDs(+)/TMC QDs(-)]n (TMC QDs: CdTe, CdSe, CdS), by a simple and general bottom-up self-assembly route. Tailor-made intrinsically oppositely charged TMC QDs are alternately deposited on the highly ordered MO via a generic ligand-triggered electrostatic interaction to craft heterostructured photoanodes. The charge-transfer pathway stimulated by the photosensitization of TMC QDs is finely tuned by the assembly sequence. The advantageous multilayered nanoarchitecture renders the MO/[TMC QDs(+)/TMC QDs(-)]n photoanodes exhibit substantially enhanced PEC performances under light irradiation, owing to the applicable energy-level configuration and peculiar combination fashion between building blocks and considerably boosted interfacial charge separation resulting from generating spatial tandem charge transport. Furthermore, photosensitization efficiency comparison among TMC QDs is comprehensively performed with PEC mechanisms elucidated.

RESUMO

As the heat load within the central core of chirally-coupled-core (CCC) fibers will change the pre-designed refractive index profile through the thermo-optic effect, its impact on the laser performance of CCC fibers is investigated. Analysis and simulation results on two typical CCC fibers show that the effects of the heat load include the modal loss reduction and the transmission spectrum drift. The former comes from the thermal lensing effect in the central core, and the latter is caused by the change in the refractive index difference between the central core and the side core. Considering the non-uniform axial heat distribution in the actual laser operation, the overall laser performance of CCC fibers with different pump power is simulated. It is found that, because of the high pre-designed high-order mode loss, the single-mode operation of CCC fibers will be maintained but the slope efficiency may reduce dramatically if the fundamental mode loss is strongly dependent on the heat load.

RESUMO

Mode instabilities (MI) threshold in the Yb:YAG crystalline fiber amplifier is simulated by a full numerical model. The propagation of signal fields is simulated by the finite-difference beam-propagation method combined with the rate equations, and the time-dependent heat equation is solved by the alternating-direction-implicit method. Considering the strong temperature-dependent laser performance of Yb:YAG, an iterative method is applied to reach the steady state of Yb:YAG, the crystalline fiber amplifier, before the simulation of MI behavior. The simulated MI thresholds in Yb:YAG crystalline fiber amplifiers are found to be at least 28 times of those in Yb-doped silica-glass fiber amplifiers, up to tens of kilowatts. Simulation results show that, in addition to the expected higher thermal conductivity and lower thermo-optic coefficient, strong gain saturation also plays an important role in the high MI threshold of the Yb:YAG crystalline fiber.

RESUMO

A new three-dimensional finite-difference (FD)-based beam propagation method (BPM) is proposed for simulating optical propagation in weakly guiding waveguides with torsional birefringence, which cannot be simulated by existing FD-BPM algorithms. We also demonstrate that this new BPM algorithm is capable of obtaining eigenmode solutions in a helically-symmetric z-dependent waveguide structure with torsional birefringence.

RESUMO

An all-in-fiber method of generating orbital angular momentum (OAM) is proposed. A simple device composed with a section of helically symmetric fiber and another section of regular fiber is designed to convert input light to optical vortices. Finite element method calculation of first- and second-order OAM generation based on the coordinates transformation technique is taken to show that the eigenmodes of the helically symmetric fiber structures carry orbital and spin angular momentum. Simulation using the self-developed beam propagation method algorithm is also performed to verify the orbital angular momentum generation and evaluate the performance of the OAM generator.

RESUMO

Both the intestine and liver develop from the endoderm, yet little is known how these two digestive organs share and differ in their developmental programs, at the molecular level. A classical forward genetic screen, with no gene bias, is an effective way to address this question by examining the defects of the intestine and liver in obtained mutants to assess mutated genes responsible for the development of either organ or both. We report here such a screen in zebrafish. ENU was used as the mutagen because of its high mutagenic efficiency and no site preference. Embryos were collected at 3.5 dpf for RNA whole mount in situ hybridization with a cocktail probe of the intestine marker ifabp and the liver marker lfabp to check phenotypes and determine their parental heterozygosis. A total of 52 F2 putative mutants were identified, and those with general developmental defects were aborted. To rule out non-inheritable phenotypes caused by high mutation background, F2 putative mutants were outcrossed with wild type fish and a re-screen in F3 generations was performed. After complementation tests between F3 mutants with similar phenotypes originating from the same F2 families, a total of 37 F3 mutant lines originated from 22 F2 families were identified after screening 78 mutagenized genomes. Classification of mutant phenotypes indicated that 31 out of the 37 mutants showed defects in both the intestine and liver. In addition, four "intestine specific mutants" and two "liver specific mutants" showed selectively more severe phenotype in the intestine and liver respectively. These results suggested that the intestine and liver share a substantial number of essential genes during both organs development in zebrafish. Further studies of the mutants are likely to shed more insights into the molecular basis of the digestive system development in the zebrafish and vertebrate.

RESUMO

A core-shell nanostructure of gold nanoparticles@covalent organic framework (COF) loaded with palladium nanoparticles (AuNPs@COF-PdNPs) was designed for the rapid monitoring of catalytic reactions with surface-enhanced Raman spectroscopy (SERS). The nanostructure was prepared by coating the COF layer on AuNPs and then in situ synthesizing PdNPs within the COF shell. With the respective SERS activity and catalytic performance of the AuNP core and COF-PdNPs shell, the nanostructure can be directly used in the SERS study of the catalytic reaction processes. It was shown that the confinement effect of COF resulted in the high dispersity of PdNPs and outstanding catalytic activity of AuNPs@COF-PdNPs, thus improving the reaction rate constant of the AuNPs@COF-PdNPs-catalyzed hydrogenation reduction by 10 times higher than that obtained with Au/Pd NPs. In addition, the COF layer can serve as a protective shell to make AuNPs@COF-PdNPs possess excellent reusability. Moreover, the loading of PdNPs within the COF layer was found to be in favor of avoiding intermediate products to achieve a high total conversion rate. AuNPs@COF-PdNPs also showed great catalytic activities toward the Suzuki-Miyaura coupling reaction. Taken together, the proposed core-shell nanostructure has great potential in monitoring and exploring catalytic processes and interfacial reactions.

RESUMO

Supernumerary teeth are common developmental anomalies of dentition. However, the factors and mechanisms driving their formation remain largely unknown. Here, we report that conditional knockout of Fst, encoding an antagonist for the transforming growth factor ß (TGF-ß) signaling pathway, in both oral epithelium and mesenchyme of mice (Fst CKO ) led to supernumerary upper incisor teeth, arising from the lingual dental epithelium of the native teeth and preceded by an enlarged and split lingual cervical loop. Fst-deficiency greatly activated TGF-ß signaling in developing maxillary incisor teeth, associated with increased epithelium cell proliferation. Moreover, Fst CKO teeth exhibited increased expression of Tbx1, Sp6, and Sox2, which were identified as direct targets of TGF-ß/SMAD2 signaling. Finally, we show that upregulation of Tbx1 in response to Fst-deficiency was largely responsible for the formation of extra teeth in Fst CKO mice. Taken together, our investigation indicates a novel role for Fst in controlling murine tooth number by restricting TGF-ß signaling.

RESUMO

To investigate difference in the quality of the different parts (back, tail muscles, and fish skin) of Opsariichthys bidens from pond and rice field cultures, a comparative study was conducted in terms of nutritional composition, volatile flavor profiles and gut microbiota. In detail, the texture, free amino acids, fatty acids were further assessed. The results suggested that the moisture content, crude protein and crude fat content in the skin of O. bidens are higher than those in the back and tail muscles, regardless of breeding modes. The fish cultured in the rice field had a higher protein content than those from the pond culture, while the fat content of the rice field-cultured fish was significantly low compared to the fish from the pond culture, especially in the back and tail parts. A total of 43 volatile components were detected by Gas Chromatography-Mass Spectrometry (GC-MS), with a maximum of 18 types of aldehydes and the highest concentration being nonanal. Compared to pond cultures, the fish from the rice field cultures showed more abundant flavor composition and odor-active compounds. The total content of DHA (Docosahexaenoic Acid) and EPA (Eicosapentaenoic Acid) in the rice field-cultured fish was higher than that of the pond group, while no significant disparity in amino acid composition was observed (p > 0.05). Comparative and clustering analyses of gut microbiota revealed notable discrepancies in the gut microbiota of O. bidens from two aquaculture systems. However, an inherent correlation between the gut microbiome and meat quality would be further emphasized in further studies. This study can offer a theoretical reference for the development of high-quality aquatic products by selecting the appropriate aquaculture models.

RESUMO

Hydrogen sulfide (H2S), an important gas signal molecule, participates in intercellular signal transmission and plays a considerable role in physiology and pathology. However, in-situ monitoring of H2S level during the processes of material transport between cells remains considerably challenging. Herein, a cell membrane-targeted surface-enhanced Raman scattering (SERS) nanoprobe was designed to quantitatively detect H2S secreted from living cells. The nanoprobes were fabricated by assembling cholesterol-functionalized DNA strands and dithiobis(phenylazide) (DTBPA) molecules on core-shell gold nanostars embedded with 4-mercaptoacetonitrile (4-MBN) (AuNPs@4-MBN@Au). Thus, three functions including cell-membrane targeted via cholesterol, internal standard calibration, and responsiveness to H2S through reduction of azide group in DTBPA molecules were integrated into the nanoprobes. In addition, the nanoprobes can quickly respond to H2S within 90 s and sensitively, selectively, and reliably detect H2S with a limit of detection as low as 37 nM due to internal standard-assisted calibration and reaction specificity. Moreover, the nanoprobes can effectively target on cell membrane and realize SERS visualization of dynamic H2S released from HeLa cells. By employing the proposed approach, an intriguing phenomenon was observed: the other two major endogenous gas transmitters, carbon monoxide (CO) and nitric oxide (NO), exhibited opposite effect on H2S production in living cells stimulated by related gas release molecules. In particular, the introduction of CO inhibited the generation of H2S in HeLa cells, while NO promoted its output. Thus, the nanoprobes can provide a robust method for investigating H2S-related extracellular metabolism and intercellular signaling transmission.

RESUMO

Glutathione S-transferases are important detoxification enzymes involved in insecticide resistance. Sequencing the Tribolium castaneum genome provides an opportunity to investigate the structure, function, and evolution of GSTs on a genome-wide scale. Thirty-six putative cytosolic GSTs and 5 microsomal GSTs have been identified in T. castaneum. Furthermore, 40, 35, 13, 23, and 32 GSTs have been discovered the other insects, Drosophila, Anopheles, Apis, Bombyx, and Acyrthosiphon, respectively. Phylogenetic analyses reveal that insect-specific GSTs, Epsilon and Delta, are the largest species-specific expanded GSTs. In T. castaneum, most GSTs are tandemly arranged in three chromosomes. Particularly, Epsilon GSTs have an inverted long-fragment duplication in the genome. Other four widely distributed classes are highly conserved in all species. Given that GSTs specially expanded in Tribolium castaneum, these genes might help to resist poisonous chemical environments and produce resistance to kinds of different insecticides.

RESUMO

Oxidative stress is involved in various signaling pathways and serves a key role in inducing cell apoptosis. Therefore, it is significant to monitor oxidative stress upon drug release for the assessment of therapeutic effects in cancer cells. Herein, a glutathione (GSH)-responsive surface-enhanced Raman scattering (SERS) nanoplatform is proposed for ultra-sensitively monitoring the substance related with oxidative stress (hydrogen sulfide, H2S), depleting reactive sulfur species and releasing anticancer drugs to amplify oxidative stress for tumor apoptosis. The Au@Raman reporter@Ag (Au@M@Ag) nanoparticles, where a 4-mercaptobenzonitrile molecule as a Raman reporter was embedded between layers of gold and silver to obtain sensitive SERS response, were coated with a covalent organic framework (COF) shell to form a core-shell structure (Au@M@Ag@COFs) as the SERS nanoplatform. The COF shell loading doxorubicin (DOX) of Au@M@Ag@COFs exhibited the GSH-responsive degradation capacity to release DOX, and its Ag layer as the sensing agent was oxidized to Ag2S by H2S to result in its prominent changes in SERS signals with a low detection limit of 0.33 nM. Moreover, the releasing DOX can inhibit the generation of H2S to promote the production of reactive oxygen species, and the depletion of reactive sulfur species (GSH and H2S) in cancer cells can further enhance the oxidative stress to induce tumor apoptosis. Overall, the SERS strategy could provide a powerful tool to monitor the dynamic changes of oxidative stress during therapeutic processes in a tumor microenvironment.

RESUMO

The core factors affecting the efficiency of photocatalysis are predominantly centered on controllable modulation of anisotropic spatial charge separation/transfer and regulating vectorial charge transport pathways in photoredox catalysis, yet it still meets with limited success. Herein, we first conceptually demonstrate the rational design of unidirectional cascade charge transfer channels over transition metal chalcogenide nanosheets (TMC NSs: ZnIn2S4, CdS, CdIn2S4, and In2S3), which is synergistically enabled by a solid-state non-conjugated polymer, i.e., poly(diallyldimethyl ammonium chloride) (PDDA), and MXene quantum dots (MQDs). In such elaborately designed photosystems, an ultrathin PDDA layer functions as an intermediate charge transport mediator to relay the directional electron transfer from TMC NSs to MQDs that serve as the ultimate electron traps, resulting in a considerably boosted charge separation/migration efficiency. The suitable energy level alignment between TMC NSs and MQDs, concurrent electron-withdrawing capabilities of the ultrathin PDDA interim layer and MQDs, and the charge transport cascade endow the self-assembled TMC/PDDA/MQD heterostructured photosystems with conspicuously improved photoactivities toward anaerobic selective reduction of nitroaromatics to amino derivatives and photocatalytic hydrogen evolution under visible light irradiation. Furthermore, we ascertain that this concept of constructing a charge transfer cascade in such TMC-insulating polymer-MQD photosystems is universal. Our work would afford novel insights into smart design of spatial vectorial charge transport pathways by precise interface modulation via non-conjugated polymers for solar energy conversion.

RESUMO

Purpose: This research focused primarily on the impact of the SARS-CoV-2 Vaccine (VeroCell) on Chinese physical education (PE) students' health and physical activity (PA) performance. Methods: This study used quantitative methods and phenomenological procedures to collect and analyze data. Survey techniques were the main method used for collecting data from Chinese university students, using a self-designed questionnaire with a Cronbach's alpha α value of 0.76. To ensure the quality of the study, confirmatory factor analyses (CFA) were conducted, and the internal consistency reliability of the instrument was measured (alpha coefficient = 0.82). The determined sample size was 490 and around 90% as the minimum sample size was determined with the help of a sample size calculator. The author using factor loadings with h2 and an independent-sample t-test analyzed the responses of the remaining valid participants (n = 443 with a response rate of 90.40). Results: Most participants (around 94%) did not experience any adverse reactions that impacted their daily life activities, health, or performance during physical activity. However, about 30-40% of students felt lethargy, weakness, muscle pain, or swelling. Regarding the impact of the vaccine on daily life, there was no difference in the responses between participants who had only received one shot of the coronavirus disease 2019 (COVID-19) vaccine and those who had received two shots (p > 0.05 in most cases). Conclusion: The study concluded that the COVID-19 vaccine had no significant effect on PE students' daily activities, health, and PA performance. The results of this study could be used by policymakers to encourage people to get vaccinated and eradicate the isolation caused by COVID-19, which leads many people to develop various non-communicable diseases (NCDs).

RESUMO

Electroconductive polystyrene (PS) composites with ideal flame-retardant properties are considered as potential electromagnetic interference (EMI) shielding materials. In this work, PS/silicon wrapped ammonium polyphosphate/multi-wall carbon nanotubes (PS/SiAPP/MWCNT) composites with segregated structure were synthesized via the methods of balling mill and hot-pressing. The obtained results revealed that the SiAPP and MWCNT were successfully introduced onto PS spheres and showed uniform distribution on the PS surface. The thermogravimetric analysis showed that PS/SiAPP/MWCNT containing 7 wt% MWCNT exhibited excellent thermal stability. Furthermore, the results of cone calorimeter test indicated that the heat release rate and total heat release of the PS/SiAPP/MWCNT containing a loading of 7 wt% MWCNT were reduced by 60.5% and 33.9%, respectively. In addition, the EMI shielding performance could reach 11 dB. Above results implied that the synergistic effect between the MWCNT and SiAPP effectively enhanced the flame retardant performance of the PS by promoting the generation of dense and continuous char layer to protect the PS from burning. The multiple reflection and adsorption are responsible for improved EMI shielding effectiveness. Therefore, segregated PS/SiAPP/MWCNT hybrid is an up-and-coming candidate for satisfactory EMI shielding materials with exceptional fire retardancy for electronic devices.
