RESUMEN

Unmanned Aerial Vehicles (UAV) have revolutionized the aircraft industry in this decade. UAVs are now capable of carrying out remote sensing, remote monitoring, courier delivery, and a lot more. A lot of research is happening on making UAVs more robust using energy harvesting techniques to have a better battery lifetime, network performance and to secure against attackers. UAV networks are many times used for unmanned missions. There have been many attacks on civilian, military, and industrial targets that were carried out using remotely controlled or automated UAVs. This continued misuse has led to research in preventing unauthorized UAVs from causing damage to life and property. In this paper, we present a literature review of UAVs, UAV attacks, and their prevention using anti-UAV techniques. We first discuss the different types of UAVs, the regulatory laws for UAV activities, their use cases, recreational, and military UAV incidents. After understanding their operation, various techniques for monitoring and preventing UAV attacks are described along with case studies.

In recent years, municipal authorities especially in the developing nations are battling to select the best health care waste (HCW) disposal technique for the effective treatment of the medical wastes during and post COVID-19 era. As evaluation of various disposal alternatives of HCW and selection of the best technique requires considering various tangible and intangible criteria, this can be framed as multi-criteria decision-making (MCDM) problem. In this paper, we propose an assessment framework for the selection of the best HCW disposal technique based on socio-technical and triple bottom line perspectives. We have identified 10 criteria on which the best HCW disposal techniques to be selected based on extant literature review. Next, we use Fuzzy VIKOR method to evaluate 9 HCW disposal alternatives. The effectiveness of the proposed framework has been demonstrated with a real-life case study in Indian context. To check the robustness of the proposed methodology, we have compared the results obtained with Fuzzy TOPSIS (Technique of Order Preference Similarity to the Ideal Solution). The results help the municipal authorities to establish a methodical approach to choose the best HCW disposal techniques. Our findings indicate that incineration is the best waste disposal technique among the available alternatives. Even if the dataset indicates 'incineration' is the best method, we must not forget about the environmental concerns arising from this method. In COVID time, incineration may be the best method as indicated by the data analysis, but "COVID" should not be an excuse for causing "Environmental Pollution".

The deregulation of the epidermal growth factor receptor (EGFR) has a significant role in the progression of tumors. Despite the development of a number of EGFR-targeting agents that can arrest tumor growth, their success in the clinic is limited in several tumor types, particularly in the highly malignant glioblastoma multiforme (GBM). In this study, we generated and characterized EGFR-specific nanobodies (ENb) and imageable and proapoptotic ENb immunoconjugates released from stem cells (SC) to ultimately develop a unique EGFR-targeted therapy for GBM. We show that ENbs released from SCs specifically localize to tumors, inhibit EGFR signaling resulting in reduced GBM growth and invasiveness in vitro and in vivo in both established and primary GBM cell lines. We also show that ENb primes GBM cells for proapoptotic tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-induced apoptosis. Furthermore, SC-delivered immunoconjugates of ENb and TRAIL target a wide spectrum of GBM cell types with varying degrees of TRAIL resistance and significantly reduce GBM growth and invasion in both established and primary invasive GBM in mice. This study demonstrates the efficacy of SC-based EGFR targeted therapy in GBMs and provides a unique approach with clinical implications.

Only a subset of cancer patients inoculated with oncolytic herpes simplex virus (oHSV) type-1 has shown objective response in phase 1 and 2 clinical trials. This has raised speculations whether resistance of tumor cells to oHSV therapy may be a limiting factor. In this study, we have identified established and patient derived primary glioblastoma multiforme (GBM) stem cell lines (GSC) resistant to oHSV and also to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) that has recently shown promise in preclinical and initial clinical studies. We created a recombinant oHSV bearing a secretable TRAIL (oHSV-TRAIL) and hypothesized that oHSV-TRAIL could be used as a cancer therapeutic to target a broad spectrum of resistant tumors in a mechanism-based manner. Using the identified resistant GBM lines, we show that oHSV-TRAIL downregulates extracellular signal-regulated protein kinase (ERK)-mitogen-activated protein kinase (MAPK) and upregulates c-Jun N-terminal kinase (JNK) and p38-MAPK signaling, which primes resistant GBM cells to apoptosis via activation of caspase-8, -9, and -3. We further show that oHSV-TRAIL inhibits tumor growth and invasiveness and increases survival of mice bearing resistant intracerebral tumors without affecting the normal tissues. This study sheds new light on the mechanism by which oHSV and TRAIL function in concert to overcome therapeutic-resistance, and provides an oncolytic virus based platform to target a broad spectrum of different cancer types.

Alpha7 nicotinic acetylcholine receptors (α7nAChRs) are interesting not only because of their physiological effects, but because this receptor requires chaperones to traffic to cell surfaces (measured by alpha-bungarotoxin [αBGT] binding). While knockout (KO) animals and antibodies that react across species exist for tmem35a encoding the protein chaperone NACHO, commercially available antibodies against the chaperone RIC3 that allow Western blots across species have not been generally available. Further, no effects of deleting RIC3 function (ric3 KO) on α7nAChR expression are reported. Finally, antibodies against α7nAChRs have shown various deficiencies. We find mouse macrophages bind αBGT but lack NACHO. We also report on a new α7nAChR antibody and testing commercially available anti-RIC3 antibodies that react across species allowing Western blot analysis of in vitro cultures. These antibodies also react to specific RIC3 splice variants and single-nucleotide polymorphisms. Preliminary autoradiographic analysis reveals that ric3 KOs show subtle αBGT binding changes across different mouse brain regions, while tmem35a KOs show a complete loss of αBGT binding. These findings are inconsistent with effects observed in vitro, as RIC3 promotes αBGT binding to α7nAChRs expressed in HEK cells, even in the absence of NACHO. Collectively, additional regulatory factors are likely involved in the in vivo expression of α7nAChRs.

INTRODUCTION: Fueled by a growing global expectation of the health and medical fields, billions of dollars/euros/pounds are invested every year in the research of new biological and chemical entities. However, little interest is seen in the development of novel drug delivery systems. One such system, pre-filled syringe (PFS), was invented decades ago but is still a rare mode of delivery in many therapeutic segments. AREAS COVERED: This review comprises properties and effects of extractables, leachables and discuss the characteristics of PFS technology; its composition, glass and polymer types, configuration of PFS, advantages over glass, technical and commercial applicability; its significance against patient, industry, quality, environment and cost; and its business potential. We discuss in brief about PFS used in various major and life-threatening disorders and future prospects. It provides showers of knowledge in the field of PFS drug delivery technology to the reader's, industrialist's and researcher's point of view. EXPERT OPINION: The PFS drug delivery system offers a wonderful panorama to lifesaving drugs that are currently only available in conventional vials and ampoules in the market. A novel approach of Form Fill Seal technology can be adopted for this particular ready-to-use dosage form also, which opens the new global doors for budding researchers in the field of pre-filled drug delivery system.

Anti-coagulants are one of the most important categories in healthcare therapeutics. For healthcare professionals dealing in cases of in-vivo blood clotting problems. Heparin and low molecular weight heparins (LMWHs) would be the first choice of drugs. This review represents an overview of the LMWHs, their importance over heparin and enlightens the advancements. In addition to these, different methods used for preparation and purification are discuss in terms of production and synthesis. Worldwide availability in pre-filled syringe, market, manufacturers and suppliers drug interactions, adverse drug reactions, in-vitro study, freezing/thawing process and structural differences of LMWHs are also focused upon in this review.

Here we introduce diffusion molecular retention (DMR) tumor targeting, a technique that employs PEG-fluorochrome shielded probes that, after a peritumoral (PT) injection, undergo slow vascular uptake and extensive interstitial diffusion, with tumor retention only through integrin molecular recognition. To demonstrate DMR, RGD (integrin binding) and RAD (control) probes were synthesized bearing DOTA (for (111) In(3+)), a NIR fluorochrome, and 5 kDa PEG that endows probes with a protein-like volume of 25 kDa and decreases non-specific interactions. With a GFP-BT-20 breast carcinoma model, tumor targeting by the DMR or i.v. methods was assessed by surface fluorescence, biodistribution of [(111)In] RGD and [(111)In] RAD probes, and whole animal SPECT. After a PT injection, both probes rapidly diffused through the normal and tumor interstitium, with retention of the RGD probe due to integrin interactions. With PT injection and the [(111)In] RGD probe, SPECT indicated a highly tumor specific uptake at 24 h post injection, with 352%ID/g tumor obtained by DMR (vs 4.14%ID/g by i.v.). The high efficiency molecular targeting of DMR employed low probe doses (e.g. 25 ng as RGD peptide), which minimizes toxicity risks and facilitates clinical translation. DMR applications include the delivery of fluorochromes for intraoperative tumor margin delineation, the delivery of radioisotopes (e.g. toxic, short range alpha emitters) for radiotherapy, or the delivery of photosensitizers to tumors accessible to light.
