RESUMEN

In the present study the concentration of selected elements in tissues of domestic rabbits and of wild brown-hares (kidneys, liver, and muscle - m. quadriceps femoris) in Slovakian habitats were determined. After mineralization the elements examined were detected using flame atomic absorption spectrophotometry/graphite furnace atomic absorption spectrophotometry. For rabbits, Fe in the liver was correlated with essential (Mn, Cu) (R2 = 0.94, p < 0.05; R2 = 0.96, p < 0.05 respectively) or toxic (Pb) elements (R2 = -0.93, p < 0.05). For hares, significant correlations were found between Cd and Cu or between Cd and Mn in the kidneys (R2 = -0.96, p < 0.05; R2 = 0.92, p < 0.05 respectively), which is the target organ for Cd. Higher concentrations of the elements were found in hare tissue, and this may be linked to pollution of their wild habitats. The xenobiotic elements as well as the essential elements were accumulated in the kidneys of the hares than rabbits. For liver, differences were less pronounced and significance was only for Fe and Cu. Muscle of hares was more contaminated than of rabbits for both biogenic and toxic elements. These results show that detectable concentrations of inorganic elements. These levels may be linked to contamination of the natural habitats of wild biota due to industry, traffic, agriculture, and urban sprawl.

Caffeine content is a crucial attribute of coffee. Its concentration and thus maximum cups of Coffea arabica from Africa, Asia, Central America, and South America from different altitudes of growing areas, altitude, and process using different post-harvest processing (dry, wet, and pulped natural). Our results suggest that geographical origin might affect the alkaloid concentration in C. arabica. The caffeine concentration pattern in green samples was as follows: Central America > South America > Asia > Africa. Altitude affected the concentrations, lowlands > midlands > highlands, however, not significantly. Given caffeine is thermostable, the medium roasting process did not affect the concentration of caffeine directly, but a small increase was observed. Scientific opinion on the safety of habitual caffeine consumption of up to 400 mg per day does not raise safety concerns for non-pregnant adults. A cup (7 g coffee in 120 mL of water) was used for recalculation. Results suggest that mostly highlands and midlands coffee from Africa reached levels of caffeine that might be consumed in more than 5.5 cups a day.

This study examined for the first time whether bee bread (BB, consisting of monofloral rape bee pollen) could alleviate lipid derangements and reduced bone quality in Zucker diabetic fatty (ZDF) rats, which are considered an appropriate animal model for type 2 diabetes mellitus (T2DM) investigation. Adult ZDF rats were segregated into four groups: lean non-diabetic rats (L group), obese diabetic rats untreated (C group), and those treated with the BB at two doses (500 and 700 mg/kg body weight, respectively, B1 and B2 groups) for 10 weeks. Significantly reduced levels of total cholesterol and triglyceride were recorded in the B2 group versus the C group. In both BB-treated groups, significantly increased relative volume of trabecular bone and trabecular thickness, enhanced density of secondary osteons, accelerated periosteal bone apposition, and improved blood flow were observed. A positive effect of higher dose of BB on femoral weight and cortical bone thickness was also demonstrated. Our results suggest a promising potential of BB to ameliorate T2DM-related complications associated with lipid and bone damages.

This study was focused to determine an individual and combined effect of mycotoxin citrinin (CIT) and two compounds of the stilbene family- resveratrol (RES) and his dimethyl ether analogue pterostilbene (PTE) which have many health benefits. As a model the human adenocarcinoma cell line HT-29 was used which may exhibits the properties of small intestine cells. Viability, plasma membrane integrity, lysosomal functionality, intracellular production of superoxide anions and superoxide dismutase activity were examined. The results indicate that concentrations of 50 and 100 µg/mL of the tested compounds were cytotoxic in mostly monitored parameters and probably caused apoptosis. HT-29 cells were more sensitive to PTE than to RES with a higher antioxidant effect of PTE than RES, which may be caused by its chemical structure. Both stilbenes at medium doses act as effective superoxide anions scavengers leading to reduction of oxidative stress and consequent cell damage. The nontoxic concentration of RES (25 µg/mL) protects the HT-29 cell line faced to the toxicity of CIT at 25 µg/mL by increasing viability of cells and by reducing the superoxide production induced by CIT concentrations of 12.5 µg/mL and 25 µg/mL.

The purpose of the present study was to evaluate the effects of aflatoxin B1 (AFB1) and benzo[a]pyrene (BaP) on the heart muscle of chicken embryos of the broiler line Ross 308. The benzo[a]pyrene in the organic oil solution was injected in ovo on the 6th day of the incubation in doses of: 0.1, 0.5, and 1 mg/kg weight of eggs; the aflatoxin B1 in the organic oil solution was injected in ovo on the 6th day of the incubation into the yolk in doses of 80, 120 and 240 ng/kg weight of eggs. Multiple biochemical and hepatic parameters have been observed, including sodium, potassium, chloride, cholesterol, uric acid, total proteins, aminotransferase aspartate, and aminotransferase alanine. A low dose of AFB1 and BaP administered in ovo during early embryonic development had a significant impact on chicken embryonic development, as demonstrated by alterations in biochemical, mineral, and hepatic parameters.

The examination of various elements in the milk products is very important in the food sector in respect of food quality and safety. The aim of this study was to determine the concentrations of calcium (Ca), cobalt (Co), cadmium (Cd), copper (Cu), chromium (Cr), iron (Fe), mercury (Hg), potassium (K), magnesium (Mg), sodium (Na), nickel (Ni), phosphorus (P), lead (Pb) and zinc (Zn) in white cottage cheese or cottage cheese supplemented with various additives (white, lacto-free, chive, tzatziki, mustard + onion, chili, active protein) available on the market of Slovakia. All essential elements were within the reference range. Cottage cheese enriched with tzatziki showed higher amount of Cu, Fe, K, and Zn. Mustard + onion cheese contained high values of Ca, Co, Mg, and Ni. In white cottage cheese high amount of Cr, Mn, and P was measured. The content of xenobiotic metals was below permitted limit. The contribution to PTWI (Provisional tolerable weekly intake) suggested very low dietary exposure to heavy metals as Cd, Hg, and Pb as well as other metals (Cu, Ni, and Zn) in cottage cheese. Numerous correlations between concentrations were observed. MOE (Margin of Exposure) evaluation denoted that average consumption of cottage cheese does not pose any high cardiovascular and nephrotoxicity threat.

The total polyphenolic content and the antioxidant activity have been analyzed in ground beans of green, light, medium and dark roasted coffee by UV-VIS spectrometry. Water coffee extracts showed the highest levels of polyphenols in green and light roasted coffees where the total polyphenolic content (TPC) ranged from 49.19 ± 0.70 to 74.05 ± 0.28 and from 59.79 ± 1.45 to 38.34 ± 1.26 g GAE.kg-1, respectively. In medium roast samples it ranged from 43.90 ± 3.07 to 74.05 ± 0.28g GAE.kg-1 and in dark roast from 37.44 ± 0.63 to 47.41 ± 0.69 g GAE.kg-1. The total antioxidant capacity (TAC) reached the highest values (DPPH inhibition ranging from 69.08 ± 1.33% to 78.55 ± 0.89%) in light roasted coffees. Dark roasted coffees showed both the lowest content of polyphenols as well as the total antioxidant capacity. In case of TPC, statistically significant differences (P˂0.001) have been identified between green coffee and other roasted degrees. Also, dark coffee showed statistically noticeable differences (P˂0.001) in TPC in relation to other roasted stages. Statistically important difference (P˂0.001) has been discovered between the total antioxidant capacity of dark roasted coffee and other roasting levels. The results demonstrated that roasting process affects both the oxidative activity as well as polyphenolic content.

(1) Background: Impaired adipose tissue function leads to the development of metabolic disorders. Reactive oxygen species play a key role in the regulation of adipogenesis and insulin-stimulated glucose uptake by adipocytes. Quercetin (QCT) regulates adipogenesis by affecting the redox state of preadipocytes. Ochratoxin A (OTA) is one of the most prevalent mycotoxins contaminating food. It has cytotoxic, genotoxic, pro-inflammatory, and anti-adipogenic effects. Antioxidants are believed to protect cells from the cytotoxicity and genotoxicity induced by OTA. The aim of this study was to investigate the effect of QCT and OTA application on preadipocyte differentiation, oxidative status, and adipocyte metabolism. (2) Methods: Primary rat preadipocytes were isolated from subcutaneous adipose tissue of Wistar rats. Gene expressions were determined by qPCR. Cell viability, reactive oxygen species (ROS) production, glucose uptake, and lipid accumulation were determined using commercially available kits. (3) Results: A dose-dependent inhibitory effect of QCT on adipogenic differentiation was observed, which was accompanied by a decrease in ROS production. Reduced ROS formation is closely related to impaired glucose uptake by adipocytes. (4) Conclusions: The results of this study indicate a key role of ROS in regulating adipogenesis and metabolic pathways, which is affected by the application of QCT and/or OTA.

The goal of this study was to determined polychlorinated biphenyls (PCBs) and organochlorine pesticides in the depot fat of roe deer (Capreolus capreolus) coming from south-western Slovakia. The mutual correlations of the organic pollutants were analyzed. The study included dichlorodiphenyltrichloroethane (DDT), hexachlorobenzen (HCB), alpha-hexachlorocyclohexane and beta-hexachlorocyclohexane (α + ß-HCH), gamma-hexachlorocyclohexane (γ-HCH), and polychlorinated biphenyls (PCB-delor). The gas chromatograph with an electron capture detector ECD was used for analysis. The accumulations of organic pollutant in depot fat of roe deer were in following order: DDT > PCB-delor > α + ß-HCH > HCB > γ-HCH. Among all pollutants, DDT was accumulated significantly in the highest level in the samples. The significantly higher content of DDT, HCB, α + ß-HCH, and γ-HCH was detected in the adult animals when compared to the juveniles. Some strong positive correlations among pollutants, between HCB and DDT, α + ß-HCH and HCB, α + ß-HCH and HCB, between γ-HCH and other pollutants, and between PCB-delor and γ-HCH were found. Game animals are a part of human food chain and monitoring of the environment pollution by PCBs and other organic pollutants are worthy to study.

In the present study, we aimed to assess antioxidant status in erythrocytes in vitro after patulin (PAT) and epicatechin exposure by measuring antioxidant enzymes (superoxide-dismutase - SOD, glutathione peroxidase - GPx and catalase - CAT) and parameters associated with oxidative stress (malondialdehyde - MDA and ROS). We also investigated the effect of PAT on viability and count of lymphocytes and lymphocyte subpopulations in rabbit blood in vitro. Whole blood of rabbits was used for analysis of antioxidant changes in rabbit erythrocytes after epicatechin and PAT treatment (separately or in combination, at concentrations of 0.2; 2; 20; 200 µg mL-1 of epicatechin and 0.5; 5; 10 µg mL-1 of PAT). Whole blood of rabbits was also used for analysis of count and viability of lymphocytes after PAT treatment at concentrations of 10; 25 and 50 µg mL-1. Results from our experiment confirmed the ability of epicatechin to protect cells against oxidative stress and lipoperoxidation. Our findings indicate that mycotoxin PAT in low concentrations did not affect the activity of antioxidant enzymes in erythrocytes of rabbits significantly. Only slight non-significant changes in lymphocytes count after treatment with low doses of PAT in rabbit blood were observed.

RESUMEN

The goal of this study was to monitor the accumulation of aflatoxin B1 in the liver and kidney of brown hares (Lepus europaeus Pall) in the region of south-western Slovakia. A total of 65 samples were involved for analysis by RIA method. Brown hares were divided into the groups according to age, sex and season (month). The sex was determined visually after shooting, and the age was assigned from dried eye lens. The average concentration of AFB1 in the liver of hares was 0.54 ± 0.053 µg/kg, and lower values were measured in the kidney (0.41 ± 0.038 µg/kg). The significantly (P < 0.05) higher values were found in winter months when compared to summer months. According to the age, juvenile animals showed significant higher accumulation of B1 in both organs than adults (P < 0.05). Wild animals can serve as a good model of real environmental contamination. Thus, monitoring of risk factors such as mycotoxins in the environment is important with regard to public health, as game animals constitute an important part of food chain for humans.

The objective of this study was to determine the concentrations of Cu, Cd, Pb, Mn, Cr, Co, Ni, Zn, and Hg in the white and fruit parts of commercially available yogurts (n = 30) from Nitra markets (Slovak Republic). The results were correlated to determine their relationships. Three yogurt fruit flavors were chosen and tested, strawberry (n = 10), blueberry (n = 10), and cherry (n = 10). The elements were analyzed using atomic absorption spectrophotometry. Higher concentrations of toxic elements, such as Cd and Pb, were found in the fruit parts of the yogurt, and in some cases, the tolerable limit was exceeded. The white part of the yogurt was not contaminated by toxic elements. White yogurt is a good source of nutrients for humans, but the fruit part in yogurt requires detailed monitoring and improvements in the processing techniques.

RESUMEN

The contamination of food and feed by mycotoxins as toxic metabolites of fungi is a risk not only for consumers resulting in various embarrassment regarding health status and well-being, but also for producers, companies and export market on the ground of economic losses and ruined stability of economic trade. As it is given in historical evidence, the contamination of food by mycotoxins is a topic as old as a history of mankind, finding some evidence even in the ancient books and records. Nowadays, the mycotoxins are used in modern biotechnological laboratories and are considered an agent for targeting the specific cells (e.g., defected cells to eliminate them). However, this promising procedure is only the beginning. More concern is focused on mycotoxins as abiotic hazard agents. The dealing with them, systematic monitoring, and development of techniques for their elimination from agricultural commodities are worldwide issues concerning all countries. They can be found alone or in co-occurrence with other mycotoxins. Thus, this review aims to provide widened information regarding mycotoxins contamination in environment with the consequences on health of animals and humans. The inevitability for more data that correctly determine the risk points linked to mycotoxins occurrence and their specific reactions in the environment is demonstrated. This review includes various symptoms in animals and humans that result from mycotoxin exposure. For better understanding of mycotoxin's impact on animals, the sensitivities of various animal species to various mycotoxins are listed. Strategies for elimination and preventing the risks of mycotoxins contamination as well as economical approach are discussed. To complete the topic, some data from past as historical evidences are presented.

The aim of this study was to determine the activity of superoxide dismutase (SOD), glutathione peroxidase (GPx), production of reactive oxygen species (ROS), and total antioxidant status (TAS) in porcine ovarian granulosa cells after quercetin and T-2 toxin exposure in vitro. Porcine ovarian granulosa cells were incubated with quercetin and T-2 toxin separately or in mutual combination at the doses of 1 ng/mL, 10 ng/mL, 100 ng/mL, and the control group without any additions for 24 h. In this study T-2 toxin developed stress reaction in porcine ovarian granulosa cells and increased generation of ROS. Quercetin had no effect in elimination of ROS generation induced by T-2 toxin, but was effective in maintaining and increasing of TAS, activities of SOD and GPx in porcine granulosa cells in vitro. These results contribute towards the understanding of cellular stress and its response.

The aim of this study was to determinate the content of some elements in a specific dairy product, mozzarella, in a particular area of western Slovakia and to evaluate the estimation of the risk to the consumers based on the contribution to the provisional tolerable weekly intake. The consumption of mozzarella can contribute to the intake of important elements in the diet, such as calcium and magnesium, along with others. The contents of some toxic and trace elements were low and have not exceeded the permitted limit. In addition, the contribution to PTWI was found to be very low, which means that the consumption of mozzarella possesses no risk to humans. It is concluded that the data obtained in this study can help as a valuable addition to methodological and scientific material in the field of food safety of dairy products and their positive impact on human health.

Isorhamnetin has gained research interest for its anti-inflammatory, anti-proliferative and chemoprotective properties. In this study, human colon adenocarcinoma cells were cultured in the presence or absence of different isorhamnetin concentrations (5-150 µM) for 24 h or 48 h of cultivation to explore the impact on several parameters of viability/proliferation (mitochondrial function using an MTT test, metabolic activity, cell membrane integrity and lysosomal activity using a triple test). The intracellular generation of superoxide radicals using an NBT test and ELISA analysis was performed to observe the biosynthesis of interleukin 8 (IL-8) in cells stimulated with zymosan, as well as in basal conditions. The antiproliferative activity of isorhamnetin was demonstrated by significantly reduced values of mitochondrial and metabolic activity, integrity of cell membranes and lysosomal activity. Its high prooxidant potential was reflected by the significantly elevated generation of superoxides even in cells with low viability status. The anti-inflammatory effect of isorhamnetin was evident due to decreased IL-8 production, and the most significant decline in IL-8 concentration was observed after 24 h treatment in cells with induced inflammation. We demonstrated that isorhamnetin can suppress the proliferation of HT-29 cells, and this effect was correlated with pro-oxidative and anti-inflammatory activity of isorhamnetin.

Bisphenol A (BPA) is an endocrine-disruptive chemical that is widely utilized in the production of polycarbonate plastic and epoxy resin, which are used to make a wide range of consumer products, food and drink containers, and medical equipment. When the potential risk of BPA emerged, it was substituted by allegedly less harmful substitutes such as bisphenols S, F, B, and AF. However, evidence suggests that all bisphenols can have endocrine-disruptive effects, while the extent of these effects is unknown. This study aimed to determine effect of BPA, BPAF, BPB, BPF, and BPS on viability and steroidogenesis in human adrenocortical carcinoma cell line in vitro. The cytotoxicity of bisphenols was shown to be considerable at higher doses. However, at low concentrations, it improved viability as well as steroid hormone secretion, indicating that bisphenols have a biphasic, hormetic effect in biological systems. The results are consistent with the hypothesis that bisphenols selectively inhibit some steroidogenic enzymes. These findings suggest that bisphenols have the potential to disrupt cellular steroidogenesis in humans, but substantially more detailed and systematic research is needed to gain a better understanding of the risks associated with bisphenols and their endocrine-disrupting effect on humans and wildlife.

This study aimed at obtaining the data on the occurrence, levels and correlations of organic pollutants present in game animals (n = 75, Brown hare, Lepus europaeus Pall.) in the region of south-western Slovakia. The analyses performed included dichlorodiphenyltrichloroethane (DDT), hexachlorobenzen (HCB), alpha- and beta hexachlorocyclohexane (α+ß-HCH), gamma-hexachlorocyclohexane (γ-HCH), and polychlorinated biphenyls (PCB-delor, commercial mixture of PCB congeners). A gas chromatograph with an electron capture detector was used for the analysis. PCB-delor and DDT were accumulated significantly in the highest level (0.105 ± 0.059 mg/kg; 0.070 mg/kg) in depot fat in brown hares; however maximum permissible limits for the observed pollutants were not exceeded. Significantly higher concentrations of DDT, HCB, γ-HCH, and PCB-delor were found in adult animals when compared with juvenile hares. Gender and season had no effect on the accumulation of observed pollutants. Moderately positive correlation was found between PCB-delor and DDT (r = 0.59). Monitoring of environmental pollution with polychlorinated hydrocarbons is important with regard to public health, as game animals constitute an important part of food chain also for humans.

The aim of this study was to monitor accumulation of cadmium (Cd), mercury (Hg), zinc (Zn), copper (Cu) and cobalt (Co) in the muscle, liver and kidney of wild boar (Sus scrofa scrofa) from hunting place of western Slovakia and the correlations among the observed elements. A total of 120 samples were involved for analyses by atomic absorption spectrophotometry (AAS). The significantly highest accumulation of Cd in the kidney followed by the liver and muscles was found. Zn accumulated mainly in the liver. Significantly lower values were found in the kidney followed by the muscle. The concentration of Cu was significantly lowest in the muscle when compared to the liver and kidney. Hg and Co accumulated mainly in the kidney, followed by the liver and muscle of wild boars, but without significant differences. In the muscle of wild boar moderately positive correlation between Zn and Cu (r = 0.59), Cd and Co (r = 0.51), Cu and Co (r = 0.33), and Zn and Hg (r = 0.36) were found. In the liver moderately positive correlation between Cd and Hg (r = 0.39) was detected. Moderately positive correlation between Zn and Cu (r = 0.40) was noted for the kidney.

This study reports the effect of dietary nickel (Ni) and a combination of Ni and zinc (Zn) on the accumulation of lead (Pb), cadmium (Cd), Ni and Zn in muscles, liver and kidneys of rabbits. Female rabbits (Oryctolagus cuniculus) of experimental groups were fed a granular mixture with addition of various concentrations of Ni or Ni+Zn (E1 - 17.5 g NiCl(2) per 100 kg of feed mixture, group E2 - 35.0 g NiCl(2) per 100 kg of feed mixture, group E3 - 17.5 g NiCl(2) per 100 kg of feed mixture and 30 g ZnCl(2) per 100 kg of feed mixture, group E4 - 35.0 g NiCl(2) per 100 kg of feed mixture and 30 g ZnCl(2) per 100 kg of feed mixture). Group C without inclusion of Ni or Zn served as control. After the 90-day experimental period biological material (liver, kidney, musculus longissimus dorsi, musculus biceps femoris) was taken from the animals. Samples were analyzed by the AAS method. Ni added to the FM caused an increase in Cd concentration in the kidneys of the rabbits, significantly in the E3 group in comparison with the control group. In the liver an insignificant decrease of Cd concentration was found. Zn addition in the amount of 30 g to the diet caused an increase of Cd level in the kidney as well as in the liver. Ni and Zn treatment caused a significant decrease of Pb accumulation in the m. longissimus dorsi of rabbits. This study indicates that dietary inclusion of Ni and Zn caused specific interactions among the observed metals.
