RESUMEN

To prepare hybridomas secreting monoclonal antibodies (MoAb) against human alpha-interferon (alpha-IFN), BALB/c mice were immunized with IFN produced in Namalwa cells. Native alpha-IFN, as well as partially purified or on cellulose adsorbed alpha-IFN preparations were used for immunization. Seven hybridomas continuously secreting IgG against human alpha-IFN were prepared by fusion of splenocytes from immunized donors with the mouse myeloma cells. MoAb reacted in ELISA as well as in neutralization test with human lymphoblastoid, leukocytic and recombinant alpha-IFN.

RESUMEN

The results of a comparative study of the sensitivity of several methods for mycoplasma detection in cell cultures are presented. The most sensitive method was found to be that of electron microscopy detecting mycoplasmal contamination in 100% preparations. Seeding of the material on mycoplasma-elective nutrient medium (0.3% PPLO agar) and the method of autoradiography detect the culture contamination in 90% of the test materials. Staining of cell cultures with orsein and Schiff reagent are less sensitive methods revealing mycoplasmal contamination in 69% and 77.7% of the cultures, respectively.

RESUMEN

Respiratory syncytial virus multiplying activity in the lungs of newborn and 6-day-old cotton rats, induced production of considerable amounts of interferon. Interferon production correlated with virus multiplication. Lower interferon titers were observed in the lungs of newborn cotton rats than in the lungs of 6-day-old animals. RS virus also induced production of serum interferon upon intraperitoneal inoculation, its titers being 2 times as low as after inoculation of equal doses of Newcastle disease virus. Poly I: poly C was shown to be a quite active interferon inducer in cotton rats and to inhibit RS virus reproduction in their lungs.

RESUMEN

The symplast-forming capacity of oncornavirus of continuous J-96 cells was studied by KS-test. Syncytia were formed upon combined cultivation of KS cells with J-96 cells and cells of human embryo skin-muscle tissue infected with J-96 virus as well as upon direct inoculation of KS cells with oncornavirus J-96. Formation of syncytia of KS cells correlated with the amount of J-96 cell oncornavirus. The possibility of using KS-test for titration of J-96 virus and antibody to it has been demonstrated.

RESUMEN

Changes in the interferon reaction of leukocytes, bactericidal property of the blood serum against the E. coli and streptococcus, the activity of beta-lysins in experimental nephrotoxic glomerulonephritis reproducible with the aid of heterogenous nephrotoxic serum in mongrel male rats were studied. This model permitted to trace the changes in the nonspecific protection mechanisms depending on the pathological process in the kidneys. In nephrotoxic glomerulonephritis the most pronounced changes were seen in the interferon reaction of leukocytes: it decreased up to 90% in comparison with the normal depending on the duration of the disease. In difference from this the changes in the anti-bacterial immunity indices were observed at the period of marked nephrotic process activity.
