This study examined sequence variability in internal transcribed spacers (ITS) of nuclear ribosomal DNA among Syphacia obvelata and Aspiculuris tetraptera isolates from laboratory mice from different geographical locations in China. ITS1, 5.8S and ITS2 rDNA were amplified separately from adult S. obvelata and A. tetraptera individuals by polymerase chain reaction (PCR), and the amplicons were subjected to sequencing from both directions. The lengths of the sequences of ITS1, 5.8S and ITS2 rDNA from both nematodes were 314 bp and 456 bp, 157 bp, and 273 bp and 419 bp, respectively. The intraspecific sequence variations in S. obvelata ITS1 were 0-0.3%. For A. tetraptera they were 0-0.7% in ITS1 and 0-1.0% in ITS2. However, the interspecific sequence differences among members of the infraorder Oxyuridomorpha were significantly higher, being 54.0-65.5% for ITS1 and 55.3-64.1% for ITS2. Phylogenetic analysis based on the combined partial sequences of ITS1 and ITS2 using three inference methods - Bayesian inference, maximum likelihood and maximum parsimony - revealed that all the S. obvelata and A. tetraptera samples formed independent monophyletic groups. Syphacia obvelata was closer to Syphacia muris than to A. tetraptera, consistent with morphological classification. These results demonstrate that ITS1 and ITS2 rDNA sequences are useful markers for population genetic studies of oxyurid nematodes.

OBJECTIVE: To conduct analysis and prenatal diagnosis on 11 couples carrying Thailand deletion (--(THΑI)) α-thalassemia 1, so as to provide information for clinical genetic counseling on α-thalassemia 1. METHODS: Altogether 11 Thailand deletion (--(THΑI)) α-thalassemia 1 families were collected from Fujian Maternal and Children Health Hospital from May 2009 to September 2015. Gap-polymerase chain reaction (gap-PCR) and reverse dot blot (RDB) technology were used to detect the thalassemia mutations in the couples and fetuses. RESULTS: In one family, Thailand deletion α-thalassemia 1 was detected in both the pregnant woman and her husband. In 10 families, Thailand deletion α-thalassemia 1 was detected in either the pregnant women or the husband, while the spouses had α-thalassemia heterozygote (1 combined with ß thalassemia heterozygote). Thailand deletion α-thalassemia 1 family members all had lower mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH). In prenatal diagnosis of the 12 fetuses, 4 fetuses were found with hemoglobin(Hb) Bart's hydrops fetalis syndrome, 5 were with α-thalassemia heterozygote, and 3 were normal. CONCLUSIONS: For couples with positive hematological phenotype but normal results in routine genetic examination of α-thalassemia, attention should be paid especially for with a history of having babies of hydrops fetalis syndrome or hemoglobin H disease. It is necessary to consider the possibility of the rare Thailand deletion (--(THΑI)) α-thalassemia 1. Prenatal diagnosis for high-risk families plays an important role.

A serodiagnostic ELISA (rL-ELISA) using recombinant truncated leukotoxin protein PL2 (aa 311-644) of Fusobacterium necrophorum as antigen was developed for detection of antibodies against F. necrophorum from cattle footrot. In rL-ELISA, the recombinant diagnostic antigen showed no cross-reaction with antisera against bovine foot and mouth disease virus, bovine rhinotracheitis virus, bovine viral diarrhea virus, bovine rotavirus type A, bovine Escherichia coli, and bovine Salmonella. The rL-ELISA could confirm the existence of antibodies against F. necrophorum at day 7 after infection. Detection of the field samples indicated relative sensitivity of rL-ELISA to nL-ELISA using the purified native leukotoxin A as antigen was 96.43%, and relative specificity of rL-ELISA to nL-ELISA was 94.26%. These data demonstrated the rL-ELISA would have a potential use for early diagnosis of cattle footrot caused by F. necrophorum.

A review of 18 cases of adenomatoid tumours in the female genital tract was carried out. There are 14 cases in uterus, 3 in fallopian tubes and 1 in ovary. The incidence of adenomatoid tumour in uterus, fallopian tubes and ovary is 3.1%, 0.5% and 0.2% respectively. The result showed that the uterus adenomatoid tumour is more common than fallopian tube and ovary (P < 0.01). All of the 18 cases are found in internal genital tract. The incidence of adenomatoid tumors is increased with the size of surfaces serosa or germinal epithelium in different part. Majority adenomatoid tumour are closing to the folding region of coelomic epithelium or midline of uterus. These findings suggest that adenomatoid tumour may be originated from mesothelia.

Radix Polygalae ("Yuan Zhi", the roots of Polygala tenuifolia Willd., YZ) is an important herb used in traditional Chinese medicine to mediate depression. The present study was designed to verify the antidepressant effects of the standardized YZ ethanol extract (YZE) and its four fractions YZ-30, YZ-50, YZ-70 and YZ-90 on the tail suspension (TST) and forced swimming test (FST). Furthermore, the standardization of the fractions obtained from the separation procedures was carried out by high-performance liquid chromatography (HPLC)-fingerprint. The YZ-50 fraction (Oligosaccharide esters--enriched, oral (200 mg/kg) showed a significant anti-immobility like effects. The data of YZ-50 on the corticosterone-induced injure of SH-SY5Y human neuroblastoma cell indicated that YZ-50 may have biological effects on neuroprotection. Proliferation of cell lines was assessed by dimethylthiazoldiphenyltetrazoliumbromide (MTT) and 5-bromo-2'-deoxyuridine (BrdU) incorporation assays. It was found that YZ-50 and its two bioactive compounds, 3,6'-di-o-sinapoyl-sucrose (DISS) and tenuifoliside A(TEA) showed protection activities in SY5Y cells from the lesion. By using bioassay-screening methods, our results indicate that the presence of oligosaccharide esters such as DISS and TEA in this herb may be responsible for the cytoprotective activity effects.

To analyze immunodominant regions of leukotoxin protein of Fusobacterium necrophorum strain H05, a series of truncated forms of leukotoxin gene were expressed in Escherichia coli using the vector pGEX-6p-1 or pPROEX HTa. The results of SDS-PAGE showed the truncated forms PL1, PL2, PL4, and PL5 were expressed in Escherichia coli using the vector pGEX-6p-1, and the truncated forms PL3 was expressed in Escherichia coli using the vector pPROEX HTa. These recombinant proteins were able to react with antisera against Fusobacterium necrophorum strain A25. In five recombinant proteins, the recombinant proteins PL1, PL3 and PL4 as vaccine were able to elicit formation of the better protective effects on mice against infection of Fusobacterium necrophorum strain A25.

A method for the quantitative determination of 11 organic acids (oxalic acid, citric acid, tartaric acid, malic acid, ascorbic acid, lactic acid, succinic acid, formic acid, acetic acid, glutaric acid and fumaric acid) in fruit juice was developed successfully. It was based on an ion exclusion chromatographic separation under the conditions of isocratic elution with 17 mmol/L sulphuric acid solution with the ICE-ION-300 ion exclusion column, and the UV detection at 210 nm. The precision of the method was investigated and the relative standard deviations were from 1.5% to 9.8% (n = 10).
