RESUMEN

Anisotropic microstructures are utilized in various fields owing to their unique properties, such as reversible shape transitions or on-demand and sequential release of drug combinations. In this study, anisotropic multicompartmental microfibers composed of different polymers are prepared via charge reversal electrohydrodynamic (EHD) co-jetting. The combination of various polymers, such as thermoplastic polyurethane, poly(D,L-lactide-co-glycolide), poly(vinyl cinnamate), and poly(methyl methacrylate), results in microfibers with distinct compositional boundaries. Charge reversal during EHD co-jetting enables facile fabrication of multicompartmental microfibers with the desired composition and tunable inner architecture, broadening their spectrum of potential applications, such as functional microfibers and cell scaffolds with multiple physical and chemical properties.

RESUMEN

This study explored the impact of re-acetylation on induced electric field (IEF)-assisted hydrolysis of chitosan. Chitosan with different degree of deacetylation (DD) exhibited different charge content but similar electrical conductivity and output voltage of the electrolyte system, thus theoretically resulting in equivalent electro-processing efficiency. However, molecular and rheological measurements suggested that the hydrolysis rate decreased with an increase of DD. Re-acetylation had no significant effect on the molecular weight and viscosity of chitosan but damaged its granular and crystal structure, making chitosan chains more susceptible to the acid. The IEF-assisted hydrolysis of dissolved chitosan confirmed that the structural compactness of chitosan played a more important role on the hydrolysis than the charge content. This study extended current knowledge regarding the effect of charge content and structural compactness on IEF-assisted hydrolysis process, which contributed to the applications of IEF in biopolymer modification.

RESUMEN

A water-soluble polysaccharide (LLPS) from tiger lily was extracted by ultrasonic wave-assisted extraction. The LLPS, which was isolated by alcohol precipitation, was further purified by DEAE Sepharose Fast Flow and Sephadex G-100 chromatography, which resulted in LLPS fractions in LLPS-1, LLPS-2 and LLPS-3, with molecular weights of 350.5, 403.3 and 146.2kDa, respectively. LLPS-1 and LLPS-2 primarily consisted of glucose and mannose in a molar ratio of nearly 1:2 and 1:1, respectively. In contrast, LLPS-3 was primarily composed of arabinose, galactose, glucose and mannose in a molar ratio of nearly 2:2:2:1. LLPS fractions could stimulate the proliferation of macrophages. The in vitro immunomodulatory activity of the fractions was evaluated. The results showed that treatment with 25-400 µg/mL of LLPS fractions could increase phagocytic activity and nitric oxide production of macrophages in a dose-dependent manner.

A total of nine lipases and three proteases were tested for enzymatic regioselective acylation(s) of cordycepin with vinyl acetate in organic media. The highest conversion with better initial reaction rate was achieved with immobilized Candida antarctica lipase B (Novozym 435). An eco-friendly solvent 2-methyltetrahydrofuran (MeTHF) was thought to be the most suitable reaction medium. Novozym 435 was found to be a useful biocatalyst for the 25-g scale syntheses of cordycepin acetate (96.2% isolated yield), and the biocatalyst displayed excellent regioselectivity and high operational stability during the transformation. The 5'-substituted cordycepin derivative was the sole detectable product from each acylation reaction. Novozym 435 could be recycled for the synthesis of cordycepin derivative on a 25-g scale and 63% of its original activity was maintained after being reused for 7 batches. MeTHF could be considered as an eco-friendly solvent for the large scale use in biotransformation.

The submerged fermentation of Cordyceps militaris for cordycepin production and mycelial growth was investigated in this study. Three natural materials of brown rice paste (BRP), beerwort (B), and soybean meal juice (SMJ) were used for fermentation of C. militaris in shaking flasks. The effects of the ratio of three natural materials on dry mycelium weight (DMW) and on cordycepin yield (CY) were analyzed. D-Optional mixture design was used to optimize the ratio of these materials. Compared with the signal culture, the higher mycelial growth and cordycepin production were obtained in mixture. The analysis of Design Expert 6.0 indicated that BRP, B, and SMJ very significantly influenced (P < 0.001) DMW and CY of C. militaris, respectively. The highest DMW (18.96 g/l) and CY (2.17 mg/g) were both obtained at a ratio of 53:6:42. The experiments' results indicated that the above mixture of these natural materials by D-optional mixture design can be used as a proper medium for the growth of mycelium and the production of cordycepin.

A comparative study was made of enzymatic acylation of konjac glucomannan with vinyl esters under ultrasonic irradiation and shaking in organic solvent tert-butanol. Among the 13 enzymes selected, Novozym 435 exhibited the highest acylation activity towards KGM whether under ultrasonic irradiation or shaking. The application of ultrasonic irradiation instead of shaking during the acylation led to improvement in the initial reaction rate, yield and degree of substitution of the modified KGM. Appropriate ultrasound power (100 W) and water activity (0.75) were found to accelerate enzymatic reaction. The acceleration effect of ultrasound on Novozym 435-catalyzed acylation decreased with an increase in the chain length of the acyl donors from C2 to C18. Moreover, the acylation of KGM in tert-butanol was proved to be a regioselective one, with C6-OH being acylated. Compared with shaking, ultrasound did not change regioselectivity of Novozym 435 in the acylation.
