RESUMEN

Medical attention was sought for a 23-month-old toddler because of anorexia, weight loss, irritability, profuse sweating, peeling and redness of his fingers and toes, and a miliarial rash. The diagnosis was mercury poisoning, and an investigation of his environment disclosed that he had been exposed to mercury from broken fluorescent light bulbs. Acrodynia resulting from fluorescent bulbs has not been previously reported.

OBJECTIVE: To assess whether increased collagenolysis precedes severe chronic lung disease (CLD). METHODS: Matrix metalloproteinase-1 (MMP-1) and MMP-8 (enzymes that degrade type I collagen, the main structural protein of lung extracellular matrix) were measured by enzyme linked immunosorbent assay in 100 bronchoalveolar lavage samples taken during the first 6 postnatal days from 45 ventilated preterm babies < 33 weeks gestation. The median value for each baby was calculated. CLD was defined as an oxygen requirement after the 36th week after conception. RESULTS: MMP-8 levels in bronchoalveolar lavage fluid were higher (median 13 ng/ml) in 20 babies who developed CLD than in 25 without CLD (median 2 ng/ml). No MMP-1 was detected in any sample. CONCLUSIONS: MMP-8 can be detected in bronchoalveolar lavage fluid from preterm babies, and higher levels are found in those who later develop CLD. MMP-8 may contribute to lung injury that occurs as a prelude to CLD.

The dithiothreitol test was evaluated as a substitute for the 2-mercaptoethanol test for determining Brucella immunoglobulin G agglutinating antibody in bovine serum. The tests were compared on 207 card-positive sera that showed a standard tube-agglutination titer of incomplete 1:50 or higher. The tests agreed within one dilution with 182 of the 207 sera tested for an 87.9% rate of agreement. When titers were not the same, those obtained with the dithiothreitol test were more frequently lower than higher than those obtained with the 2-mercaptoethanol test. Sixteen sera that showed a titer with the 2-mercaptoethanol test were negative with the dithiothreitol test and two that showed a titer with the dithiothreitol test were negative with the 2-mercaptoethanol test. Results suggest that the dithiothreitol test is not a reliable substitute for the 2-mercaptoethanol test to detect immunoglobulin G agglutinating antibody in bovine serum.

RESUMEN

An agar-gel immunodiffusion test was compared with other serological tests for detection of antibodies in the sera of 48 cattle vaccinated with Brucella abortus strain 19. Two hundred forty-two sera were tested over a 12 month period, and addition of positive reactions for each test resulted in totals of 170 for the card test, 74 for the standard tube-agglutination test, 71 for the 2-mercaptoethanol test, 64 for the dithiothreitol test and 22 for the agar-gel immunodiffusion test.

The noise exposures of 274 printing production workers in 34 establishments in the New York city area were monitored. Results showed that 43% were exposed to 8-hr time-weighted average (TWA) noise exposures of 85 dBA or greater and that 14% were exposed to 8-hr TWAs of 90 dBA or greater. Within the press department, web press workers were exposed to significantly greater mean 8-hr TWAs than sheetfed press workers. In general, a greater percentage of the workers in the bindery departments were exposed to potentially harmful noise than workers in the press departments. Results of this study indicate that many workers in the printing industry may be at risk of occupational hearing loss. Further research is needed to determine the extent of hearing impairment in this group of workers.

Lyophilized cells of Bacillus popilliae were protected from moisture when suspended in pellets of tung oil polymer which were then coated with paraffin wax. The survival of the protected cells at various levels of relative humidity (RH) and under various storage conditions was determined. During 6 months of storage, moisture appeared to have little effect on survival of the cells when the RH level was 22% or less; but, at higher RH levels, survival declined upon storage. Viable cells were recovered when pellets were stored for 3 months at 33% RH, 2 months at 42% RH, 1 month at 50% RH, and 4 days in distilled water. Under field conditions, some cells survived at least 1 week of storage.

RESUMEN

Survival of lyophilized vegetative Bacillus popilliae in dry soil and in soil kept in atmospheres with different relative humidities (RH) was determined. Cells survived for at least 1 year when the RH was 22% or lower, but an RH of 33% or higher reduced viability. At 33% RH, no bacteria were recovered after 5 months of storage; at 42% or above, no bacteria were recovered after 1 month of storage.

RESUMEN

Counterimmunoelectrophoresis and enzyme-linked immunosorbent assay specific for rotavirus were performed on 253 calf gut and fecal specimens from eastern North Dakota. Rotavirus antigen was detected in 29 specimens by counterimmunoelectrophoresis and in 52 specimens by enzyme-linked immunosorbent assay. All of the counterimmunoelectrophoresis-positive specimens were positive by enzyme-linked immunosorbent assay. The counterimmunoelectrophoresis and the enzyme-linked immunosorbent assay, using a rabbit antirotavirus antibodies produced a golf ball-induced granuloma, were rapid and specific tests for the detection of rotavirus.

The per cent viability of Bacillus popilliae after lyophilization of liquid nitrogen frozen cells was determined. Lyophilization of 9- to 12-hr cells which had been suspended in 5% sodium glutamate plus 0.5% gum tragacanth, frozen in liquid nitrogen vapor, and dried 4 to 5 hr with the ampoules exposed to room temperature resulted in survival of 64.6% of the original cells. After storage of these lyophilized preparations for 6 months at room temperature, 10.5% of the original cells were still viable.

A comparison was made of results obtained with a Brucella agar--gel immunodiffusion (AGID) test and the standard tube-agglutination test on 612 human sera. Agreement between the tests was 97% when the titer was 1:160 or higher. Of 448 sera that showed no agglutination titer, 447 were negative with the AGID test. Results of the AGID test were also compared to those obtained with the 2-mercaptoethanol (2-ME) agglutination test on 148 sera that demonstrated a standard tube-agglutination titer of 1:20 or higher. All sera with a 2-ME-agglutination titer of 1:40 or higher were positive with the AGID test. Of 123 sera that showed no 2-ME-agglutination titer, 21 were positive with the AGID test. Two of these 21 sera were obtained from patients with bacteriologically proven brucellosis, and eight were from abattoir employees with suspected but not bacteriologically proven brucellosis.

BACKGROUND: Matrix degradation in emphysema has long been attributed to the action of neutrophil elastase (NE). More recently a role for other proteases, particularly the matrix metalloproteinases (MMPs), in the pathogenesis of this disease has been proposed. To date, however, the presence of MMPs in the lungs of patients with emphysema has not been demonstrated. METHODS: Samples of bronchoalveolar lavage (BAL) fluid from 10 patients with emphysema and from control subjects matched for sex and current smoking status were assessed for collagenase, gelatinase, and NE activity. Pulmonary function tests and computed tomographic (CT) scans were carried out on all study subjects. RESULTS: Collagenase activity was detected in BAL fluid samples from all emphysematous patients but in only one smoking control (p < 0.001). Gelatinase B was present in six patients and in two smoking controls (p < 0.03). The concomitant presence of gelatinase B in complex with lipocalin (NGAL) in the gelatinase positive samples suggests that the neutrophil is a significant source of the gelatinase B observed. NE was detected in six of the 10 patients with emphysema and in two smoking controls (p < 0.01), indicating that collagenase was more useful in discriminating between disease and control groups than either NE or gelatinase B. No relationship was observed between any of the enzymes measured and pulmonary function or CT density score. CONCLUSIONS: This study demonstrates, for the first time, the presence of increased levels of matrix metalloproteinases in the lungs of patients with emphysema and suggests that, in BAL fluid, collagenase activity may be a better indicator of the presence of emphysema than elastase.
