RESUMEN

Strategies to alleviate heat stress on live performance - dry matter intake (DMI), average daily gain (ADG), and feed conversion efficiency (FCE) - and on physiological parameters - respiratory rate (RR) and panting score (PS) - of feedlot-finished cattle were evaluated by systematic review-meta-analysis. Using the PICO framework were identified 3813 records, with 31 publications and 6729 cattle being considered in the meta-analysis (MA). The comparison most reported was sun vs. shade (n = 24 publications), followed by shade vs. shade (area per animal and effectivity in reduce solar load) (n = 7 publications) and sun vs. sprinkling (n = 4 publications). Cattle with access to shade with total reduction in solar load improved ADG (p = 0.008) and FCE (p = 0.024), and reduced RR (p < 0.001) compared to unshaded animals. Cattle with access to shade with a height between 3 and 4 m increased ADG (p < 0.001), tended to improve FCE (p = 0.054), and reduced RR (p < 0.001) compared to unshaded animals. An area of shade from 2 to 4 m2/animal increased ADG (p = 0.002), and higher than 4 m2/animal reduced RR (p < 0.001) compared to unshaded animals. Animals that received sprinkling volume below 1 L/animal/min improved ADG (p = 0.008) compared to unshaded animals. Cattle with access to shade with a high area per animal improved DMI (p = 0.023) and ADG (p < 0.001) compared to animals with a low area. In univariate meta-regression, it was observed that the variables significantly (P < 0.05) influencing DMI were THI category; influencing ADG were cattle gender, THI category, and coat color; influencing FCE were cattle age and hemisphere; and influencing RR were country, THI category and climate. Performance and physiological parameters of feedlot-finished cattle under heat stress depend on animal and environmental characteristics.

RESUMEN

The impact of heat stress on dairy cattle leads to significant economic losses and a negative impact on the welfare of the animals. The objective of this research was to evaluate the effect of the nutritional additive (Thermoplus®) in dairy cows under postpartum heat stress conditions, and its effects on the metabolic profile, production and quality of milk. Eighteen lactating Holstein cows (8 multiparous and ten primiparous), in a free-stall system, with a mean body condition score (BCS) of 3.14 ± 0.05, live weight of 624.55 ± 18, 61 kg, with initial mean days in milk (DIM) of 90 ± 10.11, were selected. The animals were grouped into a control (CG, n = 9) and a treatment (TG, n = 9). Both groups underwent 14 days of diet adaptation, the TG received the basal diet supplemented with 50 g of the additive, once a day, individually, while the control group received only the total diet. Data collection of metabolic and productive parameters were evaluated on days -14 (before adaptation), 1 (after the diet adaptation period), 16, 30, and 44. Milk, blood, and body condition score (BCS) were collected once a day, and heart rate, respiratory rate, and rectal temperature were collected twice a day. Serum concentrations of albumin, calcium, magnesium, glucose, gamma-glutamyl transferase (GGT), beta-hydroxybutyrate (BHBA), non-esterified fatty acids (NEFAs), and paraoxonase-1 (PON-1) were evaluated. In the milk, the percentage of fat, protein, lactose, and total solids were determined in each sampling. Milk yield was measured daily. Humidity and ambient temperature values were collected on the days of the collection every 30 min, from 5:30 am to 5:00 pm, to calculate the temperature-humidity index (THI). Statistical analyzes were performed using the SAS software (version 9.3, SAS Institute Inc., Cary, NC, USA). The THI ranged from 62.22 to 79.47. Our findings showed that when the THI was greater than 72, the animals in the TG were able to maintain milk yield (Odds ratio (OD) = -0.0577,), and the animals in the CG had a greater chance of reducing it (OD = -0.2301). Multiparous cows in the TG had higher milk yield than CG (32.57 ± 0.34 vs 30.50 ± 0.36 kg per day; P = 0.0078) and lower SCC (34.110 ± 6,940 vs 665.50 ± 214.41 cells per ml; P = 0.03), with the same percentages of total solids (P > 0.05). In multiparous metabolic markers, TG when compared CG had higher albumin concentrations (2.50 ± 0.07 vs 2.12 ± 0.07 g/dl; < 0.001), equal PON-1 (P > 0.05), and higher BHBA levels (0.49 ± 0.03 vs 0.39 ± 0.04 mmol/l). Primiparous from the CG had higher concentrations of NEFA (0.18 ± 0.02 mmol/l) than multiparous from the same group (0.09 ± 0.02 mmol/l) P = 0.0265. The use of the plant polyphenol extract in postpartum Holstein cows challenged by heat stress had beneficial effects on the production and health of the mammary gland in multiparous cows without decreasing milk solids. The non-reduction of the activities of the acute phase proteins indicates an immunomodulatory and inflammatory-reducing effect of the product used.

RESUMEN

The intensification of ewe production, focusing on increasing prolificity, results in a higher incidence of metabolic disorders, especially in the transition period, which can lead to production losses with consequences for the health of the animals. The objective of this study was to evaluate the metabolic profile in transition period Lacaune ewes according to the parity and litter size born and its influence on the transference of passive immunity. Nineteen Lacaune ewes, with initial mean body weight of 76.36 ± 10.37 kg (SD), kept in a semi-extensive system and receiving the same diet, were used. Ewes were classified according to parity (primiparous and multiparous) and the litter size born at parturition (1, 2, and 3). On days - 14, - 7, and - 4 of the expected date of parturition and on days 1, 4, 7, and 14 days postpartum, blood sampling, the evaluation of the body condition score (BCS), and the weighing of animals were performed. Blood samples were taken from the lambs 48 h after birth. Colostrum was collected up to 6 h postpartum to determine the brix percentage. Serum concentrations of total plasma proteins (TPP), albumin, aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), beta-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA), paraoxonase (PON1), calcium (Ca), and magnesium (Mg) were evaluated. In lambs, the blood parameters evaluated were TPP, albumin, GGT, and PON1. Lamb plasma was used to estimate colostrum efficiency, brix percentage immunity, and total plasma protein (PPT). Productive data, pre- and postpartum metabolic variables for ewes, lambs, and those related to colostrum brix were analyzed. The BCS had a reduction (p < 0.0001) in the postpartum period, which is evidenced on days 7 and 14 (2.88 and 2.73, respectively). Serum concentration of BHB presented the highest concentrations in the prepartum period for ewes that gave birth to triplets, from day - 14, and for ewes that gave birth to twins on day - 4 (p = 0.0245). Serum concentration of NEFA demonstrated the highest value on day 14 in primiparous (1.07; p = 0.0402). Calcium had the lowest concentrations on day 1 (8.85 mg/dl, p < 0.0001) and magnesium on day 3 (1.79 mg/dl, p < 0.0001) postpartum. The multiparous with two lambs had higher brix percentages (30.5%), and the lowest value was observed in primiparous ewes that gave birth to a single lamb (27.06%; p = 0.0395). Triplet lambs had the lowest weight (3.73 kg, p = 0.0007), and the best brix percentage in plasma was observed in twins (10.29%, p = 0.0174). Regardless of the parity, the ewes that presented the greatest metabolic challenge were those that gave birth to triplets, and these lambs presented the worst immunity and the lowest live weight. The quality of colostrum was influenced by the parity, and multiparous ewes that gave birth to triplets had the lowest brix percentage in the colostrum.

RESUMEN

KEY MESSAGE: Efficiency of light interception, Radiation use efficiency and harvest index can be used as targets to improve grain yield potential in soybean. Grain yield (GY) production can be expressed as the result of three main efficiencies: light interception (Ei), radiation use (RUE), and harvest index (HI). Although dissecting GY through these three efficiencies is not entirely new, there is a lack of knowledge about the phenotypic variation, the genetic architecture, and the relative contribution of these three efficiencies on GY in soybean. This knowledge gap coupled with laborious phenotyping prevents the active consideration of these efficiencies into breeding programs. This study aims to reveal the phenotypic variation, heritability, genetic relationships, genetic architecture, and genomic prediction for Ei, RUE, and HI in soybean. We evaluated a maturity control panel of 383 Recombinant Inbred Lines (RILs) selected from the soybean nested association mapping (SoyNAM) population. Dry matter ground measured along with canopy coverage (CC) from UAS imagery were collected in three environments. Light interception was modeled through a logistic curve using CC as a proxy. The total above-ground biomass collected during the growing season and its respective cumulative light intercepted were used to derive RUE through linear models fitting. Additive-genetic correlations, genome-wide association (GWA) and whole-genome regressions (WGR) were performed to evaluate the relationship between traits, their association with genomic regions, and the feasibility of predicting these efficiencies with genomic information. Correlation analyses considered three groups: the entire data set, and the high- and low-yielding RILs to determine association as a function of the GY. Our results revealed moderate to high phenotypic variation for Ei, RUE, and HI with ranges of 8.5%, 1.1 g MJ-1, and 0.2, respectively. Additive-genetic correlation revealed a strong relationship of GY with HI and moderate with RUE and Ei when whole data set was considered, but negligible contribution of HI on GY when just the top 100 was analyzed. The GWA analyses showed that Ei is associated with three SNPs; two of them located on chromosome 7 and one on chromosome 11 with no previous quantitative trait loci (QTLs) reported for these regions. RUE is associated with four SNPs on chromosomes 1, 7, 11, and 18. Some of these QTLs are novel, while others are previously documented for plant architecture and chlorophyll content. Two SNPs positioned on chromosome 13 and 15 with previous QTLs reported for plant height and seed set, weight and abortion were associated with HI. WGR showed high predictive ability for Ei, RUE, and HI with maximum correlation ranging between 0.75 and 0.80. Future improvements in GY can be expected through strategies prioritizing Ei for short-term results when using high yielding germplasm and RUE for medium- and long-term outcomes. This work is a pioneer attempt to integrate traditional physiological traits into the breeding process in the context of physiological breeding.

RESUMEN

The environmental impact of microplastics is a challenging theme, especially under realistic experimental conditions. We investigated physiological responses to 0.1-1.0 µm PVC particles intake by the mussel Perna perna after a relative long-term exposure (90 days) at a less extreme concentration compared with previous studies (0.125 g/L). Microplastic intake was inferred by the presence of PVC in the feces of mussels, and physiological damages were assessed through ingestion rate, assimilation efficiency, growth rate, cellular and molecular biomarkers (lysosomal integrity, lipid peroxidation, and DNA damage), and condition index. All physiological responses showed nonsignificant effects of the microplastics on the exposed mussels. We suggest that, despite the experimental concentration of microplastics, mussels were able to acclimate to the exposure through their abilities for long-term recovery and tolerance to stresses. These data have positive implications for environmental health and in terms of human food resource because mussel farming is a worldwide practice that heavily relies on plastic materials, increasing the chances of microplastic exposure and mussels contamination.

RESUMEN

The effect of different percentages of Duroc genes in crossbreeding (5, 50 and 100%), the gender (immunocastrated males - ICM and females) and their interaction was evaluated on growth, carcass and meat quality traits for pigs. Crossbreds (50% Duroc) had greater average daily feed intake, hot carcass weight and backfat thickness but lower meat yield when compared to 5% Duroc crossbreds and purebred (100% Duroc) animals (P < 0.05). Purebred (100% Duroc) animals had the lowest backfat thickness and subcutaneous fat thickness, and the greatest muscle depth and meat yield (P < 0.05). ICM animals had better feed conversion, greater average daily gain, hot carcass weight and amount of meat, and lower hot carcass yield, cold carcass weight, Longissimus thoracis depth, rib with belly and ham weights compared to females (P < 0.05). Marbling scores were greater in purebred (100%) animals (P < 0.05). The meat from 50% Duroc crossbreds and 100% Duroc purebred pigs was more reddish pink in color than 5% Duroc crossbreds (P < 0.05). Also, marbling scores were greater for females compared to ICM (P < 0.05). Purebred (100% Duroc) animals required more medications during production (P < 0.05).

RESUMEN

Antimicrobial substitutes are being used in pig production systems, to maintain the health of the animals without compromising their performance. The aim of this study is to evaluate the impact of either the removal of in feed antimicrobials or their substitution for feed additives, at the nursery and growing/finishing stages. At weaning, 1091 piglets were sexed, vaccinated, homogenized by weight and allocated to six treatments during the nursery stage (26-63 d): T1- feed with no antimicrobials nor additives; T2 - feed with antimicrobials; T3 - feed with prebiotic; T4 - feed with probiotic; T5 - feed with essential oils; T6 - feed with organic acids. The same treatments were applied to 840 pigs during the growing/finishing stages (64-167 d). There was no effect of the treatments on feed conversion at the nursery (P = 0.222) and the growing/finishing (P = 0.809) stages. The average daily gain did not differ across treatments in the nursery (P = 0.342) and in growing/finishing (P = 0.050). The cost of the interventions with injectable drugs was not different between the treatments neither at the nursery (P = 0.990) nor at the growing/finishing (P = 0.310). However, the pneumonia and pleurisy index for all treatments was equal or above 1.0, which indicates a respiratory challenge. There was an increase in the cost with antimicrobials or additives per kg of feed produced, which impacts the cost per kg of pig produced. In conclusion, the removal of antimicrobials in pig diets is financially feasible and their substitution by additives did not impact growing performance.

RESUMEN

Soybean grain yield has steadily increased during the last century because of enhanced cultivars and better agronomic practices. Increases in the total biomass, shorter cultivars, late maturity, and extended seed-filling period are frequently reported as main contributors for better soybean performance. However, there are still processes associated with crop physiology to be improved. From the theoretical standpoint, yield is the product of efficiency of light interception (Ei), radiation use efficiency (RUE), and harvest index (HI). The relative contribution of these three parameters on the final grain yield (GY), their interrelation with other phenological-physiological traits, and their environmental stability have not been well established for soybean. In this study, we determined the additive-genetic relationship among 14 physiological and phenological traits including photosynthesis (A) and intrinsic water use efficiency (iWUE) in a panel of 383 soybean recombinant inbred lines (RILs) through direct (path analyses) and indirect learning methods [least absolute shrinkage and selection operator (LASSO) algorithm]. We evaluated the stability of Ei, RUE, and HI through the slope from the Finley and Wilkinson joint regression and the genetic correlation between traits evaluated in different environments. Results indicate that both supervised and unsupervised methods effectively establish the main relationships underlying changes in Ei, RUE, HI, and GY. Variations in the average growth rate of canopy coverage for the first 40 days after planting (AGR40) explain most of the changes in Ei. RUE is primarily influenced by phenological traits of reproductive length (RL) and seed-filling (SFL) as well as iWUE, light extinction coefficient (K), and A. HI showed a strong relationship with A, AGR40, SFL, and RL. According to the path analysis, an increase in one standard unit of HI promotes changes in 0.5 standard units of GY, while changes in the same standard unit of RUE and Ei produce increases on GY of 0.20 and 0.19 standard units, respectively. RUE, Ei, and HI exhibited better environmental stability than GY, although changes associated with year and location showed a moderate effect in Ei and RUE, respectively. This study brings insight into a group of traits involving A, iWUE, and RL to be prioritized during the breeding process for high-yielding cultivars.

RESUMEN

Understanding temporal accumulation of soybean above-ground biomass (AGB) has the potential to contribute to yield gains and the development of stress-resilient cultivars. Our main objectives were to develop a high-throughput phenotyping method to predict soybean AGB over time and to reveal its temporal quantitative genomic properties. A subset of the SoyNAM population (n = 383) was grown in multi-environment trials and destructive AGB measurements were collected along with multispectral and RGB imaging from 27 to 83 days after planting (DAP). We used machine-learning methods for phenotypic prediction of AGB, genomic prediction of breeding values, and genome-wide association studies (GWAS) based on random regression models (RRM). RRM enable the study of changes in genetic variability over time and further allow selection of individuals when aiming to alter the general response shapes over time. AGB phenotypic predictions were high (R 2 = 0.92-0.94). Narrow-sense heritabilities estimated over time ranged from low to moderate (from 0.02 at 44 DAP to 0.28 at 33 DAP). AGB from adjacent DAP had highest genetic correlations compared to those DAP further apart. We observed high accuracies and low biases of prediction indicating that genomic breeding values for AGB can be predicted over specific time intervals. Genomic regions associated with AGB varied with time, and no genetic markers were significant in all time points evaluated. Thus, RRM seem a powerful tool for modeling the temporal genetic architecture of soybean AGB and can provide useful information for crop improvement. This study provides a basis for future studies to combine phenotyping and genomic analyses to understand the genetic architecture of complex longitudinal traits in plants.

RESUMEN

Wildfire effects go beyond direct impact in terrestrial ecosystems. Specifically, the periphytic communities of aquatic ecosystems standing within and downstream the burnt areas are relevant ecological receptors of post-fire runoff contamination. Nevertheless, the off-site impacts of wildfires in these communities are limitedly studied so far. The present study aimed to assess the effects of river water contaminated with ash-loaded runoff in the growth benthic diatom Navicula libonensis (Schoeman 1970). Four surface water samples were collected approximately one year after the wildfire for laboratory testing with the diatom: one was collected from a site upstream the burnt area, within the Unhais river (UU); three were collected from sites standing within the burnt area, one in the Unhais river (UB) and two in the Zêzere river (Z1 and Z2), reflecting different hydrological regimes. N. libonensis was proven able to discriminate among river sites affected and unaffected by wildfire runoff, reflecting, in general, the expected trends considering the physico-chemical characterization of the water samples. The water samples from the sites standing within the burnt area inhibited the biomass yield and growth rate of the tested diatom, ranking the samples regarding toxicity as follows: Z1 > UB > Z2 > UU. However, UB rather than Z1 presented the highest contaminant burden, namely metal elements, and some were found above widely accepted safety benchmarks (polycyclic aromatic hydrocarbons were not detected). This inconsistency can be linked to unknown interactions among metals within each water sample, to differential nutrient enrichment of samples, as well as hydrological factors. Overall, our results suggest that monospecific laboratory assays with sensitive diatoms can be valuable as cost-effective screening tools to prioritize sites affected by wildfires runoff requiring in-depth monitoring of negative effects in benthic producer communities.

RESUMEN

Although it is well documented that leptin signals the body nutritional status to the brain, mechanisms of leptin regulation at the ovary are not well understood. This study was conducted to determine whether there was leptin and the receptor for leptin (LEPR) in cattle ovarian follicles and to investigate potential actions of leptin on follicular growth in vivo and on regulation of granulosa cell functions in vitro. There was leptin and LEPR in granulosa and theca cells of dominant and subordinate follicles, with greater immunostaining for leptin in granulosa cells of subordinate follicles. There was a lesser relative abundance of leptin receptor gene-related protein (LEPROT) and of the adiponectin receptors 1 (ADIPOR1) and 2 (ADIPOR2) mRNA transcripts in granulosa cells of subordinate than dominant follicles (P < 0.05). Intrafollicular injection of either 100 or 1000 ng/mL leptin did not affect the diameter and the growth of dominant follicles (P> 0.05). Supplementation of in vitro culture medium with different leptin concentations did not affect (P > 0.05) the relative abundance of hydroxy-delta-5-steroid dehydrogenase, 3 beta- and steroid delta-isomerase 1 (HSD3B1), cytochrome P450 family 11 subfamily A member 1 (CYP11A1), signal transducer and activator of transcription 3 (STAT3) and X-linked inhibitor of apoptosis protein (XIAP) mRNA transcripts in granulosa cells. These findings indicate that leptin and LEPR are present in the follicular cells of cattle ovaries, but leptin apparently does not have essential functions in steroidogenesis and growth of dominant follicles.

RESUMEN

The rapid development of remote sensing in agronomic research allows the dynamic nature of longitudinal traits to be adequately described, which may enhance the genetic improvement of crop efficiency. For traits such as light interception, biomass accumulation, and responses to stressors, the data generated by the various high-throughput phenotyping (HTP) methods requires adequate statistical techniques to evaluate phenotypic records throughout time. As a consequence, information about plant functioning and activation of genes, as well as the interaction of gene networks at different stages of plant development and in response to environmental stimulus can be exploited. In this review, we outline the current analytical approaches in quantitative genetics that are applied to longitudinal traits in crops throughout development, describe the advantages and pitfalls of each approach, and indicate future research directions and opportunities.

RESUMEN

Enterohaemorrhagic Escherichia coli (EHEC) and enteropathogenic E. coli (EPEC) translocate dozens of type III secretion system effectors, including the WxxxE effectors Map, EspM and EspT that activate Rho GTPases. While map, which is carried on the LEE pathogenicity island, is absolutely conserved among EPEC and EHEC strains, the prevalence of espM and espT is not known. Here we report the results of a large screen aimed at determining the prevalence of espM and espT among clinical EPEC and EHEC isolates. The results suggest that espM, detected in 51 % of the tested strains, is more commonly found in EPEC and EHEC serogroups that are linked to severe human infections. In contrast, espT was absent from all the EHEC isolates and was found in only 1.8 % of the tested EPEC strains. Further characterization of the virulence gene repertoire of the espT-positive strains led to the identification of a new zeta2 intimin variant. All the espT-positive strains but two contained the tccP gene. espT was first found in Citrobacter rodentium and later in silico in EPEC E110019, which is of particular interest as this strain was responsible for a particularly severe diarrhoeal outbreak in Finland in 1987 that affected 650 individuals in a school complex and an additional 137 associated household members. Comparing the protein sequences of EspT to that of E110019 showed a high level of conservation, with only three strains encoding EspT that differed in 6 amino acids. At present, it is not clear why espT is so rare, and what impact EspM and EspT have on EPEC and EHEC infection.

RESUMEN

BACKGROUND: In the early stages of plant breeding programs high-quality phenotypes are still a constraint to improve genetic gain. New field-based high-throughput phenotyping (HTP) platforms have the capacity to rapidly assess thousands of plots in a field with high spatial and temporal resolution, with the potential to measure secondary traits correlated to yield throughout the growing season. These secondary traits may be key to select more time and most efficiently soybean lines with high yield potential. Soybean average canopy coverage (ACC), measured by unmanned aerial systems (UAS), is highly heritable, with a high genetic correlation with yield. The objective of this study was to compare the direct selection for yield with indirect selection using ACC and using ACC as a covariate in the yield prediction model (Yield|ACC) in early stages of soybean breeding. In 2015 and 2016 we grew progeny rows (PR) and collected yield and days to maturity (R8) in a typical way and canopy coverage using a UAS carrying an RGB camera. The best soybean lines were then selected with three parameters, Yield, ACC and Yield|ACC, and advanced to preliminary yield trials (PYT). RESULTS: We found that for the PYT in 2016, after adjusting yield for R8, there was no significant difference among the mean performances of the lines selected based on ACC and Yield. In the PYT in 2017 we found that the highest yield mean was from the lines directly selected for yield, but it may be due to environmental constraints in the canopy growth. Our results indicated that PR selection using Yield|ACC selected the most top-ranking lines in advanced yield trials. CONCLUSIONS: Our findings emphasize the value of aerial HTP platforms for early stages of plant breeding. Though ACC selection did not result in the best performance lines in the second year of selections, our results indicate that ACC has a role in the effective selection of high-yielding soybean lines.

RESUMEN

Microplastics are important vectors for the transport and accumulation of persistent organic contaminants in coastal and marine environments. We determined the concentration of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) associated with microplastic pellets collected along a 39-km stretch of Brazil's South Atlantic coastline to understand the spatial dynamics and potential risk posed by these contaminants. Total PAH concentrations ranged from 1,454 to 6,002 ng g-1 and regularly exceeded the threshold effect level (TEL) for sediments defined by the United States Environmental Protection Agency. Sampling stations, spaced evenly along the coastline (i.e., 3-km intervals) exhibited a general north-to-south decline in the concentrations of some PAHs, but this spatial gradient was complicated by small-scale differences in the concentrations and composition of associated contaminants. Similarly, analysis of individual isomer ratios revealed further complexity driven by differences in the contribution of petrogenic versus pyrolytic inputs which pose different levels of risk to marine organisms. PCB concentrations ranged from 0.8 to 104.6 ng g-1 and were dominated by low chlorinated congeners likely to have originated from major industrial areas to the north. Overall, this study highlights the challenge of directly linking microplastic pollution with the potential toxicological effects of organic contaminants in coastal waters. We recommend that monitoring programs should explicitly consider both the origin (i.e., pellet sources and dispersal pathways) and nature of organic contamination (i.e., concentration and composition) when assessing the risks for biota and human health.

RESUMEN

Omega-3 PUFA may benefit sow reproductive performance, but effects on weaned gilts are unknown. This study evaluated the effects of supplementing omega-3 PUFA to gilts after weaning on growth, metabolic markers, and gene expression of steroidogenic enzymes and hormone receptors. For 52 d, gilts in the control group were fed 100 g/d of regular diets, whereas gilts in the omega-3 group were fed 75 g/d of such diets plus 25 g/d of the microalgae Schizochytium sp. (3.5 g/d of omega-3 PUFA; n = 8 gilts/group). Blood samples were collected at day 0, day 21, and day 52. Total serum cholesterol levels were lower for the omega-3 group than for the control group (P < 0.05), but high-density lipoprotein-cholesterol levels were reduced at day 52 for both groups (P < 0.05). Gilts in the omega-3 group presented lower feed intake, better feed conversion, and less-intense immunolabeling for leptin and its receptor in the cytoplasm of oocytes included in primordial/primary follicles than gilts in the control group (P < 0.05). The expression of genes coding for cholesterol side-chain cleavage and aromatase enzymes and the LH receptor in follicular cells was lower for supplemented gilts (P < 0.05). Compared with controls, supplemented gilts presented decreased serum cholesterol levels and better feed conversion, but leptin presence and gene expression for steroidogenic enzymes and for the LH receptor were lower at ovarian level.

RESUMEN

Placental tissues from humans, rodents, and farm animal contain leptin and its receptor. Expression of leptin has already been described in horses, although there is no description about immunolocalization of leptin and its receptor. The aim of the present study was to investigate the presence and distribution of leptin and ObR-b in the equine placenta at term by immunofluorescence staining, and the changes on plasma leptin concentrations during late gestation. The present study involved eight Criollo-type mares carrying healthy pregnancies. Blood samples were collected during the third trimester of gestation, at foaling, and at 24 hours after foaling. Leptin concentrations were analyzed via radioimmunoassay. Plasma leptin concentrations did not change from the 8th to the 10th month of gestation and displayed a discrete decrease 24 hours after parturition (P = .07). The expression of leptin and ObR-b was observed in the cytoplasm of pseudostratified epithelial cells in the areolar region and in the epithelium of microcotyledons. Also, leptin receptor was allocated in the apical surface of the cells. The presence of leptin and its receptor (ObR-b) in the placenta of mares at term supports an endocrine and autocrine/paracrine action of leptin within this organ.
