RESUMEN

Aiming to evaluate the potential of Passerine birds as bioindicators of forest fragmentation, we studied the avifauna in the mountain region of the state of Rio de Janeiro by mist-netting between 2001 and 2005. We sampled six sites, including four small fragments (from 4 to 64 ha) in an agricultural area (Teresópolis), one second-growth forest (440 ha - Miguel Pereira) and a continuous forest (10,600 ha, Serra dos Orgãos National Park - SONP). Indicator Species analysis and a Monte Carlo test were run to detect associations between species and sites, considering at least 30% of perfect indication and a significant value for the statistical test, only considering species with at least 10 captures. A total of 30 Passerine birds were sampled, and due to their association to the largest area (SONP), we considered Sclerurus scansor, Mionectes rufiventris, Chiroxiphia caudata and Habia rubica as the best indicators for this area. Five species were more captured not by chance in the 440 ha second-growth: Conopophaga melanops, Myiobius barbatus, Myrmeciza loricata, Philydor atricapillus and Schiffornis virescens and no species were related to any small fragment. This analysis has identified specialized Passerine species in many aspects (foraging substract, needs for nesting, rarity status), and it can be a valuable tool for detecting possible bioindicators.

Lippia alba is empirically used for infusions, teas, macerates, and hydroalcoholic extracts because of its antispasmodic, analgesic, sedative, and anxiolytic effects. Citral is a mixture of trans-geranial and cis-neral and is the main constituent of L. alba essential oil and possesses analgesic, anxiolytic, anticonvulsant, and sedative effects. The present study evaluated the effects of the essential oil of L. alba (EOLa) and citral on compound action potentials (CAPs) in Wistar rat sciatic nerves. Both drugs inhibited CAP in a concentration-dependent manner. The calculated half-maximal inhibitory concentrations (IC50) of peak-to-peak amplitude were 53.2 µg/mL and 35.00 µg/mL (or 230 µM) for EOLa and citral, respectively. Peak-to-peak amplitude of the CAP was significantly reduced by 30 µg/mL EOLa and 10 µg/mL citral. EOLa and citral (at 60 and 30 µg/mL, values close to their respective IC50 for CAP blockade) significantly increased chronaxy and rheobase. The conduction velocity of the first and second CAP components was statistically reduced to ∼86% of control with 10 µg/mL EOLa and ∼90% of control with 3 µg/mL citral. This study showed that EOLa inhibited nerve excitability and this effect can be explained by the presence of citral in its composition. Both EOLa and citral showed inhibitory actions at lower concentrations compared with other essential oils and constituents with local anesthetic activity. In conclusion, these data demonstrate that EOLa and citral are promising agents in the development of new drugs with local anesthetic activity.

Neste estudo, 67 ejaculados foram avaliados, antes e depois da técnica de swim-up, em relação à qualidade seminal e à presença do CAEV. Das 67 amostras testadas por PCRn, antes do swim-up, 47 (70,15%) foram positivas para o DNA pró-viral. No entanto, quatro amostras adicionais foram positivas ao RT-nested PCR após o swim-up, o que permite dizer que, pelo menos, 76,12% (51/67) delas estavam infectadas antes da lavagem. Todavia, em 23,88% (16/67) das amostras não foi detectada a presença do CAEV. Após a aplicação da técnica de swim-up, constatou-se, pela PCRn e RT-nested PCR, que houve uma redução significativa (²= 9,078, p<0,001) da presença do CAEV nas amostras seminais, pois 28 de 51 amostras positivas resultaram livres do vírus (54,90%), tanto para DNA pró-viral quanto para o vírus livre. Em relação à motilidade individual progressiva (MIP) e vigor espermático obtidos antes e depois da técnica de swim-up, observou-se uma diminuição significativa em suas médias, sendo o MIP de 86,42% para 71,49%, já o vigor espermático de 4,16 para 3,93. Conclui-se que a eliminação do CAEV no sêmen é de caráter intermitente, e que a associação da PCRn e RT-nested PCR é uma opção segura para a certificação sanitária individual das amostras seminais quanto à presença ou ausência do CAEV. Finalmente, a técnica de swim-up promove uma redução na infectividade de amostras de sêmen contaminadas, e, além disso, é possível promover a recuperação de espermatozoides viáveis.

Furthermore, four additional samples were positive for RT-nested PCR after swim-up, which allows us to affirm that at least 76.12% (51/67) were infected before washing. However, 23.88% (16/67) of the samples did not detect the presence of CAEV. After application of the swim-up technique it was found, by Nested PCR and RT-nested PCR, that there was a significant decrease (² = 9.078, p <0.001) in the presence of CAEV in semen samples, once 28 of 51 positive samples were free from the virus (54.90%) for both proviral DNA and the free form of the virus. Regarding individual progressive motility (IPM) and spermatic vigor obtained before and after the swim-up technique, a significant decrease was observed in the average, being 86.42% of the IPM to 71.49% and the spermatic vigor from 4.16 for the 3.93. It is concluded that the removal of CAEV in semen has an intermittent character, and the combination of PCR and RT-nested PCR is a safe option for health certification of individual semen samples for the presence or absence of CAEV. Finally, the swim-up technique promotes a reduction in the infectivity of contaminated semen samples, and it is possible to promote the recovery of high individual progressive motility sperm and sperm vigor.

Para avaliar o potencial de Passeriformes como bioindicadores de fragmentação florestal na Mata Atlântica, estudamos a avifauna da região serrana do Rio de Janeiro, utilizando capturas com redes de neblina entre 2001 e 2005. Foram amostradas seis áreas, sendo quatro pequenos fragmentos (de 4 a 64 ha) em áreas agrícolas (Teresópolis), uma mata secundária com cerca de 40 anos (440 ha - Miguel Pereira) e uma área de mata contínua de 10600 hectares (Parque Nacional da Serra dos Órgãos - PARNASO). A análise de espécies indicadoras e o teste de Monte Carlo foram empregados para que se verificassem as espécies mais associadas a cada local amostrado, considerando-se um mínimo de 30 por cento de indicação e valor significativo para o teste mencionado, utilizando-se somente espécies com 10 ou mais capturas. Foi amostrado um total de 30 espécies e, pela sua associação com a maior área (PARNASO), consideramos Sclerurus scansor, Mionectes rufiventris, Chiroxiphia caudata e Habia rubica como as melhores indicadoras. Cinco espécies foram mais capturadas não ao acaso na área de 440 ha: Conopophaga melanops, Myiobius barbatus, Myrmeciza loricata, Philydor atricapillus e Schiffornis virescens. Nenhuma espécie foi associada aos pequenos fragmentos. Como esta análise identificou espécies de Passeriformes especialistas em diversos aspectos (substrato de forrageio, locais de nidificação, status de raridade), ela pode ser uma ferramenta útil na detecção de possíveis bioindicadores.
