RESUMEN

A brown, moderately halophilic, photoautotrophic bacterium designated strain JA363T was purified from a photoheterotrophic enrichment obtained from sediment from Chilika lagoon, Odisha, India. Cells of the isolate were coccoid, motile by means of single polar flagellum and Gram-stain-negative. Strain JA363T had an obligate requirement for NaCl and could tolerate up to 7 % (w/v) NaCl. Strain JA363T had complex growth factor requirements. Internal photosynthetic membranes were present as vesicles. Strain JA363T contained bacteriochlorophyll a and spirilloxanthin series carotenoids with rhodopin as a major (>85 %) component. C16 : 1ω7c/C16 : 1ω6c, C18 : 1ω7c and C16 : 0 were the major fatty acids and phosphatidylglycerol and phosphatidylethanolamine were the major polar lipids. Q8 was the predominant quinone system of strain JA363T. The DNA G+C content was 64 mol%. The highest 16S rRNA gene sequence similarity of strain JA363T was found with the type strains of Thiorhodococcus kakinadensis (98.7 %), Thiohalobacter thiocyanaticus (98.2 %), Thiophaeococcus fuscus (97.4 %) and Thiorhodococcus bheemlicus (96.3 %). However, the phylogenetic trees generated firmly placed strain JA363T in the genus Thiorhodococcus, which was further supported by phenotypic and chemotaxonomic evidence. Consequently, strain JA363T is described as representing a novel species of the genus Thiorhodococcus as Thiorhodococcus fuscus sp. nov. The type strain is JA363T ( = KCTC 5701T = NBRC 104959T).

RESUMEN

An anoxygenic, phototrophic gammaproteobacterium designated strain JA418(T) was isolated from a sediment sample collected from the Baitarani River, Orissa, India. The bacterium was a Gram-negative, motile rod with a single polar flagellum. Bacteriochlorophyll a and rhodopin were the major photosynthetic pigments. The organism grew best at slightly alkaline pH (8-8.5) and lacked chemotrophic growth. The major fatty acids were C(16 : 0), C(16 : 1)omega7c/C(16 : 1)omega6c and C(18 : 1)omega7c. A phylogenetic tree based on 16S rRNA gene sequence analysis showed that strain JA418(T) clusters with species of the genus Marichromatium belonging to the class Gammaproteobacteria. The highest 16S rRNA gene sequence similarities of strain JA418(T) were found with the type strains of Marichromatium gracile (95.9 %), Marichromatium indicum (95.6 %), Marichromatium purpuratum (95.5 %) and Marichromatium bheemlicum (95.6 %). The DNA base composition of strain JA418(T) was 71.4 mol% G+C (by HPLC). Based on the 16S rRNA gene sequence analysis and physiological and chemotaxonomic characteristics, strain JA418(T) is sufficiently different from other Marichromatium species to merit the description of a novel species, Marichromatium fluminis sp. nov., to accommodate it. The type strain is JA418(T) (=KCTC 5717(T) =NBRC 105221(T)).

A Gram-negative, motile, rod-shaped, facultatively anaerobic bacterium was isolated from sediment of Chilika Lagoon, a brackish water lagoon in India. The strain, designated JC5(T), was able to grow in the presence of 0-8.0 % NaCl and at pH 7.0-10.0. The isolate was positive for oxidase and catalase and exhibited alpha-haemolysis. The major fatty acids were iso-C(15 : 0) (18.3 %), C(16 : 0) (11.3 %), C(17 : 1)omega8c (13.1 %) and a summed feature, C(16 : 1)omega7c and/or C(16 : 1)omega6c (15.1 %). The genomic DNA G+C content was 54.6 mol%. A phylogenetic tree based on the 16S rRNA gene sequences showed that strain JC5(T) forms a lineage within the genus Shewanella and is closely related to Shewanella haliotis DW01(T) (98.0 %), Shewanella algae ATCC 51192(T) (97.6 %) and Shewanella marina C4(T) (95.8 %). Further, genomic DNA-DNA hybridization of strain JC5(T) with S. haliotis DW01(T) and S. algae ATCC 51192(T) showed relatedness of only 42 and 23 %, respectively. On the basis of phenotypic and molecular genetic evidence, strain JC5(T) represents a novel species of the genus Shewanella, for which the name Shewanella chilikensis sp. nov. is proposed. The type strain is JC5(T) (=CCUG 57101(T) =NBRC 105217(T) =KCTC 22540(T)).
