RESUMEN

In primary coculture of endothelial and smooth muscle cells, the muscle cells preserve their specialized phenotype (high myosin content) contrary to a pure smooth muscle cell culture. The addition of thrombin to the coculture-system results in loss of specialized phenotype of smooth muscle cell i.e. the relative myosin content decreases.

RESUMEN

In order to contribute to the verification of the hypothesis of Furchgott et al. that the effect of acetylcholine on blood vessels is mediated by factors produced by endothelial cells, the authors tried to provide evidence for the existence of muscarinic receptors on these cells. Using cultivated cells of the endothelium of calve aortas, a specific saturable binding of the potent muscarinic antagonist 3H-Quinuclidinyl benzilate and therewith the existence of muscarinic receptors on the cells could be demonstrated. Long time cultivation leads to the disappearance of the receptors.

RESUMEN

Heparin, sodium butyrate, and the absence of platelet derived factors in the nutrient medium are able to inhibit the decrease of the content of myosin in cultivated arterial smooth muscle cells. Factor VIII and thrombin have no significant influence on this process.

RESUMEN

Endothelial cells were able to accomplish at least 120 cell doublings (60 passages) without any decrease of division speed. After that, the doubling time begins to increase. At passage 80, the number of cells was not doubled even after a period of 4 weeks. Temperature stress (41 degrees C for 3 d) leads the cells to normal doubling times again and allows the cultures to reach passage 124. The cells were able to produce factor VIII antigen, but in less quantities, still at passage 124.

RESUMEN

The mean relative content of myosin of cells from smooth muscular organs was estimated using immunocytochemical methods after enzymatical tissue disintegration. The cells were classified according to myosin content. Aortas and urinary bladders of newborn rats and chicken amnions were used as models. Cells of a permanent cell strain, being free of smooth muscular myosin served as negative controls. It could be shown, that the content of myosin, estimated by immunocytochemical methods correlates with the state of development of smooth muscular organs.

RESUMEN

Smooth muscle myosin was demonstrated in cryostat sections, isolated and cell cultures from aortas of newborn rats by the method of indirect immunofluorescence. The content of myosin in the cells was compared by measuring the fluorescence intensity of single cells. Well differentiated smooth muscle cells present in suspensions of the whole aorta and of the aortic media as well as in aortic cell cultures, are shown to have a higher myosin content than fibroblasts of adventitia and smooth muscle cells with a lower degree of differentiation.

RESUMEN

Investigations with cyclophosphamide were done to answer the question of participation of the endothelium in biotransformation of xenobiotics. Cyclophosphamide inhibits the proliferation of cultivated endothelial cells only in high concentrations. Also after induction of the cells by phenobarbital or 2,5-diphenoxazole no increased toxicity of cyclophosphamide was observed. Though the endothelium disposes of cytochrome-P-450-dependent aryl-4-monooxygenases it does not take part in biotransformation of cyclophosphamide apparently. Further possibilities of biotransformation by endothelial cells will be discussed.

RESUMEN

Eight drugs with vasodilatory effect of 32 vasoactive substances tested induce a significant inhibition of uptake of 3H-adenosine by cultivated calf aortic endothelial cells. These results are discussed as potentiation of the vasodilation activity of the drugs by adenosine.

RESUMEN

Heparin inhibits the proliferation of aortic smooth muscle cells cultured in serum containing culture medium and leads to an increase of their relative myosin content. Using plasma as medium supplement the effect of heparin is not observed. Thrombin is able to inhibit the increase of myosin content caused by heparin. Platelet mitogens are not the point of attack of heparin. The possible mechanism of action of heparin on smooth muscle cells will be discussed.

RESUMEN

Assuming that the immigration of endothelial cells from existing blood vessels into tissues to be vascularized is the first step of neoangiogenesis, we examined the potential antiangiogenic activity of some synthetic inhibitors of serine proteinases on the migratory rate of isolated endothelial cell using Boyden technique. The 4-amidinoanilide of 2-tosylamino-4-phenylbutyric acid was shown to be the most effective inhibitor. Its effect is probably caused by inhibition of the plasminogen activators of tissue type produced by the cells.

Using the muscarinic ligand 3H-quinuclidinyl benzilate the presence of muscarinic receptors on cells isolated from aortas of newborn rats and cultivated for 6 d could be shown. The number of receptors per cell was estimated with 17,400 on cells of confluent and with 87,100 on cells of nonconfluent cultures. Incubation of cells with increasing concentrations of acetylcholine in the range of 10(-13)-10(-7) mol.l-1 resulted in the reaction of growing numbers of cells following the all-or-nothing-law. Physiological concentrations of acetylcholine lead to contractions, showing that dilatations of blood vessels after application of acetylcholine in situ are caused by reaction products of endothelial cells, being absent in the cultures.

The influence of cholesterol added as cholesterol containing liposomes [sphingomyelin/cholesterol (1: 1; mol/mol)] or as cholesterol suspensions on cultured bovine aortic endothelial cells was investigated. 10(-6) mol/l cholesterol enhanced membrane potential, 10(-5) mol/l caused depolarisation. The proliferation of the cells was dependent on the concentration influenced in opposite directions too. The proliferation was stimulated by 10(-6) mol/l cholesterol and inhibited by 10(-5) mol/l. The migratory rate of the cells was increased by 10(-6) mol/l and 10(-5) mol/l cholesterol. Our results suggest that exogenous cholesterol is integrated in membranes of the endothelial cells and causes in this way changes of membrane potential, proliferation rate and migratory activity.

Cancerostatical active synthetic alkyl-lysophospholipids were examined with regard to their effects on membrane potential, proliferation and migration of isolated endothelial cells. In sublytic concentrations all alkyl-lysophospholipids tested caused a hyperpolarization of the membrane of endothelial cells. The effect of alkyl-lysophospholipids on proliferation of endothelial cells was dependent on the serum supplement. The migration of endothelial cells was strongly inhibited by 1-O-octadecyl-2-O-methyl-glycero-phosphocholine. Possible mechanisms of action are discussed.

In the methanolic extract of Meripilus giganteus a mixture of saturated and unsaturated fatty acids (among them palmitic, oleic and linoleic acid) and ergosterol peroxide were identified as immunosuppressive components. Furthermore a homologous series of straight-chained saturated hydrocarbons with 22 to 26 and 29 to 32 C-atoms could be found.

RESUMEN

The relative myosin content of aortic cells of newborn rats was estimated cytophotometrically in the course of cultivation using indirect immunofluorescence staining. It should be shown that the average myosin content of the cell population and the number of cells with high myosin content is greatly enhanced when confluency of the cells is reached and proliferation stops. Following the amount of 14C-labelled leucine incorporated in immunoprecipitated myosin during the same period we found that increasing myosin content of the cells results from an enhanced rate of myosin biosynthesis.

10 Ergostan-3-on derivatives presumably in the majority ergosta-1,4-dien-3-on derivatives could be isolated from Petunia hybrida VILM. Two were identified as the known compounds petuniasterone A and petuniasterone-C-22-O-acetate. The structures of two other compounds named petuniasterone-D-diacetate and 30-hydroxypetuniasterone A could be elucidated. The molecular formulas of the remaining substances were ascertained by mass spectroscopy. The petuniasterones inhibited the proliferation of stimulated human lymphocytes in non-cytotoxic concentrations, possessing therefore possibly immunosuppressive effects. Extracts of the plant and fractions containing a synergistically effective blue fluorescing substance retarded the multiplication speed of influenza viruses; pure petuniasterones were ineffectiv under our conditions in this regard.

Canine transmissible venereal tumors were observed in the nasal passages of three dogs from Dakar, Senegal. Genital tumors were not present in these dogs. These observations, combined with those of few previous reports, stress the necessity to include this neoplasm in the differential diagnosis of nasal tumors in the dog.

Following the oral administration of a magnet to a cow, a perforation of the pharyngeal mucosa occurred. The magnet underwent encapsulation in the vicinity of the stylohyoid bone. The subsequent osteitis gradually extended to the temporal bone, where another abscess developed. The cardiac signs resulting from the irritation of the vagus nerve by the first abscess and the nervous signs associated with the intracranial pressure due to the second abscess are described.

RESUMEN

Multiple osteochondromatosis in a female BoxerA young female Boxer was presented for a problem of lameness affecting the right hind leg. The lameness was associated with a ruptured anterior cruciate ligament. Following the radiographic examination, lesions compatible with a diagnosis of osteochondromatosis were found. There was no pain associated with this condition and it was an accidental finding. Macroscopic and microscopic finds are discussed and there are a few observations about the pathogeny, treatment and prognosis.

RESUMEN

A PULMONARY ANAPLASTIC EPITHELIOMA IN A BITCH: An anaplastic epithelioma of the lungs was diagnosed in a 26 month old female Airedale. The authors describe signs of bronchopneumonia and lameness involving the right fore and hind limbs. The lameness occurred following metastases to the long bones. The clinical signs, the radiological appearance, the postmortem lesions encountered with that neoplasm and differential diagnosis are discussed.
