RESUMEN

The matched filter hypothesis proposes a close match between senders and receivers and is supported by several studies on variation in signal properties and sensory-processing mechanisms among species and populations. Importantly, within populations, individual variation in sensory processing may affect how receivers perceive signals. Our main goals were to characterize hearing sensitivity of Pacific treefrogs (Pseudacris regilla), assess patterns of individual variation in hearing sensitivity, and evaluate how among-individual variation in hearing sensitivity and call frequency content affect auditory processing of communication signals. Overall, males and females are most sensitive to frequencies between 2.0 and 2.5 kHz, which matches the dominant frequency of the call, and have a second region of high sensitivity between 400 and 800 Hz that does not match the fundamental frequency of the call. We found high levels of among-individual variation in hearing sensitivity, primarily driven by subject size. Importantly, patterns of among-individual variation in hearing differ between males and females. Cross-correlation analyses reveal that among-individual variation in hearing sensitivity may lead to differences on how receivers, particularly females, perceive male calls. Our results suggest that individual variation in sensory processing may affect signal perception and influence the evolution of sexually selected traits.

RESUMEN

Albert Feng was a pioneer in the field of auditory neuroethology who used frogs to investigate the neural basis of spectral and temporal processing and directional hearing. Among his many contributions was connecting neural mechanisms for sound pattern recognition and localization to the problems of auditory masking that frogs encounter when communicating in noisy, real-world environments. Feng's neurophysiological studies of auditory processing foreshadowed and inspired subsequent behavioral investigations of auditory masking in frogs. For frogs, vocal communication frequently occurs in breeding choruses, where males form dense aggregations and produce loud species-specific advertisement calls to attract potential mates and repel competitive rivals. In this review, we aim to highlight how Feng's research advanced our understanding of how frogs cope with noise. We structure our narrative around three themes woven throughout Feng's research-spectral, temporal, and directional processing-to illustrate how frogs can mitigate problems of auditory masking by exploiting frequency separation between signals and noise, temporal fluctuations in noise amplitude, and spatial separation between signals and noise. We conclude by proposing future research that would build on Feng's considerable legacy to advance our understanding of hearing and sound communication in frogs and other vertebrates.

Anuran advertisement calls play a fundamental role in social interactions related to reproduction, like territory defense and mate attraction. As such, advertisement calls are often shaped by natural and sexual selection. Therefore, to understand the evolution, function, and mechanisms of signal production and perception, it is important to characterize sources and patterns of variation in call properties. We recorded and analyzed the two types of advertisement calls produced by Pacific tree frogs, Hyliola regilla: diphasic and monophasic calls. Specifically, we examined (i) the relationship between call properties and both body size and physical condition, (ii) patterns of within- and among-individual variation in call properties, and (iii) acoustic differences between diphasic and monophasic calls. While only one property of diphasic calls correlates with physical condition, spectral properties of both types of calls correlate with body size. Patterns of within- and among-individual variation were similar between call types and, overall, consistent with patterns reported for closely related species. Monophasic calls have lower frequencies, are delivered faster, and have higher call effort and duty cycle than diphasic calls. We discuss our results in relation to sexual selection and formulate hypotheses about the evolution, function, and mechanisms of acoustic communication in Pacific tree frogs.

BACKGROUND: The BRCA1 c.3331_3334delCAAG founder mutation has been reported in hereditary breast and ovarian cancer families from multiple Hispanic groups. We aimed to evaluate BRCA1 c.3331_3334delCAAG haplotype diversity in cases of European, African, and Latin American ancestry. METHODS: BC mutation carrier cases from Colombia (n = 32), Spain (n = 13), Portugal (n = 2), Chile (n = 10), Africa (n = 1), and Brazil (n = 2) were genotyped with the genome-wide single nucleotide polymorphism (SNP) arrays to evaluate haplotype diversity around BRCA1 c.3331_3334delCAAG. Additional Portuguese (n = 13) and Brazilian (n = 18) BC mutation carriers were genotyped for 15 informative SNPs surrounding BRCA1. Data were phased using SHAPEIT2, and identical by descent regions were determined using BEAGLE and GERMLINE. DMLE+ was used to date the mutation in Colombia and Iberia. RESULTS: The haplotype reconstruction revealed a shared 264.4-kb region among carriers from all six countries. The estimated mutation age was ~ 100 generations in Iberia and that it was introduced to South America early during the European colonization period. CONCLUSIONS: Our results suggest that this mutation originated in Iberia and later introduced to Colombia and South America at the time of Spanish colonization during the early 1500s. We also found that the Colombian mutation carriers had higher European ancestry, at the BRCA1 gene harboring chromosome 17, than controls, which further supported the European origin of the mutation. Understanding founder mutations in diverse populations has implications in implementing cost-effective, ancestry-informed screening.

The ability to localize communication signals plays a fundamental role in social interactions. For signal localization to take place, the sensory system of the receiver must extract information about distance and direction to the sender from physical characteristics of the signal. In many sensory systems, information from multiple peripheral receptors must be integrated by central sensory pathways to determine the sender location. Here, we asked whether evolutionary divergence in the electrosensory and visual systems of mormyrid fish is associated with signal localization behavior. In mormyrids, differences in the distribution of electroreceptors on the surface of the skin are associated with differences in the midbrain exterolateral nucleus (EL). Species with electroreceptors clustered in three rosettes on both sides of the head have a small and undifferentiated EL. In contrast, EL is enlarged and subdivided into anterior (ELa) and posterior (ELp) regions in species that have electroreceptors broadly -distributed throughout the body. Interestingly, species with EL and clustered electroreceptors also have larger visual systems and higher visual acuity than species with ELa/ELp and broadly distributed electroreceptors. Species with broadly distributed electroreceptors and ELa/ELp approached a simulated conspecific by following the curved electric field lines generated by the electrosensory stimulus. In contrast, a species with small EL and clustered electroreceptors, but an enlarged visual system, followed shorter and straighter paths to the stimulus source. In the central electrosensory system, evoked field potentials in response to stimuli delivered from the left versus the right differed more in EL than in ELa/ELp. Our results suggest that signal localization behavior is associated with differences in sensory specializations. We propose that the distribution of electroreceptors on the body affects the ability of individuals to align parallel to electric field lines and maintain such alignment while approaching the signal source. The spatial resolution of sensory information relayed from the periphery to the midbrain in species with clustered electroreceptors may allow for gross, but not fine, processing of sender location. Furthermore, visual information may play an important role in localizing signaling individuals in species with small EL and clustered electroreceptors. In line with previous studies, we suggest that the physiological and behavioral differences associated with signal localization reflect adaptations to different habitats and social environments.

Many animals communicate acoustically in large social aggregations. Among the best studied are frogs, in which males form large breeding choruses where they produce loud vocalizations to attract mates. Although chorus noise poses significant challenges to communication, it also possesses features, such as comodulation in amplitude fluctuations, that listeners may be evolutionarily adapted to exploit in order to achieve release from masking. This study investigated the extent to which the benefits of comodulation masking release (CMR) depend on overall noise level in Cope's gray treefrog (Hyla chrysoscelis). Masked signal recognition thresholds were measured in response to vocalizations in the presence of chorus-shaped noise presented at two levels. The noises were either unmodulated or modulated with an envelope that was correlated (comodulated) or uncorrelated (deviant) across the frequency spectrum. Signal-to-noise ratios (SNRs) were lower at the higher noise level, and this effect was driven by relatively lower SNRs in modulated conditions, especially the comodulated condition. These results, which confirm that frogs benefit from CMR in a level-dependent manner, are discussed in relation to previous studies of CMR in humans and animals and in light of implications of the unique amphibian inner ear for considerations of within-channel versus across-channel mechanisms.

Hypertrophic cardiomyopathy (HCM) is a heterogeneous group of diseases related to sarcomere gene mutations exhibiting heterogeneous phenotypes with an autosomal dominant mendelian pattern of inheritance. The disorder is characterized by diverse phenotypic expressions and variable natural progression, which may range from dyspnea and/or syncope to sudden cardiac death. It is found across all racial groups and is associated with left ventricular hypertrophy in the absence of another systemic or cardiac disease. The management of HCM is based on a thorough understanding of the underlying morphology, pathophysiology, and clinical course. Imaging findings of HCM mirror the variable expressivity and penetrance heterogeneity, with the added advantage of diagnosis even in cases where a specific mutation may not yet be found. The diagnostic information obtained from imaging varies depending on the specific stage of HCM-phenotype manifestation, including the prehypertrophic, hypertrophic, and later stages of adverse remodeling into the burned-out phase of overt heart failure. However, subtle or obvious, these imaging findings become critical components in diagnosis, management, and follow-up of HCM patients. Although diagnosis of HCM traditionally relies on clinical assessment and transthoracic echocardiography, recent studies have demonstrated increased utility of multidetector computed tomography (CT) and particularly cardiac magnetic resonance (MR) imaging in diagnosis, phenotype differentiation, therapeutic planning, and prognostication. In this article, we provide an overview of the genetics, pathophysiology, and clinical manifestations of HCM, with the spectrum of imaging findings at MR imaging and CT and their contribution in diagnosis, risk stratification, and therapy.

Exposure to arsenic is a global health concern. We previously documented an inhibitory effect of inorganic Arsenite on IgE-mediated degranulation of RBL-2H3 mast cells (Hutchinson et al., 2011; J. Appl. Toxicol. 31: 231-241). Mast cells are tissue-resident cells that are positioned at the host-environment interface, thereby serving vital roles in many physiological processes and disease states, in addition to their well-known roles in allergy and asthma. Upon activation, mast cells secrete several mediators from cytoplasmic granules, in degranulation. The present study is an investigation of Arsenite's molecular target(s) in the degranulation pathway. Here, we report that arsenic does not affect degranulation stimulated by either the Ca(2) (+) ionophore A23187 or thapsigargin, which both bypass early signaling events. Arsenic also does not alter degranulation initiated by another non-IgE-mediated mast cell stimulant, the G-protein activator compound 48/80. However, arsenic inhibits Ca(2) (+) influx into antigen-activated mast cells. These results indicate that the target of arsenic in the degranulation pathway is upstream of the Ca(2) (+) influx. Phospho-Syk and phospho-p85 phosphoinositide 3-kinase enzyme-linked immunosorbent assays data show that arsenic inhibits early phosphorylation events. Taken together, this evidence indicates that the mechanism underlying arsenic inhibition of mast cell degranulation occurs at the early tyrosine phosphorylation steps in the degranulation pathway. We examined temporal processing of harmonic tone complexes in two woodland species (tufted titmice and white-breasted nuthatches) and two open-habitat species (house sparrows and white-crowned sparrows). Envelope and fine-structure processing were quantified using the envelope following response (EFR) and frequency following response (FFR). We predicted stronger EFRs in the open-habitat species based on broader auditory filters and greater amplitude modulation of vocal signals in this group. We predicted stronger FFRs in woodland species based on narrower auditory filters. As predicted, EFR amplitude was generally greatest in the open habitat species. FFR amplitude, in contrast, was greatest in white-crowned sparrows with no clear difference between habitats. This result cannot be fully explained by species differences in audiogram shape and might instead reflect greater acoustic complexity of songs in the white-crowned sparrow. Finally, we observed stronger FFRs in woodland species when tones were broadcast with the next higher harmonic in the complex. Thus, species such as nuthatches that have songs with strong harmonics may process these sounds using enhanced spectral processing instead of enhanced amplitude-envelope processing. The results suggest coevolution between signal design and temporal processing of complex signals and underscore the need to study auditory processing with a diversity of signals.

In this contribution, we implement two techniques to reinforce optical encryption, which we restrict in particular to the QR codes, but could be applied in a general encoding situation. To our knowledge, we present the first experimental-positional optical scrambling merged with an optical encryption procedure. The inclusion of an experimental scrambling technique in an optical encryption protocol, in particular dealing with a QR code "container", adds more protection to the encoding proposal. Additionally, a nonlinear normalization technique is applied to reduce the noise over the recovered images besides increasing the security against attacks. The opto-digital techniques employ an interferometric arrangement and a joint transform correlator encrypting architecture. The experimental results demonstrate the capability of the methods to accomplish the task.

BACKGROUND: The lower gastrointestinal (GI) tract, formed from the midgut and hindgut, encompasses the colon, rectum and anal canal. AIM: The aim of this review is to provide an overview of the anatomy and physiology of the lower GI tract. METHODS: Literature review on anatomy and physiology of the lower GI tract, including normal motility and phases of defecation. It derives its blood supply from the superior and inferior mesenteric arteries while it is innervated by the extrinsic autonomic (the thoracolumbar and sacral nerves) and the intrinsic enteric nervous system. The colon has four layers: mucosa, submucosa, muscularis externa and serosa. The anal canal ends in the internal and external anal sphincters (EASs) involved in continence and defecation. The lower GI tract is predominantly involved in digestion, absorption, defecation and protection. Defecation is a complex process that requires inter-neural (enteric and autonomic nervous systems), neurohormonal and neuromuscular coordination. It has four phases which include basal, pre-expulsive, expulsive and end phase. High-propagating contractions in the colon propel stool to the rectum leading to rectal distention and the recruitment of the recto-anal inhibitory reflex. Once able, the EAS, under full conscious control, is then relaxed allowing stool to be evacuated. Other defecation reflexes include the gastrocolic, gastroileal and coloanal reflexes. CONCLUSIONS: Recent advances provide novel techniques to investigate motility patterns including high-resolution manometry protocols with automated assessments, magnetic resonance imaging techniques for defecography, wireless motility capsules and fecobionics.

INTRODUCTION: Pediatric prucalopride studies for treatment of gastrointestinal (GI) disorders have reported mixed results. We aimed to assess the safety and effectiveness of prucalopride in functional constipation (FC) with and without upper GI symptoms. METHODS: Retrospective data on patients with FC receiving combined prucalopride and conventional therapy was compared with those receiving conventional therapy alone within 12 months. Thirty patients on combined therapy and those on conventional therapy were each matched on the basis of age, gender, race, and presence of fecal soiling. Response (complete, partial, or no resolution) was compared. Similarly, response to concurrent functional upper GI symptoms (postprandial pain, bloating, weight loss, vomiting, early satiety, or nausea) and dysphagia, as well as adverse effects, were evaluated in the combined group. RESULTS: Mean age of 57 cases was 14.7 ± 4.9 years and 68% were female. Comorbidities included functional upper GI (UGI) symptoms (84%), dysphagia (12%), mood disorders (49%), and hypermobility spectrum disorder (37%). Unmatched cases reported 63% improvement to FC; response did not differ between the matched cohorts (70% versus 76.6%, p = 0.84). Cases showed a 56% improvement in functional UGI symptoms and 100% in dysphagia. Adverse effects were reported in 30%, abdominal cramps being most common. Four (7%) patients with a known mood disorder reported worsened mood, of which two endorsed suicidal ideation. CONCLUSION: Prucalopride efficaciously treated concurrent UGI symptoms and dysphagia in constipated pediatric patients and was overall well tolerated. Preexisting mood disorders seemed to worsen in a small subset of cases.

Toxoplasmosis is the most prevalent parasitic zoonosis worldwide, causing ocular and neurological diseases. No vaccine has been approved for human use. We evaluated the response of peripheral blood mononuclear cells (PBMCs) to a novel construct of Toxoplasma gondii total antigen in maltodextrin nanoparticles (NP/TE) in individuals with varying infectious statuses (uninfected, chronic asymptomatic, or ocular toxoplasmosis). We analyzed the concentration of IFN-γ after NP/TE ex vivo stimulation using ELISA and the immunophenotypes of CD4+ and CD8+ cell populations using flow cytometry. In addition, serotyping of individuals with toxoplasmosis was performed by ELISA using GRA6-derived polypeptides. Low doses of NP/TE stimulation (0.9 µg NP/0.3 µg TE) achieved IFN-γ-specific production in previously exposed human PBMCs without significant differences in the infecting serotype. Increased IFN-γ expression in CD4+ effector memory cell subsets was found in patients with ocular toxoplasmosis with NP/TE but not with TE alone. This is the first study to show how T-cell subsets respond to ex vivo stimulation with a vaccine candidate for human toxoplasmosis, providing crucial insights for future clinical trials.

In Cope's gray treefrog (Hyla chrysoscelis), thresholds for recognizing conspecific calls are lower in temporally modulated noise backgrounds compared with unmodulated noise. The effect of modulated noise on discrimination among different conspecific calls is unknown. In quiet, females prefer calls with relatively more pulses. This study tested the hypotheses that noise impairs selectivity for longer calls and that processes akin to dip listening in modulated noise can ameliorate this impairment. In two-stimulus choice tests, female subjects were allowed to choose between an average-length call and a shorter or longer alternative. Tests were replicated at two signal levels in quiet and in the presence of chorus-shaped noise that was unmodulated, modulated by a sinusoid, or modulated by envelopes resembling natural choruses. When subjects showed a preference, it was always for the relatively longer call. Noise reduced preferences for longer calls, but the magnitude of this reduction was unrelated to whether the noise envelope was modulated or unmodulated. Together, the results are inconsistent with the hypothesis that dip listening improves a female gray treefrog's ability to select longer calls in modulated compared with unmodulated noise.

We documented two stages of bone involvement due to syphilis in two adult patients infected with human immunodeficiency virus. Bony lesions of secondary versus tertiary syphilis cannot be differentiated on clinical or radiologic grounds alone. Given the rarity of this clinical presentation, there is no consensus on treatment duration and related outcomes.

Las lesiones óseas de la sífilis secundaria y de la sífilis terciaria no se pueden diferenciar únicamente por características clínicas o radiológicas. The degradability of the biocomposite produced from a binary mixture of thermoplastic banana starch (TPS) and polycaprolactone (PCL) reinforced with fique fibers (Fs) was evaluated in three different environments (soil, compost, water). An experimental design with two factors (soil and compost) and three levels (5, 10, and 20 cm) was used, with additional tests for a third aqueous environment (water from the lake of the Universidad del Valle) at a depth of 20 cm. The biocomposite was prepared from the implementation of a twin-screw extrusion process of the binary mixture TPS/PCL and fique fibers (54, 36, and 10% composition, respectively), followed by hot compression molding, and after that, generating ASTM D638 type V specimens using a stainless-steel die. The specimens were dried and buried according to the experimental design, for a total experimental time of 90 days, and removing samples every 30 days. After 90 days, all samples showed signs of degradation, where the best results were obtained in the compost at a depth of 20 cm (34 ± 4% mass loss and a decrease in tensile strength of 77.3%, which indicates that the material lost mechanical properties). TPS was the fastest disappearing component and promoted the degradation of the composite material as it disappeared. Finally, the aqueous media presented the lowest degradation results, losing only 20% of its initial mass after 90 days of the experiment, being the least effective environment in which the biocomposite can end up.

Triclosan is a broad-spectrum antibacterial agent, which has been shown previously to alleviate human allergic skin disease. The purpose of this study was to investigate the hypothesis that the mechanism of this action of triclosan is, in part, due to effects on mast cell function. Mast cells play important roles in allergy, asthma, parasite defense, and carcinogenesis. In response to various stimuli, mast cells degranulate, releasing allergic mediators such as histamine. In order to investigate the potential anti-inflammatory effect of triclosan on mast cells, we monitored the level of degranulation in a mast cell model, rat basophilic leukemia cells, clone 2H3. Having functional homology to human mast cells, as well as a very well defined signaling pathway leading to degranulation, this cell line has been widely used to gain insight into mast-cell driven allergic disorders in humans. Using a fluorescent microplate assay, we determined that triclosan strongly dampened the release of granules from activated rat mast cells starting at 2 µM treatment, with dose-responsive suppression through 30 µM. These concentrations were found to be non-cytotoxic. The inhibition was found to persist when early signaling events (such as IgE receptor aggregation and tyrosine phosphorylation) were bypassed by using calcium ionophore stimulation, indicating that the target for triclosan in this pathway is likely downstream of the calcium signaling event. Triclosan also strongly suppressed F-actin remodeling and cell membrane ruffling, a physiological process that accompanies degranulation. Our finding that triclosan inhibits mast cell function may explain the clinical data mentioned above and supports the use of triclosan or a mechanistically similar compound as a topical treatment for allergic skin disease, such as eczema.

Despite the importance of perceptually separating signals from background noise, we still know little about how nonhuman animals solve this problem. Dip listening, an ability to catch meaningful 'acoustic glimpses' of a target signal when fluctuating background noise levels momentarily drop, constitutes one possible solution. Amplitude-modulated noises, however, can sometimes impair signal recognition through a process known as modulation masking. We asked whether fluctuating noise simulating a breeding chorus affects the ability of female green treefrogs (Hyla cinerea) to recognize male advertisement calls. Our analysis of recordings of the sounds of green treefrog choruses reveal that their levels fluctuate primarily at rates below 10 Hz. In laboratory phonotaxis tests, we found no evidence for dip listening or modulation masking. Mean signal recognition thresholds in the presence of fluctuating chorus-like noises were never statistically different from those in the presence of a non-fluctuating control. An analysis of statistical effects sizes indicates that masker fluctuation rates, and the presence versus absence of fluctuations, had negligible effects on subject behavior. Together, our results suggest that females listening in natural settings should receive no benefits, nor experience any additional constraints, as a result of level fluctuations in the soundscape of green treefrog choruses.

An important aspect of hearing and acoustic communication is the ability to discriminate differences in sound level. Little is known about level discrimination in anuran amphibians (frogs and toads), for which vocal communication in noisy social environments is often critical for reproduction. This study used two-choice phonotaxis tests to investigate the ability of females of Cope's gray treefrog (Hyla chrysoscelis) to discriminate between two advertisement calls differing only in sound pressure level by 2, 4, or 6 dB. Tests were conducted in the presence and absence of chorus-shaped noise (73 dB) and using two different ranges of signal levels (73-79 dB and 79-85 dB). Females discriminated between two signals differing by as little as 2-4 dB. In contrast to expectations based on the "near miss to Weber's law" in birds and mammals, level discrimination was slightly better at the lower range of signal amplitudes, a finding consistent with earlier studies of frogs and insects. Realistic levels of background noise simulating a breeding chorus had no discernable effect on discrimination at the sound level differences tested in this study. These results have important implications for studies of auditory masking and signaling behavior in the contexts of anuran hearing and sound communication.
