RESUMEN

To investigate the roles and explore the altered expression of microRNAs (miRNAs) and mRNAs in chicken embryos in response to Newcastle disease virus (NDV) infection, deep sequencing was performed. Then, a conjoint analysis of small RNA-seq and mRNA-seq was performed to screen interactional miRNA⁻mRNA pairs during NDV infection. In total, 15 and 17 up- and downregulated miRNAs were identified that potentially targeted 4279 and 6080 mRNAs in NDV-infected chicken embryonic tissues, respectively; in addition, 595 upregulated and 480 downregulated mRNAs were identified. The conjoint analysis of the obtained data identified 1069 miRNA⁻mRNA pairs. Among these pairs, 130 pairs were related to immune or inflammatory responses. The relationship between gga-miR-203a and its target transglutaminase 2 (TGM2) was confirmed using a dual-luciferase reporter system and a real time quantitative polymerase chain reaction (RT-qPCR) assay. Overall, the discovery of miRNAs, mRNAs, and their potential pairing relationships, which may be involved in the regulation of NDV infection, will facilitate our understanding of the complex regulatory relationship between the host and the virus.

According to the spectral absorption characteristics of polluting gases and fluorescence characteristics, a time-division multiplexing detection system is designed. Through this system we can detect Methane (CH4) and sulfur dioxide (SO2) by using spectral absorption method and the SO2 can be detected by using UV fluorescence method. The system consists of four parts: a combination of a light source which could be switched, the common optical path, the air chamber and the signal processing section. The spectral absorption characteristics and fluorescence characteristics are measured first. Then the experiment of detecting CH4 and SO2 through spectral absorption method and the experiment of detecting SO2 through UV fluorescence method are conducted, respectively. Through measuring characteristics of spectral absorption and fluorescence, we get excitation wavelengths of SO2 and CH4 measured by spectral absorption method at the absorption peak are 280 nm and 1.64 µm, respectively, and the optimal excitation wavelength of SO2 measured by UV fluorescence method is 220 nm. we acquire the linear relation between the concentration of CH4 and relative intensity and the linear relation between the concentration of SO2 and output voltage after conducting the experiment of spectral absorption method, and the linearity are 98.7%, 99.2% respectively. Through the experiment of UV fluorescence method we acquire that the relation between the concentration of SO2 and the voltage is linear, and the linearity is 99.5%. Research shows that the system is able to be applied to detect the polluted gas by absorption spectrum method and UV fluorescence method. Combing these two measurement methods decreases the costing and the volume, and this system can also be used to measure the other gases. Such system has a certain value of application.

Sodium methylparaben as one kind of preservatives is widely used in our life, but it will do harm to health if it is eaten too much. So there are strict rules on the dosage of sodium methylparaben in every country. The fluorescence spectral properties of sodium methylparaben in aqueous solution and orange juice solution are analyzed with FS920 fluorescence spectrometer. The research result shows that the fluorescence characteristic peak of sodium methylparaben solution is in λ(ex)/λ(em) = 380/5 10 nm, while sodium methylparaben orange juice solution has two fluorescence characteristic peaks which are in λ(ex)/λ(em) = 440/520 nm and 470/530 nm, and its best excitation wavelength is 440 nm. So it can be concluded from the result that there is a significant change between the characteristic peaks of sodium methylparaben in the two solution. Compared with the fluorescence characteristic peak of sodium methylparaben solution, thoses of sodium methylparaben orange juice solution are changed significantly, which are caused by the interference of orange juice fluorescence characteristics. In order to determine the content of sodium methylparaben in the fresh orange juice, a detection model of sodium methylparaben content in orange juice is built based on GA-BP neural network, according to the relationship between fluorescence intensity in λ(ex) = 440 nm and the content of sodium methylparaben orange juice solution. When the accuracy of the mean square error in the process of network training reaches 10(-3), the correlation coefficient of network output and that of the expected is 0.996. At the same time, a better prediction result can be obtained that the average recovery of the forecast samples is 98.67% and the average relative standard deviation is 0.86%. When the concentration ranges from 0.02 to 1.0 g x L(-1), the results testify that detection method based on fluorescence spectroscopy and GA-BP neural network can accurately determine the content of sodium methylparaben in orange juice. This method has the features of novelty and simplicity and it is expected to be applied to the determination of sodium methylparaben in other kinds of drink.

In this paper, fluorescence spectra properties of potassium sorbate in aqueous solution and orange juice are studied, and the result.shows that in two solution there are many difference in fluorescence spectra of potassium sorbate, but the fluorescence characteristic peak exists in λ(ex)/λ(em) = 375/490 nm. It can be seen from the two dimensional fluorescence spectra that the relationship between the fluorescence intensity and the concentration of potassium sorbate is very complex, so there is no linear relationship between them. To determine the concentration of potassium sorbate in orange juice, a new method combining Particle Swarm Optimization (PSO) algorithm with Back Propagation (BP) neural network is proposed. The relative error of two predicted concentrations is 1.83% and 1.53% respectively, which indicate that the method is feasible. The PSO-BP neural network can accurately measure the concentration of potassium sorbate in orange juice in the range of 0.1-2.0 g · L⁻¹.

OBJECTIVE: To investigate the effect of NaCl stress on the growth and physiological characteristics of Physalis alkekengi var. franchetii, and to discuss the mechanism of salt tolerance. METHODS: The test-tube plantlet was cultured on the stress medium with NaCl (0.2%~1.0%), the content of proline, soluble sugar, soluble protein, chlorophyll and malondialdthyde (MDA), and membrane relative permeability, as well as the activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) were determined and analyzed. RESULTS: The average height, stem diameter, root length, number of root and chlorophyll content gradually decreased at salinity less than or equal 0.6% NaCl, then decreased rapidly at salinity greater than or equal 0.8% NaCl, while the root formation was significantly inhibited. The content of free proline and SOD activities always increased with NaCl concentration increasing (0.2%~ 1.0%). Nevertheless, the contents of soluble sugar, soluble protein and the activities of CAT and POD increased firstly and then decreased, but the contents of soluble sugar and soluble protein were higher than those of the control group, whereas the content of MDA and membrane relative permeability accumulated slowly at salinity less than or equal 0.6% NaCl, then increased rapidly at salinity greater than 0.8% NaCl. CONCLUSION: The test-tube plantlet has an adaptability to salty environment at salinity less than or equal 0.6% NaCl, and the test-tube plantlet can alleviate the salty injury by increasing the contents of osmotic regulation substances, such as soluble sugar and soluble protein, decreasing the osmotic potential, and increasing the activities of antioxidant enzymes.

OBJECTIVE: Teprotumumab plays an important role in thyroid eye disease pathogenesis and progression. We intend to mine the adverse event (AE) signals from a relevant database, thereby contributing to the safe use of teprotumumab. METHODS: The data obtained from the ASCII data packages in the FAERS database from January 2020 to the second quarter of 2023 were imported into the SAS software (version 9.4) for data cleaning and analysis. Disproportionality analysis was performed using the reporting odds ratio (ROR) in conjunction with the United Kingdom Medicines and Healthcare Products Regulatory Agency (MHRA) omnibus standard method to detect positive signals. PARTICIPANTS: This retrospective observational study relied on adverse drug reactions reported to the FDA through FAERS, which is a standard public system for spontaneous reporting. RESULTS: Collectively, 2171 AE reports for teprotumumab were collected, among which 108 significant signals were identified involving 17 system organ classes. The SOC of ear and labyrinth disorders included the most AE signals and reports. Muscle spasms, fatigue, headache, nausea, diarrhea, alopecia, blood glucose increased, hypoacusis, tinnitus, and diabetes mellitus were the top ten PTs ranked by the frequency of reporting, meanwhile, the two high-strength signals of thyroid-stimulating immunoglobulin increase (ROR 662.89, 95% CI 182.40-2409.19) and gingival recession (ROR 125.13, 95% CI 79.70-196.45) were not documented in the drug instruction. Meanwhile, we found a higher risk of increased blood glucose, deafness, and decreased appetite for male patients, and headache for female patients. CONCLUSIONS: Clinical application of teprotumumab should be closely monitored for ototoxicity, nail abnormalities, and menstrual changes, as well as for AEs not mentioned in the drug instruction, including gingival recession, thyroid-stimulating immunoglobulin increase, and so on.

Background: Currently, there is no standard treatment for relapsed/refractory NK/T-cell lymphoma (NKTCL). Liposomal mitoxantrone (Lipo-MIT) showed good anti-tumor effect in patients with NKTCL, breaking the limitation of natural resistance of NKTCL to anthracyclines. To further improve the efficacy, we tried a combination therapy based on Lipo-MIT in patients with relapsed/refractory NKTCL. Methods: 12 patients with relapsed/refractory NKTCL were enrolled in this retrospective study, all of whom had previously received pegaspargase-based treatments. The salvage treatment was a combination regimen based on Lipo-MIT. The efficacy was evaluated after every two cycles. Results: 11 patients had stage IV NKTCL, and all but one patients had an NRI score of ≥3. The median previous lines of treatment was two (range, 1-4), and five patients were refractory to their last line of treatment. The best response rates were as follows: complete response (CR) in five (41.7%) patients, partial response in five (41.7%) patients, stable disease in one (8.3%) patient, and progressive disease in one (8.3%) patient. At a median follow-up of four months (range, 2-14), seven patients died, with a median PFS of five months and a median OS of seven months. The six-month PFS and OS rate was 44.4% and 52.1%, respectively. All patients had suffered from side effects, among which myelosuppression was most reported. Nine patients had grade three or more myelosuppression, and the median recovery time from myelosuppression was 14 days (2-35 days). Five patients had obvious skin hyperpigmentation, and the CR rate was significantly higher compared with those without skin hyperpigmentation (80% vs. 14.3%, p=0.023). Other side effects included liver insufficiency (N=4), coagulation dysfunction (N=4), acute pancreatitis (N=2), and immunotherapy-related adverse effects (irAEs, N=2). Conclusion: Combination therapy based on Lipo-MIT has a high remission rate for relapsed/refractory NKTCL, but the duration of remission needs to be further extended. Lipo-MIT has obvious myelosuppression toxicity, and active supportive therapy should be given when combined with other cytotoxic drugs.

Interferon-induced protein-35 kDa (IFI35) was an antiviral protein induced by interferon (IFN)-γ, which plays an important role in the IFN-mediated antiviral signaling pathway. Here, we cloned and identified IFI35 in the chicken for the first time. Chicken IFI35 (chIFI35) contains an open reading frame (ORF) of 1,152 bp encoding a protein of 384 amino acids containing two conserved Nmi/IFI35 domain (NID) motifs. Tissue distribution analysis of chIFI35 in healthy and Newcastle disease (ND) virus-infected chickens indicated a positive correlation between chIFI35 mRNA transcription and ND viral loads in various tissues. The role of chIFI35 in regulation NDV replication were further assessed by up- or down-regulated chIFI35 expression in DF-1 cells transfected with plasmid harboring chIFI35, pCMV-3HA-chIFI35 or shRNA targeting chIFI35, pshRNA-chIFI35 plasmids. NDV replications in DF-1 cells were significantly reduced or slightly increased by over- or under-expression of the chIFI35 protein, respectively, indicating the role of chIFI35 in anti-NDV infection. Moreover, chIFI35 also involved in regulation of viral gene transcription and IFNs expression. The collected data were meaningful for research of chicken antiviral immunity and shed light on the pleiotropic antiviral effect of chIFI35 during NDV infection.

Background: The prevalence of diabetes mellitus remains high in China, and more cardiovascular and cerebrovascular adverse events due to diabetes mellitus are likely to occur in the future. Objective: To analyze the gap between the current pharmacotherapy management and the guidelines for inpatients with type 2 diabetes mellitus from the perspective of pharmacists so as to provide a reference for optimal pharmacotherapy management methods and models for patients with type 2 diabetes mellitus. Methods: The study was a cross-sectional observational study. The study was conducted by investigating and analyzing the use of glucose-lowering drugs, adjustment of blood pressure management strategy, lipid management, weight management, and application of antiplatelet drugs in type 2 diabetes inpatients. Results: A total of 1086 patients with type 2 diabetes were included. Metformin, glycosidase inhibitors, and basal insulin were the most used among type 2 diabetes inpatients. The use of SGLT-2, GLP-1 RAs, DPP-4, and metformin all showed significant increase. SGLT-2 inhibitors (SGLT-2i) showed the fastest increase from 2020 to 2021 (14.5% vs. 39.6%); However, the application rate of SGLT-2i was low among patients with combined ASCVD, renal insufficiency, and diabetic nephropathy (46.4%, 40.9%, and 45.8% respectively). For patients with substandard blood pressure at admission, the average rate of intervention by endocrinologists for adjusting the antihypertensive regimen during hospitalization was 55.6%, and the application rate of ACEI/ARB drugs reached 64.4%. The application rate of statins among patients with type 2 diabetes was still relatively high, at 78.8%. However, the overall intervention rate for patients with suboptimal LDL-c was only 24.1%. The application rate of antiplatelet agents for patients with ASCVD was 77.6%, which was higher than that for patients without ASCVD. Conclusion: There is still a gap between the practice of medication treatment management of Chinese inpatients with type 2 diabetes and the guidelines, especially in the application of GLP-1RAs and SGLT-2i in patients with concomitant ASCVD, diabetic nephropathy, and renal insufficiency. Meanwhile, physicians and pharmacists should pay more attention on achieving blood pressure and LDL-c standards in type 2 diabetic patients and provide timely interventions.

The purpose of this study was to investigate the presence of endolymphatic hydrops (EH) in both affected and unaffected ears of patients with pantonal unilateral idiopathic sudden sensorineural hearing loss (ISSNHL) using three-dimensional fluid-attenuated inversion recovery magnetic resonance imaging (3D-FLAIR MRI) and further evaluate the significance of EH in this disorder. Twenty-seven ISSHL patients were enrolled in this study. 3D-FLAIR MRI was performed 24 h after intratympanic injection of gadolinium-diethylenetriaminepentaacetic acid (Gd-DPTA). The incidences of EH in the affected ears and contralateral unaffected ears were compared and the correlations of EH with vertigo or prognosis were analyzed using the Chi-square test. The results showed that the incidence of EH was 68.0% (17/25) in the affected ears and 34.8% (8/23) in the unaffected ears. There was a statistically significant difference between affected ears and unaffected ears in regard to the incidence of EH (P<0.05). There were no significant correlations of EH with vertigo (P=1.000) or with prognosis (P=0.359) in the affected ears. In conclusion, there is EH in the inner ear of patients with pantonal ISSNHL; EH is not related to vertigo, a concomitant symptom of ISSNHL, and the prognosis of this condition. The presence of EH may be a secondary reaction following the impairment of the inner ears with pantonal ISSNHL.

Mitochondrial dysfunction is an early feature of Alzheimer's disease (AD). Accumulated damaged mitochondria, which are associated with impaired mitophagy, contribute to neurodegeneration in AD. We show levels of Disrupted-in-schizophrenia-1 (DISC1), which is genetically associated with psychiatric disorders and AD, decrease in the brains of AD patients and transgenic model mice and in Aß-treated cultured cells. Disrupted-in-schizophrenia-1 contains a canonical LC3-interacting region (LIR) motif (210 FSFI213 ), through which DISC1 directly binds to LC3-I/II. Overexpression of DISC1 enhances mitophagy through its binding to LC3, whereas knocking-down of DISC1 blocks Aß-induced mitophagy. We further observe overexpression of DISC1, but not its mutant (muFSFI) which abolishes the interaction of DISC1 with LC3, rescues Aß-induced mitochondrial dysfunction, loss of spines, suppressed long-term potentiation (LTP). Overexpression of DISC1 via adeno-associated virus (serotype 8, AAV8) in the hippocampus of 8-month-old APP/PS1 transgenic mice for 4 months rescues cognitive deficits, synaptic loss, and Aß plaque accumulation, in a way dependent on the interaction of DISC1 with LC3. These results indicate that DISC1 is a novel mitophagy receptor, which protects synaptic plasticity from Aß accumulation-induced toxicity through promoting mitophagy.

The clinical value of MR diffusion tensor imaging (DTI) in grade diagnosis of gliomas was investigated. A total of 31 patients with glioma were administered 3.0T MR convention and DTI examination, with quantitative measurement of anisotropy coefficient fractional anisotropy (FA) and apparent dispersion coefficient (ADC) value, and the comparison of quantitative parameters of glioma between low- and high-grade, which was detected by Mann-Whitney U test. The receiver operation characteristic (ROC) curve was drawn to take the value of ADC and FA in tumor ROI as a critical point, to calculate the area under the curve and to confirm the diagnosis threshold value and evaluate its diagnostic efficiency. The FA value of 14 low-grade glioma cases was 139.4±81.3, with an ADC value of (1.36±0.21) ×10-3 mm2/sec. The FA value of 17 high-grade glioma cases was 103.1±41.5, with ADC value of (1.09±0.28)-3 mm2/sec; the difference between the two groups was statistically significant (P<0.05). The ADC value was taken as the critical point to judge tumor grade and draw the ROC curve; the area under the curve was 0.79. As the diagnosis threshold value, the ADC value of 1.11×10-3 mm2/sec was used to distinguish between low- and high-grade tumor with a sensitivity of 58.8% and specificity of 92.9%. The FA value was taken as a critical point to judge tumor grade and draw the ROC curve; the area under the curve was 0.62. As the diagnosis threshold value, the FA value of 178.9 was applied to distinguish between low- and high-grade tumor sensitivity of 94.1% and specificity of 35.7%. Therefore, the FA value and ADC value in DTI has an important estimated value for the pathological grade of glioma.

OBJECTIVE: To explore the value of the self-decompression bone block in interbody fusion. METHODS: From April 2014 to May 2015, 42 patients with degenerative lumbar instability and spinal stenosis were treated by posterior vertebral lamina decompression and pedicle nail-rod fixation and unilateral modified transforaminal lumbar interbody fusion, including 18 males and 24 females. The treatment group had 24 cases with autologous pure decompression bone block as single interbody fusion material and the control group had 18 cases with cage and autologous bone as interbody fusion material. Clinical data, bone healing time, interbody fusion rate, intervertebral height and curative effect were analyzed in two groups. RESULTS: All the patients were followed up for 12 to 24 months with an average of 16 months. There was no significant difference in age, sex ratio, degree of lumbar instability, or follow-up time between two groups(P>0.05); and there was no significant difference in curative effect, intervertebral height loss, or interbody fusion rate between two groups(P>0.05). CONCLUSIONS: Using self-decompression bone block fusion can get high fusion rate, maintain good intervertebral height, obtain satisfactory curative effect. Its design was scientific and reasonable with less complication, which provide an effective, economic, and practical method for degenerative lumbar instability and spinal stenosis.

The morphological parameters, root wad indexes and site conditions of 127 uprooting trees from 76 plots (20 mx20 m) in Lesser Khingan coniferous-broad leaved Korean pine mixed forest were measured. Then the influencing factors of uprooting differences and the relationship between uprooting trees and disturbed soil were analyzed. Results showed that the number of uprooting trees varied significantly among species. Abies nephrolepis suffered the most serious uprooting damage, then Pinus koraiensis, and Ulmus spp. the least. Deciduous species had a stronger uprooting-resistant capacity than broad-leaved species. With the increase of tree DBH and height, tree' s uprooting resistance declined rapidly first and then was gradually enhanced, and finally reached the minimum at diameter class of 20 cm and height class of 14 m, respectively. The smaller the taper degree and projected area of crown were, the stronger the uprooting resistance was. Uprooting rate was negatively correlated with stand density. Trees lying in wet ground, flat terrain, medium low altitude area and windward slope had a greater risk of uprooting. There were significant positive correlation between the depth, area and volume of disturbed soil and the DBH, height, volume of uprooting trees.

A field experiment was conducted to study the effects of planting pattern (ridge culture, flatten culture, furrow culture) and film mulching on the distribution of spring maize root system and their influence on the yield of spring maize in the hilly area of central Sichuan basin. The results showed that ridge and film mulching had great influence on root morphology and root distribution of maize. The root length, root surface area and root volume of film mulching was 42.3%, 50.0%, 57.4% higher than those of no film mulching at jointing stage. The film mulching significantly increased the dry mass of root in vertical and horizontal distribution, and increased the root allocation ratio in deeper soil layer (20-40 cm) and the allocation ratio of wide row (0-20 cm) in horizontal direction. The effects of planting pattern on root growth and root distribution differed by film mulching. With film mulching, the ridge culture significantly increased the root dry mass in each soil layer and enlarged the distribution percentage of wide row (20-40 cm) in horizontal direction, as well as the dry mass of root in horizontal distribution and the root allocation ratio of wide row. The root mass under film mulching was in the order of ridge culture>flatten culture>furrow culture. Without film mulching, the furrow culture significantly increased root dry mass of narrow row (0-40 cm), and the root mass under no film mulching was in the order of furrow culture > ridge culture >flatten culture. As for the spike characteristics and maize yield, the filming mulching mea-sures reduced the corn bald length while increased the spike length, grain number, 1000-grain mass and yield. The yield under film mulching was in the order of ridge culture>flatten culture> furrow culture, while it was furrow culture > flatten culture > ridge culture under no film mulching. The reason for yield increase under ridge culture with film mulching was that it increased root weight especially in deep soil, and promoted the root allocation ratio in deeper soil and wide row (20-40 cm) in horizontal direction. The ridge-furrow culture without film mulching was helpful to root growth and increased the maize yield.

A new LOS2 gene was cloned from C. bursa-pastoris by rapid amplification of cDNA ends (RACE). The full-length cDNA of C. bursa-pastoris LOS2 gene (designated as Cblos2) was 1694 bp containing a 1332 bp open reading frame (ORF) encoding a 444 amino acid protein. The predicted CbLOS2 protein contained enolase-N domain, enolase domain, conserved putative DNA-binding and repression domains like LOS2 from A. thaliana. Bioinformatic analysis indicated that CbLOS2 had similarity with other enolase proteins. Cold acclimation assay revealed that Cblos2 expressed constitutively in C. bursa-pastoris and was involved in the cold acclimation process, implying CbLOS2 was a bi-functional enolase.

Brucellosis is an infectious disease affecting humans and animals worldwide. Effective methods of control include inducing immunity in animals by vaccination and elimination. Brucella abortus S19 is one of the popular vaccines for control of cattle brucellosis, as it has low virulence. In this paper, allelic exchange plasmids of wzm and wzt genes were constructed and partially knocked out to evaluate the effects on the induction of immunity to Brucella abortus S19 mutants. Cytokine secretion in vitro, INF-γ induction in vivo and antibody dynamics were evaluated. These data suggested that the immunity-eliciting ability of the wzm and wzt gene deletion mutants was similar, although reduced compared with the S19 strain. The results demonstrated that the wzt gene may be more important in the regulation of the induction of immunity than the wzm gene.

Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the important pathogens causing serious economic losses to swine industry worldwide. PRRSV is genetically and pathologically heterogenous. PRRSV NT0801 strain was isolated in a pig farm with clinical signs and had high pathogenesis in piglets. But its NSP2 gene did not have 30 amino acids deletion as highly pathogenic JXA1 strain. To elucidate the genetic characteristics of PRRSV NT0801 strain, the full-length genome of NT0801 isolate was sequenced and analyzed. The results showed that the genome of PRRSV NT0801 was 15439bp in length, including 29nt Poly(A) tail. Compared with the highly pathogenic JXA1 strain, it had the nucleotide sequence identity of 96.7%, amino acid sequence homology of 97.2% and 98.5% in GP3 and GP5, respectively. Phylogenetic analysis indicated that NT0801 isolate was located between the traditional strain and the highly pathogenic strain. But no obvious recombination signal was observed, compared with other PRRSV isolates with different virulence. The alignment of amino acid sequence of NT0801 with other PRRSV isolates demonstrated that three out of nine sites, being consistent with the highly pathogenic strain, were different from those in highly pathogenic while same as those in traditional strains and JXA1 vaccine strain. And one out of 9 sites was same as that of JXA1 vaccine strain exclusively, two out of 9 sites were different from all the strains. These results indicated that PRRSV NT0801 strain is closely related to highly pathogenic PRRSV, although there has no 30 amino acids deletions in NSP2 region. The epidemic PRRSV strains variation results from the gene mutation. It should be useful for studying on the virulence genes located in different ORFs of PRRSV in the future.
