OBJECTIVE: To determine the minimum effective concentration (MEC) of an imbibing and soluble nonoxynol-9 (N-9) diaphragm (ISND) required for immobilisation of all spermatozoa in vitro and in vivo. The speed of semen absorbance, time of ISND to dissolution, and the antifertility effects were also investigated in rabbits. METHODS: In vitro spermicidal tests with ISND were conducted using fresh semen from humans and rabbits. Spermicidal and antifertility effects were observed in vivo after the ISND was placed directly into the vagina of rabbits. RESULTS: The MEC of N-9 required in the ISND to totally immobilise sperm within 20 seconds was 0.15 mg/ml for human sperm, and 0.5 mg/ml for rabbit sperm. The human semen was absorbed into the ISND in 45 minutes; the diaphragm dissolved in the vagina 3.5 hours later. In vivo, in rabbits, the MEC of N-9 required to immobilise sperm within five minutes of mating was 1 mg/kg in the ISND, and 10 mg/kg for the nonoxynol-9 film. The median effective dose of N-9 in the ISND was 1.07 mg/kg, whereas for the film it was 3.30 mg/kg. CONCLUSION: The spermicidal and antifertility activities of a low dose N-9 in the ISND were high, with properties of imbibition and solubility confirmed.

Objective:To explore the association between sibling violence experience and emotional and behavioral problems.Methods:A Multistage stratified cluster sampling was used in Hunan province, 1735 participants from multiple-child family were recruited to survey the prevalence of sibling violence and the occurrence of emotional and behavioral problems with the Strength and Difficulties Questionnaires (SDQ).Results:According to SDQ total score, the rate of abnormal emotion and behavior in children and adolescents involved in sibling violence was higher than that in students who were not experienced sibling violence [45.6% (236/518) vs.26.8% (326/1217) ], the rate in bully-victim was higher than that in pure-victim and pure-bully [52.8% (132/250) vs.37.9% (74/195) vs.41.1% (30/73) ].The rates in children and adolescents who were bullied and perpetrated more than 4 sub-types of sibling violence were 63.4% (53/93) and 76.9% (60/78).Conclusion:There is an association between sibling violence experience and emotional and behavioral problems among children and adolescents in rural areas.

ObjectiveTo compare children's resilience and peer relationship in divorced and complete family,and explore the moderating effect of psychological resilience between family structure and peer relationship.Methods208 children from divorced family and 465 children from complete family were surveyed.Results(1)There were significant differences in children's resilience(divorced family (3.58±0.73),complete family(3.83±0.59),t=-2.765,P=0.006) and peer relationship(divorced family (3.47±0.72),complete family(3.73±0.59),t=-2.069,P=0.042) between complete and divorced family.(2) Family structure had remarkable negative correlation with resilience and peer relationship(r1=-0.19,r2=-0.20),and resilience had positive correlation with peer relationship(r=0.43).(3)The moderating effect of resilience between family structure and peer relationship was significant(β=0.641,P<0.05).ConclusionParent divorce has negative influence on children's resilience and peer relationship.Furthermore,the relationship of family structure and peer relationship is mediated by resilience.The negative influence of parent divorce on peer relationship is not significant among children with high resilience,while it is significant on children with low resilience.

Objective:To determine the prevalence of dyslipidemia in students from the primary and middle schools and the influential factors,and to provide evidence for the prevention and control of dyslipidemia and relevant chronic diseases in primary and middle schools.Methods:A total of 2 028 students aged 7 to 17 from the primary and middle schools in Changsha were selected by stratified random cluster sampling.The contents of the study included questionnaire survey,physical measurement and blood tests for fasting blood triglyceride (TG),total cholesterol (TC),low-density lipoprotein cholesterol (LDL-C),and high-density lipoprotein cholesterol (HDL-C) levels.Non-high-density lipoprotein cholesterol (non-HDL-C) level was calculated by TC level minus HDL-C level.Chi-square test and non-conditional logistic regression model were used to analyze the factors that contributed to dyslipidemia.Results:The dyslipidemia rate was 18.6%,and the abnormal rates of TG,TC,LDL-C,non-HDL-C and HDL-C were 6.8%,5.6%,2.3%,4.2% and 8.6%,respectively.By chi-square test,the dyslipidemia rate in students with different ages,home locations,BMI groups,central obesity,time on watching TV or playing computer per day,and daily sleep time was statistically significant.Non-conditional logistic regression analysis showed that home location for the city (OR=1.332),overweight (OR=1.548),obesity (OR=2.201),central obesity (OR=1.695),watching TV or playing computer for more than 2 hours per day (OR=1.357),daily sleep time longer than 11 hours (OR=2.518) were the risk factors for dyslipidemia in students from the primary and middle schools.Conclusion:Nearly 1/5 primary and middle school students show dyslipidemia,which is associated with obesity and other bad behaviors.

Objective To explore the related factors of anemia during pregnancy and provide scientific evidence for the primary prevention of anemia during pregnancy.Methods The pregnant women (≤ 12 pregnant weeks) who received the first pregnancy care in a local medical institution in Hunan province from June 2013 to November 2014 were included in this cohort study,and for them anemia had been excluded by physical examination.Baseline survey and follow up till childbirth were conducted for them.A queue-based nested case-control study (1 ∶ 2) was conducted (380 pregnant women with anemia detected in this study as case group,760 pregnant women without anemia randomly selected and matched by age,habitual residence during pregnancy as control group.And t test,x2 test and logistic regression analysis were conducted to identify related factors of anemia during pregnancy.Results Multivariate logistic regression analysis indicated that low family annual income level (net income) (OR=2.08,95%CI:1.22-3.59),low educational level (OR=2.09,95%CI:1.22-3.59),pre-pregnancy perm/hair dye (OR=2.23,95% CI:1.63-3.05),early pregnancy vomiting (OR=2.51,95% CI:1.56-4.03) were the risk factors for anemia during pregnancy.Intake of vitamin and trace element supplements (OR=0.69,95% CI:0.50-0.94),frequent meat,fish,shrimp,egg intakes (OR=0.68,95%CI:0.49-0.92),frequent soy milk,milk intakes (OR=0.51,95%CI:0.27-0.95) were the protective factors for anemia during pregnancy.Conclusion A number of factors,such as family annual income level,education level,poisonous and harmful material contact,pregnancy reaction,nutrition,are related to the incidence of anemia during pregnancy,it is necessary to take preventive measures to reduce the incidence of anemia during pregnancy.

<p><b>OBJECTIVE</b>To explore genetic mutation and clinical treatment for a patient with globozoospermia.</p><p><b>METHODS</b>Histomorphology of the sperms was studied by Wright-Giemsa staining and transmission electron microscopy. Potential mutation of the DPY19L2 gene was detected by PCR amplification and Sanger sequencing.</p><p><b>RESULTS</b>Wright-Giemsa staining showed that all spermatozoa from the patient were round-headed and lacked the acrosome, with the nuclei of sperm head stained in dark and full. Transmission electron microscopy revealed large round sperm heads, with an even layer of unit membrane surrounding the nuclei and dispersed cytoplasmic vacuoles but no acrosomal structure. The patient has harbored a homozygous deletion of the DPY19L2 gene. With intracytoplasmic sperm injection (ICSI) treatment, fertilization rate of the oocytes has reached 28.6%, which resulted in a successful pregnancy. A healthy male was born.</p><p><b>CONCLUSION</b>The homozygous deletion of DPY19L2 probably underlies the globozoospermia in this case, for which ICSI has provided an effective treatment. However, there is still a risk of low oocyte fertilization rate or fertilization failure. Further studies are required.</p>

Objective To investigate the effect of silencing of ataxia-telangiectasia mutated (ATM) expression by plasmid-mediated RNA interference on the radiosensitivity of human lung adenocarcinoma A549 cells.Methods Eukaryotic expression plasmid containing ATM small interfering RNA (siRNA) (pSilencer2.1-ATM),as well as pSilencer2.1-nonspecific,was constructed.Lung adenocarcinoma A549 cells were divided into positive group,negative group,and control group to be transfected with pSilencer2.1-ATM,pSilencer2.1-nonspecific,and no plasmid,respectively.The mRNA and protein expression of ATM was measured by RT-PCR and Western blot,respectively.The change in cell radiosensitivity was observed by colony-forming assay.Cell cycle and cell apoptosis were analyzed by flow cytometry.Results The eukaryotic expression plasmid containing ATM siRNA was successfully constructed.The RT-PCR and Western blot demonstrated that the expression of ATM was down-regulated in the positive group.The sensitization enhancement ratios (D0 ratios) for the positive group and negative group were 1.50 and 1.01,respectively.The flow cytometry revealed that the proportions of A549 cells in G1 and G2/M phases were significantly lower in the positive group than in the control group (51.27% vs 61.85%,P =0.012;6.34% vs 10.91%,P =0.008) and that the apoptosis rate was significantly higher in the positive group than in the control group and negative group (49.31% vs 13.58%,P=0.000;49.31% vs 13.17%,P=0.000).Conclusions Silencing of ATM expression may increase the radiosensitivity of human lung adenocarcinoma A549 cells,probably by affecting the cell cycle and promoting cell apoptosis.

Objective To investiGate the efficacy of three-dimensional conformal radiotherapy( 3-DCRT)combined with concurrent S-l for locally advanced esophaGus carcinoma in the elderly. Methods Fifty-four elderly patients with locally advanced esophaGus carcinoma were randomly divided into test Group(27 cases treated with 3-DCRT combined with S-l)and control Group(27 cases treated with 3-DCRT alone). DiaGnosis in all cases was confirmed by patholoGy. Results The complete remission rate and overall efficiency rate in test Group were 62. 96% and 85. l9% respectively,and 33. 33% and 59. 26% in control Group(χ2 =4. 75,P=0. 029;χ2 =4. 52,P=0. 033). All 23 patients with dysphaGia in test Group were in remission,and l6 of 22 patients with dysphaGia in control Group were in remission,and there was statistical siGnificant difference between two Groups(χ2 =7. 24,P=0. 007). The one-and two-year survival rates in test Group were 74. l% and 54. 8%respectively,and 55. 6% and 36. l% in control Group(χ2 =3. 9l,P=0. 047;χ2 =3. 98,P=0. 046). Toxicities of Grade 0-Ⅱ in both Groups were similar,and there was no statistical siGnificant difference(χ2 =l. 08,P=0. 299). Conclusion The complete remission rate,overall efficiency rate and one-and two-year survival rates of 3-DCRT combined with concurrent S-l were siGnificant hiGher than 3-DCRT alone in the treatment for the elderly patients with locally advanced esophaGus carcinoma. And ti’s adverse effects were well tolerated.

Objective To investigate the relationship between the effect of unmethylated cytosine phosphate-guanine oligodeoxynucleotide ( CpG ODN ) 7909 on X-ray-induced G2/M phase arrest and apoptosis and the phosphorylation of ATM kinase.Methods Human lung adenocarcinoma A549 cells were randomly classified into five groups,control,CpG group,irradiation group,CpG + irradiation group and ATM siRNA + CpG + irradiation group.Cell survival fraction was evaluated by clonogenic assay.Cell cycle and apoptosis were analyzed by flow cytometry.The expressions of ATM,checkpoint kinase-2(Chk2) and p53 were detected with Western blot.Results Compared to the situation under X-ray irradiation alone,decreased cell clonogenic survival,prolonged G2/M arrest,and increased cell apoptosis were observed after the combination treatment with CpG ODN7909 and X-rays ( t =13.41,17.32 and 7.71,P < 0.05).Moreover,the phosphorylations on ATM,Chk2,and p53 were increased in the irradiated A549 cells that had been pre-treated with CpG ODN7909.After ATM siRNA interfering,abrogation of G2/M arrest,reduction of apoptosis,and decrease of Chk2 and p53 phosphorylation were found in A549 cells treated with CpG ODN7909 and X-rays ( t =26.84,2.98,47.24 and 67.47,P <0.05).Conclusions CpG ODN7909 can enhance the X-ray-induced phosphorylation of ATM kinase in human lung adenocarcinoma cells in vitro,which might be involved in regulating G2/M phase arrest and apoptosis.

CD4 + CD25 + regulatory T cells(Treg) are thought to be a subgroup of mature CD4 + T cells.Forkhead winged helix transcription factor-3 (Foxp3)is specifically expressed on them and plays a key role in their development and function. CD4 + CD25 + Treg cells can maintain the stabilization of internal environment by two principal pathways to suppress the immunological function: the direct suppression of the target cells by cell-contact and the secretion of suppressor cytokines.At present,it has been considered that decreased number and dysfunction of CD4+ CD25+ Treg cells are closely related to pathogenesis of autoimmune disease. Recent findings show that CD4+ CD25+ Treg cells play an important role in pathogenesis of idiopathic thrombocytopenic purpura.
