In the context of the rapid rise of the village revitalization in China, the study examines the impact of grassroots autonomous organizations embedding level and farmer non-agriculture employment on the farmer homestead exit behavior. Based on 421 household surveys in Qionglai, this study obtained three results through empirical analysis. (1) The system embedding level of grassroots autonomous organization and the interests embedding level of grassroots autonomous organization have a significant positive impact on the farmer homestead withdrawal behavior, but the emotion embedding level of grassroots autonomous organizations has a significant negative impact on the farmer homestead withdrawal behavior. (2) The farmer non-agriculture employment level is beneficial for the farmer homestead exit behavior. (3) Further mechanism analysis shows that the non-agricultural employment level plays an inhibitory role in the positive influence of grassroots organizations to mobilize farmers to exit homestead. Therefore, it is suggested that when mobilizing farmers to voluntarily withdraw from the homestead, grassroots organizations should actively improve their embedding level, improve the construction of farmers' social network, and accelerate the accumulation of farmers' material capital.

<p><b>OBJECTIVE</b>To analyze whether Vitapan Classical shade guide could be used to evaluate effectively the effect of tooth whitening.</p><p><b>METHODS</b>A total of 67 patients underwent Beyond cold light tooth whitening between February 2009 and July 2014. The effect of treatment was evaluated by Vitapan Classical shade guide. The percentage of discolored teeth that could not be matched by the aforementioned shade guide was calculated. By contrast, the color difference between discolored teeth and corresponding shade tabs was calculated for discolored teeth that could be matched by the shade guide.</p><p><b>RESULTS</b>Approximately 64% (16/25) of tetracycline teeth and 28% (17/61) of mottled-enamel teeth could not be matched using the Vitapan Classical shade guide, but yellow teeth were all matched, and the difference between any pair of teeth was statistically significant (P=0.000). For discolored teeth that could be matched, statistically significant difference was found in the ΔL*, Δb*, and ΔE between tetracycline and yellow teeth, as well as between tetracycline and mottled-enamel teeth. However, no statistically significant difference was found between mottled-enamel and yellow teeth. Moreover, no statistically significant difference was found in the Δa* among the three groups.</p><p><b>CONCLUSION</b>Vitapan Classical shade guide could not be used to evaluate effectively the effect of tooth whitening, especially for tetracycline and mottled-enamel teeth. The evaluating method should be improved.</p>

<p><b>OBJECTIVE</b>To investigate a method of designing an accurate and scientific shade guide, especially used for judging the effect of tooth whitening, by analyzing the colorimetric values of discolored teeth statistically.</p><p><b>METHODS</b>One hundred thirty-six pictures of patients who had been receiving the Beyond cold light whitening treatment from February 2009 to July 2014 were analyzed, including 25 tetracycline teeth, 61 mottled-enamel teeth, and 50 yellow teeth. The colorimetric values of discolored teeth were measured. The L* values of shade tabs were calculated by hierarchical clustering of those of discolored teeth. The a* and b* values of shade tabs were the mean of those observed for discolored teeth. Accordingly, different shade guides were designed for each type of discolored teeth, and the effects were evaluated.</p><p><b>RESULTS</b>A statistically significant difference in colorimetric values was found among the three types of discolored teeth. Compared with the Vitapan Classical shade guide, the shade guides designed through the present method were more scientific and accurate in judging the effect of tooth whitening. Moreover, the arrangement of shade tabs was more logical, and the color difference between shade tabs and discolored teeth was smaller.</p><p><b>CONCLUSION</b>The proposed designing method is theoretically feasible, although its clinical effect has yet to be proven.</p>

<p><b>OBJECTIVE</b>To investigate the predictive value of metastatic lymph node ratio for postoperative distant metastasis in patients with colorectal cancer.</p><p><b>METHODS</b>The clinicopathological data were collected from 180 patients with colorectal cancer who underwent surgical resection in General Hospital of PLA between from January, 2007 to January, 2012. The patients were divided into 2 groups according to the presence of distant organ metastasis and the clinicopathological factors were analyzed with Chi-square test and logistic regression.</p><p><b>RESULTS</b>Of the 118 surgical patients enrolled, 118 were free of distant metastasis and 62 had distant metastasis involving the liver (39 cases), lungs (12 cases), and multiple organs (11 cases). The gross types, differentiation, ELN and LNR of the tumors differed significantly between the two groups. Logistic regression analysis showed that LNR was an independent factor correlating to distant metastasis of colorectal cancer.</p><p><b>CONCLUSION</b>LNR is independently correlated with distant organ metastasis of colorectal cancer and serves as an important predicative factor for estimating the prognosis of colorectal cancer.</p>

<p><b>OBJECTIVE</b>To evaluate the impact of preoperative platelet-lymphocyte ratio (PLR) on the prognosis in patients with operable colorectal cancer.</p><p><b>METHODS</b>The clinical data of 140 patients with operable colorectal cancer were analyzed retrospectively. According to preoperative PLR, the patients were divided into low PLR group (PLR<250) and high PLR group (PLR≥250). The overall 5-year survival rates and the clinicopathological factors in the two groups were analyzed using univariate analysis and COX regression model.</p><p><b>RESULTS</b>The patients with a high PLR had a significantly lower overall 5-year survival rate than those with a low PLR (71.4% vs 51.5%, P=0.045). Univariate analyses identified tumor location, the depth of tumor invasion, lymph nodes metastasis, TMN stage, and PLR as significant risk factors for reduced overall 5-year survival. In COX regression model, a high PLR was identified as an independent risk factor for poorer prognosis in patients with resectable colorectal cancer (RR: 2.213; 95%CI: 1.007-4.863, P=0.0048).</p><p><b>CONCLUSION</b>Preoperative PLR can be a clinically significant factor for assessment of prognosis of resectable colorectal cancer.</p>

The population of the very elderly is growing steeply with prolonged life expectancy.Age is an independent risk factor for stroke.Naturally,the very elderly become the high risky population.Hypertension is the most common and modifiable risk factor of stoke in the very elderly.Results of studies conducted during these 20 years have been reported to explore and investigate the relationship between stroke primary prevention and antihypertensive treatment,some of which are regarded as landmarks trials,providing essential evidences for blood pressure management in the very elderly.

<p><b>OBJECTIVE</b>To establish a nude mouse model of orthotopic engineered gastric tumor for in vivo fluorescence imaging studies.</p><p><b>METHODS</b>An engineered gastric tumor was constructed in vitro using collagen as the scaffold and the human gastric cancer cell line BGC823-EGFP cells expressing enhanced green fluorescence protein (EGFP) as the seed cells. The engineered tumor was then implanted into the stomach of nude mice, and the tumor growth was observed with in vivo fluorescence imaging. The nude mice were sacrificed 6 weeks after the transplantation to assess the tumor growth and metastasis, and the tumor histology was evaluated.</p><p><b>RESULTS</b>The tumor cells in the engineered tumor model grew well in three-dimensional culture. The success rate of orthotopic gastric tumor implantation was 100% (10/10) in nude mice with metastasis in the abdominal organs. The isolated tumor mass, weighing 1.719∓0.349 g, showed a histological characteristic of poorly differentiated adenocarcinoma. In vivo fluorescence imaging detected EGFP-expressing tumors in the abdominal cavity of the nude mice, but not accurately.</p><p><b>CONCLUSION</b>The nude mouse model bearing orthotopic engineered gastric tumor provides a simple animal model for the study of gastric cancer, but a stronger fluorescence than green fluorescence is more desirable for more effective observation in in vivo fluorescence imaging.</p>

ObjectiveTo investigate the clinical characteristics,3-month outcome and predictive factors in the very elderly patients with ischemic stroke.MethodsA total of 305 acute ischemic patients aged 65 years and over were enrolled in the study.They were divided into two subgroups by age:80 years old and over(n =78),65-79 years old(n =227).The clinical outcome was assessed by the modified Rankin Scale (mRS) on (90 ± 7 ) days after stroke,and categorized as good ( scoring 0-2 ) or poor( scoring 3-6) outcome.Results Significantly lower BMI [ ( 23.62 ± 4.92 ) kg/m2 vs ( 25.08 ± 3.69 ) kg/m2,P =0.005],lower frequency of dyslipidemia(56.41% vs 71.13%,P =0.006) and alcohol intake (0% vs 6.61%,P =0.043 )were found in the very elderly group.The rates of poor functional outcome in the ≥ 80years group and the 65-79 years old group were 56.41% (44/76) and 41.40% (94/224) respectively,with a P value of 0.015.Multivariate logistic regression analysis showed that higher National Institute of Health stroke scale (NItISS) total score (OR 1.48,95% CI 1.19-1.83 ) and lower albumin level (OR 0.73,95 % CI 0.55 -0.95 ) were associated with poor outcome in ≥ 80 year old,whereas higher NIHSS total score ( OR 1.38,95% CI 1.24-1.53) and complications during hospital stay ( OR 2.58,95% CI 1.07-6.19 ) were predictive factors in the 65-79 years old group.ConclusionOur study suggests that NIHSS scores,albumin level and complications during hospitalization are useful predictive factors for the short-term poor functional outcome in the patients of ≥65 years old and ≥80 years old patients have a worse prognosis.

Mounting evidence has demonstrated that CD4(+) T cells play an important role in anti-tumor immune responses. Thus, adoptive transfer of these cells may have great potential for anti-cancer therapy. However, due to the difficulty to generate sufficient tumor-specific CD4(+) T cells, the use of CD4(+) T cells in tumor therapy is limited. It has been found that IL-15 transfection enhances the proliferation and anti-tumor activity of tumor-specific CD8(+) T cells, but the effect of IL-15 transfection on CD4(+) T cells remains unknown. Here, the effects of retrovirus-mediated IL-15 expression in Ova-specific CD4(+) T cells from Do11.10 mice were evaluated and it was discovered that IL-15 transfected CD4(+) T cells expressed both soluble and membrane-bound IL-15. Retrovirus-mediated IL-15 expression led to a selective expansion of antigen-specific CD4(+) T cells by inhibiting their apoptosis. In vivo IL-15 transfected CD4(+) T cells were more effective in suppressing tumor growth than control retroviral vector transfected ones. To ensure the safety of the method, the employment of thymidine kinase gene made it possible to eliminate these transgenic CD4(+) T cells following ganciclovir treatment. Together, we show that IL-15 transfection induced a selective expansion of antigen-specific CD4(+) T cells ex vivo and enhanced their tumor-suppression effects in vivo. This has an important significance for improving the efficacy of adoptive T cell therapy.

RESUMEN

Objective To explore the preparation of specific immune RMA(iRNA) on anti-rabies and further study immunotherapy of rabies virus exposure. Methods Horses were immunized with the rabi-es virus and their livers were isolated from the horse of antiserum, from which total RNA was extracted and purified by sodium lauryisulfonate, phenol, chloroform, ethyiene glycol monomethyl ether, cetyltrimethyam-moniumbromide and alcohol. Results Pure preparation physico-chemical characterization was analyzed, and it's weight was 0.15% of weight of liver. The RNA contained 2.86% DNA and 1.16% protein. The iRNA with a maximum UV absorbance at 258 nm and A_(258/280) about 2.0. The test of RNA was positive, which had a relative molecular mas of 13.7×10~3 by high performance liquid chromatography(HPLC), and its hy-perchromic effect was 50.67%. The vesults of biological activity was showed that the rate of leucocyte adher-ence inhibition(LAI) was 41.73%, The protective rate was 50% and prolonging the life was 31.62%. Conclusion The results obtained with the practical value were identical and provide a basis on medicines of anti-rabies.

Objective To identify the changes of mesenteric lymph fluid in cases of uncontrolled hemor-rhagic shock (UHS), which is most often occurred in modem trauma and battle injury. The lymph of gut plays an important role during shock and MODS. The objective of this experiment is to identify the change of mesenteric lymph fluid in UHS. Method Thirty-eight SD rats were canulated with PE 50 catheters separately into right carotid artery,left femoral artery and femoral vein. A serf-made steel catheter was inserted into main mesenteric lymph duct and the mesenteric lymph fluid was collected. The rots were divided into three groups: control group (n=10), controlled hemorrhagic shock (CHS) group (n=10) and UHS group (n=10). After the mean blood pressure was dropped to 40 mmHg by exsanguination, hemorrhage was made by amputation of 75% tail in length and let the stump keep open to bleed. The volume of mesenteric lymph fluid was recorded, and the protein and endotoxin in it were examined, too. Data were analyzed using ANOVA and Chi-square test. Results The mesenteric lymph fluid was decreasing remarkably during shock until it was below 1/4 of normal. After hemorrhage was controlled, the mesentefic lymph fluid increased slowly. However, it kept decreasing persistently in cases of UHS. So did the protein content. The eudotoxin in lymph fluid increased 2 hours after CHS and one hour after UHS. There was significant difference in endotoxin between CHS and UHS 2 hours after shock (P<0.05). Con-dusions The volume and protein content of mesenteric lymph fluid decreased mare significantly in UHS than that in CHS. And contrarily, the endotoxin in lymph fluid increased more significantly in UHS than that in CHS. The change of the mesenteric lymph fluid may be an important cause of MODS after UHS.

BACKGROUND: Neonatal rat liver cells are moderately differentiated cells with the characterization and function of both liver progenitors and hepatocytes, thus, it is an ideal cell source for the study of the hepatocyte characterization. OBJECTIVE: To explore the isolation and in vitro culture of neonatal rat liver cells.DESIGN, TIME AND SETTING: An in vitro cytology trial was carried out at the Institute of General Surgery, General Hospital of Chinese PLA from March to August 2008.MATERIALS: Neonatal SD rats with 3 months old were used, irrespective of genders.METHODS: Liver cells from neonatal rat were isolated by tissue piece-cold trypsin digestion combining with multi-step low centrifuge, and cultured onto the plate in HepatoZYME-SFM supplemented with 10% fetal serum. The growth and function of the cultured liver cells was evaluated by contrast microscopy, MTT assay, PAS staining and urine enzyme test. MAIN OUTCOME MEASURES: Cell morphology and viability, content of glycogen, as well as urea level in the supernatant.RESULTS: Totally 1.0×10~6-2×10~6 cells per whole liver could be obtained with viability above 90%. The cells displayed round or orbicular-ovate shapes with big nuclei, and cell body was smaller than mature cells. More than 95% purity achieved after removal erythrocyte, nonparenchymal cells and dead cells with multi-step low-speed centrifugalization. The viability of cells were gradually increased at the beginning of culture, noticeably alleviated at the 3 days, and reached a peak at the 11 days, and then gradually decreased. The difference between day 1 and days 11, as well as days 15 and days 11 had significance (P=0). Liver cells cultured in HepatoZYME-SFM attached and kept hepatocyte-specific morphology. PAS staining showed the cultured liver cells at day 7 were strongly positive and then the positive cells decreased gradually, until 15 days, only few positive cells could be seen. Urea level in the supernatant remained stable at the initial time and dramatically decreased after 7-day culture. CONCLUSION: The tissue piece-cold trypsin digestion and HepatoZYME-SFM is a simple and efficient isolation and culture system for neonatal rat liver cells.

Obiective To set up a method of scaffold evaluation using human cell line as seed cells and screen appropriate scaffold for live tissue engineering, Methods HepG2 cells were plated onto biodegradable polymer scaffolds: PLGA, 3% chitosan (3%CS) and 2% silk fibroin (2%SF), and cultured in vitro. The growth, distribution and function of HepG2 cells in the scaffolds were evaluated using MTT assay, H.E. staining, and urea assay kit. Results HepG2 cells plated on the three scaffolds maintained a proliferative state. In contrast, the cells on the 2%SF proliferated strongly, while the cells on the PLGA and chitin proliferated poorly. Histological examination showed that HepG2 cells distributed evenly on the 2%SF scaffold with a high amount, while few cells could be found on the PLGA and ehitin at day 7. Cell function assay showed that HepG2 cells on the 2%SF and PLGA exhibited slower decrease of urea synthesis compared to HepG2 cells on the chitosan. Conclusion The three scaffolds have good biocompatibility. In contrast, 2%SF scaffold is more appropriate for liver tissue engineering. This method may be used for scale screening of scaffolds for liver tissue engineering.

RESUMEN

Objective To study the early resuscitation with intravenous administration of liquor in uncontrolled hemorrhagic shock model by four-staged abdominal aorta injury in rats in order to settle the controversy over the issue of volume resuscitation.Method Forty Sprague-Dawley rats were canulated with catheters into the left ventricle,femoral artery and femoral vein,respectively.Animals were subjected to vaseular injury of infrarenal pierced with a 25G needle leading to uncontrolled hemorrhagic shock.After aorta injury,the models were processed into 4 stages:early injured stage,early rescue stage,delayed rescue stage and observation stage.The rescue with rapid infusion of 45 ml/kg lactated Ringer's solution in 15 minutes in the early stage and the followed rescue in the late stage;and(4)sham-rescued.The MAP,CVP,lactic acid and Hct were monitored.The volume of blood loss and the survival time of rats were recorded too.Results After aorta injury,The MAP dropped from 94.3 mmHg to 25.8 mmHg and rised quickly after early resuscitation with rapid infusion of liquor and then again fell soon,and CVP did likewise.Lactic acid was increased and reached 3-4 times of baseline at 4 h in all group except Sham-resuced group.The Hct levels decreased quickly after resuscitation with liquor in early rescue stage and delayed rescue stage.The blood loss in abdomen increased after rapid infusion of lactated Ringer's solution in early rescue stage(blood loss increased from 22.8 ml/kg to 27.7ml/kg,P<0.05).There Was no significant difference in mortality between those groups.Conclusions Rapid volume resuscitation at early stage of abdominal aorta injured with uncontrolled hemorrhagic shock cannot increase the circulatory blood volume for maintaining stable blood pressure norminimize the elevation of blood lactic acid.On the contrary,resuscitation with rapid administration of liquor at early stage can enhance blood loss and decrease the level of Hct.The survival time and survival rate can't be increased by early rescue with rapid liquor infusion.

Objective Protein chip for mucin detection was prepared to detect the serum levels of mucin1(MUC1),mucin2(MUC2)and micin5ac(MUC5AC)of colorectal cancer patients,and to discuss the relationship between colorectal cancer and the expression of MUC1,MUC2 and MUC5AC in serum.Methods The protein chip was prepared with monoclone and polyclone antibodies for the measuement of the serum levels of MUC1,MUC2 and MUC5AC in 30 patients with colorectal cancer and 30 healthy adults as control,using the method of immuno-fluorescence.Results The serum levels of mucin and CEA in colorectal cancer group were higher than that in control group.The differences were significant between the two groups concerning MUC1(5.25?2.37 vs 3.65?1.82U/ml,P0.05).The positive expression of mucin was related with the cancer differentiation.The positive expression of MUC1 and MUC5AC was higher when cancer differentiation was low as compared with high and median differentiation,but the positive expression of MUC2 was higher in high and middle cancer differentiation.When the serum level of mucin was used as an indicator for the diagnosis of colorectal cancer,the sensibility and specificity were both higher.The sensibility and specificity of MUC1 were 82.1% and 78.1%,respectively,while of combined mucins(at least 2 in 3 mucins were positive)were 96.2% and 85.3%,respectively.Conclusion The application of protein chip for mucin detection on the measurement of serum levels of MUC1,MUC2 and MUC5AC is of higher sensibility and specificity in the diagnosis of colorectal cancer.The establishment of protein chip has provided a new approach to early diagnosis of colorectal cancer,and help estimate the prognosis.

Objective To establish an abdominal-trauma-induced uncontrolled hemorrhagic shock (UHS) model, and explore the effect of staged resuscitation. Methods Indwelling catheters were inserted into left ventricle, femoral artery and femoral vein of 50 Sprague-Dawley rats, and then laparotomy was performed. 42 out of the 50 animals were subjected to vascular injury to simulate UHS by infra-renal arterial puncture with 25G needles, and the model was successfully established in 32 rats, for which the animals were divided into 3 groups: control group (n=10), with no resuscitation; hospital-resuscitation group (n=11), resuscitated in early injured stage; and staged-resuscitation group (n=11), resuscitated pre-hospital and then in-hospital. 8 out of the 50 rats, which were not subjected to vascular injury, served as sham-operated group. The mean aortic pressure (MAP), central venous pressure (CVP), serum lactic acid, alanine aminotransferase (ALT) and creatinine (Cr) were determined, and the volume of blood loss and survival time were recorded. Results The MAP decreased from 94.7mmHg to 28.9mmHg 15min after aortic injury in control group and rose slowly after continuous transfusion in staged-resuscitation group. Similar change was found in CVP. Serum lactic acid, ALT and Cr were increased in control group and hospital-resuscitation group at early injured stage, but the increase was delayed in staged-resuscitation group. The mean survival time in control group, hospital-resuscitation group and staged-resuscitation group were 76, 846 and 2480 min, respectively. The 72 h mortalities of the three groups were 100%, 72.7% and 36.4%, respectively. Conclusion Staged resuscitation which is characterized by slowly continuous transfusion at early aid stage cannot increase the blood pressure and circulating blood volume to normal level in UHS, but can reduce the level of serum lactic acid, ALT and Cr, and dose not increase the blood loss. The survival rate and mean survival time can be increased by this method.

RESUMEN

Objective To explore the effect of adjunct intraperitoneal resuscitation (IR) on bioactivity of mesenteric lymph in hemorrhagic shock (HS) rats. Method The rat model of HS was reproduced by bleeding the rats from femoral artery. The rats were resuscitated by conventional resuscitation (CR) or CR plus adjunct IR 60 minutes after HS or sham shock (SS).The rats were then divided into six groups as follows:SS group (n=4),SS+CR group (n=4),SS+CR+IR group (n=4),HS group (n=4),HS+CR group (n=5) and HS+CR+IR group (n=5). The mean arterial pressure (MAP) of rats in HS group was maintained at 40mmHg without resuscitation. The rats in SS group underwent only artery cannulation without blood letting for pressure monitoring. The rats were resuscitated with Ringer's lactate (80ml/kg) in CR. When treated with IR,each rat was given peritoneal injection of 20ml peritoneal dialysis solution after CR. Two hours after resuscitation,mesenteric lymph was collected by laparotomy for the measurement of neutrophil respiratory burst activity after being incubated with lymph. At the end of experiment,rats were sacrificed,the intestinal tissues were collected and the ileum mucosal injury was evaluated by light microscopy. Results The bioactivity of lymph of HS group was significantly higher than that of SS group. RESUMEN

To investigate the role of endothelial cell derived cytokines in sepsis induced liver and lung injury. Sepsis was induced in mice by cecal ligation and puncture(CLP). Sham operation group underwent the same manipulation but without CLP. The gene expressions of pro inflammatory cytokines (TNF?, IL 1? and IL 6) in pulmonary and hepatic microvascular endothelial cells were assessed by RT PCR. The results showed that TNF? and IL 1? mRNA levels markedly increased at 3h after CLP in both pulmonary and hepatic microvascular endothelial cells. Though at 12h after CLP both of them declined, the levels were still higher than those in sham group. IL 6 mRNA expression also increased significantly at 3h after CLP in both pulmonary and hepatic microvascular endothelial cells. At 12h after CLP, the expression of IL 6 mRNA in hepatic microvascular endothelial cells remained at a high level, while the level in pulmonary microvascular endothelial cells became higher. It suggested that endothelium is an important source of pro inflammatory cytokines in the liver and lung tissues during sepsis.

Objective To investigate the role of cytokines genes expression of liver sinusoidal endothelial cells in liver damage by sepsis.Methods Septic mice models were established with cecal ligation and perforation (CLP), while sham operation group received the same procedure exclusive CLP. The genes expression of TNF?, IL 1? and IL 6 in liver sinusoidal endothelial cells were assessed by RT PCR. Results A significant increase of TNF?, IL 1? genes expression was observed at 3h, and a slight decline at 12h after operation, but still significantly higher than that in the sham group; while IL 6 gene expression showed signficantly higher at 3h and remained at the high level at 12h. Conclusions Liver sinusoidal endothelial cell is an important source of cytokine production in mice with sepsis.
