RESUMEN

We present the integration of a natural protein into electronic and optoelectronic devices by using silk fibroin as a thin film dielectric in an organic thin film field-effect transistor (OFET) ad an organic light emitting transistor device (OLET) structures. Both n- (perylene) and p-type (thiophene) silk-based OFETs are demonstrated. The measured electrical characteristics are in agreement with high-efficiency standard organic transistors, namely charge mobility of the order of 10(-2) cm(2)/Vs and on/off ratio of 10(4). The silk-based optolectronic element is an advanced unipolar n-type OLET that yields a light emission of 100nW.

Droplets microfluidics is broadening the range of Lab on a Chip solutions that, however, still suffer from the lack of an adequate level of integration of optical detection and sensors. In fact, droplets are currently monitored by imaging techniques, mostly limited by a time-consuming data post-processing and big data storage. This work aims to overcome this weakness, presenting a fully integrated opto-microfluidic platform able to detect, label and characterize droplets without the need for imaging techniques. It consists of optical waveguides arranged in a Mach Zehnder's configuration and a microfluidic circuit both coupled in the same substrate. As a proof of concept, the work demonstrates the performances of this opto-microfluidic platform in performing a complete and simultaneous sequence labelling and identification of each single droplet, in terms of its optical properties, as well as velocity and lengths. Since the sensor is realized in lithium niobate crystals, which is also highly resistant to chemical attack and biocompatible, the future addition of multifunctional stages into the same substrate can be easily envisioned, extending the range of applicability of the final device.

We have designed an original approach for efficient Second Harmonic Generation of tailored V-shape benzo[b]thiophene molecular systems enabling versatile and flexible one-step, dry and technologically friendly thin film processing. The designed moieties show chi((2)) values at least as high as the reference LiNbO(3) single crystal, without poling processing and matching the constrains of integrated optical configuration for nonlinear optical devices. This may open the way to a new class of organic materials exploitable for photonic applications.

The technological quest for flexible devices to be interfaced with the biological world has driven the recent reinvention of bioderived polymers as multifunctional active and passive constituent elements for electronic and photonic devices to use in the biomedical field. Keratin is one of the most important structural proteins in nature to be used as biomaterial platform in view of the recently reported advances in the extraction and processing from hair and wool fibers. In this article we report for the first time the simultaneous use of naturally extracted keratin as both active ionic electrolyte for water ions sensing and as bendable and insoluble substrate into the same multielectrode array-based device. We implemented the multifunctional system exclusively made by keratin as a bendable sensor for monitoring the humidity flow. The enhancement of the functional and structural properties of keratin such as bendability and insolubility were obtained by unprecedented selective chemical doping. The mechanisms at the basis of the sensing of humidity in the device were investigated by cyclic voltammetry and rationalized by reversible binding and extraction of water ions from the volume of the keratin active layer, while the figures of merit of the biopolymer such as the ionic conductivity and relaxation time were determined by means of electrical impedance and dielectric relaxation spectroscopy. A reliable linear correlation between the controlled-humidity level and the amperometric output signal together with the assessment on measure variance are demonstrated. Collectively, the fine-tuned ionic-electrical characterization and the validation in controlled conditions of the free-standing insoluble all-keratin made microelectrode array ionic sensor pave the way for the effective use of keratin biopolymer in wearable or edible electronics where conformability, reliability and biocompatibility are key-enabling features.

Salmonelose é uma doença causada por bactérias do gênero Salmonella, com importância para saúde pública e animal. Dentre os sorotipos hospedeiro-específicos, destaca-se o Gallinarum, que possui os biovares Gallinarum e Pullorum adaptados às aves e amplamente difundidos pelo mundo. Os dados sobre a ocorrência de Salmonella spp. em criações avícolas alternativas no Brasil são escassos. O objetivo deste estudo foi pesquisar a ocorrência de Salmonella spp. em galinhas coloniais encaminhadas para necropsia ao LRD/FV/UFPel. Foram realizadas análises histopatológicas, microbiológicas e moleculares das colônias bacterianas isoladas de 12 amostras de órgãos de galinhas domésticas dos municípios de Pelotas e Piratini, no Rio Grande do Sul. Na análise microbiológica, foram isoladas bactérias do gênero Salmonella sorotipo Gallinarum das 12 amostras, sendo 10/12 bioquimicamente compatíveis com biovar Gallinarum e 2/12 com biovar Pullorum. Na análise molecular PCR 11/12, 91,7% foram identificadas genotipicamente como Salmonella spp. O presente estudo demonstrou uma elevada frequência de isolamento de Salmonella Gallinarum biovar Gallinarum em aves sintomáticas criadas em regime extensivo. Além disso, os dados epidemiológicos das aves analisadas demonstram que a infecção por Salmonella Gallinarum nesses casos está associada ao contato com aves silvestres e falhas de manejo sanitário.(AU)

Members of the caspase family of cysteine proteases are known to be key mediators of mammalian inflammation and apoptosis. To better understand the catalytic properties of these enzymes, and to facilitate the identification of selective inhibitors, we have systematically purified and biochemically characterized ten homologues of human origin (caspases 1 - 10). The method used for production of most of these enzymes involves folding of active enzymes from their constituent subunits which are expressed separately in E. coli, followed by ion exchange chromatography. In cases where it was not possible to use this method (caspase-6 and -10), the enzymes were instead expressed as soluble proteins in E. coli, and partially purified by ion exchange chromatography. Based on the optimal tetrapeptide recognition motif for each enzyme, substrates with the general structure Ac-XEXD-AMC were used to develop continuous fluorometric assays. In some cases, enzymes with virtually identical tetrapeptide specificities have kcat/Km values for fluorogenic substrates that differ by more than 1000-fold. Using these assays, we have investigated the effects of a variety of environmental factors (e.g. pH, NaCl, Ca2+) on the activities of these enzymes. Some of these variables have a profound effect on the rate of catalysis, a finding that may have important biological implications.

In this study we describe the epidemiology, clinical signs, and pathology of an outbreak of avian aspergillosis in alternative breeding in the southern region of Rio Grande do Sul, Brazil. Between the fifth and tenth day of life, 360 chicks from a flock of 4000 developed unspecific clinical signs and died. The birds were housed in a reused aviary litter, without previous treatment. In 11 six-day-old female ISA Brown chicks (Gallus gallus domesticus), necropsy revealed firm, yellowish-white, multinodular lesions extending from the pleura to the lung parenchyma. Histologically, a granulomatous, multifocal to coalescent pneumonia was observed. Granulomas were characterized by central necrosis, with heterophil and epithelioid macrophage infiltration and presence of countless Y-shaped intralesional septate hyphae morphologically compatible with Aspergillus spp. The diagnosis through isolation confirmed Aspergillus fumigatus. We highlight the importance of aspergillosis as a primary cause of diseases in the respiratory tract of young birds in alternative breeding. Measures to prevent aspergillosis mainly regarding the reuse of aviary litter are essential in poultry husbandry to prevent economic losses, reduce environmental contamination and mitigate the potential risk to public health.(AU)


Descrevem-se os aspectos epidemiológicos e patológicos de um surto de aspergilose aviária em criação alternativa na região sul do Rio Grande do Sul, Brasil. De um lote de 4000 pintainhas, entre o quinto e o 10º dia de vida, 360 aves apresentaram sinais clínicos inespecíficos e morreram. As aves foram alojadas em cama reutilizada do aviário, sem tratamento prévio. Na necropsia de 11 pintainhas (Gallus gallus domesticus), fêmeas, seis dias de idade da linhagem Isa Brown, foram observadas no pulmão lesões multinodulares, branco-amareladas e firmes, que se estendiam da pleura ao parênquima. Histologicamente foi observada pneumonia granulomatosa, multifocal a coalescente. Os granulomas eram caracterizados por necrose central, com infiltrado inflamatório de heterófilos, macrófagos, células epitelioides com presença de inúmeras hifas septadas intralesionais, semelhantes à letra "Y", morfologicamente compatíveis com Aspergillus spp. O diagnóstico foi confirmado pelo isolamento de Aspergillus fumigatus. Alerta-se para a importância da aspergilose como causa primária de afecções no trato respiratório de aves jovens em criações alternativas. Medidas preventivas relacionadas ao manejo dessas aves são indispensáveis principalmente quanto à reutilização da cama dos aviários, a fim de evitar perdas econômicas, reduzir a contaminação ambiental e o potencial risco à saúde pública.(AU)

Meningiomas são os principais tumores primários do sistema nervoso central (SNC) que afetam cães e gatos. Na maioria dos casos, são neoplasias benignas, geralmente expansivas, causando compressão do SNC, e raramente fazem metástase para outros órgãos. O presente trabalho tem como objetivo relatar a ocorrência de um meningioma microcístico com metástase pulmonar em um canino de 11 anos de idade, com sinais clínicos de andar cambaleante, compressão da cabeça contra objetos, agitação, salivação e agressividade. Na necropsia, foram observadas, no encéfalo, massas bem delimitadas pardo-avermelhadas, firmes, de aspecto granular, localizadas no córtex parietal e nos núcleos da base. Inúmeras micronodulações de aspecto semelhante foram observadas no pulmão. Histologicamente observaram-se nódulos formados por células neoplásicas fusiformes, com núcleos grandes e alongados e nucléolos evidentes, dispostas de forma frouxa, formando vacúolos e microcistos. À imuno-histoquímica, o meningioma apresentou marcação fortemente positiva para citoqueratina e negativa para vimentina. Por meio da histopatologia e da imuno-histoquímica, foi possível estabelecer a classificação histológica de meningioma microcístico, bem como diferenciá-lo de outras doenças que cursam com sinais nervosos.(AU)


Meningiomas are the main tumors of the central nervous system (CNS) affecting dogs and cats. In most of the cases they are benign neoplasms, usually expansive, causing compression of the CNS and rarely metastasize to other organs. We describe the occurrence of a microcystic meningioma with pulmonary metastasis in an 11 - year - old canine with clinical signs of staggering gait, head compression against objects, agitation, salivation and aggressiveness. At necropsy, well-defined, firm, granular-looking masses located in the parietal cortex and nuclei of the base were observed in the encephalon. Numerous micronodulations of similar appearance were observed in the lung. Histologically, nodules formed by spindle neoplastic cells with large, elongated nuclei and evident nuclei were loosely arranged, forming vacuoles and microcysts. Immunohistochemistry were strongly positive for cytokeratin and negative for vimentin. Through the histopathology and immunohistochemistry, it was possible to establish the histological classification of microcystic meningioma, as well as to differentiate from other diseases that present with nervous signals.(AU)

O objetivo deste estudo foi identificar as principais doenças de felinos na região sul do Rio Grande do Sul. Foram revisados os protocolos de necropsia e das amostras biológicas de felinos encaminhados ao Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas (LRD/UFPel), no período de 1978 a 2018. Nesse período foram recebidas 1633 amostras de felinos, sendo 363 (22%) entre os anos de 1978 e 1999 e 1270 (78%) entre os anos de 2000 e 2018. Com relação aos diagnósticos, 457 felinos (28%) apresentaram tumores benignos ou malignos, sendo os tegumentares e os mamários os mais frequentes. As doenças bacterianas, fúngicas, virais, parasitárias, sem agente definido e as intoxicações totalizaram 554 casos (33,9%), destacando-se a esporotricose, com 12,8% dos diagnósticos. Concluiu-se que, na região sul do RS, o encaminhamento de felinos para diagnóstico aumentou significativamente após o ano 2000, comprovando que a espécie passou a ter maior importância como animal de companhia. Concluiu-se, também, que as neoplasias têm papel relevante entre as doenças de felinos e que a esporotricose é uma das mais importantes zoonoses na região.(AU)


The goal of this paper was to identify the main disease affecting felines in the southern region of Rio Grande do Sul. The necropsy protocols and feline biological materials submitted to the Regional Diagnostic Laboratory of the Veterinary College of the Federal University of Pelotas (LRD / UFPel) were reviewed, from 1978 to 2018. During this period 1633 feline samples were received, 363 (22%) between 1978 and 1999 and 1270 (78%) between 2000 and 2018. 59% of felines did not present a defined breed. As for diagnoses, 457 felines (28%) presented benign or malignant tumors, the most common being the integumentary and mammary tumors. Bacterial, fungal, viral, parasitic or undefined agent infections and intoxications were observed in 554 cases (33.9%), especially sporotrichosis with 12.8 % of the diagnoses. It was concluded that in southern RS the referral of cats for diagnosis increased significantly after the year 2000, proving that they became more significant as companion animals. It was also concluded that neoplasia play a relevant role among feline diseases, and that sporotrichosis is one of the most important zoonoses in the region.(AU)

Human leukotriene C4 synthase specific activity in the human monocytic leukemia cell line THP-1 (0.302 +/- 0.062 nmol LTC4 formed.min-1 x mg-1) was 7.6-fold higher than in U937 cells (0.040 +/- 0.017 nmol LTC4 formed.min-1 x mg-1) and comparable to dimethylsulfoxide-differentiated U937 cells (0.399 +/- 0.084 nmol LTC4 formed.min-1 x mg-1). Using the photoaffinity probe, azido[125I]-LTC4, a single polypeptide with a molecular mass of 18 kDa was specifically labelled in THP-1 microsomal membranes. The rank order of potencies for competition of azido[125I]-LTC4 photolabelling of the 18 kDa protein by glutathione, leukotrienes and their analogs was found to be LTC2 > (azido[127I]-LTC4 approximately LTC4) > (LTD4 approximately LTE4) > (LTA4 approximately LTB4) > S-hexyl glutathione > glutathione, corresponded with the rank order of potencies for inhibition of LTC4 synthase activity but not inhibition of microsomal glutathione S-transferase activity. The 18 kDa protein specifically labelled by azido[125I]-LTC4 had high specificity for LTC4 and closely related leukotrienes and was separable from microsomal glutathione S-transferase. We conclude that azido[125I]-LTC4 specifically photolabels LTC4 synthase which is an 18 kDa polypeptide or contains an 18 kDa subunit.

Based on LTD4 receptor antagonist activity of 3-(2-quinolinyl-(E)-ethenyl)pyridine (2) found in broad screening, structure-activity studies were carried out which led to the identification of 3-[[[3-[2-(7-chloro-2-quinolinyl)-(E)-ethenyl]phenyl][[3- (dimethylamino)-3-oxopropyl]thio]methyl]thio]propionic acid (1, MK-571) as a potent and orally active LTD4 receptor antagonist. These studies demonstrated that a phenyl ring could replace the pyridine in 2 without loss of activity, that 7-halogen substitution in the quinoline group was optimal for binding, that the (E)-ethenyl linkage was optimal, that binding was enhanced by incorporation of a polar acidic group or groups in the 3-position of the aryl ring, and that two acidic groups could be incorporated via a dithioacetal formed from thiopropionic acid and the corresponding styrylquinoline 3-aldehyde to yield compounds such as 20 (IC50 = 3 nM vs [3H]LTD4 binding to the guinea pig lung membrane). It was found that one of the acidic groups could be transformed into a variety of the amides without loss of potency and that the dimethylamide 1 embodied the optimal properties of intrinsic potency (IC50 = 0.8 nM on guinea pig lung LTD4 receptor) and oral in vivo potency in the guinea pig, hyperreactive rat, and squirrel monkey. The evolution of 2 to 1 involves the increase of > 6000-fold in competition for [3H]LTD4 binding to guinea pig lung membrane and a > 40-fold increase in oral activity as measured by inhibition of antigen-induced dyspnea in hyperreactive rats.

Tardive dyskinesia (TD) is the most feared and troublesome extrapyramidal side-effect of prolonged neuroleptic (NL) treatment. We present a review of TD. Its pathophysiology remains elusive, although extrapyramidal symptoms (EPS) increase the liability for TD. Nowadays, therefore, avoidance of all EPS remains the best preventive strategy, as it is not possible to predict which liable patients will develop TD, or of what type or severity. TD frequently includes dystonic features, and is more disabling when these dystonias are present. Clozapine (CLZ) has been reported to be effective in suppressing nearly 60% of TD syndromes, specially those with dystonic features. Based on the few reports in the literature on CLZ and TD by the early 1980s, we started to videotape the first severe TD patient treated with CLZ in 1984. We present the first three case reports of severe TD, with prominent disabling dystonic features, treated with CLZ and videotaped since pretreatment and then periodically for 12, 8 and 5 years of follow-up, respectively. The patients' current diagnosis, gender and age are: Case 1, DSM-IV Schizophrenia Residual Type, male, 39 years; Case 2, DSM-IV Polysubstance Related Disorder, Borderline Personality Disorder, female, 28 years; Case 3, DSM-IV Schizoaffective Disorder, male, 40 years. Two of them presented with a recurrence of TD because of CLZ interruption within the first 2 months of treatment, with no further breakthrough to date. The first two cases have complete remission of TD; the third case is still improving after 5 years of CLZ treatment, with only minor dystonic features persisting that constitute no impairment for work or daily routines at present. All patients, independent of their psychiatric primary diagnosis, have shown significant and progressive improvement in both motor and psychosocial aspects. None of them has been rehospitalized. Long-term treatment and follow-up is required to avoid TD recurrence and to assure full assessment of treatment effectiveness. Ideally, periodic video recording with standardized examination is advisable for long-term follow-up and outcome assessment. At present, CLZ could be regarded as the drug of choice for patients with TD, specially for those with disabling and or dystonic features and who require ongoing NL therapy. The use of novel antipsychotic agents for TD treatment and prevention, with their low EPS liability, is promising, but has yet to be tested.

For the determination of antimony by hydride generation techniques a pretreatment procedure has been developed for the reduction of Sb(V) to Sb(III) in order to remove the effect of hydrofluoric acid which strongly interferes with the reduction of pentavalent antimony to the trivalent state. It is based on the combined action of l-cysteine and boric acid at 80 degrees C. The pretreatment is effective in both nitric and hydrochloric acid media. Quantitative recoveries are obtained in less than 60 min. Under these conditions antimony is reduced to the trivalent state in acid media containing both nitric and hydrochloric acid. The method has been applied to the determination of total antimony in certified reference materials of sediments after pressurized microwave digestion with HNO(3)-HCl-HF. Good agreement is obtained by using both analytical techniques: continuous flow hydride generation atomic fluorescence spectrometry and flow injection electrothermal atomic absorption spectrometry with in-situ trapping of stibine in a graphite atomizer.

At the request of the Geriatric Residences Management Committees, the Prevention and Safety at the Workplace Service of Mantua Hospital coordinated the practical application of Chapter V of Law 626/94 with special reference to patient handling. Assessment of exposure and clinical identification of disorders and impairment of the lumbar spine carried out using the methods proposed by the EPM Research Centre involved 15 residences and a total of 31 departments analyzed and 435 workers submitted to health surveillance by the respective certified occupational physicians. With the exception of one department, the exposure levels were found to be medium-high (MAPO Index > 1.51) considering the number of disabled patients, the significant lack of lifting equipment and the lack of training for this risk factor. Acute low back pain reported in the previous 12 months revealed a prevalence of 10%, more than 4 times that of non-exposed workers. The prevalence of functional spondylarthropathies also showed high values (16%), as also the number of degenerative diseases of the lumbosacral spine (16%). 11% of the males and 17% of the females were judged unfit for patient handling, which created problems of job reallocation. By concentrating the results at the Prevention and Safety at the Workplace Service it was possible to plan interventions for improvement, using as referents for the various residences a working group where the presence of a qualified physiotherapist proved to be particularly useful. Coordination of the certified occupational physicians also improved the quality of health surveillance.
