RESUMEN

The activity for ATP-ase was investigated in cells of rabbit and guinea pig epidermis and human oral mucosa. Observations both in the light- and electron microscope indicate that the ATP-ase positive cells of guinea pig and human epithelia are Langerhans cells while in the rabbit epidermis the reacting cells have the features of keratinocytes. A calcium containing mixture of paraformaldehyde and glutaraldehyde is recommended as the fixative of choice for demonstration of ATP-ase activity in Langerhans cells.

RESUMEN

Four of the five kinds of cells constituting the walls of thyroid cysts can be identified in the SEM. These are cuboidal cells, mucous cells, cells with large granules and ciliated cells. A correlation between SEM and TEM observations is attempted.

RESUMEN

The ultimobranchial cysts in the thyroid of the adult guinea pig are identified by a cytochemical method. The ultimobranchial tissue has the shape of an irregular cyst, with non-specific esterase activity that is resistent to HgCl2 inhibition. The TEM reveals five different types of epithelial cell in the cyst wall: 1. cells with deeply invaginated nuclei and of varying shape, from flat to cylindrical but most of them cuboidal. 2. Mucous cells that are tall and similar to goblet cells. 3. Tall cells with big granules that have a dense core. 4. Ciliated cells similar to those in the respiratory tract. 5. C-cells crowded with the small dense granules typical for the cell type. The cyst is enveloped by rather coarse collagen fibers and many wide sinusoids.

RESUMEN

After 1 hours of aldehyde fixation, 4 to 20 hours of soaking in 2% BSA (bovine serum albumin) solution and another 17 hours in the same fixative, the vibratome will produce smooth, even sections from agar embedded guinea pig thyroid, skeletal muscle or larynx with the same ease as it will from soft, homogeneous tissues such as liver or spleen.

RESUMEN

A mitochondrial pellet, prepared from rat skeletal muscle, contained a number of carboxylic ester hydrolase isoenzymes. The esterases which split alpha-naphthyl acetate were organophosphate sensitive, whereas two out of three indoxyl acetate hydrolysing enzymes were resistant to both organophosphate and organomercury. The activity of the indoxyl acetate esterases was enhanced by the non-ionic detergents Tween-40 and Lubrol. After freezing, thawing and high speed centrifugation most of the alpha-naphthyl acetate splitting enzymes were found in the supernatant, indicating that the enzymes are loosely bound to mitochondrial membranes.
