RESUMEN

Melanized focal changes (MFCs) in the fillet of farmed Atlantic salmon is a major quality concern. The changes are thought to initially appear as acute red focal changes (RFCs) that progress into chronic MFCs. Recent findings have indicated that hypoxia may be important in their development, possibly leading to necrosis affecting not only myocytes but also adipocytes. Thus, the aim of this study was to investigate possible hypoxic conditions in RFCs and the subsequent inflammatory responses and lesions in the adipose tissue in RFCs and MFCs. A collection of RFCs, MFCs and control muscle samples from several groups of farmed salmon was studied. Using immunohistochemistry, we found induction of the hypoxia-inducible factor 1 pathway in RFCs. Histological investigations of RFCs and MFCs revealed different stages of fat necrosis, including necrotic adipocytes, a myospherulosis-like reaction and the formation of pseudocystic spaces. Accumulations of foamy macrophages were detected in MFCs, indicating degradation and phagocytosis of lipids. Using in situ hybridization, we showed the presence of tyrosinase- and tyrosinase-related protein-1-expressing amelanotic cells in RFCs, which in turn became melanized in MFCs. In conclusion, we propose a sequence of events leading to the formation of MFCs, highlighting the pivotal role of adiposity, hypoxia and fat necrosis.

Road accidents, mostly on national highways, pose a significant public health and economic burden in Bangladesh, requiring in-depth analysis for road safety measures. This study comprehensively analyzes accident trends, characteristics, causes, and consequences by identifying the accident black spots on the Kushtia-Jhenaidah National Highway (N704). Accident records from 2017 to 2021 were collected from nearby police stations. Additionally, using a cluster random sampling approach, a questionnaire survey with 100 respondents (50% drivers and 50% general road users) was also conducted to capture diverse perceptions and behaviors. The study utilizes descriptive methods, such as trends analysis and frequency distributions, alongside spatial analysis techniques, including severity index, Kernel Density Estimation, and hotspot analysis. Findings indicate a decrease in accidents from 2018 to 2021, yet a concerning rise in fatalities in 2021. Trucks (47.9%) emerge as the primary contributor among 169 vehicles involved in accidents. Head-on collisions (36%) are prevalent, attributed to both human and environmental factors, including driver inexperience (56%), mobile phone use while driving (78%), lack of proper training (12%), overspeeding (28.3%), and nighttime driving (54%) influenced by seasons and land use. Mostly, victims aged from 20 to 40, where men are more affected by fatalities (70.7%) and women by injuries (86.3%). Out of 35 identified accident spots, including Battail, Bittipara Bazar, Laxmipur Bazar, Modhupur Bazar, IU Main Gate, Sheikhpara Bazar, and DM College Front, are designated as blackspot zones based on the frequency of accidents, deaths, and injuries. The study concludes by recommending targeted interventions, driver training, infrastructure improvements, regulatory measures, and victim assistance in collaboration with local and national agencies to enhance road safety and mitigate accident risks.

Identifying black spots effectively and accurately is a pivotal and challenging task to improve road traffic safety. A novel black spot identification model is proposed by integrating the GIS-based processing with hierarchical density-based spatial clustering of applications with noise. Additionally, the optimal clustering parameters are determined based on an internal validation indicator called the density-based clustering validation index to minimize the impact of subjectivity in parameter selection. The model is validated by collecting 3536 accident data from 1 August to 31 October 2020 in Hangzhou, China, and eventually identifies 39 black spots. The results show that: (1) The number of accidents contained in black spots account for 75% of all accidents, while the length of network in the black spots only account for 23.26% of the total road network length. (2) Compared with the conventional density-based spatial clustering of applications with noise model and K-means model, the proposed model achieves the best performance with more accidents gathered per unit road length. (3) The sample survey with 6 onsite of the identified black spots indicates that the proposed model has high recognition accuracy and recommend these sites for further investigation.

In recently developed organic semiconductors, the continuously improving sample purity makes the stability of the chemical bonds of organic materials themselves become a key factor in device stability, which provides greater uncertainty for the generation of "zeroth defect", and the spatial resolution of performance at different positions becomes particularly important. In this work, complete maps of electroluminescent, photoluminescent, and Raman scattering in the same area on an organic light-emitting diode during its operation have been achieved with a confocal spectrometer with multiple laser sources. The different spectral characters help to establish different regions and suggest the mechanism of degradation. In particular, Raman scattering has been shown to be very sensitive in a multilayer device to a change in thickness of several nanometers. In amorphous films with few defects, the very weak film uniformity, including the thickness and degree of aggregation, would induce dramatic degradation. The relatively thin and/or loosely textured region easily locally overheats and has the highest probability of "zeroth defect" generation. This method has high spatial resolution, a low level of damage to samples, good reproducibility, and multiple interconnected pieces of information, which is significant for online quality prediction and mechanistic analysis.

Melanized focal changes in white skeletal muscle of farmed Atlantic salmon, "black spots", is a quality problem affecting on average 20% of slaughtered fish. The spots appear initially as "red spots" characterized by hemorrhages and acute inflammation and progress into black spots characterized by chronic inflammation and abundant pigmented cells. Piscine orthoreovirus 1 (PRV-1) was previously found to be associated with macrophages and melano-macrophages in red and black spots. Here we have addressed the inflammatory microenvironment of red and black spots by studying the polarization status of the macrophages and cell mediated immune responses in spots, in both PRV-1 infected and non-infected fish. Samples that had been collected at regular intervals through the seawater production phase in a commercial farm were analyzed by multiplex fluorescent in situ hybridization (FISH) and RT-qPCR methods. Detection of abundant inducible nitric oxide synthase (iNOS2) expressing M1-polarized macrophages in red spots demonstrated a pro-inflammatory microenvironment. There was an almost perfect co-localization with the iNOS2 expression and PRV-1 infection. Black spots, on the other side, had few iNOS2 expressing cells, but a relatively high number of arginase-2 expressing anti-inflammatory M2-polarized macrophages containing melanin. The numerous M2-polarized melano-macrophages in black spots indicate an ongoing healing phase. Co-localization of PRV-1 and cells expressing CD8+ and MHC-I suggests a targeted immune response taking place in the spots. Altogether, this study indicates that PRV-1 induces a pro-inflammatory environment that is important for the pathogenesis of the spots. We do not have indication that infection of PRV-1 is the initial causative agent of this condition.

Fatal hypothermia represents a diagnostic challenge in veterinary pathology. Wischnewski spots (WS) and black oesophagus (BO) have been described in human cases of fatal hypothermia but rarely in animals. We now describe WS and BO in suspected fatal hypothermia in a free-ranging brown howler monkey (Alouatta guariba clamitans) and a pet rabbit (Oryctolagus cuniculus). Both animals had dark spots resembling WS on the gastric mucosa and the monkey also had BO with haematin deposition. In both cases, stress factors and relatively cold environmental conditions were present prior to death.

INTRODUCTION: Currently, spatial and temporal distribution of safety resources in Iran is entirely based on expert opinions, regardless of network priorities. Considering the lack of resources for implementing safety treatments, prioritizing unsafe points is an important and complicated issue where the effectiveness of each safety treatment option should be thoroughly investigated. The political, social, and environmental aspects should also be taken into consideration, including social and political pressures and officials talks on less important topics. Obviously, this inappropriate resource allocation poses a serious challenge to the expected goals. In this study, a methodology based on economic and social issues is proposed to optimize the annual budget allocation for eliminating or reducing the risk of accident-prone points. In this methodology, the spatial and temporal distribution of budget is determined using a mathematical model aimed to maximize the benefits of reducing the accidents after deducting the costs of implementing the safety countermeasures. The outputs of this model include the safety countermeasure alternatives and a five-year time schedule for implementing them, or the alternative of no action with regard to budget, social, and judicial constraints. In order to evaluate the proposed method, it is applied to the road network of Iran and the results are compared with those of the conventional method that is currently used for resource allocation in this country. The results show that the proposed method leads to 15% higher benefits compared to the conventional method. Moreover, this method makes 641 safe points, which is about 17% more than the safe points resulted from the existing method. Therefore, the proposed method brings about a safer network as a result of the optimal allocation of available resources.

Objective The recently reported endoscopic finding of black spots is defined as black pigmentation in gastric mucosa. We attempted to clarify the relationship between the Helicobacter pylori infection status and black spot occurrence. Methods The study subjects were 1,600 individuals who underwent an annual medical checkup and whose H. pylori status could be determined. Upper endoscopic examinations were performed in all, and the presence of black spots in the stomach as well as the degree of gastric mucosal atrophy were determined. Results Among the 1,600 enrolled subjects, 784 underwent eradication for H. pylori, of whom 144 were originally H. pylori-positive and 672 H. pylori-negative. Black spots in the stomach were observed in 156 (9.8%). The rate of prevalence of black spots in the H. pylori-positive and H. pylori-negative subjects was 2.1% and 1.5%, respectively, while that in subjects after undergoing eradication of H. pylori was 18.2%. A multiple logistic regression analysis demonstrated that an older age and post-eradication status were significant factors for black spot occurrence, while proton pump inhibitor treatment showed a tendency to be a risk factor. In subjects with post-eradication status, a higher grade of gastric mucosal atrophy was a significant risk factor for the occurrence of black spots. Conclusion H. pylori post-eradication status and an older age were significant factors related to the appearance of black spots, and a higher grade of gastric mucosal atrophy was also a significant risk factor in subjects who had undergone successful eradication.

Neascus-type metacercariae, which cause the black spot disease, are described in specimens collected from 33 samples of Geophagus brasiliensis from the Peixe river, in Monte Verde, Juiz de Fora, state of Minas Gerais, Brazil, during March and May 2006. The main morphological features of these metacercariae are: genital primordia composed of three cell masses located in the hind body, and a median dorsal vessel of the reserve bladder around the acetabulum. Thirty fish specimens (90.9%) were parasitized by metacercariae, with mean infection intensity of 15.6 ± 16.1 (1-75). Also, a positive correlation was detected between G. brasiliensis total length and the parasite abundance (r = 0.459; p 0.01).


A metacercária de Neascus sp., causadora da doença dos pontos pretos, é descrita em espécimes de Geophagus brasiliensis coletados no Brasil. Entre março e maio de 2006, foram capturados 33 espécimes de G. brasiliensis, coletados com o auxílio de vara e anzol no Rio do Peixe, Monte Verde, município de Juiz de Fora, Estado de Minas Gerias. Dos 33 espécimes examinados, 30 (90,9%) apresentaram-se parasitados por metacercárias causadoras dos pontos pretos, com intensidade média de infecção de 15,6 ± 16,1 (1 75). Foi observada correlação significativa entre o comprimento total de G. brasiliensis e a abundância parasitária (r = 0,459; P 0,01). As principais características morfológicas desta metacercária foram: primórdio genital representado por três massas celulares, localizadas posteriormente na parte posterior do corpo e vaso mediano dorsal da bexiga de reserva se estendendo entorno do acetábulo.
