Hierarchical porous hypercrosslinked monoliths (PolyHIPE-HCP) with ultrahigh specific surface areas are prepared via a solvent knitting strategy. Compared to previous work, the solvent knitting strategy is carried out in a relatively low air-controlled atmosphere with gradient heating starting from low temperature while using DCM (Dichloromethane) as both a solvent and a cross-linker, allowing for a slow and controlled cross-linking process, thereby achieving a BET surface area ranging from 514 to 728 m2 g-1. Scanning electron microscopy (SEM) shows that the knitting process does not affect the presence of macroporous structure in the PolyHIPE. With the introduction of mesopores and micropores, these hierarchical porous monoliths exhibit significant potential for applications in gas adsorption and water treatment. Hence, a universal, simple and low-cost method to synthesize polymeric monoliths with hierarchically porous structure and higher surface area is proposed, which has fascinating prospects in industrialization.

This work proposes a rigorous mathematical model capable of reproducing the adsorption process in dynamic regime on advanced monoliths geometries. For this, four bed geometries with axisymmetric distribution of channels and similar solid mass were proposed. In each geometry a different distribution of channels was suggested, maintaining constant the bed dimensions of 15 cm high and 5 cm radius. The mathematical modeling includes mass and momentum transfer phenomena, and it was solved with the COMSOL Multiphysics software using mass transfer parameters published in the literature. The overall performance of the column was evaluated in terms of breakthrough (CA/CA0 = 0.1) and saturation times (CA/CA0 = 0.9). The mass and velocity distributions obtained from the proposed model show good physical consistency with what is expected in real systems. In addition, the model proved to be easy to solve given the short convergence times required (2-4 h). Modifications were made to the bed geometry to achieve a better use of the adsorbent material which reached up to 80%. The proposed bed geometries allow obtaining different mixing distributions, in such a way that inside the bed a thinning of the boundary layer is caused, thus reducing diffusive effects at the adsorbent solid-fluid interface, given dissipation rates of about 323 × 10-11 m2/s3. The bed geometry composed of intersecting rings deployed the best performance in terms of usage of the material adsorbent, and acceptable hydrodynamical behavior inside the channels (maximum fluid velocity = 35.4 × 10-5 m/s and drop pressure = 0.19 Pa). Based on these results, it was found that it is possible to reduce diffusional effects and delimit the mass transfer zone inside the monoliths, thus increasing the efficiency of adsorbent fixed beds.

The objective of this work was to develop an actinide-specific monolithic support in capillary designed to immobilize precise Pu:Am ratios and its coupling to inductively coupled plasma mass spectrometry (ICP-MS) for immobilized metal affinity chromatography applications. This format offers many advantages, such as reducing the sample amount and waste production, which are of prime importance when dealing with highly active radioelements. Four organic phosphorylated-based monoliths were synthesized in situ through UV photo-polymerization in capillary and characterized. The capillary coupling to ICP-MS was set up in conventional laboratory using Th and Sm as chemical analogues of Pu and Am. A dedicated method was developed to quantify online Th and Sm amounts immobilized on the monolithic capillaries, allowing to select the best monolith candidate poly(BMEP-co-EDMA)adp. By precisely adjusting the elemental composition in the loading solutions and applying the developed quantification method, the controlled immobilization of several Th:Sm molar ratios onto the monolith was successful. Finally, the capillary ICP-MS coupling was transposed in a glove box and by applying the strategy developed to design the monolithic support using Th and Sm, the immobilization of a 10.5 ± 0.2 (RSD = 2.3%, n = 3) Pu:Am molar ratio reflecting Pu ageing over 48 years was achieved in a controlled manner on poly(BMEP-co-EDMA)adp. Hence, the new affinity capillary monolithic support was validated, with only hundred nanograms or less of engaged radioelements and can be further exploited to precisely determine differential interactions of Pu and Am with targeted biomolecules in order to better anticipate the effect of Am on Pu biodistribution.

Haptoglobin (Hp) is a positive acute phase protein, synthesized in the liver, with four N-glycosylation sites carrying mainly complex type N-glycans. Its glycosylation is altered in different types of diseases but still has not been extensively studied mainly due to analytical challenges, especially the lack of a fast, efficient, and robust high-throughput Hp isolation procedure. Here, we describe the development of a high-throughput method for Hp enrichment from human plasma, based on monolithic chromatographic support in immunoaffinity mode and downstream Hp N-glycome analysis by hydrophilic interaction ultrahigh-performance liquid chromatography with fluorescent detection (HILIC-UHPLC-FLR). Chromatographic monolithic supports in a 96-well format enable fast, efficient, and robust Hp enrichment directly from diluted plasma samples. The N-glycome analysis demonstrated that a degree of Hp deglycosylation differs depending on the conditions used for N-glycan release and on the specific glycosylation site, with Asn 241 being the most resistant to deglycosylation under tested conditions. HILIC-UHPLC-FLR analysis enables robust quantification of 28 individual chromatographic peaks, in which N-glycan compositions were determined by UHPLC coupled to electrospray ionization quadrupole time of flight mass spectrometry. The developed analytical approach enables fast evaluation of total Hp N-glycosylation and is applicable in large-scale studies.

Polymer monoliths are promising materials for sample preparation due to their high porosity, pH stability, and simple preparation. The use of melamine formaldehyde foams has been reported as an effective support to prepare highly robust silica and polymer monoliths. Herein, divinylbenzene monoliths based on a 50:50 (%, w/w) crosslinker/porogen ratio have been nested within a melamine-formaldehyde sponge, resulting in monoliths with a surface area higher than 400 m2 /g. The extraction performance of these monoliths was evaluated for the extraction of endocrine-disrupting bisphenols from aqueous solutions. We evaluated for the first time the versatility of sponge-nested polymer monoliths by comparing three different extraction modes (vortex mixing, magnetic stirring, and orbital shaking). Vortex mixing showed a comparable recovery of bisphenols (39%-81%) in a shorter extraction time (30 min, instead of 2 h). In addition, the robustness of the sponge-nested polymer monoliths was demonstrated for the first time by reshaping a larger monolithic cube (0.125 cm3 ) into four smaller pieces (4 × 0.03125 cm3 ) leading to a 16%-21% increase in extraction efficiency. This effect was attributed to an increase in the effective contact area with the sample, obtaining a higher analyte extraction capacity.

In this study, an adaptable end-column platform was fitted to a commercially available monolith, which enabled the column to be fitted with a flow-splitting device. A variety of flow-splitting adapters could be incorporated into the platform, and in this study, a radial flow stream splitter was utilized. The advantage of the radial flow stream spitter was that it overcame issues relating bed density variations that could cause bands to distort in the radial cross-section of the column. Using propylbenzene as a test standard in isocratic elution mode, height equivalent to a theoretical plate curves were constructed across ten flow rates, and it was found that the column efficiency improved by as much as 73%. Furthermore, the dual outlet flow splitter enabled a very substantial reduction in column back pressure, with the decrease being consistently between 20 to 30% depending on the column length. Additionally, sensitivity increased by 45%, consistent with the observed increase in efficiency. The adaptable end-column platform could be retrofitted to almost any commercial column with the expectation of gaining efficiency, sensitivity, and reducing back pressure.

We characterized thermally polymerized organo-silica hybrid monolithic capillaries to test their applicability in the gradient elution of peptides. We have used a single-pot approach utilizing 3-(methacryloyloxy)propyltrimethoxysilane (MPTMS), ethylene dimethacrylate (EDMA), and n-octadecyl methacrylate (ODM) as functional monomers. The organo-silica monolith containing MPTMS and EDMA was compared with the stationary phase prepared by adding ODM to the original polymerization mixture. Column prepared using a three-monomer system provided a lower accessible volume of flow-through pores, a higher proportion of mesopores, and higher efficiency. We utilized isocratic and gradient elution data to predict peak widths in gradient elution. Both protocols provided comparable results and can be used for peptide peak width prediction. However, applying gradient elution data for peak width prediction seems simpler. Finally, we tested the effect of gradient time on achievable peak capacity in the gradient elution of peptides with a column prepared with a three-monomer system providing a higher peak capacity. However, the performance of hybrid organo-silica monolithic stationary phases in gradient elution of peptides must be improved compared to other monolithic stationary phases. The limiting factor is column efficiency in highly aqueous mobile phases, which needs to be focused on.

Novel chiral capillary electrochromatography (CEC) microsystems were constructed based on Aspergillus sp. CM96. As a newly discovered intrinsic characteristic of the cell, cell chirality occupies an essential position in life evolution. Aspergillus sp. CM96 spore (CM96s) was chosen as a proof of concept to develop chiral capillary columns. Interestingly, various types of amino acid (AA) enantiomers were baseline separated under the optimized conditions. Furthermore, the time-dependent chiral interactions between AAs and CM96s were explored in a wider space. Pectinases generated from Aspergillus sp. CM96 fermentation were immobilized onto graphene oxide-functionalized capillary silica monoliths for separating AA enantiomers. Molecular docking simulations were performed to explore chiral separation mechanisms of pectinase for AA enantiomers. These results indicated that Aspergillus sp. CM96-based CEC microsystems have a significant advantage for chiral separation.

Messenger RNA (mRNA) is becoming an increasingly important therapeutic modality due to its potential for fast development and platform production. New emerging RNA modalities, such as circular RNA, drive the need for the development of non-affinity purification approaches. Recently, the highly efficient chromatographic purification of mRNA was demonstrated with multimodal monolithic chromatography media (CIM® PrimaS), where efficient mRNA elution was achieved with an ascending pH gradient approach at pH 10.5. Here, we report that a newly developed chromatographic material enables the elution of mRNA at neutral pH and room temperature. This material demonstrates weak anion-exchanging properties and an isoelectric point of 5.3. It enables the baseline separation of mRNA (at least up to 10,000 nucleotides (nt) in size) from parental plasmid DNA (regardless of isoform composition) with both a NaCl gradient and ascending pH gradient approach, while mRNA elution is achieved in a pH range of 5-7. In addition, the basic structure of the novel material is a chromatographic monolith, enabling convection-assisted mass transfer of large RNA molecules to and from the active surface. This facilitates the elution of mRNA in 3-7 column volumes with more than 80% elution recovery and uncompromised integrity. This is demonstrated by the purification of a model mRNA (size 995 nt) from an in vitro transcription reaction mixture. The purified mRNA is stable for at least 34 days, stored in purified H2O at room temperature.

Aqueous zinc ion hybrid capacitors (aZHCs) are of great potential for large-scale energy storage and flexible wearable devices, of which the specific capacity and energy density need to be further enhanced for practical applications. Herein, a urea-mediated foaming strategy is reported for the efficient synthesis of monoliths consisting of nitrogen-enriched mesoporous carbon nanosheets (NPCNs) by prefoaming drying a solution made of polyvinylpyrrolidone, zinc nitrate, and urea at low temperatures, foaming and annealing at high temperatures, and subsequent acid etching. NPCNs have a large lateral size of ≈40 µm, thin thickness of ≈55 nm, abundant micropores and mesopores (≈3.8 nm), and a high N-doping value of 9.7 at.%. The NPCNs as the cathode in aZHCs provide abundant zinc storage sites involving both physical and chemical adsorption/desorption of Zn2+ ions, and deliver high specific capacities of 262 and 115 mAh g-1 at 0.2 and 10 A g-1 , and a remarkable areal capacity of ≈0.5 mAh cm-2 with a mass loading of 5.3 mg cm-2 , outperforming most carbon cathodes reported thus far. Moreover, safe and flexible NPCNs based quasi-solid-state devices are fabricated, which can withstand drilling and mechanical bending, suggesting their potential applications in wearable devices.

The rise of biosimilar monoclonal antibodies has renewed the interest in monoclonal antibody (mAb) charge variants composition and separation. The sample displacement chromatography (SDC) has the potential to overcome the low separation efficiency and productivity associated with bind-elute separation of mAb charge variants. SDC in combination with weak cation exchanging macroporous monolithic chromatographic column was successfully implemented for a separation of charge variants and aggregates of monoclonal IgG under overloading conditions. The charge variants composition was at-line monitored by a newly developed, simple and fast analytical method, based on weak cation exchange chromatography. It was proven that basic charge variants acted as displacers of IgG molecules with lower pI, when the loading was performed 1 to 1.5 pH unit below the pI of acidic charge variants. The efficiency of the SDC process is flow rate independent due to a convection-based mass transfer on the macroporous monolith. The productivity of the process at optimal conditions is 35 mg of purified IgG fraction per milliliters of monolithic support with 75-80% recovery. As such, an SDC approach surpasses the standard bind-elute separation in the productivity for a factor of 3, when performed on the same column. The applicability of the SDC approach was confirmed for porous particle-based column as well, but with 1.5 lower productivity compared to the monoliths.

Phosphorus (P) losses from flooded soils and subsequent transport to waterways contribute to eutrophication of surface waters. This study evaluated the effectiveness of MnO2 and a zeolite Y amendment in reducing P release from flooded soils and explored the underlying mechanisms controlling P release. Unamended and amended (MnO2 or zeolite, surface-amended at 5 Mg ha-1) soil monoliths from four clayey-alkaline soils were flooded at 22 ± 2 °C for 56 days. Soil redox potential and dissolved reactive P (DRP), pH, and concentrations of major cations and anions in porewater and floodwater were analyzed periodically. Soil P speciation was simulated using Visual MINTEQ at 1, 28, and 56 days after flooding (DAF) and P K-edge X-ray absorption near-edge structure spectroscopy and sequential fractionation at 56 DAF. Porewater DRP increased with DAF and correlated negatively with pe+pH and positively with dissolved Fe. Reductive dissolution of Fe-associated P was the dominant mechanism of flooding-induced P release. The MnO2 amendment reduced porewater DRP by 30%-50% by favoring calcium phosphates (Ca-P) precipitation and delaying the reductive dissolution reactions. In three soils, the zeolite amendment at some DAF increased porewater and/or floodwater DRP through dissolution of Ca-P and thus was not effective in reducing P release from flooded soils.

Processable microporous organic polymers (MOPs) attract incomparable research interest because their vairous types, such as monoliths and membranes are for practical application. Most processable MOPs usually need harsh conditions such as the use of expensive metal catalysts, specialized stereospecific monomers, etc., which restrict the sustainable and real applications of processable MOPs. Therefore, the economical mass production of processable MOPs remains a formidable challenge. Herein, a novel strategy is reported for constructing processable hypercrosslinked polymers (HCPs) need two steps synthesis of pre-crosslinking and deep-crosslinking using divinylbenzene (DVB) as a self-crosslinking monomer under the catalysis of a small amount of FeCl3 . The resulting HCPs monoliths possess high BET surface area of 1033-1056 m2 g-1 with hierarchical porosity, and show excellent mechanical strength up to 65 MPa. It is, to the best of authors' knowledge, the first report of using aromatic vinyl monomers as self-crosslinking monomers to generate HCPs monoliths with high surface area, yielding no by-products, and high mechanical strength.

In this work, we report two novel solid-state opto-chemosensors that proffer exclusive selectivity and excellent sensitivity for the naked-eye detection of ultra-trace Co2+ ions. The opto-chemosensors are concocted using structurally engineered porous silica and polymer monolith templates that are uniformly arranged with a chromoionophoric probe i.e., (Z)-2-mercapto-5-(quinolin-8-yldiazenyl)pyrimidine-4,6-diol (AQTBA). The probe anchored monolithic opto-chemosensors induces sequential color transitions, from yellowish-orange to dark brown, with incremental addition of Co2+ ions. The optimized ground state structure of the AQTBA probe and its AQTBA-Co2+ complex are analyzed using a gaussian 16 program at B3LYP level, with a 6-311+ G (d, p) basis set. The structural and surface morphology of the opto-sensors are characterized using various microscopic, spectroscopic, and diffraction techniques, which discloses a uniform pattern of pore network that proffers rapid ion diffusion kinetics to the probe chelating sites. The proposed monolithic sensors exhibit a high degree of tolerance towards various foreign cations and anions, thus revealing its exclusive selectivity in targeting ultra-trace concentrations of Co2+. The silica and polymer monolithic sensors exhibit a broad linear response range of 0-200 ppb, with a detection limit of 0.35 and 0.07 ppb for Co2+ ions, respectively. The unique features of the proposed sensors are their faster response kinetics (120 s), greater reusability (nine cycles), excellent chemical and thermal durability (pH ≤ 12.0; T ≤ 200 °C), with reliable data reproducibility (recovery ≥99.3 %; RSD ≤2.3 %). The proposed solid-state opto-chemosensors paves way for maximum waste reduction strategy, along with the feasibility for real-time monitoring of environmental and industrial water samples.

Highly crosslinked monolithic capillary columns with inner diameters in the range of 50-530 µm were prepared by radical polymerization of pentaerythritol tetraacrylate, polyhedral oligomeric silsesquioxane-methacrylate, and n-octadecyl methacrylate in the presence of methanol, dodecyl alcohol, and polyethylene glycol lauryl ether. Columns were evaluated by inverse size-exclusion chromatography employing a set of polystyrene standards of narrow molecular-size distribution and by scanning electron microscopy. Chromatographic performance under reversed-phase conditions was also evaluated. The combination of two effective crosslinkers as pentaerythritol tetraacrylate and polyhedral oligomeric silsesquioxane-methacrylate in the polymerization mixture allows for the preparation of robust and efficient monolithic capillary columns within a fairly wide range of internal diameters.

A novel acetylcholinesterase Nano liquid chromatography capillary column (75 µm i.d. × 50 mm length) was developed for the fast screening of acetylcholinesterase inhibitors and the evaluation of their molecular recognition mechanism. Biotinylated acetylcholinesterase was immobilized on a streptavidin Nano liquid chromatography capillary column. Because of the very strong streptavidin-biotin interaction, the acetylcholinesterase immobilization step performed by frontal analysis is very fast (required less than 10 min), and the amount of immobilized acetylcholinesterase was in the microgram range (1 µg). The yellow anion obtained from the enzymatic reaction detected at 412 nm was achieved within 60 s, and the immobilized acetylcholinesterase retained 96% of the initial activity beyond 90 days. This column was successfully applied for the discrimination of weak affinity ligands to acetylcholinesterase from nonbinders, which is the heart of fragment-based drug design. This column was used for the determination of the IC50 values of a series of inhibitor molecules. In addition, it was demonstrated that competitive experiments could be performed with our miniaturized system to confirm the existence and binding pocket of a ligand to acetylcholinesterase contained in a methanol plant extract. The results revealed that our acetylcholinesterase Nano liquid chromatography capillary column developed in this work represents a useful tool for the rapid screening of inhibitor candidates and evaluation of the action mechanism.

An efficient multiple fibers solid-phase microextraction method based on porous monolith was established for Se(IV) and Se(VI) analysis. Poly(4-vinylphenylboronic acid/styrene-co-ethylene dimethacrylate/divinylbenzene) monolith was fabricated and employed as the extraction phase for efficient entrapment of Se(IV) complexed with o-phenylenediamine, followed by elution with a methanol/formic acid (99/1.0, v/v) mixture and quantification by high-performance liquid chromatography with diode array detector. The Se(VI) species was measured by the difference between total inorganic Se and Se(IV) after pre-reduction. Different characterization techniques were employed to inspect the structure and morphology of prepared adsorbent. A series of key extraction factors were optimized so as to achieve the expected extraction performance. Under the optimized separation and capture parameters, the linear range and limit of detection for Se(IV) in water sample were 0.050-200 and 0.013 µg/L, respectively. For beer sample, the corresponding values were 0.010-300 and 0.032 µg/L. The developed microextraction approach was successfully utilized to detect trace Se(IV) and Se(VI) in environmental water and beer samples with satisfactory fortified recovery and repeatability. Results well reveal the attractive merits of the established method in the analysis of Se species, including simple preparation of adsorbent, convenient extraction procedure, good sensitivity, high cost-effectiveness, and eco-friendliness.

Advances in the development of column-based analytical separations are strongly linked to the development of novel materials. Stationary phases for chromatographic separation are usually based on silica and polymer materials. Nevertheless, recent advances have been made using porous crystalline reticular materials, such as metal-organic frameworks and covalent organic frameworks. However, the direct packing of these materials is often limited due to their small crystal size and nonspherical shape. In this review, recent strategies to incorporate porous crystalline materials as stationary phases for liquid-phase separations are covered. Moreover, we discuss the potential future directions in their development and integration into suitable supports for analytical applications. Finally, we discuss the main challenges to be solved to take full advantage of these materials as stationary phases for analytical separations.

Biomarkers are significant indicators to assist the early diagnosis of diseases and assess the therapeutic response. However, due to the low abundance of biomarkers in complex biological fluids, it is highly desirable to explore efficient techniques to attain their selective recognition and capture before the detection. Molecularly imprinted monoliths integrate the high selectivity of imprinted polymers and the rapid convective mass transport of monoliths, and as a result, are promising candidates to achieve the specific enrichment of biomarkers from complex samples. This review summarizes the various imprinting approaches for the preparation of molecularly imprinted monoliths. The state-of-art advances as an effective platform for applications in the selective capture of biomacromolecules related biomarkers were also outlined.

The degradation of toluene from a gas stream by the heterogeneous Fenton process was evaluated over a carbon-coated monolith impregnated or not with iron as catalyst in a bubble column reactor (BCR). The carbon-coated monolith support (CM) was prepared by chemical vapor deposition and the catalyst (CM impregnated with iron - herein called CM-Fe) by adsorption. In the screening of processes (absorption, adsorption and reaction), it was shown that the heterogeneous Fenton process catalyzed by CM-Fe presents the best efficiency (toluene transfer (η) = 10 × 10-3 mol, for 300 mL of liquid solution and 0.69 g of catalyst). Finally, the stability of CM and CM-Fe was evaluated, wherein ten consecutive runs were carried out, the results showing a considerable deactivation of CM during the first five cycles. In contrast, the CM-Fe sample only slightly decreases its activity from the 1st to 2nd cycle (due to a small amount of iron leached from the monolith, 0.7%), remaining stable after that, which is important for applying this technology at the industrial level. This work showed for the first time that the treatment of gaseous effluents containing organic compounds by the Fenton process (which takes place in the liquid phase) using a carbon-coated monolith impregnated with iron is plausible, so the proof of concept was successfully accomplished.
