RESUMEN

The arsenic (As) release from litter decomposition of As-hyperaccumulator (Pteris vittata L.) in mine areas poses an ecological risk for metal dispersion into the soil. However, the effect of atmospheric nitrogen (N) deposition on the litter decomposition of As-hyperaccumulator in the tailing mine area remains poorly understood. In this study, we conducted a microcosm experiment to investigate the As release during the decomposition of P. vittata litter under four gradients of N addition (0, 5, 10, and 20 mg N g-1). The N10 treatment (10 mg N g-1) enhanced As release from P. vittata litter by 1.2-2.6 folds compared to control. Furthermore, Streptomyces, Pantoea, and Curtobacterium were found to primarily affect the As release during the litter decomposition process. Additionally, N addition decreased the soil pH, subsequently increased the microbial biomass, as well as hydrolase activities (NAG) which regulated N release. Thereby, N addition increased the As release from P. vittata litter and then transferred to the soil. Moreover, this process caused a transformation of non-labile As fractions into labile forms, resulting in an increase of available As concentration by 13.02-20.16% within the soil after a 90-day incubation period. Our findings provide valuable insights into assessing the ecological risk associated with As release from the decomposition of P. vittata litter towards the soil, particularly under elevated atmospheric N deposition.

The aim of the research was to determine the protective effect and mechanism of Pteris wallichiana J. Agardh extract (PWE) on DSS-induced ulcerative colitis (UC) in mice. In this research, PWE is rich in flavonoids and diterpenoids by UPLC-MS/MS analysis. In LPS-induced RAW264.7 cells, PWE reduced the productions of inflammatory factors (i.e., NO, TNF-α, IL-6, and IL-1ß). In DSS-induced UC in mice, PWE improved disease activity index (DAI) score, attenuated oxidative stress by decreasing MPO and MDA activities and activating GSH and SOD levels, and inhibited TNF-α, IL-6, and IL-1ß expressions in the colonic tissues. PWE also improved the intestinal barrier by upregulating the expressions of tight junction proteins, including occludin and ZO-1. Moreover, PWE extract alleviated intestinal inflammation by suppressing the TLR4/MyD88/NF-κB signaling pathway. Conclusion: PWE can alleviate DSS-induced UC in mice by increasing the expressions of intestinal tight junction proteins and inhibiting the TLR4/NF-κB inflammatory pathway.

The objective of this work was the development of a detailed, extensive and reliable database of the metabolomes of P. vittata. Using an ultra-performance liquid chromatography-triple quadrupole tandem mass spectrometry system (UPLC-QqQ-MS/MS) and based on the knowledge of retention time and mass spectral characteristics of an in-house collection of authentic standards, we screened for the presence of a large collection of natural compounds. The database represents 359 authenticated metabolites, comprising 220 primary and 139 secondary metabolites (70 flavonoids, 16 phenylpropanoic acid derivatives, five coumarins, two stilbenoids, 14 benzoic acids, nine phenols, 20 alkaloids and three terpenoids). Comparison of the accumulation of these compounds in two tissues showed that the aerial parts were enriched in flavonols, whereas the subterranean parts were enriched in anthocyanins. The comprehensive database developed here will be beneficial in improving the understanding of the chemical basis of plant therapeutic profile using multivariate analysis, with a particular example of antioxidant activity.

Within the domain of phytoremediation research, the proper disposal of harvestable plant parts, that remove pollutants from contaminated soil, has been attracted extensive attention. Here, the bioenergy generation capability of trace metals (Cu, Pb, Zn, Cd, Mn, and As) polluted plants was assessed. The biogas production potential of accumulators or hyperaccumulator plants, Elsholtzia haichowensis, Sedum alfredii, Solanum nigrum, Phytolacca americana and Pteris vittata were 259.2 ± 1.9, 238.7 ± 4.2, 135.9 ± 0.9, 129.5 ± 2.9 and 106.8 ± 2.1 mL/g, respectively. The presence of Cu (at approximately 1000 mg/kg) increased the cumulative biogas production, the daily methane production and the methane yield of E. haichowensis. For S. alfredii, the presence of Zn (≥500 mg/kg) showed a significant negative impact on the methane content in biogas, and the daily methane production, which decreased the biogas and methane yield. The biogas production potential increased when the content of Mn was at 5 000-10,000 mg/kg, subsequently, decreased when the value of Mn at 20,000 mg/kg. However, Cd (1-200 mg/kg), Pb (125-2000 mg/kg) and As (1250-10,000 mg/kg) showed no distinctive change in the cumulative biogas production of S. nigrum, S. alfredii and P. vittata, respectively. The methane yield showed a strong positive correlation (R2 =0.9704) with cumulative biogas production, and the energy potential of the plant residues were at 415-985 kWh/ton. Thus, the anaerobic digestion has bright potential for the disposal of trace metal contaminated plants, and has promising prospects for the use in energy production.

Wheat (W) and accumulators (A) were planted in plots (arsenic amended soil and without arsenic) designed with ecotoxicological concern for arsenic safe-grains. For the study sixteen plots of 2 × 2 × 0.5 m (l × b × h) size were prepared. Arsenic (As) in the form of sodium arsenate was applied at 50 mg/kg in plots. Out of these sixteen plots eight plots had arsenic amended soil and rest 8 without any arsenic (C). Accumulator's viz. Pteris vittata (PV), Phragmites australis (PA) and Vetiveria zizanioides (VZ) were planted along with wheat in combination (W + PV, W + PA and W + VZ) in twelve plots (6 AWAs plots and 6 AWC plots). In the rest 4 plots (2 WAs plots and 2 WC plots), only wheat was planted. The study was conducted for two cropping seasons, where accumulators were left in the plots between the cropping seasons except that before 2nd cropping accumulators were properly pruned and extra tillers were removed. The germination % of wheat in WAs in 1st and the 2nd cropping season was found to be 55 and 57%, while in AWAs and AWC plots it was between 86 and 92% (W + VZ, 56 and 73%). The physiological activity was found to be reduced in WAs plots compared to AWAs (except for vetiver combination) and AWC plots in both cropping seasons. The antioxidant activity was enhanced in WAs compared with AWAs. The arsenic concentration in grains of wheat was within the permissible limit set by WHO and GOI in AWAs plots while it exceeded the limit in W + VZ (in 1st cropping) and WAs in both cropings.

Phytochemical investigation of the aerial parts of Pteris cretica led to the isolation and elucidation of nine pterosins, including four new pterosins, creticolacton A (1), 13-hydroxy-2(R),3(R)-pterosin L (2), creticoside A (3), and spelosin 3-O-ß-d-glucopyranoside (4), together with five known pterosins 5-9. Their structures were identified mainly on the basis of 1D and 2D NMR spectral data, ESI-MS and literature comparisons. Compounds 1 and 3 were new type of petrosins with a six membered ring between C-14 and C-15. The new compounds were tested in vitro for their cytotoxic activities against four human tumor cell lines (SH-SY5Y, SGC-7901, HCT-116, Lovo). Results showed that compounds 1 and 2 exhibited cytotoxic activity against HCT-116 cells with IC50 value of 22.4 µM and 15.8 µM, respectively.

Pot experiments were conducted to assess the effects of monoammonium phosphate (NH4H2PO4) and citric acid (CA) on the arsenic uptake of Chinese brake fern (Pteris vittata L. in two typical arsenic-contaminated soils i.e. fluvo-aquic and brown) from Jiyuan (JY) City and Baoding (BD) City in Northern China. NH4H2PO4 improved the biomass of P. vittata, whereas CA exerted no significant influence. NH4H2PO4 and CA both increased the arsenic uptake of P. vittata by 6.08 and 2.72 times, respectively, in fluvo-aquic soil and 4.20 and 2.52 times, respectively, in brown soil. Moreover, CA, but not NH4H2PO4, promoted the transfer of arsenic from the root to the frond. NH4H2PO4 and CA increased Olsen's arsenic contents in the soils and promoted the transformation of residual arsenic and crystalline Fe/Al oxide-bound arsenic to nonspecifically and specifically sorbed arsenic. This study proved that P. vittata can be used in Northern China. Applying NH4H2PO4 and CA can enhance the effectiveness of P. vittata in the phytoremediation of arsenic-contaminated soils.

Two new pterosin glycosides, (2S,3S)-pterosin C 3-O-ß-d-(4'-(E)-caffeoyl)-glucopyranoside (1) and (2S,3S)-pterosin C 3-O-ß-d-(6'-(E)-p-coumaroyl)-glucopyranoside (2), were isolated from Pteris multifida (Pteridaceae) roots along with ten known pterosin compounds (3-12). The chemical structures of the isolated compounds were elucidated by extensive analysis of the 1D, 2D NMR, HRESIMS, and CD spectroscopic data. The cytotoxicities of 1-12 against HCT116 human colorectal cancer cell line were evaluated. Among the isolates, compound 1 showed moderate antiproliferative activity in HCT116 cells with an IC50 value of 8.0±1.7µM. Additionally, 1 induced the upregulation of the caspase-9 and procaspase-9 levels in Western blots and increased the annexin V/propidium iodide (PI)-positive cell population in flow cytometry.

A pot experiment was conducted to compare the content of endogenous trans-zeatin (Z), plant arsenic (As) uptake and physiological indices in the fronds of As-hyperaccumulator (Pteris cretica var. nervosa) and non-hyperaccumulator (Pteris ensiformis). Furthermore, a stepwise regression method was used to study the relationship among determined indices, and the time-course effect of main indices was also investigated under 100mg/kg As stress with time extension. In the 100-200mg/kg As treatments, plant height showed no significant difference and endogenous Z content significantly increased in P. cretica var. nervosa compared to the control, but a significant decrease of height and endogenous Z was observed in P. ensiformis. The concentrations of As (III) and As (V) increased significantly in the fronds of two plants, but this increase was much higher in P. cretica var. nervosa. Compared to the control, the contents of chlorophyll and soluble protein were significantly increased in P. cretica var. nervosa but decreased in P. ensiformis in the 200mg/kg As treatment, respectively. A significant positive correlation was found between the contents of endogenous Z and total As in P. cretica var. nervosa, but such a correlation was not found in P. ensiformis. Additionally, in the time-course effect experiment, a peak value of each index was appeared in the 43rd day in two plants, except for chlorophyll in P. ensiformis, but this value was significantly higher in P. cretica var. nervosa than that in P. ensiformis. In conclusion, a higher endogenous Z content contributed to As accumulation of P. cretica var. nervosa under As stress.

RESUMEN

Phytochemistry investigation on Pteris ensiformis led to the isolation of a new ent-kaurane diterpenoid, ent-kaurane-6ß,16α-diol-3-one (1), along with five known diterpenoids (2-6) and three known sesquiterpenes (7-9). Their structures were established by means of spectroscopic methods. The ethanol extract and the isolated compounds (1-9) were evaluated for their antitumor activity against three cancer cell lines.

Two new dihydrochalcone enantiomers (+)-1 and (-)-1, along with eight known compounds 3-10, were obtained from Pteris ensiformis. The planar structures were determined on the basis of extensive 1D and 2DNMR and HRESIMS. The resolution of (+)-1 and (-)-1 was achieved by chiral HPLC analysis. The absolute configurations of (+)-1 and (-)-1 were established by the bulkiness rule using Rh2(O2CCF3)4-induced circular dichroism (ICD) method. Compounds (+)-1, (-)-1, 8, 9 and 10 exhibited the inhibitory assay of NO production in mouse macrophages stimulated by LPS, with IC50 values of 2.0, 2.5, 8.0, 9.5 and 5.6 µM, respectively. Otherwise, compound 10 showed moderate cytotoxic activity against HCT-116, HepG-2 and BGC-823 cell lines with IC50 values of 3.0, 10.5 and 6.3 µM, respectively.

The dried whole plant of Pteris dispar were milled and extracted with 95% EtOH. The resulting dried extract was isolated by kinds of chromatographic column, including polyamide, Sephadex LH-20, preparative HPLC. As a result, ten diterpenes were isolated from the plant. By analyzing of ESI-MS and NMR spectroscopic data, the structures were established as geopyxin B(1), geopyxin E(2), ent-11α-hydroxy-18-acetoxykaur-16-ene(3), ent-14ß-hydroxy-18-acetoxykaur-16-ene(4), neolaxiflorin L(5), ent-3ß,19-dihydroxy-kaur-16-ene(6), ent-3ß-hydroxy-kaur-16-ene(7), 7ß,17-dihydroxy-16α-ent-kauran-19-oic acid 19-O-ß-D-glucopyranoside ester(8), crotonkinin C(9)and crotonkinin C(10). Compounds 1-10 were obtained from P. dispar for the first time. Compounds 1 and 2 showed moderate activities against Bel-7402 with IC50 values of 7.50 and 10.60 µmol•L⁻¹, and against HepG2 with IC50 values of 6.68,11.80 µmol•L⁻¹, respectively.

Four new pterosin sesquiterpenoids (1-4), a new ent-kaurane diterpenoid (17), and 18 known compounds were isolated from the aerial parts of Pteris cretica L. The structures of the isolates were elucidated based on spectroscopic data analysis, and their absolute configurations were determined by comparison of experimental and calculated electronic circular dichroism spectra. The compounds were evaluated for lipid-lowering effects in 3T3-L1 adipocytes. Compounds 4, 8, 17, and 22 were more potent than the positive control, berberine, in decreasing triglycerides activity, with compound 4 exerting the most potent activity. Compound 4 activated LXRα/ß in a HEK 293T cell-based reporter gene assay. Molecular dynamic simulations revealed that compound 4 activates liver X receptors (LXRs) through hydrogen bonding with the residues of LXRα/ß, suggesting that compound 4 reduces total triglycerides through the regulation of LXRα/ß.

Activated microglia are known to be a major source of cellular neuroinflammation which causes various neurodegenerative diseases, including Alzheimer's disease. In our continuing efforts to search for new bioactive phytochemicals against neuroinflammatory diseases, the 80% methanolic extract of Pteris multifida (Pteridaceae) roots was found to exhibit significant NO inhibitory activity in lipopolysaccharide (LPS)-activated BV-2 microglia cells. Three new ent-kaurane diterpenoids, pterokaurane M1 2-O-ß-d-glucopyranoside (4), 2ß,16α-dihydroxy-ent-kaurane 2,16-di-O-ß-d-glucopyranoside (10), and 2ß,16α,17-trihydroxy-ent-kaurane 2-O-ß-d-glucopyranoside (12), were isolated along with nine other known compounds from P. multifida roots. The chemical structures of the new compounds were determined by 1D- and 2D-NMR, HR-ESI-MS, and CD spectroscopic data analysis. Among the isolates, compounds 1 and 7 significantly inhibited NO production in LPS-stimulated BV-2 cells reducing the expression of the cyclooxygenase-2 (COX-2) protein and the level of pro-inflammatory mediators such as prostaglandin E2 (PGE2), tumor necrosis factor (TNF)-α, interleukin (IL)-1ß, and IL-6. These results suggest that ent-kaurane diterpenes from P. multifida could be potential lead compounds that act as anti-neuroinflammatory agents.

RESUMEN

The materials were extracted by 95% ethanol, and the extracting solution was isolated by kinds of chromatographic columns including polyamide, MCI, preparative MPLC, and preparative HPLC. Eight diterpenes and two sesquiterpenes were isolated from the plant. On analysis of ESI-MS and NMR spectroscopic data, the structures were established as ent-3ß-hydroxy-kaur-16-en-19-al (1), 4-epi-kaurenic acid (2), mitrekaurenone (3), 7ß,16α,17-trihydroxy-ent-kauran-19-oic acid (4), crotonkinin E (5), crotonkinin F (6), pterisolic acid A (7), pterisolic acid C (8), (2R)-pterosin P (9), and dehydropterosin B (10). Compounds 1-6 were obtained from Pteris for the first time, and compounds 7-10 were obtained from P. ensiformis for the first time. Compounds 5-8 showed moderate activity against HCT-116, HepG2 and BGC-823 cell lines, separately.

RESUMEN

An unusual 5-C-methylated-dihydrobenzofuroisocoumarin, named multifidarin A (1), and two new ent-kaurane diterpenoids, named multikauranes A (2) and B (3), together with three known ent-kaurane diterpenoids, were isolated from the whole plants of Pteris multifida. The structures of 1-3 were elucidated by spectroscopic methods. The cytotoxic activities of all new compounds were evaluated against five human tumor cell lines. A possible biosynthetic process for the formation of 1 is proposed.

RESUMEN

Henrin A (1), a new ent-kaurane diterpene, was isolated from the leaves of Pteris henryi. The chemical structure was elucidated by analysis of the spectroscopic data including one-dimensional (1D) and two-dimensional (2D) NMR spectra, and was further confirmed by X-ray crystallographic analysis. The compound was evaluated for its biological activities against a panel of cancer cell lines, dental bacterial biofilm formation, and HIV. It displayed anti-HIV potential with an IC50 value of 9.1 µM (SI = 12.2).

Synchrotron radiation X-ray absorption near edge structure was employed to study the arsenic (As) speciation in 13 Pteris vittata L. populations collected from 7 provinces, cities, and autonomous regions in China. As in roots of P. vittata was mainly combined with oxygen (O), with a small amount of As combined with glutathione (GSH). Populations from Hunan and Guangxi provinces showed higher percentages of As-GSH in soots. As in roots of P. vittata was predominated with As(V), with the percentage of As(V) to the total As being 59.6±0.6%~83.8±3.8%. The As(V) percentage was in the order of HN5HN3>HN1>TW>CQ>AH>FJ>HN5>HN2>GX2>GX3>HN4>GX1, within the range of 2.4%~12.9%. Different from that in roots, As in shoots was predominated with As(III), with no As(V) detected. The disclosure of As speciation in the roots and shoots of P. vittata contributes to the future research on As accumulation mechanism.

The quality control method and standard were established to control the quality of Pteris multifida in this paper. The tests of water content, total ash, acid-unsoluble ash and ethanol-soluble extractives of P. multifida were carried out according to the methods recorded in appendix of Chinese Pharmacopeia (2010 edition, volume 1) . The TLC method was established by using rhoifolin as references, and a mixture of CHCl3 -MeOH-HAc (6: 1: 1) as the developing solvent system on GF254 thin layer plate. The contents of rhoifolin was determined by HPLC on a Diamonsil C18 (4.6 mm x 250 mm, 5 µm) column, using acetonitrile-water (containing 0.15% formic acid) (16: 84) as mobile phase at a flow rate of 1.0 mL x min(-1). The column temperature was 30 degrees C and the detection wave-length was 350 nm. As a result, pterosin C 3-O-ß-D-glucosidede and the other constituents were well separated on TLC detected under the UV light at 254 nm . The methodology validation for the assay of rhoifolin presented that it was in good linear correlation in the ranges of 0.025 5-5.1 µg with the regression equations of Y = 1 092.4X + 9.503 5 (r = 0.999 8), and the average recoveries were 100.3% (RSD 1.3%). The content range of rhoifolin from 16 different batches of Pteris multifida was 0.08-5.06 mg x g(-1). The water content, total ash, acid-unsoluble ash and ethanol-soluble extractives of 16 samples varied in the ranges of 7.35% - 12.96%, 6.90% - 16.33%, 2.07% -11.38% and 13.29% -23.87%, respectively. The suggesting limes in the quality standard for water content, total ash, acid-unsoluble ash, ethanol-soluble extractives and rhoifolin content were ≤ 12% , ≤ 15% , ≤ 8.5% , ≥ 14% and ≥ 0.040%, respectively. The result proved that the established quality of control method was specific and accurate, which can be used for the quality control of P. multifida.

On March 11, 2011, one of the negative effects of the tsunami phenomenon that devastated the Pacific coast of the Tohoku district in Japan was the deposition of a wide range of arsenic (As) contamination to the soil. To remediate such a huge area of contamination, phytoremediation by Pteris vittata, an As-hyperaccumulator, was considered. To evaluate the efficacy of applying P. vittata to the area, the salt tolerance of P. vittata and the phytoextraction of As from soil samples were investigated. For the salt tolerance test, spore germination was considerably decreased at an NaCl level of more than 100 mM. At 200 mM, the gametophytes exhibited a morphological defect. Furthermore, the growth inhibition of P. vittata was observed with a salinity that corresponded to 66.2 mS/m of electric conductivity (EC) in the soil. A laboratory phytoremediation experiment was conducted using As-contaminated soils for 166 days. P. vittata grew and accumulated As at 264 mg/kg-DW into the shoots. Consequently, the soluble As in the soil was evidently decreased. These results showed that P. vittata was applicable to the phytoremediation of As-contaminated soil with low salinity as with the contamination caused by the 2011 tsunami.
