RESUMO

Knowledge of the developmental ontogeny of the digestive system and nutritional requirements of marine fish larvae is a primary requisite for their successful rearing under an optimal feeding regime. In this context, we assessed the activity profile of key digestive enzymes viz., trypsin, chymotrypsin, leucine aminopeptidase, lipase, amylase, and alkaline phosphatase during the early ontogeny of milkfish, Chanos chanos (0 day, 3 days, 6 days, 9 days, 12 days, 15 days, 18 days, 21 days, 25 days, and 30 days post-hatch). Larvae for this study were obtained from the successful breeding of milkfish at ICAR-Central Institute of Brackishwater Aquaculture, India. Growth curves (length and weight) of the larvae indicated a positive morphological development under a standardized feeding regime that comprised Chlorella salina, Brachionus plicatilis, Artemia salina nauplii, and commercial weaning feed for different larval stages. With respect to protein digestion, the specific activity of pancreatic enzymes trypsin and chymotrypsin and intestinal brush border leucine aminopeptidase showed two peaks at 3 dph and 15 dph, following the introduction of rotifer and Artemia nauplii. Similar bimodal peaks were observed for alkaline phosphatase and amylase activities, with the first peak at 3 dph and the second peak at 18 dph and 21 dph, respectively. Whereas in the case of lipase, high activity levels were observed at 0 dph, 3 dph, and 18 dph, with subsequent decreases and fluctuations. Overall, as most of the enzymes were found to have peak activities at 15 to 21 dph, this period can be potentially considered as the developmental window for weaning larvae from live to formulated feeds in milkfish hatcheries.

A 45 days feeding trial was conducted to study the effect of Jerusalem artichoke (JA) on growth performance, body composition, biochemical, immuno-hematological parameters and disease resistance in Asian seabass (Lates calcarifer) fingerlings against Aeromonas hydrophila. JA was supplemented at three different levels viz., control 0, 5, 10, and 20 g kg-1 in the commercial diet (403 g kg-1protein and 89 g kg -1lipid) in L. calcarifer. The results showed that there were no significant (P > 0.05) differences in various growth parameters, while the whole body composition showed significant differences (P < 0.05) between control and treatment groups. Hematological parameters showed that red blood cells (RBC), white blood cells (WBC), hemoglobin (Hb), pack cell volume (PCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) were not significantly (P > 0.05) affected by dietary supplementation of JA at different concentration. However, the mean corpuscular volume (MCV) was significantly (P < 0.05) higher in the fish fed with 20 g kg-1 JA supplemented diet. Biochemical parameters revealed that glucose, urea, cholesterol, and triglyceride showed significant (P < 0.05) differences between control and treatments groups. Interestingly, 20 g kg-1 JA supplemented diet significantly modulates the innate immune response and disease resistance against Aeromonas hydrophila compared with control and other treatment groups. The results of the study revealed that 20 g kg-1 JA supplementation has a beneficial effect in the biochemical, immunological and disease resistance in L. calcarifer juveniles.

The amelioration effect of water-soluble vitamin pyridoxine against stress was evaluated in milkfish, Chanos chanos exposed to endosulfan. Two hundred and twenty-five fish were distributed randomly into five treatments, each with three replicates. Four isocaloric and isonitrogenous diets with graded levels of pyridoxine feed were as follows: normal water and fed with control diet (En0/PY0); endosulfan-treated water and fed with control diet (En/PY0); and endosulfan-treated water and fed with 50 (En/PY 50 mg/kg), 75 (En/PY 75 mg/kg) and 100 mg/kg (En/PY 100 mg/kg) pyridoxine-supplemented feed. The endosulfan in treated water was maintained at a level of 1/40th of LC50 (0.52 ppb). The effect of dietary pyridoxine supplementation was studied in terms of antioxidative enzymes (catalase, superoxide dismutase, glutathione-S-transferase), stress markers [heat-shock protein 70, caspase-3, cortisol, acetylcholine esterase (AChE), blood glucose], immunohaematological parameters (total protein, albumin, globulin and A/G ratio, nitroblue tetrazolium, RBC, WBC, Hb), gill histopathology and a subsequent challenge study with Vibrio parahaemolyticus. The antioxidative enzymes, stress markers, albumin and A/G ratio were significantly (p < 0.01) elevated, brain AChE and immunohaematological parameters were significantly (p < 0.01) decreased, and chromosome aberration and gill histopathology were also altered due to endosulfan exposure. The relative survival % was reduced due to the combined effect of endosulfan stress and bacterial challenge. Fish fed the diet supplemented with pyridoxine at 75 and 100 mg/kg was found to restore the studied parameter towards normal compared with control and indicated protection against endosulfan-induced stress significantly (p < 0.01). Results obtained in the present study indicate that the supplementation of 75 and 100 mg/kg of pyridoxine in the diet has a definitive role in the mitigation of the endosulfan-induced stress in milkfish, C. chanos fingerlings.
