RESUMO

B cell receptor-associated protein 31 (BAP31) is a transmembrane protein that is widely expressed and primarily located in the endoplasmic reticulum (ER). B cells play a crucial role in the immune system, and BAP31 significantly contributes to the functions of various immune cells. However, the specific role of BAP31 in B lymphocytes development remains unknown. In this study, we utilized a mouse model with BAP31 deleted from B cells to investigate its effects. Our findings reveal a block in early B cell development in the bone marrow and a significant decrease in the number of B cells in peripheral lymphoid organs taken from BAP31 B cell conditional knockout (BAP31-BCKO) mice. B cell receptor (BCR) signaling is crucial for the normal development and differentiation of B lymphocytes. BAP31, an endoplasmic reticulum membrane protein, directly regulates the BCR signaling pathway and was shown to be significantly positively correlated with B cell activation and proliferation. These findings establish BAP31 as a crucial regulator of early B cell development.

Constructing a synergistic effect with different structural fillers is an important strategy for improving the comprehensive properties of polymeric composites. To improve the comprehensive properties of two-component additive liquid silicon rubber (SR) materials used in electronics packaging, the synergistic effect of granular aluminum nitride (AlN) and tubular carbon nanotube (CNT)-reinforced SR nanocomposites was investigated. AlN/CNT/SR composites with different AlN/CNT ratios were fabricated with two-component additive liquid SR via the thermal curing technique, and the influence of AlN/CNT hybrid fillers on the hardness, strength, elongation at break, surface resistivity, thermal conductivity, and thermal decomposition was investigated in detail. With the incorporation of AlN/CNT hybrid fillers, the comprehensive properties of the obtained AlN/CNT/SR composites are better than those of the AlN/SR and CNT/SR composites. The synergistic thermal conductive mechanism of AlN/CNT hybrid fillers was proposed and demonstrated with the fractural surface morphology of the obtained composites. The obtained AlN/CNT/SR composites show promising applications in electronic packaging, where necessary mechanical strength, electrical insulating, thermal conductivity, and thermal stable materials are needed.

The differentiation of CD4+T cells is a crucial component of the immune response. The spleen and thymus, as immune organs, are closely associated with the differentiation and development of T cells. Previous studies have suggested that BAP31 may play a role in modulating T cell activation, but the specific impact of BAP31 on T cells through macrophages remains uncertain. In this study, we present evidence that BAP31 macrophage conditional knockout (BAP31-MCKO) mice display an enlarged spleen and thymus, accompanied by activated clustering and disrupted differentiation of CD4+T cells. In vitro co-culture studies were conducted to investigate the impact of BAP31-MCKO on the activation and differentiation of CD4+T cells. The examination of costimulatory molecule expression in BMDMs and RAW 264.7 cells, based on the endoplasmic reticulum function of BAP31, revealed an increase in the expression of antigen presenting molecules, particularly MHC-II molecule, in the absence of BAP31 in BMDMs or RAW264.7 cells. These findings suggest that BAP31 plays a role in the activation and differentiation of CD4+T cells by regulating the MHC class II molecule on macrophages. These results provide further support for the importance of BAP31 in developing interaction between macrophages and CD4+T cells.

PURPOSE: To investigate correlations between clinical and histopathologic characteristics of idiopathic epiretinal membrane (ERM). DESIGN: Retrospective interventional case series. PARTICIPANTS: In total, 87 eyes from 87 patients with idiopathic ERM who underwent pars plana vitrectomy with peeling of the ERM from 2019 to 2020 were included. METHODS: The outcomes of clinical ophthalmic examination, including measurement of best-corrected visual acuity (BCVA) and spectral-domain OCT (SD-OCT), before and after surgery were reviewed. Surgical specimens were fixed in formalin and embedded in paraffin for histologic and immunohistochemical analysis. MAIN OUTCOMES MEASURES: The association between morphological characteristics revealed on SD-OCT images and the cellular composition of the surgically excised ERM demonstrated with immunohistochemical staining were the main outcome measures. Changes in the BCVA and central macular thickness (CMT) were assessed through a comparison of preoperative and postoperative measurements. RESULTS: Based on SD-OCT morphological characteristics in the foveal area, 15 cases were classified into group 1A (mainly outer retinal thickening), 39 into group 1B (more tenting of the outer retina and distorted inner retina), and 33 into group 1C (prominent inner retina thickening). Overall, postoperative final BCVA and CMT at 1 year improved in all groups. Patients who presented with a better initial BCVA exhibited a more favorable final BCVA. Epiretinal membranes in group 1C demonstrated the greatest decrease in CMT compared with those in groups 1B and 1A, but the final CMT did not differ among the groups. A negative correlation between the density of hyalocytes (P = 0.003) and myofibroblasts (P = 0.047) was noted between the 3 groups. Total cell density and glial cell density of the ERMs were strongly associated with poor final BCVA and BCVA improvement. CONCLUSIONS: The present study provides new histopathologic information regarding the formation and progression of idiopathic ERM. Glial cell proliferation plays a predominant role in these processes. Epiretinal membranes with high cellularity and glial cell density may cause damage to the retina structure, resulting in poor postoperative visual outcomes. These findings provide additional evidence supporting early surgical intervention in patients with idiopathic ERM reported with visual disturbance.

Mapping distortion can be introduced in null testing for off-axis aspheric surfaces, from which the testing results with coordinate deviation cannot be used to guide deterministic optical manufacturing. We propose a correction method for mapping distortion in this study based on the imaging distortion analysis of a null lens and iteration algorithm. We use imaging distortion data to fit the distortion polynomial function by the least-squares method, and we design an iteration algorithm to determine the rotation angle and the position of the null distortion point of the testing results. Finally, the mapping distortion can be accomplished without any markers on the mirror under test. The 280mm×180mm primary mirror and 234mm×176mm tertiary mirror of the hyperspectral camera were tested with a null compensator, and the distortion correction results were obtained using the proposed method. After magnetorheological finishing based on the correction results, the rms of the primary mirror and tertiary mirror reduced from 0.263λ to 0.013λ and 0.059λ to 0.018λ (λ=632.8nm), respectively.

Sepsis, the syndrome of infection complicated by acute organ dysfunction, is a serious and growing global problem, which not only leads to enormous economic losses but also becomes one of the leading causes of mortality in the intensive care unit. The detection of sepsis-related pathogens and biomarkers in the early stage plays a critical role in selecting appropriate antibiotics or other drugs, thereby preventing the emergence of dangerous phases and saving human lives. There are numerous demerits in conventional detection strategies, such as high cost, low efficiency, as well as lacking of sensitivity and selectivity. Recently, the aptamer-based biosensor is an emerging strategy for reasonable sepsis diagnosis because of its accessibility, rapidity, and stability. In this review, we first introduce the screening of suitable aptamer. Further, recent advances of aptamer-based biosensors in the detection of bacteria and biomarkers for the diagnosis of sepsis are summarized. Finally, the review proposes a brief forecast of challenges and future directions with highly promising aptamer-based biosensors.

Jerusalem cherry (Solanum pseudocapsicum), which belongs to the genus Solanum and the family Solanaceae, possesses high ornamental value and is widely cultivated as an indoor ornament due to its bright red berries at maturity (Xu et al., 2018). In September 2019, leaf spot was detected on jerusalem cherry plants in Yuxiu Park, Shizhong district, Jinan, Shandong Province. Field surveys were done in a 1/15 ha park. Disease incidence was estimated at approximately 18% across the survey area. Foliar symptoms began as small white spots. As the disease progressed, lesions expanded and merged, and developed into large irregular white spots, with pale grey edge. At last, lesions were densely distributed throughout the leaves. To isolate the pathogen, twenty leaf tissues (5 × 5 mm) were cut from the border between diseased and healthy tissue, surface disinfected in 75% alcohol for 15 s, soaked in 0.1% mercuric chloride for 1 min, washed with sterile distilled water three times, and cultured on potato dextrose agar (PDA) at 25°C. Nineteen fungal isolates were obtained and were single-spored to obtain pure cultures. The colony of LCL7, a representative isolate, on PDA was initially white to orange, but turned black after 3 to 4 days incubation with black conidial masses. Conidia were single-celled, hyaline, straight, cylindrical, apex obtuse, and ranged from 13.4 to 18.3 × 3.2 to 4.9 µm (n = 50) (Diao et al., 2017). To validate the species identification, rDNA internal transcribed spacer (ITS) region (White et al., 1990), and the partial sequences of glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), ß-tubulin (TUB2), and chitin synthase (CHS-1) (Damm et al., 2019; He et al., 2019), were amplified and sequenced. The ITS, GAPDH, ACT, TUB2, and CHS-1 sequences of isolate LCL7 were submitted to GenBank (MW221320, MW227217, MW227218, MW227219, and MW266988, respectively). ITS, ACT, TUB2, and CHS-1 BLAST showed 99-100% homology with sequences of Colletotrichum liaoningense (ITS, 100% to MH636504; ACT, 100% to MH622582; TUB2, 99.56% to MH622714, CHS-1, 99.33% to MH622446, respectively), although GAPDH showed 93.98% homology with sequence MH681383 (234/249bp). Neighbor-joining tree based on concatenated sequences of the five genes was constructed using MEGA7.0. The results showed the isolate was closely related to C. liaoningense. Based on morphological and molecular characteristics, the isolate LCL7 was identified as C. liaoningense. Pathogenicity tests were performed by spraying a conidial suspension (1 × 105 conidia/mL) on ten two-year-old healthy jerusalem cherry plants. Ten other plants with sterile water served as controls. All samples were incubated in a growth chamber at 25±2°C and transparent plastic bags to keep relative humidity high for 2 days. All inoculated plants showed symptoms similar to those observed in the field after 21 days, but no disease occurred on control plants. The same fungus was successfully reisolated from inoculated leaves and reidentified based on morphology and molecular characteristics, and the fungus was not isolated from the control plants, thus confirming Koch's postulates. Pathogenicity tests were repeated twice. C. liaoningense can cause anthracnose in chili and mango in China (Diao et al., 2017; Li et al., 2019).To our knowledge, this is the first report of anthracnose on jerusalem cherry caused by C. liaoningense in China, which influences ornamental value and reduces market value. Identification of the causes of the disease will help develop effective strategies for managing this disease.

The combination of diagnostic and therapeutic functions in a single theranostic nanoagent generally requires the integration of multi-ingredients. Herein, a cytotoxic near-infrared (NIR) dye (IR-797) and its nanoassembly are reported for multifunctional cancer theranostics. The hydrophobic IR-797 molecules are self-assembled into nanoparticles, which are further modified with an amphiphilic polymer (C18PMH-PEG5000) on the surface. The prepared PEG-IR-797 nanoparticles (PEG-IR-797 NPs) possess inherent cytotoxicity from the IR-797 dye and work as a chemotherapeutic drug which induces apoptosis of cancer cells. The IR-797 NPs are found to have an ultrahigh mass extinction coefficient (444.3 L g-1 cm-1 at 797 nm and 385.9 L g-1 cm-1 at 808 nm) beyond all reported organic nanomaterials (<40 L g-1 cm-1 ) for superior photothermal therapy (PTT). In addition, IR-797 shows some aggregation-induced-emission (AIE) properties. Combining the merits of good NIR absorption, high photothermal energy conversion efficiency, and AIE, makes the PEG-IR-797 NPs useful for multimodal NIR AIE fluorescence, photoacoustic, and thermal imaging-guided therapy. The research exhibits the possibility of using a single ingredient and entity to perform multimodal NIR fluorescence, photoacoustic, and thermal imaging-guided chemo-/photothermal combination therapy, which may trigger wide interest from the fields of nanomedicine and medicinal chemistry to explore multifunctional theranostic organic molecules.

Child unintentional injuries are one of the most prominent global health threats and parents may play a vital role in these injuries. This study thus aims to explore the associations of parents teaching safety rules with preschool children's safety behaviors and unintentional injuries. A total of 62,922 children registered at 182 kindergartens in Longhua District of Shenzhen, China during the fall semester of 2016, were included in this cross-sectional study. Their parents were invited to complete a self-administered questionnaire covering information about socio-demographics, parents teaching safety rules to children, child safety behaviors and unintentional injuries. Logistic and linear regression models were carried out to test the associations among parents teaching safety rules, child safety behaviors, and child unintentional injuries. Whether child safety behaviors mediated the relationship between parents teaching safety rules and child unintentional injuries was assessed using Hayes' PROCESS macros for SPSS. Regression analyses revealed that the higher scores of both mothers' and fathers' teaching safety rules to children were significantly associated with the reduced risks of child unintentional injuries and the modest improvements in child safety behaviors, after adjusting for potential confounders. Furthermore, mediation analysis illustrated that child safety behaviors mediated 18.1% of the association between mothers teaching safety rules and child unintentional injuries and 30.3% of the association between fathers teaching safety rules and child unintentional injuries, respectively. These findings suggest that parents teaching safety rules to children is beneficial for mitigating unintentional injury risks among Chinese preschool children through improving child safety behaviors.

A three-dimensional metal-organic framework (MOF) {[Eu(L)(H2O)2]·DMF} n (1) (H3L = 2'-nitro-3,4',5-biphenyl tricarboxylic acid) was obtained by hydrothermal methods and was characterized in detail. Compound 1 contains [Eu2] binuclear units, and [Eu2] units are further linked by H3L, forming a three-dimensional structure. Stability tests suggest that compound 1 exhibits high solvent, pH, and thermal stability. Besides, we also explored the influence of pH values in the luminescence emission of 1 which shows that pH has a negligible effect on the luminescence intensity with the range of pH = 3-11. Importantly, luminescent investigations reveal that 1 can selectively detect aspartic acid or histidine among 14 amino acids with good regenerability in water. Meanwhile, the concentration of aspartic acid can be quantitatively determined. We also discuss the mechanism of luminescence quenching and enhancement in detail. Importantly, this Eu-MOF provides a MOF-based luminescence sensor to detect two amino acids for the first time.

Two isostructural 2D MOFs ([Ln7(CDA)6(HCOO)3(µ3-OH)6(H2O)8] n, abbreviated as 1-Gd and 2-Dy) were successfully synthesized under solvothermal conditions. The self-assembly of lanthanide(III) nitrate and 1,1'-cyclopropane-dicarboxylic acid (H2CDA) resulted in wheel-like Ln18 cluster second building units (SBU), which are further linked to six neighboring wheels to generate a 2D ordered honeycomb array. Both 1-Gd and 2-Dy exhibit high thermal stability and decompose above 330 °C. Moreover, they have good solvent stability in ten common solvents and pH stability with pH values from 1 to 13. Magnetic studies reveal that 1-Gd exhibits weak antiferromagnetic coupling between adjacent Gd3+ ions and has a large magnetocaloric effect of 47.30 J kg-1 K-1 (Δ H = 7.0 T at 2 K), while 2-Dy shows ferromagnetic interaction between adjacent Dy3+ ions. Interestingly, 1-Gd and 2-Dy can catalyze the cycloaddition of CO2 to epoxides under mild conditions and can be reused at least five rounds with negligible loss of catalytic performance.

The novel synthesis of a dual-modality, pentamethine cyanine (Cy5) fluorescent, 18F positron emission tomography (PET) imaging probe is reported. The probe shows a large extinction coefficient and large quantum yield in the biologically transparent, near-infrared window (650-900 nm) for in vivo fluorescent imaging. This fluorophore bears the isotope, 18F, giving a 18F-PET/near-infrared fluorescent (NIRF), bi-modal imaging probe, that combines the long-term stability of NIRF and the unlimited penetration depth of PET imaging. The bi-modal probe is labeled with 18F in a quick, one-step reaction, which is important in working with the rapid decay of 18F. The bi-modal probe bears a free carboxyl group, highlighting a PET/NIRF synthon that can be conjugated onto many advanced biomolecules for biomarker-specific in vivo dual-modal PET/NIR tumor imaging, confocal histology, and utility in multi-fluorophore, fluorescence-guided surgery. Its potential in vivo biocompatibility is explored in a quick proof-of-principal in vivo study. The dye is delivered to A549 xenograft flank-tumors to generate PET and NIRF signals at the tumor site. The tumor distribution is confirmed in ex vivo gamma counting and imaging. Pentamethine cyanine (Cy5) has the ability to preferentially accumulate in tumor xenografts. We substitute the PET/NIRF probe for Cy5, and explore this phenomenon.

OBJECTIVE: The purpose of this article is to summarize advances in PET fluorescence resolution, agent design, and preclinical imaging that make a growing case for clinical PET fluorescence imaging. CONCLUSION: Existing SPECT, PET, fluorescence, and MRI contrast imaging techniques are already deeply integrated into the management of cancer, from initial diagnosis to the observation and management of metastases. Combined positron-emitting fluorescent contrast agents can convey new or substantial benefits that improve on these proven clinical contrast agents.

Stripe rust, caused by Puccinia striiformis f. tritici, is an important wheat disease in China. P. striiformis f. sp. tritici overwintering and nonoverwintering regions based on the temperature were described elsewhere ( Shi et al. 2005 ). The temperature limit for P. striiformis f. sp. tritici overwintering is derived from field observations. However, P. striiformis f. sp. tritici has recently been observed to overwinter at sites where overwintering is predicted to be unlikely. We studied P. striiformis f. sp. tritici overwintering across several sites in regions close to or further away from the current P. striiformis f. sp. tritici "overwintering boundary" in China. Plants with P. striiformis f. sp. tritici symptoms and uredinia were tagged in late autumn and moved to the laboratory in early spring the following year for quantification of P. striiformis f. sp. tritici biomass via a quantitative reverse-transcription polymerase chain reaction method and for assessment of P. striiformis f. sp. tritici symptoms and sporulation after incubation in a greenhouse. The molecular method detected P. striiformis f. sp. tritici in leaves and sheath in most samples, much greater than the observed incidence of P. striiformis f. sp. tritici symptoms and sporulation after incubation. Thus, further refinement may been necessary to calibrate this molecular method in order to avoid overestimating P. striiformis f. sp. tritici overwintering potential. Active sporulation (hence, successful overwintering) was observed for all sites except one. Increasing altitude led to decreasing incidence of visible P. striiformis f. sp. tritici symptoms and sporulation; in addition to lower temperatures in high altitudes, wind chill may also explain this negative relationship between P. striiformis f. sp. tritici overwinter potential and altitude. P. striiformis f. sp. tritici sporulation on plants subjected to different treatments (control, two oldest leaves, or all leaves removed) indicated that P. striiformis f. sp. tritici overwinters in young green leaves as latent infection established in late autumn. The present study suggests that using only temperature to predict overwintering potential of P. striiformis f. sp. tritici at a given site is insufficient for mountainous regions.

Pure nanodrugs (PNDs), nanoparticles consisting entirely of drug molecules, have been considered as promising candidates for next-generation nanodrugs. However, the traditional preparation method via reprecipitation faces critical challenges including low production rates, relatively large particle sizes, and batch-to-batch variations. Here, for the first time, we successfully developed a novel, versatile, and controllable strategy for preparing PNDs via an anodized aluminum oxide (AAO) template-assisted method. With this approach, we prepared PNDs of an anticancer drug (VM-26) with precisely controlled sizes reaching the sub-20 nm range. This template-assisted approach has much higher feasibility for mass production comparing to the conventional reprecipitation method and is beneficial for future clinical translation. The present method is further demonstrated to be easily applicable for a wide range of hydrophobic biomolecules without the need of custom molecular modifications and can be extended for preparing all-in-one nanostructures with different functional agents.

Thinopyrum chromosomes 7el1, 7el2, 7E(e), and 7E(i), homoeologous to group 7 chromosomes of common wheat (Triticum aestivum), were determined to have many useful agronomical traits for wheat improvement. To analyze the genetic relationships among the 4 Thinopyrum 7E chromosomes, the conserved orthologous set markers, genomic in situ hybridization (GISH), and meiotic chromosome pairing were used in this study. The unweighted pair-group method with arithmetical averages (UPGMA) analysis indicated that 7el1, derived from T. ponticum, and 7E(i), derived from T. intermedium, were the most closely related. 7el2, derived from T. ponticum, was relatively distant from the 7el1-7E(i) complex. While 7E(e), derived from T. elongatum, was more distantly related to 7el1, 7el2, and 7E(i). This is the first report showing that 7el1 and 7E(i) may be similar, which could be explained by the similar chromosome signal distribution revealed by GISH as well as UPGMA analysis revealed by both molecular markers and the highest frequency of meiotic pairing. The newly developed genome-specific molecular markers may be useful for marker-assisted selection of Lr19, Bdv3, and Fhblop.

BACKGROUND: Obstructive sleep apnea (OSA) is characterized by repeated episodes of obstruction of the upper airway. Numerous studies have indicated a relationship between OSA and metabolic syndrome (MS), but the results remain debatable. We aimed to perform a systematic review and meta-analysis to evaluate the association between OSA and MS. METHODS: We searched electronic databases (PubMed, EMBASE, and ISO Web of Knowledge) up to September 2014 with English-language restriction. Cross-sectional, case-control, and cohort studies in which the presence of OSA was assessed by objective measurements, the exposure of interest was OSA, and the outcome of interest was the presence (or incidence) of MS were included. The adjusted odds ratios (ORs) (or relative risk) and 95 % confidence intervals (CIs) were extracted and pooled. Sensitivity analyses were conducted, and heterogeneity and publication bias were assessed. RESULTS: Overall, 15 cross-sectional (2456 patients with OSA and 1705 subjects without OSA), five case-control (1156 OSA patients and 404 controls), and no cohort studies were included. The pooled ORs of MS in individuals with OSA for cross-sectional and case-control studies were 2.87 (95 % CI: 2.41-3.42) and 2.56 (95 % CI: 1.98-3.31), respectively. There was clinically unimportant (I (2) = 20 %) and moderate (I (2) = 35 %) between-study heterogeneity of the analysis. The pooled crude ORs of MS in individuals with mild and moderate-to-severe OSA was 2.39 (95 % CI: 1.65-3.46) and 3.45 (95 % CI: 2.33-5.12), respectively, and there was substantial heterogeneity in the meta-analyses (I (2) = 53 % and I (2) = 63 %, respectively). However, no evidence of publication bias was detected. CONCLUSIONS: OSA is shown to be associated with MS, although causality between these two factors has not been demonstrated yet. Future cohort and randomized controlled studies are needed.

Introduction: Emotional words are often used as stimulus material to explore the cognitive and emotional characteristics of individuals with depressive disorder, while normal individuals mostly rate the scores of affective words. Given that individuals with depressive disorder exhibit a negative cognitive bias, it is possible that their depressive state could influence the ratings of affective words. To enhance the validity of the stimulus material, we specifically recruited patients with depression to provide these ratings. Methods: This study provided subjective ratings for 501 Chinese affective norms, incorporating 167 negative words selected from depressive disorder patients' Sino Weibo blogs, and 167 neutral words and 167 positive words selected from the Chinese Affective Word System. The norms are based on the assessments made by 91 patients with depressive disorder and 92 normal individuals, by using the paper-and-pencil quiz on a 9-point scale. Results: Regardless of the group, the results show high reliability and validity. We identified group differences in three dimensions: valence, arousal, and self-relevance: the depression group rated negative words higher, but positive and neutral words lower than the normal control group. Conclusion: The emotional perception affected the individual's perception of words, to some extent, this database expanded the ratings and provided a reference for exploring norms for individuals with different emotional states.

The utilization of a heterogeneous nanojunction design has shown significant enhancements in the gas sensing capabilities of traditional metal oxide gas sensors. In this study, a novel room temperature H2S gas sensor employing Fe2O3 functionalized Ti3C2 MXene as the sensing material has been developed. This sensor exhibits a broad detection range (0.01-500 ppm), low detection limit (10 ppb), and rapid response/recovery times (10 s/15 s), making it ideal for ppb-level H2S detection. The exceptional gas sensitivity of Fe2O3/Ti3C2 composite to H2S can be attributed to several key factors. First, the unique layered frame structure of Fe2O3/Ti3C2 significantly amplifies the surface area of the hybrid material, enhancing the absorption and diffusion capabilities of H2S molecules. Second, the abundance of functional groups (-O, -OH, and -F) on the surface of Ti3C2 MXene nanosheets provides additional active sites for H2S adsorption, The density functional theory calculation confirms that the adsorption energy of the Fe2O3/Ti3C2 composite for H2S (-2.93 eV) is significantly lower than that of pure Fe2O3 (-2.37 eV) and Ti3C2 (-0.2 eV). Lastly, the remarkable metal conductivity of Ti3C2 MXene ensures efficient electron transfer, thereby enhancing overall sensing performance.
