RESUMO

Parasitic dinoflagellates of the genus Hematodinium are known to infect various marine crustaceans worldwide, especially crabs and several species of shrimp and lobster. Some of these species are new host species and components of commercial fishery products. These parasitic species are predominantly found in the hemolymph of the host and cause pathological changes and functional damage to organs and tissues, leading to death. In recent years, these parasites have infected important commercially valuable species, particularly in European waters, US waters, Australian waters, and recently in Shandong Peninsula in China. These Hematodinium pathogens were also reported to affect wild shrimp in Chinese waters and in the English North Sea. These rapid spreads affect crustacean aquaculture industries, where they are indeed a significant threat to the sustainability of the aquaculture of important crustaceans. The fishery products industries are also under pressure from the invasion of this pathogen, as the crab meat produced has a bitter taste, which may reduce its marketability. In response to these threats, this review was aimed at providing a broader understanding of the development of parasite distribution and ecological aspects of Hematodinium. In addition, the interaction of these pathogens with their hosts, the environmental drivers of Hematodinium disease, and future research perspectives were discussed.

Anisakis spp. (Nematoda: Anisakidae) parasitize a wide range of marine animals, mammals serving as the definitive host and different fish species as intermediate or paratenic hosts. In this study, 18 fish species were investigated for Anisakis infection. Katsuwonus pelamis, Euthynnus affinis, Caranx sp., and Auxis thazard were infected with high prevalence of Anisakis type I, while Cephalopholis cyanostigma and Rastrelliger kanagurta revealed low prevalence. The mean intensity of Anisakis larvae in K. pelamis and A. thazard was 49.7 and 5.6, respectively. A total of 73 Anisakis type I larvae collected from K. pelamis and A. thazard were all identified as Anisakis typica by PCR-RFLP analysis. Five specimens of Anisakis from K. pelamis and 15 specimens from A. thazard were sequenced using ITS1-5.8S-ITS2 region and 6 specimens from A. thazard and 4 specimens from K. pelamis were sequenced in mtDNA cox2 region. Alignments of the samples in the ITS region showed 2 patterns of nucleotides. The first pattern (genotype) of Anisakis from A. thazard had 100% similarity with adult A. typica from dolphins from USA, whereas the second genotype from A. thazard and K. pelamis had 4 base pairs different in ITS1 region with adult A. typica from USA. In the mtDNA cox2 regions, Anisakis type I specimens from A. thazard and K. pelamis showed similarity range from 94% to 99% with A. typica AB517571/DQ116427. The difference of 4 bp nucleotides in ITS1 regions and divergence into 2 subgroups in mtDNA cox2 indicating the existence of A. typica sibling species in the Makassar Strait.

This study describes recent infestations of ectoparasites on tilapia fingerlings in several hatcheries. High mortality of fingerlings and juvenile fish often occurs in hatcheries but the causative agents remain unclear. Nile tilapia fingerlings were obtained from three hatcheries in South Sulawesi, Indonesia. The fish were euthanized in clove oil, length and weight were measured, and the whole body and gills were examined for ectoparasites. Trichodinids were silver stained with 2% AgNO3, whereas monogeneans were placed in glycerin-alcohol and examined under a microscope. Other protozoans were placed directly on fresh mounts and identified under a compound microscope. The monogenean parasites were cleaned and placed in a microtube containing 70% alcohol for molecular identification. Trichodinid (Trichodina centrostrigeata, T. magna, Paratrichodina africana, Trichodinella sp., and Tripartiella sp.), and monogenean (Gyrodactylus sp. and Cichlidogyrus spp.) parasites were found in highly prevalent. The Cichlidogyrus specimens were morphologically identified as C. sclerosus, C. halli, and C. thurston, but only C. sclerosus and C. halli were confirmed based on partial 28S rRNA. The Gyrodactylus specimens were morphologically identified as G. cichlidarum and confirmed based on the ITS region. This is the first time that T. centrostrigeata and P. africana have been confirmed on fish from hatcheries and is also the first time that C. sclerosus, C. halli, and G. cichlidarum have been verified in Indonesian hatcheries based on a molecular technique. The high infestation rates of these parasites were likely caused by rearing the fish under stressful conditions, indicating low biosecurity and poor health management practices in the aquaculture facilities.

Infections with Streptococcus spp. were observed in Nile tilapia cultured in net cages in Lake Sentani, Papua, Indonesia. Clinical signs included exophthalmia, erratic swimming, ascites in abdominal cavity, and external hemorrhages. Four types of bacterial colonies (SK, K10, P20, and M12) were isolated from the brain, kidney, and eyes. Based on phenotypic and genetic (16S rDNA sequencing) characteristics, the isolates were identified as Streptococcus iniae (SK), Streptococcus agalactiae (K10 and P20) and Lactococcus garvieae (M12). The latter species has not been previously isolated or reported from fish streptococcosis in Indonesia. Intraperitoneal injection of healthy tilapia with the bacterial species caused significant morbidity (70%) within 3 days and 100% mortality at 6 days post injection. Experimental infections and reisolation of the bacteria from morbid and dead fish suggest they are the causative agents of streptococcosis, which rendered high mortality among cage cultured Nile tilapia in Lake Sentani. Our results suggest the need for developing diagnostic tools for accurate identification of the agents of streptococcosis. As tilapia aquaculture continues to expand as a means of food production and livelihood in Indonesia, it becomes crucial to ensure that fish resources are monitored and protected from the adverse effects of infectious diseases.

BACKGROUND: The Caligidae is a family of parasitic copepods containing over 30 recognised genera. They are commercially important parasites as they cause disease in numerous finfish aquaculture facilities globally. Morphological features are used to distinguish between the genera and Pseudocaligus has traditionally been differentiated from Caligus solely by the presence of a much reduced form of the fourth thoracic leg. Currently there are numerous DNA sequences available for Caligus spp. but only the type species, Pseudocaligus brevipedis, has molecular data available, so systematic studies using molecular phylogenetic analyses have been limited. METHODS: Three gene regions, SSU rDNA, 16S and CO1, for Pseudocaligus fugu from puffer fish from Japan and Pseudocaligus uniartus from rabbit fish from Indonesia are sequenced and molecular phylogenetic analyses performed in order to infer phylogenetic relationships between Pseudocaligus and other caligid copepods. RESULTS: The analysis revealed that there was no discrete grouping of Pseudocaligus spp. and that they had a polyphyletic distribution within Caligus taxa. Pseudocaligus fugu grouped with Caligus elongatus and contained a unique synapomorphy in the SSU rDNA region only seen in members of that clade. Pseudocaligus uniartus formed a well-supported group, in the SSU rDNA analyses, with a Caligus sp. that also infects rabbit fish, but was unresolved in the other analyses. Pseudocaligus brevipedis consistently and robustly grouped with Caligus curtus and C. centrodonti in all analyses. The majority of Lepeophtheirus spp. form a monophyletic sister group to the Caligus clade; however, L. natalensis is unresolved in all analyses and does not form part of the main Lepeophtheirus clade. CONCLUSIONS: These findings do not support the morphological-based distinction between Pseudocaligus and Caligus, suggesting that the reduced fourth leg is a feature that has evolved on multiple occasions throughout caligid evolution. Congruent molecular phylogenetic data support groupings based on the presence of morphological features, such as lunules, geography and host fish type rather than appendage morphology. Therefore, we support the synonymy of Pseudocaligus with Caligus.
