RESUMO

An innovative charge-transfer complex between the Schiff base 2-((2-hydroxybenzylidene) amino)-2-(hydroxymethyl) propane-1,3-diol [SAL-THAM] and the π-acceptor, chloranilic acid (CLA) within the mole ratio (1:1) was synthesized and characterized aiming to investigate its electronic transition spectra in acetonitrile (ACN), methanol (MeOH) and ethanol (EtOH) solutions. Applying Job`s method in the three solvents supported the 1:1 (CLA: SAL-THAM) mole ratio complex formation. The formation of stable CT- complex was shown by the highest values of charge-transfer complex formation constants, KCT, calculated using minimum-maximum absorbance method, with the sequence, acetonitrile > ethanol > methanol DFT study on the synthesized CT complex was applied based on the B3LYP method to evaluate the optimized structure and extract geometrical and reactivity parameters. Based on TD-DFT theory, the electronic properties, 1H and 13C NMR, IR, and UV-Vis spectra of the studied system in different solvents showing good agreement with the experimental studies. MEP map described the possibility of hydrogen bonding and charge transfer in the studied system. Finally, a computational approach for screening the antiviral activity of CT - complex towards SARS-CoV-2 coronavirus protease via molecular docking simulation was conducted and confirmed with molecular dynamic (MD) simulation.

BACKGROUND: Rheumatoid arthritis (RA) is a chronic inflammatory condition that impacts not only the musculoskeletal system but also various other systems in the body, including the cutaneous, ocular, respiratory, cardiovascular, and circulatory systems. MicroRNAs (miRNAs) are a class of naturally occurring and highly conserved transcripts that primarily function in the regulation of gene expression. They accomplish this by facilitating the degradation of messenger RNA (mRNA) or by repressing mRNA translation. miRNAs are well-known regulators of a variety of cellular processes. Therefore, we aimed to detect the impact of miR-155 rs767649 polymorphism on RA activity. METHODS: This case-control study included 66 Egyptian patients with RA who visited Al-Zhraa University Hospital, Internal Medicine Department, Cairo, Egypt, and 50 apparently healthy control subjects matched for age and sex. The participants were subjected to full clinical evaluation, including assessments of the disease activity score (DAS), erythrocyte sedimentation rate (ESR), liver and kidney function, anti-cyclic citrullinated peptide antibody (anti-CCP), and miR-155 polymorphism using real-time polymerase chain reaction (PCR). RESULTS: In the RA group, the majority (98.5%) were female, with a mean age of 43 years, while in the control group, 94% were female, with a mean age of 43.4 years. Comparison of laboratory parameters indicated significantly lower hemoglobin levels, higher ESR, and higher serum creatinine and anti-CCP levels in the RA group than in the control group. The RA group had a significantly higher frequency of TT genotypes and significantly lower frequencies of TA and TT genotypes than the control group. Considering the TT genotype and T allele as references, TA, AA, and TA/AA genotypes in the dominant model; AA in the recessive model; and A allele were significantly associated with protective effects against RA development (p<0.05, odds ratio<1). CONCLUSION: rs767649, the functional variant of miR-155, plays an important role in susceptibility to the increased risk of RA, suggesting that miR-155 can be used as a therapeutic target for the treatment of Egyptian patients with RA.

The present study was performed to evaluate the toxic effects of imidacloprid (IMI) insecticide on the growth performance, oxidative status, and immune response of Nile tilapia, Oreochromis niloticus (L.), and the protective role of dietary supplementation of spirulina, Arthrospira platensis, (SP). Fish (20.2 ± 0.5 g) were assigned to bifactorial design (2 IMI levels x 3 SP levels) to represent 6 treatments in triplicates. Spirulina was incorporated in diets at levels of 0.0 (control), 20, and 40 g/kg diet. Under each SP level, fish were exposed to 0.0 or 0.05 µg IMI/L. Fish in each treatment were fed on the corresponding diets up to apparent satiation thrice a day for 8 weeks. Two-way ANOVA revealed a significant decline in growth indices, hepatic superoxide dismutase, catalase, and glutathione peroxidase activities in the IMI-exposed fish. Contrariwise, serum alanine and aspartate aminotransferases, alkaline phosphatase, urea, creatinine, and malondialdehyde levels were markedly higher along with significant reductions of the reduced glutathione, nitric oxide as well as lysozyme values in the IMI-exposed fish group. The dietary supplementation of SP showed stimulating effects on the growth performance, haemato-biochemical, oxidants/antioxidants, and immune biomarkers of Nile tilapia with optimum level of 20 g SP/kg diet. Interestingly, the dietary supplementation of SP to Nile tilapia attenuated the above-mentioned variables with improving the growth performance, haemato-biochemical, oxidative stress, and immunity biomarkers. Therefore, the dietary supplementation of 20 g SP /kg diet could be a valuable candidate as a natural antioxidant for ameliorating the IMI toxicity in Nile tilapia.
