RESUMO

An ectoparasiticide combining three active ingredients [dinotefuran, permethrin and pyriproxyfen (DPP)] was used in mice in an experiment designed to evaluate its anti-feeding and insecticidal efficacy against Stegomyia albopicta (= Aedes albopictus) (Diptera: Culicidae) mosquitoes. Twenty-two adult mice were randomly allocated into two groups consisting of an untreated control group and a DPP-treated group. Mice were exposed individually for 1 h to a mean ± standard deviation of 27 ± 2 starved female mosquitoes on days 1, 7, 14, 21 and 28 post-treatment. At the end of the exposure (1 h), mosquitoes were assessed for immediate survival and engorgement status. Additionally, live mosquitoes in both groups were incubated separately and observed for mortality at 24 h after the end of the exposure. The anti-feeding efficacy of DPP after the 1-h exposure period was 99.2, 100, 98.0, 89.3 and 87.4% at 1, 7, 14, 21 and 28 days, respectively. Levels of insecticidal efficacy evaluated at 1 h and 24 h after exposure on days 1, 7, 14, 21 and 28 were 36.7, 28.9, 30.8, 23.1 and 11.9%, and 68.4, 45.0, 43.3, 37.9 and 19.9%, respectively. Based on the mouse model, the present study demonstrates that the DPP combination has significant anti-feeding and insecticidal efficacy against S. albopicta for at least 4 weeks.

This study, based on the rat model, was designed to explore the anti-feeding and insecticidal efficacy of a topical ectoparasiticide, dinotefuran-permethrin-pyriproxyfen (DPP), against Triatoma infestans (Hemiptera: Reduviidae), a vector of Trypanosoma cruzi (Trypanosomatida: Trypanosomatidae), for which dogs are domestic reservoir hosts. Twenty rats were divided into two equal groups: untreated and treated. Each rat was exposed under sedation to 16 T. infestans of mixed life stages for 1 h on days 1, 7, 14, 21 and 28 post-treatment. The anti-feeding and insecticidal effects of DPP were estimated after 1 h of exposure. Insecticidal efficacy was also assessed after incubation of the insects for 24 h post-exposure. Anti-feeding efficacy was 96.7, 84.7, 80.5, 81.5 and 42.6% on days 1, 7, 14, 21 and 28, respectively. Insecticidal efficacy evaluated at 1 and 24 h after exposure on days 1, 7, 14, 21 and 28 was 100, 91.2, 82.5, 80.0 and 29.1, and 100, 100, 100, 96.0 and 49.9%, respectively. This study demonstrates that a single administration of DPP spot-on treatment at a dose equivalent to the minimal recommended dose in rats has a powerful effect against T. infestans starting from day 1 that lasts for at least 3 weeks.

Ticks and fleas are vectors for numerous human and animal pathogens. Controlling them, which is important in combating such diseases, requires accurate identification, to distinguish between vector and non-vector species. Recently, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was applied to the rapid identification of arthropods. The growth of this promising tool, however, requires guidelines to be established. To this end, standardization protocols were applied to species of Rhipicephalus sanguineus (Ixodida: Ixodidae) Latreille and Ctenocephalides felis felis (Siphonaptera: Pulicidae) Bouché, including the automation of sample homogenization using two homogenizer devices, and varied sample preservation modes for a period of 1-6 months. The MS spectra were then compared with those obtained from manual pestle grinding, the standard homogenization method. Both automated methods generated intense, reproducible MS spectra from fresh specimens. Frozen storage methods appeared to represent the best preservation mode, for up to 6 months, while storage in ethanol is also possible, with some caveats for tick specimens. Carnoy's buffer, however, was shown to be less compatible with MS analysis for the purpose of identifying ticks or fleas. These standard protocols for MALDI-TOF MS arthropod identification should be complemented by additional MS spectrum quality controls, to generalize their use in monitoring arthropods of medical interest.

BACKGROUND: Bedbugs (Cimex lectularius) have been feeding on sleeping human beings since prehistory. In Europe, bed bugs were common and endemic until World War II when improved body and home hygiene, and widespread use of insecticides led to almost complete eradication. Current evidence indicates that bedbugs are making a comeback in Europe, USA, Canada and Australia. In our practice in Southern France, we observed several cases within a period of only 1 year. OBJECTIVES: Based on this experience, we conducted an epidemiological study to evaluate the status of bedbugs in France. METHODS: During summer 2009, we mailed a short questionnaire to all hospital professors in the CEDEF (Collège des Enseignants de Dermatologie de France) asking four questions: number of suspected diagnosis of bedbugs in the year 2009, and number of certain positive diagnosis, difficulties in treatment, use of a pest control professional for treatment, and finally personal opinion on actual incidence of bedbugs, compared with past years. RESULTS: Of the 84 questionnaires sent, there were only 26 responses despite two reminders. The responses were predominantly southern France, probably as a result of intensive immigration and increased travel and trade. Difficulties encountered during diagnosis and treatment are also mentioned. Utilizing the services of entomological experts and pest control professionals is essential. CONCLUSIONS: France has the same experience regarding the resurgence of bedbugs as several European countries, USA, Canada and Australia, especially the southern regions. This emerging health problem has to be known by dermatologists. A national programme has been launched in France to assess actual incidence and study C. lectularius- related diseases.

RESUMO

Bedbugs have been man's faithful companions from his cave days to the present time. After World War II, improvements in household and personal cleanliness and above all widespread use of insecticidal treatment led to a decline in the prevalence of bedbugs so that they became almost unknown to populations in industrialized countries. However in recent years a resurgence of bedbugs has been reported in North America as well as in most European countries. The purpose of this study is to provide an update on the comeback of bedbugs in France and the world, to discuss their impact on public health, and to consider possible causes for the current resurgence.

Arthropod vectors can transmit pathogenic microorganisms from one vertebrate to another during their blood meal. Although some vector-borne diseases have been eradicated in the Mediterranean area, such as malaria and dengue, recent endemic microorganisms (Toscana virus, Rickettsia spp.) remain neglected even though they cause many more cases. New diagnostic tools and innovative tools for the identification and characterization of vector species and microorganisms have been developed at IHU Méditerranée Infection, either internally or through collaborative and integrated projects. We have detected Rickettsia slovaca as a human pathogen and have described the disease; we have shown that Rickettsia felis can be transmitted by Anopheles mosquitoes; we have emphasized the increasing importance of bedbug (Cimex lectularius) as a potential vector of Bartonella quintana; and we have described the Toscana virus, a major agent of meningitis and meningoencephalitis which was disseminated in North Africa and Central and Eastern Europe, where it frequently cocirculates with a large number of newly described phleboviruses transmitted by sand flies.

RESUMO

Triatominae are biting hematophageous insects that have been wild vectors of the parasite Trypanosoma curzi for thousands of years. The arrival of man with his cortege of domestic animals and impact on the natural environment led these insects to adapt to the human environment so well that many species are now domesticated. Insect extermination programs have allowed satisfactory control of parasite transmission but have also promoted replacement of the exterminated species by species that were once semi-domestic or wild.

This paper reports on an entomological survey performed over the period 2009-2011 in endemic focus of peri-urban TOSV in South of France located from 24km east of Marseille. Sand flies were captured using CDC light traps set in sand fly resting places overnight, and temperature, relative humidity and wind were recorded to establish possible relations between meteorological factors and vector densities. The most common species, of 5,432 specimens collected and identified, was Phlebotomus perniciosus (74%), followed by Sergentomyia minuta (6%) and Phlebotomus ariasi (1%). Male flies were highly predominant for all Larroussius species instead of S. minuta which counted (85%) of females. The results shed light on the wide population's dynamic of P. perniciosus in France showing a diphasic seasonal trend with two abundance peaks at the beginning of July and late August, when a mean temperature is from 23.3 to 25.7°C. Interestingly, these two peaks are corresponding to the peaks of occurrence of human TOSV cases. Among the 1724 females collected, 549 (32%) were blood-fed. Based on the results of blood meal analyses, P. perniciosus fed on large animal's diversity (man, chicken, rabbit, others mammalians, etc.), including bats that are the only species found naturally infected by TOSV. Results indicate that host choice was probably related to its availability than specific attractiveness. Data presented confirm that sand flies easily adapted to the periurban sites like, P. perniciosus may represent a public health concern for pathogen transmission in similar Mediterranean environments.

The discovery of two atypical specimens of Rhodnius pictipes Stål, 1872 in French Guiana and the examination of the female holotype of R. amazonicus Almeida, Santos & Sposina, 1973, the only specimen of this species so far known, lead us to propose the rehabilitation of R. amazonicus synonymized with R. pictipes. The male is described for the first time and the female redescribed. Both external characters and genitalia distinguish R. amazonicus from R. pictipes. R. amazonicus shows affinities not only with pictipes but also with R. stali Lent, Jurberg & Galvão, 1993 and R. paraensis Sherlock, Guitton & Miles, 1977. A key is provided for these four species forming, in all likelihood, a natural group, i.e. the "pictipes group".
