RESUMO

Although primary metabolism is well conserved across species, it is useful to explore the specificity of its network to assess the extent to which some pathways may contribute to particular outcomes. Constraint-based metabolic modelling is an established framework for predicting metabolic fluxes and phenotypes and helps to explore how the plant metabolic network delivers specific outcomes from temporal series. After describing the main physiological traits during fruit development, we confirmed the correlations between fruit relative growth rate (RGR), protein content and time to maturity. Then a constraint-based method is applied to a panel of eight fruit species with a knowledge-based metabolic model of heterotrophic cells describing a generic metabolic network of primary metabolism. The metabolic fluxes are estimated by constraining the model using a large set of metabolites and compounds quantified throughout fruit development. Multivariate analyses showed a clear common pattern of flux distribution during fruit development with differences between fast- and slow-growing fruits. Only the latter fruits mobilise the tricarboxylic acid cycle in addition to glycolysis, leading to a higher rate of respiration. More surprisingly, to balance nitrogen, the model suggests, on the one hand, nitrogen uptake by nitrate reductase to support a high RGR at early stages of cucumber and, on the other hand, the accumulation of alkaloids during ripening of pepper and eggplant. Finally, building virtual fruits by combining 12 biomass compounds shows that the growth-defence trade-off is supported mainly by cell wall synthesis for fast-growing fruits and by total polyphenols accumulation for slow-growing fruits.

In recent years, alongside the conventional screening procedures for the evaluation of probiotics for human usage, the pharmaceutical and food industries have encouraged scientific research towards the selection of new probiotic bacterial strains with particular functional features. Therefore, this study intended to explore novel functional properties of five Lactiplantibacillus plantarum strains isolated from bee bread. Specifically, antioxidant, antimicrobial and ß-glucosidase activities, exopolysaccharides (EPS) production and the ability to synthesize γ-aminobutyric acid (GABA) were evaluated. The results demonstrated that the investigated L. plantarum strains were effective in inhibiting the growth of some human opportunistic pathogens in vitro (Pseudomonas aeruginosa, Escherichia coli, Proteus mirabilis, Enterococcus faecalis and Staphylococcus aureus). Moreover, the evaluation of antioxidant and ß-glucosidase activity and of EPS and GABA production, revealed a different behavior among the strains, testifying how these properties are strongly strain-dependent. This suggests that a careful selection within a given species is important in order to identify appropriate strains for specific biotechnological applications. The results highlighted that the five strains of L. plantarum are promising candidates for application as dietary supplements in the human diet and as microbial cultures in specific food productions.

BACKGROUND: Drought stress is among the most serious threats jeopardizing the economic yield of crop plants in Iran. In particular, in response to withholding irrigation, the reduction in performance and quality of a precious plant such as the olive tree is remarkable. Therefore, the selection of cultivars that are resistant or tolerant to drought has been recognized as one of the most effective long-term strategies for sustainably alleviating the adverse effects of this stress. In this view, our study evaluated the response of 8 olive cultivars including 4 elite native cultivars (Zard Aliabad, Roughani, Dezful, and Shengeh) and 4 foreign cultivars (Manzanilla, Sevillana, Konservolia, and Mission) to water shortage in the Dallaho Olive Research station of Sarpole-Zahab in Kermanshah province in 2020. Olive trees underwent 3 levels of irrigation treatment including 100% full irrigation (control), 75%, and 50% deficit irrigation. RESULTS: Based on the results, 50% deficit irrigation decreased both growth and pomological traits, but determined the highest dry matter percentage. As the severity of drought stress increased, with an accumulation of sodium and malondialdehyde, an incremental increase in osmolytes was observed, as well as an enhancement of the activity of antioxidant enzymes (peroxidase and catalase). In contrast, full irrigation led to an increase in photosynthetic pigments, calcium, and potassium. Dezful and Konservolia cultivars revealed a significantly higher growth rate, correlated in the former to higher levels of chlorophyll, compatible compounds, total phenolic content, relative water content, potassium to sodium ratio, catalase, and peroxidase activities compared with other cultivars. Konservolia showed the best yield parameters under 75% and 100% irrigation regimes, correlated to higher chlorophyll, potassium, and total phenolic content (in particular at 75% ET). CONCLUSIONS: Generally, the selection of more resilient or tolerant cultivars to sustain water scarcity stress is a widely operative solution to extend rainfed orchards in semi-arid environments. Our study showed that Dezful and Konservolia had the best adaptive mechanisms to cope with the detrimental effects of drought stress.

C4 photosynthesis concentrates CO2 around Rubisco in the bundle sheath, favouring carboxylation over oxygenation and decreasing photorespiration. This complex trait evolved independently in >60 angiosperm lineages. Its evolution can be investigated in genera such as Flaveria (Asteraceae) that contain species representing intermediate stages between C3 and C4 photosynthesis. Previous studies have indicated that the first major change in metabolism probably involved relocation of glycine decarboxylase and photorespiratory CO2 release to the bundle sheath and establishment of intercellular shuttles to maintain nitrogen stoichiometry. This was followed by selection for a CO2-concentrating cycle between phosphoenolpyruvate carboxylase in the mesophyll and decarboxylases in the bundle sheath, and relocation of Rubisco to the latter. We have profiled 52 metabolites in nine Flaveria species and analysed 13CO2 labelling patterns for four species. Our results point to operation of multiple shuttles, including movement of aspartate in C3-C4 intermediates and a switch towards a malate/pyruvate shuttle in C4-like species. The malate/pyruvate shuttle increases from C4-like to complete C4 species, accompanied by a rise in ancillary organic acid pools. Our findings support current models and uncover further modifications of metabolism along the evolutionary path to C4 photosynthesis in the genus Flaveria.

Photoperiodic light quality affects flowering of long day plants, by influencing the phytochrome photoequilibria (PPE) at plant level; however, the most effective light spectrum to promote flowering is still unknown for most of the flower crops. We evaluated the influence of light spectrum of three light sources, with different induced PPE, on photosynthesis, metabolic profiling, plant growth and flowering in two hybrids of Ranunculus asiaticus L., MBO (early flowering) and MDR (medium earliness). Three photoperiodic treatments were compared to natural day length (NL): white fluorescent light (PPE 0.84), light emitting diodes (LEDs) with red:far red (R:FR) light at 3:1 ratio (PPE, 0.84) and LEDs with R:FR light at 1:3 ratio (PPE 0.63). Under natural light, net photosynthesis was higher in MDR than in MBO, while photochemistry was similar in the hybrids. Compared to NL, photoperiodic treatments did not affect net photosynthesis, while they promoted the quantum yield of PSII and reduced the non-photochemical quenching. Under NL, plant growth was greater in MBO, while flowering started earlier in MDR and flowers characteristics were similar in the hybrids. Despite the greater sensitivity of MDR plants in terms of metabolism, photoperiodic lighting improved plant growth and reduced the flowering time only in MBO, with a stronger effect under R:FR 3:1 light. MDR plants were characterized by higher soluble sugars, polyphenols, photosynthetic pigments and proteins, while MBO plants by higher starch and amino acid content. The morphological effects of photoperiodic light quality and the hybrid-specific response should be taken into account to optimize lighting protocols in commercial farms.

Consumer demand for fresh and functional horticultural products is on the rise. Perilla frutescens, L. Britt (Lamiaceae) is a potential specialty/niche crop for consumption and therapeutic uses with high contents of phenolic and volatile compounds. Plant growth, mineral composition, polyphenol profile and aroma volatile components of two perilla genotypes in response to salinity (non-salt control, 10, 20 or 30 mM NaCl) applied as chemical eustressor were assessed. Salinity suppressed growth and yield of both genotypes, although the red-pigmented genotype was less sensitive than the green-pigmented one. Mild (10 mM NaCl) and moderate (20 and 30 mM NaCl) salinity suppressed foliar potassium, magnesium, nitrate and chlorophyll a concentrations of both genotypes and increased the levels of rosmarinic acid, total polyphenols and target aroma volatile components. Green perilla showed higher yield and biomass production and higher content of protein, dry matter, calcium, magnesium, perilla ketone and cis-jasmone, whereas red perilla exhibited higher content of potassium, chlorophyll a, rosmarinic acid, total polyphenols, perilla aldehyde and benzaldehyde. Our findings support that chemical eustressors such as mild to moderate salinity offer valuable means to manipulate phytochemical and aroma profiles.

Plants are exposed to varying irradiance and temperature within a day and from day to day. We previously investigated metabolism in a temperature-controlled greenhouse at the spring equinox on both a cloudy and a sunny day [daily light integral (DLI) of 7 mol m-2 d-1 and 12 mol m-2 d-1]. Diel metabolite profiles were largely captured in sinusoidal simulations at similar DLIs in controlled-environment chambers, except that amino acids were lower in natural light regimes. We now extend the DLI12 study by investigating metabolism in a natural light regime with variable temperature including cool nights. Starch was not completely turned over, anthocyanins and proline accumulated, and protein content rose. Instead of decreasing, amino acid content rose. Connectivity in central metabolism, which decreased in variable light, was not further weakened by variable temperature. We propose that diel metabolism operates better when light and temperature are co-varying. We also compared transcript abundance of 10 circadian clock genes in this temperature-variable regime with the temperature-controlled natural and sinusoidal light regimes. Despite temperature compensation, peak timing and abundance for dawn- and day-phased genes and GIGANTEA were slightly modified in the variable temperature treatment. This may delay dawn clock activity until the temperature rises enough to support rapid metabolism and photosynthesis.

Irradiance from sunlight changes in a sinusoidal manner during the day, with irregular fluctuations due to clouds, and light-dark shifts at dawn and dusk are gradual. Experiments in controlled environments typically expose plants to constant irradiance during the day and abrupt light-dark transitions. To compare the effects on metabolism of sunlight versus artificial light regimes, Arabidopsis thaliana plants were grown in a naturally illuminated greenhouse around the vernal equinox, and in controlled environment chambers with a 12-h photoperiod and either constant or sinusoidal light profiles, using either white fluorescent tubes or light-emitting diodes (LEDs) tuned to a sunlight-like spectrum as the light source. Rosettes were sampled throughout a 24-h diurnal cycle for metabolite analysis. The diurnal metabolite profiles revealed that carbon and nitrogen metabolism differed significantly between sunlight and artificial light conditions. The variability of sunlight within and between days could be a factor underlying these differences. Pairwise comparisons of the artificial light sources (fluorescent versus LED) or the light profiles (constant versus sinusoidal) showed much smaller differences. The data indicate that energy-efficient LED lighting is an acceptable alternative to fluorescent lights, but results obtained from plants grown with either type of artificial lighting might not be representative of natural conditions.

Durum wheat plants are extremely sensitive to drought and salinity during seedling and early development stages. Their responses to stresses have been extensively studied to provide new metabolic targets and improving the tolerance to adverse environments. Most of these studies have been performed in growth chambers under low light [300-350 µmol m-2 s-1 photosynthetically active radiation (PAR), LL]. However, in nature plants have to face frequent fluctuations of light intensities that often exceed their photosynthetic capacity (900-2000 µmol m-2 s-1 ). In this study we investigated the physiological and metabolic changes potentially involved in osmotic adjustment and antioxidant defense in durum wheat seedlings under high light (HL) and salinity. The combined application of the two stresses decreased the water potential and stomatal conductance without reducing the photosynthetic efficiency of the plants. Glycine betaine (GB) synthesis was inhibited, proline and glutamate content decreased, while γ-aminobutyric acid (GABA), amides and minor amino acids increased. The expression level and enzymatic activities of Δ1-pyrroline-5-carboxylate synthetase, asparagine synthetase and glutamate decarboxylase, as well as other enzymatic activities of nitrogen and carbon metabolism, were analyzed. Antioxidant enzymes and metabolites were also considered. The results showed that the complex interplay seen in durum wheat plants under salinity at LL was simplified: GB and antioxidants did not play a main role. On the contrary, the fine tuning of few specific primary metabolites (GABA, amides, minor amino acids and hexoses) remodeled metabolism and defense processes, playing a key role in the response to simultaneous stresses.

Trehalose 6-phosphate (Tre6P), the intermediate of trehalose biosynthesis, has a profound influence on plant metabolism, growth, and development. It has been proposed that Tre6P acts as a signal of sugar availability and is possibly specific for sucrose status. Short-term sugar-feeding experiments were carried out with carbon-starved Arabidopsis thaliana seedlings grown in axenic shaking liquid cultures. Tre6P increased when seedlings were exogenously supplied with sucrose, or with hexoses that can be metabolized to sucrose, such as glucose and fructose. Conditional correlation analysis and inhibitor experiments indicated that the hexose-induced increase in Tre6P was an indirect response dependent on conversion of the hexose sugars to sucrose. Tre6P content was affected by changes in nitrogen status, but this response was also attributable to parallel changes in sucrose. The sucrose-induced rise in Tre6P was unaffected by cordycepin but almost completely blocked by cycloheximide, indicating that de novo protein synthesis is necessary for the response. There was a strong correlation between Tre6P and sucrose even in lines that constitutively express heterologous trehalose-phosphate synthase or trehalose-phosphate phosphatase, although the Tre6P:sucrose ratio was shifted higher or lower, respectively. It is proposed that the Tre6P:sucrose ratio is a critical parameter for the plant and forms part of a homeostatic mechanism to maintain sucrose levels within a range that is appropriate for the cell type and developmental stage of the plant.

The rationale for this data article is to provide resources which could facilitate the studies focussed over weed detection and segmentation in precision farming using computer vision. We have curated Multispectral (MS) images over crop fields of Triticum Aestivum containing heterogenous mix of Raphanus raphanistrum in both uniform and random crop spacing. This dataset is designed to facilitate weed detection and segmentation based on manual and automatically annotated Raphanus raphanistrum, commonly known as wild radish. The dataset is publicly available through the Zenodo data library and provides annotated pixel-level information that is crucial for registration and segmentation purposes. The dataset consists of 85 original MS images captured over 17 scenes covering various spectra including Blue, Green, Red, NIR (Near-Infrared), and RedEdge. Each image has a dimension of 1280 × 960 pixels and serves as the basis for the specific weed detection and segmentation. Manual annotations were performed using Visual Geometry Group Image Annotator (VIA) and the results were saved in Common Objects in Context (COCO) segmentation format. To facilitate this resource-intensive task of annotation, a Grounding DINO + Segment Anything Model (SAM) was trained with this manually annotated data to obtain automated Visual Object Classes Extended Markup Language (PASCAL VOC) annotations for 80 MS images. The dataset emphasizes quality control, validating both the 'manual" and 'automated" repositories by extracting and evaluating binary masks. The codes used for these processes are accessible to ensure transparency and reproducibility. This dataset is the first-of-its-kind public resource providing manual and automatically annotated weed information over close-ranged MS images in heterogenous agriculture environment. Researchers and practitioners in the fields of precision agriculture and computer vision can use this dataset to improve MS image registration and segmentation at close range photogrammetry with a focus on wild radish. The dataset not only helps with intra-subject registration to improve segmentation accuracy, but also provides valuable spectral information for training and refining machine learning models.

This article is focused on the comprehensive evaluation of alleyways to scale-invariant feature transform (SIFT) and random sample consensus (RANSAC) based multispectral (MS) image registration. In this paper, the idea is to extensively evaluate three such SIFT- and RANSAC-based registration approaches over a heterogenous mix containing Triticum aestivum crop and Raphanus raphanistrum weed. The first method is based on the application of a homography matrix, derived during the registration of MS images on spatial coordinates of individual annotations to achieve spatial realignment. The second method is based on the registration of binary masks derived from the ground truth of individual spectral channels. The third method is based on the registration of only the masked pixels of interest across the respective spectral channels. It was found that the MS image registration technique based on the registration of binary masks derived from the manually segmented images exhibited the highest accuracy, followed by the technique involving registration of masked pixels, and lastly, registration based on the spatial realignment of annotations. Among automatically segmented images, the technique based on the registration of automatically predicted mask instances exhibited higher accuracy than the technique based on the registration of masked pixels. In the ground truth images, the annotations performed through the near-infrared channel were found to have a higher accuracy, followed by green, blue, and red spectral channels. Among the automatically segmented images, the accuracy of the blue channel was observed to exhibit a higher accuracy, followed by the green, near-infrared, and red channels. At the individual instance level, the registration based on binary masks depicted the highest accuracy in the green channel, followed by the method based on the registration of masked pixels in the red channel, and lastly, the method based on the spatial realignment of annotations in the green channel. The instance detection of wild radish with YOLOv8l-seg was observed at a mAP@0.5 of 92.11% and a segmentation accuracy of 98% towards segmenting its binary mask instances.

Olive trees are known as one of the most iconic crops in the world. Considering the increasing water deficit worldwide, implementing some profitable and empirical strategies can be inevitable upon exposure to drought stress. Therefore, the present study aimed at clarifying the beneficial role of exogenously foliar application of Fe2O3 modified carbon nitride nanostructures (control, FeSO4, C3N4 and Fe2O3/g-C3N4) to "Shengeh" olive cultivars grown at different watering levels (100, 75, and 50% ET) in two experimental years (2022 and 2023) and the pomological attributes, physiological and biochemical changes happening in the treated leaves and fruits were discussed. The results indicated that drought stress caused a significant decline in pomological attributes in this experiment, and treatments could remarkably make up for this damage. Overall, Fe2O3/g-C3N4 outperformed as compared FeSO4 and C3N4 alone, which were also efficacious in conferring tolerance to the water deficit stress. Conversely, severe drought stressed-olive fruits showed higher oil content percent in the fresh matter and water use efficiency (WUE) in oil by 30% and 52.5%, respectively, as an average of results of two years, and after Fe2O3/g-C3N4, these features in olive plants subjected to severe drought improved by an average of 35% over two years. Ca2+ and K+ in olive plants under severe drought stress declined by 50% and 83% in 2022 and 46% and 24% in 2023, while Na+ increased in the plants exposed to 50%ET stress by 48% and 57% in two successive experimental years respectively. The application of Fe2O3/g-C3N4 remarkably improved the contents of Ca2+ and K+ by 101.5% and 369%, respectively, as an average of two years. Conversely, this beneficial treatment led to a significant decline in Na+ levels by 30% in 2022 and 2% in 2023 under stressful conditions. Moreover, it decreased the 'osmolytes' content, caused a smaller decline in chlorophyll levels, and resulted in higher relative water content occurring in the treated olive leaves. The reduction of oxidative markers was a result of the increased enzymatic activity after the use of Fe2O3/g-C3N4. Therefore, this treatment is a promising strategy to achieve improved resistance in olive plants in the future.

This research introduces an extensive dataset of unprocessed aerial RGB images and orthomosaics of Brassica oleracea crops, captured via a DJI Phantom 4. The dataset, publicly accessible, comprises 244 raw RGB images, acquired over six distinct dates in October and November of 2020 as well as 6 orthomosaics from an experimental farm located in Portici, Italy. The images, uniformly distributed across crop spaces, have undergone both manual and automatic annotations, to facilitate the detection, segmentation, and growth modelling of crops. Manual annotations were performed using bounding boxes via the Visual Geometry Group Image Annotator (VIA) and exported in the Common Objects in Context (COCO) segmentation format. The automated annotations were generated using a framework of Grounding DINO + Segment Anything Model (SAM) facilitated by YOLOv8x-seg pretrained weights obtained after training manually annotated images dated 8 October, 21 October, and 29 October 2020. The automated annotations were archived in Pascal Visual Object Classes (PASCAL VOC) format. Seven classes, designated as Row 1 through Row 7, have been identified for crop labelling. Additional attributes such as individual crop ID and the repetitiveness of individual crop specimens are delineated in the Comma Separated Values (CSV) version of the manual annotation. This dataset not only furnishes annotation information but also assists in the refinement of various machine learning models, thereby contributing significantly to the field of smart agriculture. The transparency and reproducibility of the processes are ensured by making the utilized codes accessible. This research marks a significant stride in leveraging technology for vision-based crop growth monitoring.

The increase in the frequency and magnitude of environmental stresses poses a significant risk to the stability of food supplies. In coastal areas of the Mediterranean, brackish water has long been considered a limitation on horticultural production. In this scenario, the use of biochar in agriculture could be considered a valuable tool to cope with the deleterious effects of salt stress. This work aimed to investigate, in a protected environment, the effects of different concentrations of biochar (0, 1, and 2% v/v) obtained from poplar (Populus L.) biomass on the yield and quality of dwarf San Marzano ecotype tomatoes irrigated with saline water at different concentrations of NaCl (0, 40 and 80 mM). The increase in salt concentration from 0 to 80 mM NaCl reduced the total yield (-63%) and the number of fruits (-25%), but improved the main quality parameters such as dry matter (+75%), total soluble solids (+56%), and polyphenol content (+43%). Compared to control conditions, biochar supplementation improved the total yield (+23%) and number of fruits (+26%) without altering the functional and organoleptic characteristics of the fruits. The promising results underscore the potential of biochar as a sustainable solution to amend soils in order to improve tomato production under unfavorable conditions such as high salinity. However, there is a need to clarify which adaptation mechanisms triggered by biochar amending improve production responses even and especially under suboptimal growing conditions.

Nursery growing as well as common landscape hydrangeas are all susceptible to leaf spot fungus Cercospora hydrangeae. Warm and rainy weather causes the fungal spores to germinate quickly and spread over the plant leaves forming small purple or brown spots. Although Hydrangea plants are not killed by leaf spot, it detracts from the value of plants through the reduction of flowering and plant vigor. The aim of our study was to isolate, characterize and investigate the expression profile of Hydrangea macrophylla resistance (R) gene transcripts under C. hydrangeae fungus infection and examine their evolutionary relationships by phylogenetic analysis. R-genes are thought to be one of the components of the genetic resistance in plants and most of them encode nucleotide binding site-leucine rich repeat (NBS-LRR) proteins. A cDNA-NBS strategy was carried out using as template cDNAs isolated from control and infected plant leaves. The cDNA-NBS profiling gave an excellent bands reproducibility. Twenty new transcripts corresponding to NBS-LRR proteins were identified only in infected plants. The extent of positivity between the aminoacid sequences at NBS region varied from 45 to 90 %, which indicates the diversity among the RGAs. The results of this paper will provide a genomic framework for the further isolation of candidate disease resistance NBS-encoding genes in Hortensia, and contribute to the understanding of the evolutionary mode of NBS-encoding genes in Hydrangeaceae crops.

Climate change and rising global average temperatures across the year may strongly affect olive fruits' development process and their oil yield and quality. There is therefore an urgency to take immediate actions to characterize the wide variability of cultivars in order to identify those with a stable response to high temperatures, particularly in areas like the west of Iran, which is characterized by a warm summer continental climate. The objective of this study is to investigate the process of fruit development and oil accumulation in response to high summer temperature conditions in a set of four Iranian olive cultivars (Shengeh, Roughani, Zard Aliabad, and Dezful) in comparison with four foreign olive cultivars (Konservolia, Sevillana, Manzanilla, and Mission) in seven various harvesting times (20 July, 5 and 20 August, 5 and 20 September, 6 and 21 October). The obtained results evidence a significant positive correlation between fruit dry matter and oil content. High temperatures reduced the oil and dry matter accumulation in the second half of the summer, with severe thermal conditions adversely affecting oil synthesis. Paramount variations were observed among the cultivars regarding oil accumulation, dry matter, and pomological attributes. All of them showed the highest oil content at the last harvest. Among all analyzed varieties, Roughani showed the highest tolerance and adaptive capacity to high temperatures as it accumulated the greatest amount of dry matter as well as oil content in all of the harvesting times, demonstrating a positive correlation between these two traits. Although Shengeh showed the lowest oil content on a dry and fresh weight basis at the first harvesting time, this cultivar generally presented higher fruit development attributes than the other cultivars, highlighting that it benefits from a high temperature.

Ranunculus asiaticus L. is an ornamental geophyte. In commercial practice, it is mainly propagated by rehydrated tuberous roots. Vernalization before planting is a common practice to overcome the natural dormancy of tuberous roots; however, little is known about the mechanisms underlying the plant's response to low temperatures. We investigated the influence of three preparation procedures of tuberous roots, only rehydration (control, C), and rehydration plus vernalization at 3.5 °C for 2 weeks (V2) and for 4 weeks (V4), on plant growth, leaf photosynthesis, flowering, and metabolism in plants of two hybrids, MBO (early flowering, pale orange flower) and MDR (medium earliness, bright orange flower), grown in pots in an unheated greenhouse. We reported the responses observed in the aerial part in a previous article in this journal. In this paper, we show changes in the underground organs in carbohydrate, amino acids, polyphenols, and protein levels throughout the growing cycle in the different plant stages: pre-planting, vegetative growth, and flowering. The metabolic profile revealed that the two hybrids had different responses to the root preparation procedure. In particular, MBO synthesized GABA and alanine after 2 weeks and sucrose after 4 weeks of vernalization. In contrast, MDR was more sensitive to vernalization; in fact, a higher synthesis of polyphenols was observed. However, both hybrids synthesized metabolites that could withstand exposure to low temperatures.

In Ranunculus asiaticus L., vernalization of propagation material is a common practice for the production scheduling of cut flowers, however little is known about the plant physiology and metabolism of this species as affected by cold treatments. We investigated the influence of two hybrids, MBO and MDR, and three preparation procedures of tuberous roots, only rehydration (control, C), and rehydration plus vernalization at 3.5 °C for 2 weeks (V2) and for 4 weeks (V4), on plant growth and flowering, leaf photosynthesis, and leaf metabolic profile in plants grown in pot in a cold greenhouse. Net photosynthesis (NP) was higher in MDR than in MBO. In the two genotypes, the NP did not change in V2 and increased in V4 compared to C in MBO, while was unaffected by vernalization in MDR. Quantum yield of PSII electron transport (ΦPSII), linear electron transport rate (ETR) and non-photochemical quenching (NPQ) did not differ in the two hybrids, whereas maximal PSII photochemical efficiency (Fv/Fm) was higher in MBO than in MDR. Fluorescence indexes were unaffected by the preparation procedure, except for ETR, which decreased in V2 compared to C and V4 in MDR. A significant interaction between genotype and preparation procedure was found in plant leaf area, which was reduced only in V4 in MBO, while decreased in both the vernalization procedures in MDR. In Control plants, flowering started in 65 days in MBO and 69 days in MDR. Compared to controls, both the vernalization treatments anticipated flowering in MDR, while they were detrimental or only slightly efficient in promoting flowering in MBO. Vernalization always reduced the quality of flower stems in both the hybrids.

In addition to be used as a plant protection agent, copper (Cu) is also an essential micronutrient for plant growth and development. The bioavailability of Cu in agricultural systems can be limited due to its specific physical-chemical characteristics, leading to imbalances in plant production. To address this issue, an experimental trial was conducted on Genovese basil (Ocimum basilicum L.) in protected conditions to comparatively evaluate the effects of a vegetable protein hydrolysate (VPH), free Cu and Cu complexed with peptides and amino acids of vegetal origin (Cu and Cu-VPH, respectively), and a combination of VPH and Cu-VPH (VPH+Cu-VPH). The study showed that the combined application of VPH+Cu-VPH led to a significant average increase of 16.3% in fresh yield compared to the untreated Control and Cu treatment. This finding was supported by an improved photosynthetic performance in ACO2 (+29%) and Fv/Fm (+7%). Furthermore, mineral analysis using ICP OES demonstrated that Cu and Cu-VPH treatments determined, on average, a 15.1-, 16.9-, and 1.9-fold increase in Cu in plant tissues compared to control, VPH, and VPH+Cu-VPH treatments, respectively. However, the VPH+Cu-VPH treatment induced the highest contents of the other analyzed ions, except for P. In particular, Mg, Mn, Ca, and Fe, which take part in the constitution of chlorophylls, water splitting system, and photosynthetic electron transport chain, increased by 23%, 21%, 25%, and 32% compared to respective controls. Indeed, this improved the photosynthetic efficiency and the carboxylation capacity of the plants, and consequently, the physiological and productive performance of Genovese basil, compared to all other treatments and control. Consistently, the untargeted metabolomics also pointed out a distinctive modulation of phytochemical signatures as a function of the treatment. An accumulation of alkaloids, terpenoids, and phenylpropanoids was observed following Cu treatment, suggesting an oxidative imbalance upon metal exposure. In contrast, a mitigation of oxidative stress was highlighted in Cu-VPH and VPH+Cu-VPH, where the treatments reduced stress-related metabolites. Overall, these results highlight an interaction between Cu and VPH, hence paving the way towards the combined use of Cu and biostimulants to optimize agronomic interventions.
