RESUMO

During a recent marine biological expedition to the Northeast Greenland shelf break (latitudes 74-77 °N), we made the first discovery of Atlantic cod (Gadus morhua), beaked redfish (Sebastes mentella) and capelin (Mallotus villosus). Our novel observations shift the distribution range of Atlantic cod >1000 km further north in East Greenland waters. In light of climate change, we discuss physical forcing and putative connections between the faunas of the Northeast Greenland shelf and the Barents Sea. We emphasise the importance of using real data in spread scenarios for understudied Arctic seas.

RESUMO

In light of ocean warming and loss of Arctic sea ice, harvested marine fishes of boreal origin (and their fisheries) move poleward into yet unexploited parts of the Arctic seas. Industrial fisheries, already in place on many Arctic shelves, will radically affect the local fish species as they turn up as unprecedented bycatch. Arctic marine fishes are indispensable to ecosystem structuring and functioning, but they are still beyond credible assessment due to lack of basic biological data. The time for conservation actions is now, and precautionary management practices by the Arctic coastal states are needed to mitigate the impact of industrial fisheries in Arctic waters. We outline four possible conservation actions: scientific credibility, 'green technology', legitimate management and overarching coordination.

The ice cod Arctogadus glacialis (Peters, 1872) is one of the few fish species endemic to the Arctic. With a circumpolar distribution, the species is confined to the fjords and shelves of the Arctic seas. Biological information on A. glacialis is scarce, with genomic information restricted to microsatellites. Within the frame of the TUNU-Programme: Arctic Ocean Fishes-Diversity, Adaptation and Conservation, we studied A. glacialis at the chromosomal level to explore fish diversity and evolutionary aspects. The analysis of over 50 individuals from the Northeast Greenland fjords between latitudes 71°09' N and 76°42' N revealed a remarkable intraspecific diversity epitomized by chromosome numbers spanning from 28 to 33, the occurrence of putative B chromosomes, and diversified patterns of distribution of heterochromatin and rDNAs. The number of B chromosomes followed a latitudinal gradient from 0-2 in the north to 2-5 in the south. Considering the benthic and rather stationary life history of this species, the observed chromosomal differences might have arisen independently, possibly driven and/or fostered by the dynamics of repetitive sequences, and are being fixed in relatively isolated fjord populations. The resulting latitudinal cline we observe today might have repercussions on the fate of local populations facing the ongoing climate-driven environmental changes.

The Greenland shark (Somniosus microcephalus, Squaliformes: Somniosidae) is a long-lived Arctic top predator, which in combination with the high historical and modern fishing pressures, has made it subject to increased scientific focus in recent years. Key aspects of reproduction are not well known as exemplified by sparse and contradictory information e.g. on birth size and number of pups per pregnancy. This study represents the first comprehensive work on Greenland shark reproductive biology based on data from 312 specimens collected over the past 60 years. We provide guidelines quantifying reproductive parameters to assess specific maturation stages, as well as calculate body length-at-maturity (TL50) which was 2.84±0.06 m for males and 4.19±0.04 m for females. From the available information on the ovarian fecundity of Greenland sharks as well as a meta-analysis of Squaliform reproductive parameters, we estimate up to 200-324 pups per pregnancy (depending on maternal size) with a body length-at-birth of 35-45 cm. These estimates remain to be verified by future observations from gravid Greenland sharks.

The olfactory organ of Chondrichthyes is characterized by a central support with several lamellae covered by a sensory olfactory epithelium. Although secondary folds are present on the lamellae in all the chondrichthyan species analyzed to date, their shape and size have not been described. We here analyze the olfactory organ of 13 elasmobranch and 1 holocephalan species, describe the shape of the secondary folds and evaluate how they contribute to the epithelial surface area. The secondary folds vary in shape and size, and they should always be considered when comparing the epithelial surface area among species; in fact, the increase of the area, due to the presence of the secondary folds, spans from 70 to 495% in the considered species. Because of the complexity of the shapes, we approach the description of the secondary folds by analyzing histological sections of the olfactory lamellae to obtain illustrative silhouettes. We introduce two indexes in order to describe a 2D-sectioned shape of the secondary folds. Considering the different numerical parameters which describe the morphology of the olfactory organ (secondary folds included), a principal component analysis elucidates the possible ecological role and phylogenetic relationship of the chondrichthyan olfactory organ.

As a result of ocean warming, the species composition of the Arctic seas has begun to shift in a boreal direction. One ecosystem prone to fauna shifts is the Northeast Greenland shelf. The dispersal route taken by boreal fauna to this area is, however, not known. This knowledge is essential to predict to what extent boreal biota will colonise Arctic habitats. Using population genetics, we show that Atlantic cod (Gadus morhua), beaked redfish (Sebastes mentella), and deep-sea shrimp (Pandalus borealis) recently found on the Northeast Greenland shelf originate from the Barents Sea, and suggest that pelagic offspring were dispersed via advection across the Fram Strait. Our results indicate that boreal invasions of Arctic habitats can be driven by advection, and that the fauna of the Barents Sea can project into adjacent habitats with the potential to colonise putatively isolated Arctic ecosystems such as Northeast Greenland.

The Arctic is a unique and fragile ecosystem that needs to be preserved and protected. Despite its remoteness, plastic pollution has been documented in this region. In the coming years, it is likely to worsen since, with climate changes and the opening of new shipping routes, the human presence is going to increase in the whole area. Here, we investigated the presence of microplastics (MPs) in sub-surface water and in two mid-trophic level Arctic fishes collected off Northeast Greenland: the demersal bigeye sculpin, Triglops nybelini, and the pelagic polar cod, Boreogadus saida. Plastics debris were found in the water samples at a concentration of 2.4 items/m3 ±0.8 SD which is higher than in most seas at lower latitudes. Both fish species had eaten MPs with different proportion among the species, 34% for T. nybelini (n = 71) and 18% for B. saida (n = 85). The significant difference in the occurrence of MPs between the two species is likely a consequence of their feeding behavior and habitat. Polyethylene was the main plastic polymer for water samples (41%, n = 17) and polyester (34%, n = 156) for fish samples as analyzed by Fourier Transformed Infrared (FT-IR) spectroscopy. Our data underscore that the Arctic regions are turning into a hotspot for plastic pollution, and this calls urgently for precautionary measures.

The Greenland shark (Somniosus microcephalus), an iconic species of the Arctic Seas, grows slowly and reaches >500 centimeters (cm) in total length, suggesting a life span well beyond those of other vertebrates. Radiocarbon dating of eye lens nuclei from 28 female Greenland sharks (81 to 502 cm in total length) revealed a life span of at least 272 years. Only the smallest sharks (220 cm or less) showed signs of the radiocarbon bomb pulse, a time marker of the early 1960s. The age ranges of prebomb sharks (reported as midpoint and extent of the 95.4% probability range) revealed the age at sexual maturity to be at least 156 ± 22 years, and the largest animal (502 cm) to be 392 ± 120 years old. Our results show that the Greenland shark is the longest-lived vertebrate known, and they raise concerns about species conservation.

The Greenland shark Somniosus microcephalus is an opportunistic feeder, a top predator, and a very long-lived species. The brain, liver, red and white muscle, gonads, fat, skin, pancreas, and spleen of Greenland sharks from NE Greenland fjords were analysed for PCBs, PCDDs/DFs, PBDEs; DDT isomers; HCH isomers; dieldrin; endrin; HCB; Cd, Hg, Pb, and Se. PCBs (2.01-103 ng/g wet wt) and PBDEs (7.9-3050 pg/g wet wt) were detected in most of the samples. PCDDs/DFs showed high values when detected. DDTs, HCB and HCHs were only detected in some tissues. The ΣTEQ was 5.76 pg/g in muscle. Cadmium mainly accumulated in the pancreas and liver (19.6 and 10.7 mg/kg dry wt, respectively); mercury in red muscle (4.10-6.91 mg/kg dry wt); selenium in the pancreas (3.57 mg/kg dry wt) and spleen (1.95 mg/kg dry wt); lead in the skin (0.358 mg/kgd ry wt). The selenium-mercury ratio in the liver was also evaluated.

Seasonality of biomarker baseline levels were studied in polar cod (Boreogadus saida), caught in Kongsfjorden, Svalbard, in April, July, September and December, 2006-2007. Physiological parameters (condition factor, gonado- and hepato-somatic indexes, energy reserves, potential metabolic activity and antifreeze activity) in polar cod were used to interpret the seasonality of potential biomarkers. The highest levels of ethoxyresorufin-O-deethylase (EROD) activity occurred concomitantly with the highest potential metabolic activity in July due to e.g. intense feeding. During pre-spawning, EROD showed significant inhibition and gender differences. Hence, its potential use in environmental monitoring should imply gender differentiation at least during this period. Glutathione S-transferase and catalase activities were stable from April to September, but changed in December suggesting a link to low biological activity. Knowledge of the biomarker baseline levels and their seasonal trends in polar cod is essential for a trustworthy interpretation of forthcoming toxicity data and environmental monitoring in the Arctic.

Polar cod Boreogadus saida were exposed weekly to two doses of dietary crude oil for 4 weeks followed by 2 weeks of depuration. Administered doses corresponded on average to 4 and 9microgSigmaPAHsg(-1)fishweek(-1). Cytochrome P4501A1 (cyp1a1) and glutathione S-transferase (gst) mRNA expression, ethoxyresorufin O-deethylase (EROD) activity and metabolites in the bile showed strong and dose-dependent inductions at 2 and 4 weeks of exposure. Following 2 weeks depuration, mRNA expression of cyp1a1 and gst and PAH metabolites returned to basal levels while EROD activity and GST activity were still induced in the high oil treatment. The mRNA expressions of antioxidant defense genes (catalase, glutathione peroxidase and cytosolic and mitochondrial superoxide dismutase) did not change significantly during the experiment. Catalase activity was significantly depressed at week 2 in the high oil treatment. We conclude that the cyp1a1 mRNA expression, EROD activities and bile metabolites were the most reliable biomarkers of exposure while gst mRNA expression and GST activity were less sensitive and are considered only as complementary. Antioxidant defenses were poor biomarkers to assess effects of crude oil exposure in polar cod.

In order to mimic the biological effects of an oil spill in Arctic waters, we examined several types of biomarkers (genes, enzymes, metabolites, and DNA damage) in polar cod Boreogadus saida experimentally exposed to the water soluble fractions of crude oil. During 4 weeks of exposure, induction of the studied biomarkers exceeded baseline levels. The mRNA expression of the cytochrome P4501A1 (cyp1a1) gene was the most promising biomarker, with glutathione S-transferase (gst) as a suitable complement. The delayed ethoxyresorufin O-deethylase (EROD) and GST activities and their persistence following 2 weeks of depuration may allow detection of previous exposures in field samples. The composition of PAH metabolites in the bile indicated the bioavailability of different PAH size-classes. Although mRNA expressions of antioxidant defense genes were induced at start of the exposure, with the strongest responses from catalase and cytosolic superoxide dismutase, they were poor for oil monitoring purposes due to their very short response times. Significant DNA damage demonstrated genotoxicity even at low PAH concentrations (<15microgL(-1)) and was correlated with benzo(a)pyrene and pyrene metabolites in the bile.

With expanding oil and gas activities into the Arctic region, there is a need to evaluate the induction capacity of polycyclic aromatic hydrocarbon (PAH) biomarkers on Arctic marine organisms and to test analytical methods that have been optimized for their temperate counterparts. Polar cod Boreogadus saida were injected intraperitoneally with cod liver oil (solvent control), 6.6+/-3.7, 85+/-48 or 378+/-190 microg kg(-1) wet weight of benzo(a)pyrene (B(a)P), or not injected (control), and liver and bile were sampled at 0 and 16 h and 1, 2, 4 and 7d. The mRNA expression of cytochrome P4501A1 (cyp1a1) and glutathione S-transferase (gst) genes showed a dose-dependent induction in the first 16 h following the injection and a return to basal levels after 4d. The aryl hydrocarbon receptor 2, however, showed no change in mRNA expression. The protein quantification of cytochrome P4501A (CYP1A), through Western blot analysis and the enzyme-linked immunosorbent assay (ELISA), presented similar but weaker and time-delayed responses (4-7d) compared to the gene (16 h to 2d). Ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase (GST) activities increased significantly at day 7 following the gene induction and increase in protein levels. Overall, these biomarkers showed dose-dependent but weak responses to B(a)P and low levels of bile metabolites. The mRNA expressions of oxidative stress genes, superoxide dismutases (sod(Cu/Zn) and sod(Mn)), catalase (cat) and glutathione peroxidase (gpx), were all up-regulated between 16 h and 2d of B(a)P exposure with cat (72-fold) and sod(Cu/Zn) (20-fold) giving the strongest responses in the highest dose. Finally, CAT protein level and enzyme activities showed less clear responses than the genes. The mRNA expression showed the earliest responses, followed by the protein levels. The enzymatic activities were the least sensitive and responded to the exposure after 7d. The study shows the induction capability of biomarkers in polar cod at very low bioavailable doses of B(a)P and provides new information on the selected biomarkers for use in oil monitoring in the Arctic.
