RESUMO

Preadult development of necrophagous flies is commonly recognized as an accurate method for estimating the minimum postmortem interval (PMImin). However, once the PMImin exceeds the duration of preadult development, the method is less accurate. Recently, fly puparial hydrocarbons were found to significantly change with weathering time in the field, indicating their potential use for PMImin estimates. However, additional studies are required to demonstrate how the weathering varies among species. In this study, the puparia of Chrysomya rufifacies were placed in the field to experience natural weathering to characterize hydrocarbon composition change over time. We found that weathering of the puparial hydrocarbons was regular and highly predictable in the field. For most of the hydrocarbons, the abundance decreased significantly and could be modeled using a modified exponent function. In addition, the weathering rate was significantly correlated with the hydrocarbon classes. The weathering rate of 2-methyl alkanes was significantly lower than that of alkenes and internal methyl alkanes, and alkenes were higher than the other two classes. For mono-methyl alkanes, the rate was significantly and positively associated with carbon chain length and branch position. These results indicate that puparial hydrocarbon weathering is highly predictable and can be used for estimating long-term PMImin.

Emerald ash borer (EAB) has killed millions of ash trees in the United States and Canada, yet impacts on terrestrial-aquatic linkages are largely unknown. Ash tree death along streams creates canopy gaps, increasing light to riparian plants and potentially affecting organic matter subsidies. Six EAB-related canopy gaps along streams across a gradient of timing of EAB invasion in Michigan were characterized for coarse woody material (CWM), terrestrial and aquatic leaf litter and their associated bacterial communities, and macroinvertebrates upstream, downstream, and at the center of the gap. Stream sites downstream of EAB-related canopy gaps had significantly lower dissolved oxygen and macroinvertebrate diversity than sites upstream and at the gaps. Yet there was no difference in CWM or aquatic leaf litter, likely due to downstream movement of organic matter from upstream riparian sources. Low abundance bacterial amplicon sequence variants unique to gap or forest were detected in leaves and leaf litter, suggesting that EAB-related canopy gaps altered leaf-associated bacterial communities. Overall, EAB invasion indirectly impacted some variables, while organic matter dynamics were resistant to change.

Buruli ulcer (BU) is an emerging, but neglected tropical disease, where there has been a reported association with disturbed aquatic habitats and proposed aquatic macroinvertebrate vectors such as biting Hemiptera. An initial step in understanding the potential role of macroinvertebrates in the ecology of BU is to better understand the entire community, not just one or two taxa, in relation to the pathogen, Mycobacterium ulcerans, at a large spatial scale. For the first time at a country-wide scale this research documents that M. ulcerans was frequently detected from environmental samples taken from BU endemic regions, but was not present in 30 waterbodies of a non-endemic region. There were significant differences in macroinvertebrate community structure and identified potential indicator taxa in relation to pathogen presence. These results suggest that specific macroinvertebrate taxa or functional metrics may potentially be used as aquatic biological indicators of M. ulcerans. Developing ecological indicators of this pathogen is a first step for understanding the disease ecology of BU and should assist future studies of transmission.
