RESUMO

Two new lanthanide-complexes based on the 5-nitropicolinate ligand (5-npic) were obtained and fully characterized. Single-crystal X-ray diffraction revealed that these compounds are isostructural to a Dy-complex, previously published by us, based on dinuclear monomers link together with an extended hydrogen bond network, providing a final chemical formula of [Ln2(5-npic)6(H2O)4]·(H2O)2, where Ln = Dy (1), Gd (2), and Tb (3). Preliminary photoluminescent studies exhibited a ligand-centered emission for all complexes. The potential antitumoral activity of these materials was assayed in a prostatic cancer cell line (PC-3; the 2nd most common male cancerous disease), showing a significant anticancer activity (50-60% at 500 µg·mL-1). In turn, a high biocompatibility by both, the complexes and their precursors in human immunological HL-60 cells, was evidenced. In view of the strongest toxic effect in the tumoral cell line provided by the free 5-npic ligand (~ 40-50%), the overall anticancer complex performance seems to be triggered by the presence of this molecule.

RESUMO

A new Y-based metal-organic framework (MOF) GR-MOF-6 with a chemical formula of {[YL(DMF)2]·(DMF)}n {H3L = 5-[(4-carboxyphenyl)ethynyl] isophthalic acid; DMF = N,N-dimethylformamide} has been prepared by a solvothermal route. Structural characterization reveals that this novel material is a three-dimensional MOF in which the coordination of the tritopic ligand to Y(III) metal ions leads to an intercrossing channel system extending over three dimensions. This material has proven to be a very efficient catalyst in the cyanosilylation of carbonyls, ranking second in catalytic activity among the reported rare earth metal-based MOFs described so far but with the lowest required catalyst loading. In addition, its electrophoretic behavior has been studied in depth, providing a zero-charge point between pH 4 and 5, a peak electrophoretic mobility of -1.553 µm cm V-1 s-1, and a ζ potential of -19.8 mV at pH 10.

RESUMO

Herein, we present the syntheses of a novel coordination polymer (CP) based on the perylene-3,4,9,10-tetracarboxylate (pery) linkers and sodium metal ions. We have chosen sodium metal center with the aim of surmising the effect that the modification of the metal ion may have on the relative humidity (RH) experimental measurements of the material. We confirm the role of the ions in the functionalization of the deposited layer by modifying their selectivity towards moisture content, paving the way to the generation of sensitive and selective chemical sensors.

In this work, we present a panoramic digital holographic system for the first time capable of obtaining 3D information of a quasi-cylindrical object by using a conical mirror. The proposed panoramic digital holographic system is able to scan the entire surface of the object to determine the amplitude and phase simultaneously. This paper demonstrates the feasibility of analyzing quasi-cylindrical objects in a short time (0.5 s) with a single camera and a minimum number of optical components. In addition, it can be applied to determine not only topographic measurement of the cylindrical surface but also measurements of radial deformations. Experimental results are presented at different magnifications, thus illustrating its capabilities and versatility.

This paper describes an optical measurement system that is capable of simultaneously measuring a three-dimensional shape and circumferential train in a thin-walled cylinder. We used the fringe projection technique to obtain a three-dimensional shape and out-of-plane displacements, which were then associated with out-of-plane strain. Although 360 deg profilometry is not new, we propose an experimental setup that requires only one camera and one projector, since two mirrors are located at 120 deg with respect to the y-z plane of the reference system to obtain three distinct, complementary views of the sample studied. A sequence of images of the fringe pattern was captured and processed using the phase-shifting method, while the out-of-plane data were determined by comparing the specimen before and after deformation. A latex tube filled with liquid was used to perform mechanical testing. The cylinder was subjected to internal hydrostatic pressure that caused deformation. Experimental results were compared with analytical solutions, and the results show that out-of-plane strains can be accurately measured using the proposed method.

A series of diclofenac N-derivatives (2, 4, 6, 8c, 9c, 10a-c) were synthesized in order to test their anti-cancer and anti-inflammatory effects. The anticarcinogen activity has been assayed against three cancer cell lines: HT29, human colon cancer cells; Hep-G2, human hepatic cells; and B16-F10, murine melanoma cells. First, we determined the cytotoxicity of the different compounds, finding that the most effective compound was compound 8c against all cell lines and both compounds 4 and 6 in human Hep-G2 and HT29 cell lines. Compounds 4 and 8c were selected for the percentage of apoptosis determination, cell cycle distribution, and mitochondrial membrane potential measure because these products presented the lowest IC50 values in two of the three cancer cell lines assayed (B16-F10 and HepG2), and were two of the three products with lowest IC50 in HT29 cell line. Moreover, the percentages of apoptosis induction were determined for compounds 4 and 8c, showing that the highest values were between 30 to 60%. Next, the effects of these two compounds were observed on the cellular cycle, resulting in an increase in the cell population in G2/M cell cycle phase after treatment with product 8c, whereas compound 4 increased the cells in phase G0/G1, by possible differentiation process induction. Finally, to determine the possible apoptosis mechanism triggered by these compounds, mitochondrial potential was evaluated, indicating the possible activation of extrinsic apoptotic mechanism. On the other hand, we studied the anti-inflammatory effects of these diclofenac (DCF) derivatives on lipopolysaccharide (LPS) activated RAW 264.7 macrophages-monocytes murine cells by inhibition of nitric oxide (NO) production. As a first step, we determined the cytotoxicity of the synthesized compounds, as well as DCF, against these cells. Then, sub-cytotoxic concentrations were used to determine NO release at different incubation times. The greatest anti-inflammatory effect was observed for products 2, 4, 8c, 10a, 10b, and 9c at 20 µg·mL-1 concentration after 48 h of treatment, with inhibition of produced NO between 60 to 75%, and a concentration that reduces to the 50% the production of NO (IC50 NO) between 2.5 to 25 times lower than that of DCF. In this work, we synthesized and determined for the first time the anti-cancer and anti-inflammatory potential of eight diclofenac N-derivatives. In agreement with the recent evidences suggesting that inflammation may contribute to all states of tumorigenesis, the development of these new derivatives capable of inducing apoptosis and anti-inflammatory effects at very low concentrations represent new effective therapeutic strategies against these diseases.

In this work, we present an optical and mechanical characterization of the behavior of an inhomogeneous biopolymer sample through the use of an in-plane electronic speckle pattern interferometer with a pulling system along the $y$y direction. The characterization of the sample subjected to stress comprised the acquisition of speckle patterns for 1360 states. Displacement maps and their corresponding strain maps were computed for every state. Since the information of the maps changes with size due to the sample being pulled at the upper end while it is clamped at the lower end, a scaling method to relate the maps to each other, point-to-point, is presented. The method allows the correct evaluation of sequential strain maps, which depicts the mechanical evolution of the material. Upon managing to relate the strain maps, it is possible to extract strain values for zones of interest from every map in order to build the respective stress-strain curves. Three stress-strain curves associated with three zones in the sample (upper, middle, and bottom) are constructed. When sequential displacement and deformation maps are optically obtained by the interferometer, we present a full-field characterization, along with the obtention stress-strain curves associated with the three zones of strain maps. The curves represent the inhomogeneous performance of the sample. Three different elastic moduli (${E_u} = 2.59\;{\rm MPa}$Eu=2.59MPa, ${E_m} = 1.97\;{\rm MPa}$Em=1.97MPa, and ${E_{b}} = 1.67\;{\rm MPa}$Eb=1.67MPa), associated with three respective zones, were obtained. The experimental results for a biopolymer sample here presented show that the technique, in conjunction with the scaling method, is a novel proposal to characterize inhomogeneous materials.

All authors who participated at the conference were contacted and invited to contribute to this special issue. Furthermore, the AO dedicated issue on OTPI was open to contributions from other practitioners in all related areas, through a call for papers published in AO.

Methods for measuring variations in diffuse surfaces using electronic speckle pattern interferometry (ESPI) are widely used and well known. In this research, we present an out-of-plane ESPI system coupled to a Michelson configuration to generate simultaneous parallel interferograms with different phase shifts. The system uses circular polarization states to generate parallel phase shifted interferograms. Due to the polarization states, the fringes do not experience a contrast reduction, thus avoiding measurement errors that affect spatial or temporal phase-shifting in interferometry. The basic operating principle of polarization modulation is described, and results that represent the temporal evolution of an aluminum plate are presented. The generation of two simultaneous patterns allows one to track the dynamic performance of the plate.

We have demonstrated a speckle out-of-plane interferometer that employs phase-stepping procedures by means of polarization modulation. The system generates circular polarization states with opposite signs at each arm of the system, which overlap at the output of the interferometer, to generate phase shifts operating a conventional linear polarizer; the emerging polarization states have been analyzed to obtain the shifts needed to process the optical phase. The phase-stepping technique is demonstrated with a two-step algorithm to measure out-of-plane displacement on a flat metal plate.

In this work the sensitivity vector is analyzed for collimated and divergent oblique illumination for an out-of-plane arrangement. In the case of collimated illumination, the variation of the sensitivity vector components was considered and a geometric model was proposed for its evaluation. The geometry of the optical setup used allowed us to find sensitivity mostly along the pulling direction; the other two components of the sensitivity vector were relatively small. We measured the displacement induced only along the pulling direction. Errors in the displacement measurement associated with divergent and collimated illumination can be predicted when the sensitivity vector is considered constant. Experimental and theoretical results are presented for the out-of-plane electronic speckle pattern interferometer. The analyzed object was an aluminum plate.

A 6-month-old female Iberian lynx (Lynx pardinus) cub that was severely affected by mange died in September 2016 in the Montes de Toledo (Spain) with crusts and fissures on its face, outer ears, nipples and footpads. The body condition of the cub was very poor, and it also had a mandibular abscess and a severely ankylosed luxation on its left knee. After confirming that the origin of the deceased cub's dermal lesions was Sarcoptes scabiei, the subsequent search for ectoparasites and a comparison of histopathological and immunohistochemical findings in all sympatric lynxes handled (n = 30) and submitted for necropsy (n = 4) during 2016 and 2017 revealed the presence of S. scabiei mites and/or milder mange compatible lesions in five members of her family group, which was treated against mange together with two exposed contiguous family groups. An ELISA developed by the authors showed the presence of antibodies against S. scabiei in the deceased female cub and one brother. The presence of concomitant immunosuppressive factors in the dead female cub and the results obtained for the other sympatric lynxes studied since 2016 suggest that S. scabiei had a limited effect on immune-competent Iberian lynxes in the local population of the Montes de Toledo. However, a different evolution and relevance of sarcoptic mange in different populations-or even in the same one in the presence of immunosuppressive factors-cannot be ruled out, thus confirming the need for further research in order to attain a complete comprehension of the epidemiology and the real threat that this ectoparasitic disease may imply for L. pardinus.

We present a multiwavelength analysis of a wavefront detected by a color camera and a lateral shear interferometer. The system employs polarization phase-shifting techniques by rotating a linear polarizer at the output and detecting the phase information through a frequency demodulation algorithm. By considering the phase modulation obtained by rotating the analyzer, a frequency filter centered on the desired peak carries the phase and amplitude information of the detected wavefront. Theoretical approach, the consideration of Jones matrices of each element, and experimental results show the feasibility of the implementation.

In order to recover the holographic object information, a method based on the recording of two digital holograms, not only at different planes but also in a slightly off-axis scheme, is presented. By introducing a π-phase shift in the reference wave, the zero-order diffracted term and the twin image are removed in the frequency domain during the processing of the recorded holograms. We show that the zero-order elimination by the phase-shifted holograms is better than working with weak-order beam and average intensity removal methods. For recording experimentally two π-shifted holograms at different planes slightly off-axis, a single cube beam splitter is used. Computer simulations and experimental results, carried out to validate our proposal, show a high accuracy of π/14 that can be comparable with phase-shifting digital holography. For high fringe spacing, our proposal could be applied in electron holography, avoiding high voltage in a biprism.

This work presents a microfluidic paper-based analytical device (µPAD) for glucose determination using a supported metal-organic framework (MOF) acting as a peroxidase mimic. The catalytic action of glucose oxidase (GOx) on glucose causes the formation of H2O2, and the MOF causes the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) by H2O2 to form a blue-green product with an absorption peak at 650 nm in the detection zone. A digital camera and the iOS feature of a smartphone are used for the quantitation of glucose with the S coordinate of the HSV color space as the analytical parameter. Different factors such as the concentration of TMB, GOx and MOF, pH and buffer, sample volume, reaction time and reagent position in the µPAD were optimized. Under optimal conditions, the value for the S coordinate increases linearly up to 150 µmol·L-1 glucose concentrations, with a 2.5 µmol·L-1 detection limit. The µPAD remains stable for 21 days under conventional storage conditions. Such an enzyme mimetic-based assay to glucose determination using Fe-MIL-101 MOF implemented in a microfluidic paper-based device possesses advantages over enzyme-based assays in terms of costs, durability and stability compared to other existing glucose determination methods. The procedure was applied to the determination of glucose in (spiked) serum and urine. Citrulline (C6H13N3O3) is an amino acid found in the body as a zwitterion. This means its carboxylic and amine groups can act as Lewis donors to chelate metal cations. In addition, citrulline possesses a terminal ureido group on its aliphatic chain, which also appears to coordinate. Here, two new mixed complexes of citrulline were made with 1,10-phenanthroline and 2,2'-bipyridine. These compounds, once dissolved in water, gave aquo-complexes that were subject to DFT studies and in vitro toxicity studies on cancer cell lines (HeLa, MDA-MB-231, HCT 15, and MCF7) showed promising results. Docking studies with DNA were also conducted, indicating potential anticancer properties.

A new family of six complexes based on 5-nitropicolinic acid (5-npic) and transition metals has been obtained: [M(5-npic)2]n (MII = Mn (1) and Cd (2)), [Cu(5-npic)2]n (3), and [M(5-npic)2(H2O)2] (MII = Co (4), Ni (5), and Zn (6)), which display 1D, 2D, and mononuclear structures, respectively, thanks to different coordination modes of 5-npic. After their physicochemical characterization by single-crystal X-ray diffraction (SCXRD), elemental analyses (EA), and spectroscopic techniques, quantum chemical calculations using Time-Dependent Density Functional Theory (TD-DFT) were performed to further study the luminescence properties of compounds 2 and 6. The potential anticancer activity of all complexes was tested against three tumor cell lines, B16-F10, HT29, and HepG2, which are models widely used for studying melanoma, colon cancer, and liver cancer, respectively. The best results were found for compounds 2 and 4 against B16-F10 (IC50 = 26.94 and 45.10 µg mL-1, respectively). In addition, anti-inflammatory studies using RAW 264.7 cells exhibited promising activity for 2, 3, and 6 (IC50 NO = 5.38, 24.10, and 17.63 µg mL-1, respectively). This multidisciplinary study points to complex 2, based on CdII, as a promising anticancer and anti-inflammatory material.

A parallel two-step polarizing phase shifting interferometer based on a Double Cyclic Shear Interferometer (DCSI) is proposed in this paper for quantitative phase imaging. The system has the advantage of retrieving the derivative phase data map directly. Due to its configuration, it presents better stability against external configurations than other types of interferometers. The DCSI generates two π-shifted interferograms, which are recorded by the CCD camera in a single-shot. The separation between parallel interferograms can be varied in the two axes for convenience. To obtain the optical phase data map, a parallel phase shift between interferograms is obtained by rotating a half wave plate retarder. We analyzed the cases of four patterns with shifts of π/2 captured in two shots; the optical phase was processed by a four-step algorithm. Related experimental results obtained for microscopic transparent samples are presented.

Alzheimer's disease (AD) is a neurodegenerative disorder affecting 44 million people worldwide. Although many issues (pathogenesis, genetics, clinical features, and pathological aspects) are still unknown, this disease is characterized by noticeable hallmarks such as the formation of ß-amyloid plaques, hyperphosphorylation of tau proteins, the overproduction of reactive oxygen species, and the reduction of acetylcholine levels. There is still no cure for AD and the current treatments are aimed at regulating the cholinesterase levels, attenuating symptoms temporarily rather than preventing the AD progression. In this context, coordination compounds are regarded as a promissing tool in AD treatment and/or diagnosis. Coordination compounds (discrete or polymeric) possess several features that make them an interesting option for developing new drugs for AD (good biocompatibility, porosity, synergetic effects of ligand-metal, fluorescence, particle size, homogeneity, monodispersity, etc.). This review discusses the recent progress in the development of novel discrete metal complexes and metal-organic frameworks (MOFs) for the treatment, diagnosis and theragnosis of AD. These advanced therapies for AD treatment are organized according to the target: Aß peptides, hyperphosphorylated tau proteins, synaptic dysfunction, and mitochondrial failure with subsequent oxidative stress.

Toxoplasmosis is a zoonotic disease caused by Toxoplasma gondii, an intracellular parasite that presents a worldwide risk. Humans can become infected by ingesting meat infected with T. gondii, and the consumption of infected sheep and goat meat is a significant public health issue. Antibodies against T. gondii have been found in sheep in Spain, indicating the presence of the parasite in the country. However, no previous studies have assessed the presence of T. gondii in sheep meat in Spain. In view of the significance of the transmission of T. gondii through meat consumption and given the lack of previous studies in Spain, we carried out an investigation to evaluate the presence of T. gondii in adult sheep meat (mutton). A total of 216 muscle samples were analyzed by digestion, and a real-time PCR assay was used to determine the presence of T. gondii DNA. A total of 24.5% of the samples were found to be parasitized, indicating that the consumption of sheep meat can present an important risk for human health.
