BACKGROUND: Eosinophils are granulocytes that rapidly increase frequency in the bloodstream during helminthic infections and allergic responses. They are found in tissue infected by Leishmania during early disease, but their role during infection is not entirely understood. OBJECTIVES: We aim to compare the disease due to Leishmania amazonensis in BALB/c and Δdbl-GATA1 mice, which lack eosinophils. METHODS: BALB/c and Δdbl-GATA1 mice infected with L. amazonensis were observed for several weeks. The parasite load and dissemination pattern were assessed. FINDINGS: The Δdbl-GATA1 mice developed an anticipated dissemination of L. amazonensis and a worsening disease. No differences were found in the lesion development or the parasite load in the footpad among Δdbl-GATA1 mice and BALB/c eight weeks after infection. However, nine weeks after infection, massive growth of metastatic lesions appeared in several parts of the skin in Δdbl-GATA1 mice, weeks earlier than BALB/c. We observed increased parasites in the bloodstream, probably an essential dissemination route. Thirteen weeks after infection, metastatic lesions were found in all Δdbl-GATA1 mice. MAIN CONCLUSION: These results suggest a protective role of eosinophils in delaying the disease caused by L. amazonensis, although several limitations of this mice strain must be considered.

Three-dimensional scanning technology has been traditionally used in the medical and engineering industries, but these scanners can be expensive or limited in their capabilities. This research aimed to develop low-cost 3D scanning using rotation and immersion in a water-based fluid. This technique uses a reconstruction approach similar to CT scanners but with significantly less instrumentation and cost than traditional CT scanners or other optical scanning techniques. The setup consisted of a container filled with a mixture of water and Xanthan gum. The object to be scanned was submerged at various rotation angles. A stepper motor slide with a needle was used to measure the fluid level increment as the object being scanned was submerged into the container. The results showed that the 3D scanning using immersion in a water-based fluid was feasible and could be adapted to a wide range of object sizes. The technique produced reconstructed images of objects with gaps or irregularly shaped openings in a low-cost fashion. A 3D printed model with a width of 30.7200 ± 0.2388 mm and height of 31.6800 ± 0.3445 mm was compared to its scan to evaluate the precision of the technique. Its width/height ratio (0.9697 ± 0.0084) overlaps the margin of error of the width/height ratio of the reconstructed image (0.9649 ± 0.0191), showing statistical similarities. The signal-to-noise ratio was calculated at around 6 dB. Suggestions for future work are made to improve the parameters of this promising, low-cost technique.

Globally, educational institutes are trying to adapt modernized and effective approaches and tools to their education systems to improve the quality of their performance and achievements. However, identifying, designing, and/or developing promising mechanisms and tools that can impact class activities and the development of students' outputs are critical success factors. Given that, the contribution of this work is to propose a methodology that can guide and usher educational institutes step by step through the implementation of a personalized package of training Toolkits in Smart Labs. In this study, the package of Toolkits refers to a set of needed tools, resources, and materials that, with integration into a Smart Lab can, on the one hand, empower teachers and instructors in designing and developing personalized training disciplines and module courses and, on the other hand, may support students (in different ways) in developing their skills. To demonstrate the applicability and usefulness of the proposed methodology, a model was first developed, representing the potential Toolkits for training and skill development. The model was then tested by instantiating a particular box that integrates some hardware to be able to connect sensors to actuators, with an eye toward implementing this system mainly in the health domain. In a real scenario, the box was used in an engineering program and its associated Smart Lab to develop students' skills and capabilities in the areas of the Internet of Things (IoT) and Artificial Intelligence (AI). The main outcome of this work is a methodology supported by a model able to represent Smart Lab assets in order to facilitate training programs through training Toolkits.

Sludge dewatering and drying are the main processes related to sludge management in wastewater treatment plants (WWTPs). Sludge disposal is a high-cost activity, and drying the sludge reduces its mass and volume, resulting in savings in storage, handling and transportation. The discoveries regarding the use of solar energy in agricultural studies provided valuable information for using in sewage sludge drying. Some studies have reported that dry sludge has met the EPA Class A requirement for biosolids using only solar energy as an energy source. The proper sludge mixture, manual or mechanical, and the dewatering process can significantly increase the drying rate, reducing drying time and the surface area needed. The environmental conditions and the sludge's type greatly influence the drying system. A solar dryer system may be suitable to dry different types of wastewater sludge. Modeling techniques can predict the behavior of the solar drying system and, thus, save time and money in experimental steps. CFD modeling of the sludge drying system is usually done by adopting specific boundary conditions and solving the Navier Stokes equations for air and sludge. There is no standard methodology for comparing solar dryers and common methodologies, such as system efficiency and thermal efficiency, disregards different dryers in different operational conditions. A SWOT (strengths, weaknesses, opportunities, and threats) analysis indicated that, in general, the chapel-type greenhouse with mixed-mode drying has higher drying rates, resulting in reduced drying time and can be scaled to any size. Thus, this type of dryer emerges as a more economical alternative to commercial solar dryers. Based on a systematic review, this work points the SWOT analysis as a useful tool for selecting solar dryers.

In the present study, the objective was to evaluate in situ interaction between Benzo[a]anthracene 7,2-dione 7-oxime (BZA) and calf thymus dsDNA (ct-dsDNA) using electroanalytical genosensor. Analytical techniques based on Ultraviolet/Visible (UV-Vis) spectroscopy and electroanalytical were used to investigate the interaction processes in solution and immobilized on carbon screen-printed electrodes modified with electrochemical mediator Meldola blue. In addition, was possible to evaluate the degree of damage caused to the genetic material by the analyte through of toxicity estimate (S%). The interaction evaluated by genosensor showed processes of intercalation, degradation, and breaks of the double strand of ct-dsDNA, suggesting that the interaction simulates highly toxic (values varying from 0.6 to 0.8 µA in 48 h of interaction), such as 8-oxoguanine (+0.48 V), which is a by-product of guanine oxidation. Furthermore, monitoring A (+1.10 V) after 1 h showed an S% value between 50 and 90%, indicative of high toxicity, and monitoring G (+0.85 V), which showed S>90%, indicated no toxicity after 10 min. Overall, the electroanalytical genosensor developed in a miniaturized system displayed good reproducibility and stability over time being a quick alternative for assesses the degree of toxicity between toxic xenobiotics and biologically electroactive molecules, such as DNA.

As food waste gets acknowledged as a global potential source of biomass, its valorization through anaerobic digestion becomes an attractive strategy. This work describes the state-of-the-art on the valorization of food waste by anaerobic digestion and the optimization of the process. The methodology used was a bibliometric and systematic review of the optimization of the process from 66 articles selected. Bibliometric mapping allowed us to identify that, until now, most studies have been focused on the: i) anaerobic co-digestion strategy in order to stabilize the process, ii) interest in the generation of biofuels to replace non-renewable fuels, iii) study of metabolic processes for a better understanding of the system iv) reactor design optimization and others facilities to increase process efficiency. The systematic analysis showed that the operational parameters has been extensively studied to optimize the process. Therefore, co-digestion has been the main strategy to improve the process. In this sense, knowledge of the substrate and co-substrate is extremely important to operate the reactors. For methane production, the ideal operating conditions indicated were: pH of 7, solids content between 4.0 and 15%, C/N ratio of 25, hydraulic retention time from 25 to 40 days and alkalinity from 2850 to 2970.5mgCaCO3/L. In addition, the ideal OLR will vary mainly according to operating temperature, number of reactor stages, and raw material characteristics. This review indicates trends and knowledge gaps that are important to guide new research on the anaerobic digestion of food waste, pointing out the potential advantages, optimization strategies, by-products of interest and challenges of the process. The results were used for the development of references of ideal operating conditions for energy production, being able to guide the design and operation of reactors.

The number of people living with dementia in the world is rising at an unprecedented rate, and no country will be spared. Furthermore, neither decisive treatment nor effective medicines have yet become effective. One potential alternative to this emerging challenge is utilizing supportive technologies and services that not only assist people with dementia to do their daily activities safely and independently, but also reduce the overwhelming pressure on their caregivers. Thus, for this study, a systematic literature review is conducted in an attempt to gain an overview of the latest findings in this field of study and to address some commercially available supportive technologies and services that have potential application for people living with dementia. To this end, 30 potential supportive technologies and 15 active supportive services are identified from the literature and related websites. The technologies and services are classified into different classes and subclasses (according to their functionalities, capabilities, and features) aiming to facilitate their understanding and evaluation. The results of this work are aimed as a base for designing, integrating, developing, adapting, and customizing potential multimodal solutions for the specific needs of vulnerable people of our societies, such as those who suffer from different degrees of dementia.

In its 30 years of existence, there are still many improvement possibilities in studies performing the life cycle assessment (LCA) of wastewater treatment plants (WWTPs). Hence, this paper aims to start a guideline development for LCA of urban WWTPs based on the information available in the scientific literature on the topic. The authors used the ProKnow-C systematic review methodology for paper selection and 111 studies were analyzed. The most significant points that can be improved are caused by missing essential information (e.g. functional unity and input data). Other important methodological aspects are covered: allocation process, functional unit choice, sensitivity analysis, and important fluxes to be considered. Many opportunities within the LCA of WWTPs were identified, such as optimization of WWTP operational aspects and resource recovery. Furthermore, LCA should be combined with other methodologies such as big data, data envelopment analysis, life cycle cost assessment, and social life cycle assessment. To achieve this potential, it is clear that the scientific and technical community needs to converge on a new protocol to ensure that LCA application becomes more reliable and transparent.

The adoption of Quality by Design (QbD) and Analytical Method Lifecycle Management (AMLM) concepts to ensure the quality of pharmaceutical products has been applied and proposed over the last few years. These concepts are based on knowledge gained from the application of scientific and quality risk management approaches, throughout method lifecycle to assure continuous improvement and high reliability of analytical results. The overall AMLM starts with the definition of the method's intended use through the Analytical Target Profile definition, including three stages: (1) Method Design, taking advantage of the well-known concept of QbD; (2) Method Performance Qualification; (3) Continued Method Performance Verification. This is intended to holistically align method variability with product requirements, increasing confidence in the data generated, a regulatory requirement that the pharmaceutical industry must follow. This approach views all method-related activities, such as development, validation, transfer, and routine use as a continuum and interrelated process, where knowledge and risk management are the key enablers. An increase in method robustness, cost reduction, and decreased risk failures are some of the intrinsic benefits from this lifecycle management. This approach is clearly acknowledged both by regulators and industry. The roadmap of the regulatory and industry events that mark the evolution of these concepts helps to capture the current and future expectation of the pharmaceutical framework.

Choosing the right nutrients to consume is essential to health and wellbeing across species. However, the factors that influence these decisions are poorly understood. This is particularly true for dietary proteins, which are important determinants of lifespan and reproduction. We show that in Drosophila melanogaster, essential amino acids (eAAs) and the concerted action of the commensal bacteria Acetobacter pomorum and Lactobacilli are critical modulators of food choice. Using a chemically defined diet, we show that the absence of any single eAA from the diet is sufficient to elicit specific appetites for amino acid (AA)-rich food. Furthermore, commensal bacteria buffer the animal from the lack of dietary eAAs: both increased yeast appetite and decreased reproduction induced by eAA deprivation are rescued by the presence of commensals. Surprisingly, these effects do not seem to be due to changes in AA titers, suggesting that gut bacteria act through a different mechanism to change behavior and reproduction. Thus, eAAs and commensal bacteria are potent modulators of feeding decisions and reproductive output. This demonstrates how the interaction of specific nutrients with the microbiome can shape behavioral decisions and life history traits.

Gut homeostasis is a process that requires a prudent balance of host responses to the beneficial enteric microbial community and the pathogenic stimuli that can arise. The lack of this balance in the intestine can result in inflammatory bowel diseases, where the immune system dysfunctions leading to exacerbated inflammatory responses. In this process, macrophages are considered to play a pivotal role. In this review, we describe the important role of macrophages in maintaining intestinal homeostasis and we discuss how altered macrophage function may lead to inflammatory bowel diseases. The plasticity of macrophages during the gut inflammatory response shows the broad role of these cells in orchestrating not only the onset of inflammation but also its termination as well as healing and repair. Indeed, the state of macrophage polarization can be the key factor in defining the resolution or the progression of inflammation and disease. Here, we discuss the different populations of macrophages and their implication in development, propagation, control and resolution of inflammatory bowel diseases.

The renin-angiotensin system (RAS) peptides play an important role in inflammation. Resolution of inflammation contributes to restore tissue homeostasis, and it is characterized by neutrophil apoptosis and their subsequent removal by macrophages, which are remarkable plastic cells involved in the pathophysiology of diverse inflammatory diseases. However, the effects of RAS peptides on different macrophage phenotypes are still emerging. Here, we evaluated the effects of angiotensin-(1-7) (Ang-(1-7)) and the most novel RAS peptide, alamandine, on resting (M0), proinflammatory M(LPS+IFN-γ), and anti-inflammatory M(IL-4) macrophage phenotypes in vitro, as well as on specific immune cell populations and macrophage subsets into the pleural cavity of LPS-induced pleurisy in mice. Our results showed that Ang-(1-7) and alamandine, through Mas and MrgD receptors, respectively, do not affect M0 macrophages but reduce the proinflammatory TNF-α, CCL2, and IL-1ß transcript expression levels in LPS+IFN-γ-stimulated macrophages. Therapeutic administration of these peptides in LPS-induced inflammation in mice decreased the number of neutrophils and M1 (F4/80lowGr1+CD11bmed) macrophage frequency without affecting the other investigated macrophage subsets. Our data suggested that both Ang-(1-7) and alamandine, through their respective receptors Mas and MrgD, promote an anti-inflammatory reprogramming of M(LPS+IFN-γ)/M1 macrophages under inflammatory circumstances and potentiate the reprogramming induced by IL-4. In conclusion, our work sheds light on the emerging proresolving properties of Ang-(1-7) and alamandine, opening new avenues for the treatment of inflammatory diseases.

Microalgae are a potential source of biomass for the production of energy, which is why the amount of research on this topic has increased in recent years. This work describes the state of the art of microalgae production from wastewater treatment plants (WWTP), its potential to generate electricity and the scale in which it is possible. The methodology used was a systematic review of the gasification of microalgae from 49 articles selected. Based on the review, a conceptual scenario for microgeneration in WWTP using as feedstock microalgae for thermal gasification was developed. The most consistent assumptions for a real scale microgeneration are microalgae production in open ponds using domestic sewage as a nutritional medium; the use of the flocculation process in process of harvesting; microalgae to energy through thermal gasification process using a downdraft gasifier. Considering a WWTP with a 3000 m3/d flux capacity, 860 kg/d of dry microalgae biomass might be produced. For which, gasification has a production potential of 0.167 kWh/m3 of treated sewage, but the energy balance is compromised by the drying process. However, when the biogas produced in anaerobic treatment enter in the model, it is possible to add a surplus of electricity of 0.14 kWh/m3 of treated sewage. Finally, a cost estimate is made for the acquisition of drying and gasification-electricity generation systems. For this scenario, the results suggest that the investments may be financially returned after five years, with additional potential for further optimization.

Owing to the cost of destination and transportation of ornamental stone processing waste, many studies focused on the reuse and recycling of this product. However, there is a scarcity of articles addressing the environmental viability of the recycling of ornamental stone. In this context, this study comprehends a comparative life cycle assessment of ornamental stone processing waste and conventional materials: sand, clay and limestone filler. The modelling software used was SimaPro 8.3.0.0 with Ecoinvent 3.2 database, employing the ReCiPe H/H methodology for impact assessment. The results show that the recycling of ornamental stone processing waste is environmentally preferable, and the artificial drying alternatives, such as flash dryer and rotary dryer, have lower environmental impact than extracting and processing clay through atomisation methods and limestone filler production. The sensitivity analysis indicated that it is possible to transport the ornamental stone processing waste 37 km after processing, so it reaches the same environmental impact as sand extracted by dredging. On the other hand, an increase of 25% in the energy consumption incremented only 7% of the environmental impact owing to the Brazilian energy mix.

Healing is a vital response important for the re-establishment of the skin integrity following injury. Delayed or aberrant dermal wound healing leads to morbidity in patients. The development of therapies to improve dermal healing would be useful. Currently, the design of efficient treatments is stalled by the lack of detailed knowledge about the cellular and molecular mechanisms involved in wound healing. Recently, using state-of-the-art technologies, it was revealed that macrophages signal via GPNMB to mesenchymal stem cells, accelerating skin healing. Strikingly, transplantation of macrophages expressing GPNMB improves skin healing in GPNMB-mutant mice. Additionally, topical treatment with recombinant GPNMB restored mesenchymal stem cells recruitment and accelerated wound closure in the diabetic skin. From a drug development perspective, this GPNMB is a new candidate for skin healing.

Pericytes are defined by their anatomical location encircling blood vessels' walls with their long projections. The exact embryonic sources of cerebral pericytes remain poorly understood, especially because of their recently revealed diversity. Yamamoto et al. (Sci Rep 7(1):3855, 2017) using state-of-the-art techniques, including several transgenic mice models, reveal that a subpopulation of brain pericytes are derived from phagocytic macrophages during vascular development. This work highlights a new possible ancestor of brain pericytes. The emerging knowledge from this research may provide new approaches for the treatment of several neurodevelopmental disorders in the future.

BACKGROUND: Despite the very low or absent parasitism in the lungs, the interstitial pneumonitis is a common lesion found in humans and dogs with visceral leishmaniasis. The lung is a neglected organ in the study of dogs and humans with visceral leishmaniasis, but interstitial pneumonitis represents an important lesion characterized by thickening of the alveolar septum due to fibrosis and inflammatory exudate, and its pathogenesis is still uncertain. In this study, the polymerase chain reaction (PCR) was used to detect Leishmania infantum in paraffin-embedded lung biopsies from naturally infected dogs from an endemic area in Minas Gerais State, Brazil; PCR was compared to histological and immunohistochemical techniques for detecting Leishmania. RESULTS: Eighteen dogs in which leishmaniasis had been diagnosed by serological tests - indirect immunofluorescence assay (IFAT), enzyme-linked immunosorbent assay (ELISA) and complement fixation tests (CFT) - were classified as asymptomatic, oligosymptomatic or symptomatic. Nine of the 18 dogs studied had a positive PCR (50%) but parasites were not detected by histopathological and immunocytochemistry methods. CONCLUSIONS: These data indicate that PCR on DNA extracted from paraffin-embedded tissue is a valuable method for detecting Leishmania infantum parasites in lungs of naturally infected dogs, despite the apparent absence of parasites from standard HE (hematoxylin and eosin) stained slides and of labeled parasites from immunocytochemical preparations.

Bacterial spot, caused by Xanthomonas spp., is one of the major bacterial diseases in pepper (Capsicum annuum L.). The infection results in reduced crop yield, particularly during periods of high rainfall and temperature, due to the low efficiency of chemical control with copper bactericides. This study evaluated the copper and zinc sulfate sensitivity of 59 pathogenic strains of Xanthomonas euvesicatoria isolated from pepper plants produced in various regions throughout Brazil. Both the respective sulfates and a mixture thereof was evaluated at 50, 100, 200 and 400 µg.mL-1. All the evaluated strains were found to be resistant to zinc sulfate (100 µg.mL-1) and 86.4% were resistant to copper sulfate (200 µg.mL-1). The mixture of copper (200 µg.mL-1) and zinc (200 µg.mL-1) sulfates inhibited the growth of all strains of X. euvesicatoria. To our knowledge this is the first study to report the resistance of X. euvesicatoria strains from pepper plants to copper and zinc sulfates in Brazil.

RESUMO

The use of sensors and actuators as a form of controlling cyber-physical systems in resource networks has been integrated and referred to as the Internet of Things (IoT). However, the connectivity of many stand-alone IoT systems through the Internet introduces numerous cybersecurity challenges as sensitive information is prone to be exposed to malicious users. This paper focuses on the improvement of IoT cybersecurity from an ontological analysis, proposing appropriate security services adapted to the threats. The authors propose an ontology-based cybersecurity framework using knowledge reasoning for IoT, composed of two approaches: (1) design time, which provides a dynamic method to build security services through the application of a model-driven methodology considering the existing enterprise processes; and (2) run time, which involves monitoring the IoT environment, classifying threats and vulnerabilities, and actuating in the environment ensuring the correct adaptation of the existing services. Two validation approaches demonstrate the feasibility of our concept. This entails an ontology assessment and a case study with an industrial implementation.
