RESUMO

The present study aims to monitor and assess the occurrence of veterinary antibiotics (VAs) in animal manure, compost, and fertilized soil, originating from different large-scale feedlots. The corresponding concentrations of 39 types of VAs in 8 large-scale feedlots of pig, dairy cow, and poultry were sampled in different seasons and analyzed using LC-MS. The results indicated that 17 types, 16 types, and 5 types of VAs were detected in the swine manure, compost, and fertilized soil with the concentrations of 0.003-17.82, 0.002-9.59, and 0.004-0.007 mg kg-1 (dry matter), respectively; 3 types, 2 types, and 1 type of VAs were detected in the dairy manure, compost, and fertilized soil with the concentrations of 0.003-1.94, 0.014-0.044, and 0.025 mg kg-1 (dry matter), respectively; 7 types, 5 types, and 1 type of VAs were detected in the poultry manure, compost, and fertilized soil with the concentrations of 0.035-1.06, 0.018-0.049, and 0.019 mg kg-1 (dry matter), respectively. The main antibiotic classes persisted in the animal manure and their composting product and fertilized soil were sulfonamides (SAs), macrolides (MAs), and tetracyclines (TCs). Thus, this study would help to adopt strategies in pollution control of VAs and environmental protection of agriculture.

The contrast sensitivity function (CSF) has shown promise as a functional vision endpoint for monitoring the changes in functional vision that accompany eye disease or its treatment. However, detecting CSF changes with precision and efficiency at both the individual and group levels is very challenging. By exploiting the Bayesian foundation of the quick CSF method (Lesmes, Lu, Baek, & Albright, 2010), we developed and evaluated metrics for detecting CSF changes at both the individual and group levels. A 10-letter identification task was used to assess the systematic changes in the CSF measured in three luminance conditions in 112 naïve normal observers. The data from the large sample allowed us to estimate the test-retest reliability of the quick CSF procedure and evaluate its performance in detecting CSF changes at both the individual and group levels. The test-retest reliability reached 0.974 with 50 trials. In 50 trials, the quick CSF method can detect a medium 0.30 log unit area under log CSF change with 94.0% accuracy at the individual observer level. At the group level, a power analysis based on the empirical distribution of CSF changes from the large sample showed that a very small area under log CSF change (0.025 log unit) could be detected by the quick CSF method with 112 observers and 50 trials. These results make it plausible to apply the method to monitor the progression of visual diseases or treatment effects on individual patients and greatly reduce the time, sample size, and costs in clinical trials at the group level.

Measurement efficiency is of concern when a large number of observations are required to obtain reliable estimates for parametric models of vision. The standard entropy-based Bayesian adaptive testing procedures addressed the issue by selecting the most informative stimulus in sequential experimental trials. Noninformative, diffuse priors were commonly used in those tests. Hierarchical adaptive design optimization (HADO; Kim, Pitt, Lu, Steyvers, & Myung, 2014) further improves the efficiency of the standard Bayesian adaptive testing procedures by constructing an informative prior using data from observers who have already participated in the experiment. The present study represents an empirical validation of HADO in estimating the human contrast sensitivity function. The results show that HADO significantly improves the accuracy and precision of parameter estimates, and therefore requires many fewer observations to obtain reliable inference about contrast sensitivity, compared to the method of quick contrast sensitivity function (Lesmes, Lu, Baek, & Albright, 2010), which uses the standard Bayesian procedure. The improvement with HADO was maintained even when the prior was constructed from heterogeneous populations or a relatively small number of observers. These results of this case study support the conclusion that HADO can be used in Bayesian adaptive testing by replacing noninformative, diffuse priors with statistically justified informative priors without introducing unwanted bias.

The occurrence of microplastics (MPs) in farmlands poses a threat to soil health and crop yield. There needs to be more research on the role of cropping patterns in the accumulation of MPs and quantizing the threat of MPs on soil health and crop yield. In this study, a field study was carried out to explore the role of cropping patterns in the accumulation of MPs in agricultural soil in Shanghai, China. Furthermore, the specific effect and importance of MPs and each soil physicochemical indicator to soil health and crop yield were clarified, and the threat of MPs in reducing soil health and crop yield was quantized. Relative lower MPs abundance was detected in Shanghai. MPs abundance in vegetable fields was significantly higher than that in orchards. The broad source of MPs, the acceleration of plastics breaking under artificial disturbance and warmer temperatures, and the block of MPs exchange could account for the quicker accumulation of MPs in vegetable fields. MPs have a negligible effect on microbial diversity and metabolic activity which plays a role in soil enzyme activity. Besides, MPs served as one of the critical factors for rice yield reduction.

OBJECTIVE: To investigate the prognostic value for predicting mortality of partial pressure of oxygen/fraction of inspired oxygen (PaO2/FiO2), the Sequential Organ Failure Assessment (SOFA) score and D-dimer in elderly patients with sepsis. METHODS: This retrospective cohort study enrolled elderly patients with sepsis admitted to the intensive care unit (ICU) between January 2019 and October 2020. Patients were divided into a survival group and a non-survival group. Biomarkers, SOFA, Acute Physiology and Chronic Health Evaluation II and Glasgow Coma Scale scores were recorded within 24 h after admission to the ICU. RESULTS: A total of 135 elderly patients with sepsis were enrolled in the study: 89 were in the survival group and 46 were in the non-survival group at 28 days. Univariate and multivariate regression analyses demonstrated that PaO2/FiO2, SOFA and D-dimer were independently associated with 28-day mortality. The predictive performance for mortality of the combination of PaO2/FiO2, SOFA score and D-dimer (area under the receiver operating characteristic curve of 0.926) was higher than the values for the individual factors (0.761, 0.745 and 0.878, respectively). CONCLUSION: The combination of PaO2/FiO2, SOFA score and D-dimer represents a promising tool and biomarker for predicting 28-day mortality of the elderly patients with sepsis.

Machine learning has the potential to facilitate the development of computational methods that improve the measurement of cognitive and mental functioning. In three populations (college students, patients with a substance use disorder, and Amazon Mechanical Turk workers), we evaluated one such method, Bayesian adaptive design optimization (ADO), in the area of delay discounting by comparing its test-retest reliability, precision, and efficiency with that of a conventional staircase method. In all three populations tested, the results showed that ADO led to 0.95 or higher test-retest reliability of the discounting rate within 10-20 trials (under 1-2 min of testing), captured approximately 10% more variance in test-retest reliability, was 3-5 times more precise, and was 3-8 times more efficient than the staircase method. The ADO methodology provides efficient and precise protocols for measuring individual differences in delay discounting.

Pot-culture experiments were carried out in Shanghai to screen crop varieties with low bioaccumulation properties with respect to cadmium (Cd). Eight common crops, such as green pepper, cucumber, cowpea, spinach, cauliflower, tomatoes, rice, and wheat, were planted in contaminated soil with different Cd concentrations of 0.23, 0.6, 1.2, 1.8, 2.4, and 3.0 mg·kg-1 to investigate the effects on biomass, Cd accumulation characteristics, and edible risk safety. The results indicated that:① With the increase in soil Cd content, the aboveground biomass of crops increased firstly and then decreased. The different crop types had different tolerance to Cd, with green pepper showed the strongest tolerance and spinach and tomato showed the least tolerance. ② The bioaccumulation factor of Cd in the edible parts of eight crops ranged in order of wheat > spinach > rice > green pepper > cauliflower > tomato > cucumber > cowpea. ③ Total Cd content in soil was significantly correlated with Cd content in the crops (P<0.05), and the order of the correlation coefficients was spinach > wheat > tomato > cucumber > green pepper > rice > cauliflower > cowpea. ④ The risk threshold value of Cd in soil based on the edible safety of different crops ranged in order of cowpea > cucumber > cauliflower > green pepper > tomato > rice > spinach > wheat. Cucumber, cowpea, and cauliflower were selected as the low-Cd-accumulating varieties according to their tolerance to soil Cd, bioaccumulation capacity, and edible risk threshold values.

Land application of animal manure has been encouraged widely in China. This presents a risk of heavy metals (HMs) accumulation in the soil due to their high contents in the feeds and additives. A 4-year field-scale study was conducted to monitor and estimate HMs accumulation in the soil with land application of swine manure. The results show a clear tendency for As, Hg, Cr, Cu, Zn and Mn to increase gradually with the application duration, yielding an average annual increase of 0.57, 0.011, 6.20, 5.64, 22.58, and 23.45 mg kg-1, respectively, at the annual application rate of about 250 t ha-1 of swine manure. The estimation from the mass balance modeling indicates the environmental risk of Cd, Cu and Zn will exceed the threshold levels for agricultural soils in China in the next 10-50 years. Determination of a suitable application rate of animal manure would be the first consideration for mitigating the environmental risk of HMs currently.

Studies of the interaction and toxicity of pollutant combinations such as heavy metals and PAHs are of practical importance in the remediation and monitoring of the industrial soil environment. This study investigated the single and combined toxicity of chromium(VI) and phenanthrene on three important higher plants: mung beans (Phaseolus aureus), pakchoi cabbage (Brassica chinensis), and rice (Oryza sativa). In experiments using artificial soil matrix, the EC10 and EC20 of the two pollutants, alone and in combination, were analyzed with respect to seed germination, stem length, and above-ground fresh weight of these higher plants. The additive index method was used to evaluate the combined biological toxicity of chromium(VI) and phenanthrene. The results showed that the EC20 of chromium(VI) on the stem lengths of mung beans, pakchoi cabbage, and rice was 289, 248, and 550 mg kg(-1), respectively. The corresponding EC20 values for the fresh weights of the three plants were 334, 307, and 551 mg kg(-1). The EC20 of phenanthrene on the stem lengths of mung beans, pakchoi cabbage, and rice was 528, 426, and 628 mg kg(-1), respectively. The corresponding EC20 values for the fresh weights of the three plants were 696, 585, and 768 mg kg(-1). The EC20 of a combination of chromium(VI) and phenanthrene on the stem lengths of mung beans, pakchoi cabbage, and rice was 192, 173, and 279 mg kg(-1), respectively, and 200, 205, and 271 mg kg(-1) for the fresh weights of the three plants. The single and combined exposure of soil to chromium(VI) and phenanthrene had deleterious effects on plants in the early stage of growth. Overall, pakchoi cabbage was more sensitive than mung beans and rice. The two pollutants exerted synergistic effects on the stem lengths and above-ground fresh weights of both mung beans and rice but antagonistic effects on pakchoi cabbage. The results of this study also suggested pakchoi cabbage as a sensitive indicator of soil pollution.

BACKGROUND: Toll-like receptor 4 participates in the process of acute heart injury. The underlying mechanisms of its protection are multifactorial, but we hypothesized that toll-like receptor-mediated autophagy control plays a vital role. The purpose of this study was to clarify the effect of autophagy on cardiac fibrosis. METHODS: Cardiac fibrosis was induced by subcutaneous isoproterenol (ISO) injection, and rapamycin was simultaneously administered orally for 14 days. Animal echocardiography was then used to evaluate the success of the cardiac fibrosis model, and the mice were killed after the echocardiography examination. RESULTS: Toll-like receptor 4 knockout (TLR4 KO) mice had better heart function than did wild-type (WT) mice (P < .05). Rapamycin treatment reduced the left ventricular ejection fraction to 23.5% (P < .05), and the collagen volume fraction of the ISO and ISO plus rapamycin groups was 5.9% and 25.9%, respectively, in TLR4 KO mice. Compared with the WT mice, Beclin 1 and autophagy were downregulated in TLR4 KO mice (P < .05); however, the ISO plus rapamycin group had higher autophagy activity than did the ISO group in TLR4 KO mice (P < .05). CONCLUSIONS: Our results suggest that TLR4 KO-induced cardioprotection against ISO-induced cardiac fibrosis is associated with reduced autophagy induction. Cardiac fibroblast autophagy participates in its own activation. The moderate inhibition of autophagic activity may be a new strategy for treating cardiac fibrosis.
