RESUMO

Photocatalytic conversion of CO2 to hydrocarbon fuel is a potential strategy to solve energy shortage and mitigate the greenhouse effect. Here, direct Z-scheme heterojunction photocatalysts (In2O3/Bi2S3) without an electron mediator are prepared by a simple hydrolysis method. The In2O3/Bi2S3 composite photocatalysts show greatly boosted photoactivity on CO2 conversion to CO compared with the pristine In2O3 and Bi2S3. The highest CO evolution rate of 2.67 µmol·g-1·h-1 is achieved by In2O3/Bi2S3-3, without any sacrificial agent or cocatalyst, which is about 3.87 times that of In2O3 (0.69 µmol·g-1·h-1). The boosted photocatalytic performance of In2O3/Bi2S3 composite catalysts can be ascribed to the establishment of a Z-scheme heterojunction, improving the photoabsorption and facilitating charge separation and transfer. This study provides a reference for designing and fabricating high-efficiency Z-scheme heterojunction photocatalysts for photocatalytic CO2 reduction.

RESUMO

The secondary active transporter NBCe1 couples the transmembrane movement of Na+ and carbonate species with an apparent stoichiometry of 1Na+ :2HCO3- (the 'influx' mode) or 1Na+ :3HCO3- (the 'efflux' mode). Here, we employed molecular biology, electrophysiology and structural biology approaches to investigate the molecular mechanism for the transport coupling of Na+ and HCO3- in NBCe1. In Xenopus oocytes, decreasing extracellular [HCO3- ] from 66 to 4 mm progressively decreases the Na+ affinity of NBCe1. However, decreasing [Na+ ] from 96 to 35 mm has little effect on the HCO3- affinity. The residues responsible for the coordination of Na+ and HCO3- in the substrate pocket of NBCe1 were respectively determined by mutational and molecular simulation studies. Mutation to the residues for HCO3- coordination decreased the affinities of NBCe1 for both Na+ and HCO3- . However, mutation to the residues for Na+ coordination decreased the affinity for Na+ but had little effect on the affinity for HCO3- . Molecular simulation showed that NBCe1 has the capacity to coordinate only two ions of HCO3- or CO32- . We propose that (1) NBCe1 has an ordered substrate-binding kinetics with the binding of HCO3- preceding that of Na+ ; (2) NBCe1 operating in the influx mode moves 1Na+  + 2HCO3- , whereas NBCe1 in the efflux mode moves 1Na+  + 1HCO3-  + 1CO32- . The substrate-binding kinetics of NBCe1 is distinct from the known kinetics models of many other Na+ -coupled transporters with Na+ binding preceding the driven solute. KEY POINTS: Under physiological conditions, the secondary active transporter NBCe1 can operate in the 'influx' mode with an apparent stoichiometry of 1Na+ :2HCO3- or in the 'efflux' mode with an apparent stoichiometry of 1Na+ :3HCO3- . NBCe1 has an ordered substrate-binding kinetics with HCO3- preceding the binding of Na+ . The kinetics of NBCe1 is distinct from the known kinetics of many other Na+ -driven cotransporters for which the binding of Na+ usually precedes the driven substrate. The residues responsible for the coordination of Na+ and those for carbonate species in the substrate-binding pocket of NBCe1 were determined by mutation and molecular simulation studies. The substrate-binding pocket of NBCe1 contains just two coordination sites for HCO3- or CO32- . It is proposed that NBCe1 in the influx mode moves 1Na+  + 2HCO3- across the plasma membrane, whereas NBCe1 in the efflux mode moves 1Na+ +1HCO3- +1CO32- .

RESUMO

Developing highly efficient photocatalysts toward synchronously removing heavy metals and organic pollutants is still a serious challenge. Herein, we depict hierarchical S-scheme heterostructured photocatalysts prepared via in situ anchoring UiO-66-NH2 nanoparticles onto the CdIn2S4 porous microsphere structures assembled with numerous nanosheets. In the mixed system of Cr(VI) and tetracycline (TC), the optimal photocatalyst (CIS@U66N-30) shows remarkable photocatalytic activities toward the synchronous removal of Cr(VI) (97.26%) and TC (close to 100% of) under visible-light irradiation for 60 min, being the best removal rates among those of the reported photocatalysts, and sustains the outstanding stability and reusability. Its reaction rate constants of Cr(VI) reduction and TC degradation are about 2.06 and 1.58 folds that in the single Cr(VI) and TC systems, respectively. The enhanced photocatalytic activities of CIS@U66N-30 mainly result from the following synergism: (1) its hierarchical structure offers abundant active sites, and the S-scheme migration mechanism of charge carriers in the heterostructure accelerates the separation and migration of the useful photoinduced electrons and holes with the high redox capability; (2) Cr(VI) and TC can serve as the electron scavenger for TC oxidation degradation and the hole and •OH scavenger for Cr(VI) reduction, respectively, further enhancing the separation and utilization efficiency of photoinduced electrons and holes. Besides, the possible TC degradation pathway and plausible S-scheme photocatalytic mechanism over CIS@U66N-30 for the concurrent elimination of Cr(VI) and TC are proposed.

RESUMO

OBJECTIVE: To investigate the current status of antibiotic use for very and extremely low birth weight (VLBW/ELBW) infants in neonatal intensive care units (NICUs) of Hunan Province. METHODS: The use of antibiotics was investigated in multiple level 3 NICUs of Hunan Province for VLBW and ELBW infants born between January, 2017 and December, 2017. RESULTS: The clinical data of 1 442 VLBW/ELBW infants were collected from 24 NICUs in 2017. The median antibiotic use duration was 17 days (range: 0-86 days), accounting for 53.0% of the total length of hospital stay. The highest duration of antibiotic use was up to 91.4% of the total length of hospital stay, with the lowest at 14.6%. In 16 out of 24 NICUs, the antibiotic use duration was accounted for more than 50.0% of the hospitalization days. There were 113 cases with positive bacterial culture grown in blood or cerebrospinal fluid, making the positive rate of overall bacterial culture as 7.84%. The positive rate of bacterial culture in different NICUs was significantly different from 0% to 14.9%. The common isolated bacterial pathogens Klebsiella pneumoniae was 29 cases (25.7%); Escherichia coli 12 cases (10.6%); Staphylococcus aureus 3 cases (2.7%). The most commonly used antibiotics were third-generation of cephalosporins, accounting for 41.00% of the total antibiotics, followed by penicillins, accounting for 32.10%, and followed by carbapenems, accounting for 13.15%. The proportion of antibiotic use time was negatively correlated with birth weight Z-score and the change in weight Z-score between birth and hospital discharge (rs=-0.095, -0.151 respectively, P<0.01), positively correlated with death/withdrawal of care (rs=0.196, P<0.01). CONCLUSIONS: Antibiotics used for VLBW/ELBW infants in NICUs of Hunan Province are obviously prolonged in many NICUs. The proportion of routine use of third-generation of cephalosporins and carbapenems antibiotics is high among the NICUs.

RESUMO

To prepare Helix aspersa muller-paeonol nanogel( PAE-HAM-Gels) with anti-proliferative scar effect, evaluate its skin penetration, retention and irritation, and to investigate its prevention and treatment effect for hypertrophic scar in rabbit ears. The dermal retention, transdermal rate and cumulative permeability of paeonol were investigated in vitro by using the modified Franz diffusion cell and the abdominal skin of suckling pigs,SD rats and KM mice, respectively, and the in vitro permeation curves were drawn. The normal skin of the back of New Zealand rabbits was continuously treated with PAE-HAM-Gels for 7 days, and the physiological state of the skin was observed under light microscope after HE staining by using homologous left and right contrast method. The hypertrophic scar model in rabbit ears was established, and the New Zealand rabbits were randomly divided into blank group, model group, positive drug group,PAE-Gels group and PAE-HAM-Gels group. After 28 days of administration, the scar hyperplasia rate and scar elevation index( SEI) of each group were calculated; the scar tissues were taken and stained with Masson for observation of collagen fibers and muscle fibers hyperplasia under light microscope, and the expression level of TGF-ß1 in each group was detected. The Qnof PAE-HAM-Gels in aqueous solution was in line with the Higuchi equation, and its transdermal rate, cumulative permeation and dermal retention in different animal skins were all higher than those of PAE-Gels. The skin of the drug-administered group was intact, without erythema, edema or other phenomena; under light microscope, the subcutaneous tissue and the epidermal cells were neatly arranged with uniform thickness, which showed no difference from the blank group. The scar hyperplasia rate of the PAE-HAM-Gels group was 62. 50%; SEI was 2. 17±0. 33 and TGF-ß1 was( 815. 4±34. 69) ng·L~(-1),significantly different from those in model group( P<0. 01). Masson staining showed that as compared with the model group, the number of collagen fibers and muscle fibers was small and the arrangement was loose and tidy in the PAE-HAM-Gels group, with regular arrangement of chondrocytes and a small number of inflammatory cells and microvessels.PAE-HAM-Gels have good transdermal properties and dermal retention without skin irritation, offering a promising therapeutic strategy for transdermal delivery during the prevention and treatment of hypertrophic scar in rabbit ears.

RESUMO

Hypertrophic scar( HS) is a very common skin fibrosis disorder after human skin injury and wound healing. The objective of this study was to investigate the efficacy of cell penetrating peptide TAT-modified liposomes loaded with salvianolic acid B( SAB-TAT-LIP) on proliferation, migration and cell cycle of human skin fibroblasts( HSF),and preliminarily evaluate its effect on prevention and treatment of HS. HSF were cultured in vitro, and MTT assay was used to detect the inhibitory effect of SAB-TAT-LIP on cell proliferation. Cell migration was assessed by Transwell chamber method and scratch method; and cell cycle change was detected by flow cytometry. In vitro cell studies showed that blank liposome basically had no toxic effect on HSF. Different concentrations of SABTAT-LIP inhibited proliferation on HSF in varying degrees after intervention for different periods in a dose and time dependent manner;meanwhile,SAB-TAT-LIP significantly inhibited the migration and invasion of HSF. At the same time,SAB-TAT-LIP could block the cell cycle at G0/G1 phase after intervention for 48 h,P<0.01 as compared with the blank control group. Conclusively, our experimental data quantitatively demonstrate that SAB-TAT-LIP has significant inhibitory effect on cells proliferation, invasion and migration, with blocking effect on G0/G1 phase. This may offer a promising therapeutic strategy for transdermal delivery in prevention and treatment of HS.

RESUMO

Reactive oxygen species (ROS) play important regulatory roles in plant growth and development, as well as in cell differentiation and stress responses. Respiratory burst oxidase homolog (RBOH) is the key enzyme in ROS production. So far, the Rboh family genes in Pyropia yezoensis have not been comprehensively characterized, and whether their function was involved in the formation of archeospores is still unknown. In this study, a total of 11 PyRboh genes were identified from the P. yezoensis genome by homology mining. Through phylogenetic analysis, it is suggested that the PyRboh genes were evolutionarily conserved among the lineages of red algae, but a few genes exhibited a species-specific manner. The treatment of P. yezoensis blades with NADPH oxidase inhibitor diphenylene iodonium (DPI) could significantly inhibit the formation of archeospores, suggesting that RBOH may be involved in the formation of archeospores. According to PyRboh gene expression analysis using the P. yezoensis strains with obvious differences in releasing archeospores, it is showed that the expression trends of most genes were consistent, with no significant difference between strains, whereas the expression pattern of the two P. yezoensis-specific genes (PyRbohJ and PyRbohK) was positively correlated with the amount of archeospores. Furthermore, as treatment of blades with allantoin resulted in a significant increase in the release of archeospores, the expression levels of PyRbohJ and PyRbohK were also consistently upregulated, further confirming the relationship between the two genes and archeospore formation. These findings provide insights into the molecular mechanism of P. yezoensis archeospore formation.

RESUMO

Achieving both rapid adsorption rate and high adsorption capacity for bisphenol micropollutants from aquatic systems is critical for efficient adsorbents in water remediation. Here, we elaborately prepared three nitrogen-rich triazine-based porous polymers (NTPs) with similar geometric configurations and nitrogen contents (41.70-44.18 wt%) while tunable BET surface areas and micropore volumes in the range of 454.7-536.3 m2 g-1 and 0.20-0.84 cm3 g-1, respectively. It was systematically revealed that the synergy of hydrogen bonding, π-π electron-donor-acceptor interaction, and micropore preservation promoted the rapid (within 5 min) and high capacity adsorption of bisphenols by NTPs. Particularly, microporous-dominated NTPs-3 with the highest micro-pore volume (0.84 cm3 g-1) displays remarkable adsorption capacity towards bisphenol A as evidenced by the adsorption capacity of 182.23 mg g-1. A simple column filter constructed by NTPs-3 also expressed good dynamic adsorption and regeneration capacity. This work provided new insight into the rational design and engineering of nitrogen-rich porous polymers for the remediation of micropollutant wastewater.

RESUMO

In this work, a dense and acid-resistant beta zeolite membrane was applied to improve the esterification of citric acid and n-butanol, for the first time. Through the continuous removal of the by-product water via pervaporation (PV), the conversion of citric acid was significantly enhanced from 71.7% to 99.2% using p-Toluenesulfonic acid (PTSA) as catalyst. PTSA was a well-known strong acid, and the membrane kept almost no change after PV-esterification, indicating the superior acid resistance of beta zeolite membrane. Compared to the use of acid-resistant MOR zeolite membrane by PV-esterification, a consistently higher conversion of citric acid was obtained using a high-flux beta zeolite membrane. The results showed that high water permeation on the beta zeolite membrane, with good acid resistance, had a strong promoting effect on esterification, leading to an improved conversion. In addition, the citric acid conversion of 97.7% could still be achieved by PV-esterification at a low reaction temperature of 388 K.

RESUMO

OBJECTIVE: To report the experiences of the diagnosis and treatment of primary lymphoma of the small intestine (PSIL). METHODS: The clinical data of 15 patients with PSIL treated from January 2003 to July 2007 was investigated retrospectively. Of the 15 cases, 9 patients were male and 6 were female, the average age was 51.6 years (range, 18 - 73 years). Data of gender, age, clinical manifestation, laboratory examination, imageology examination, diagnosis and treatment of the patients was reviewed. RESULTS: The most common clinical manifestations were as follow: abdominal pain, abdominal lump, bowel obstruction, gastrointestinal hemorrhage and athrepsy. Serum tumor markers were checked normal. All the 15 cases were found with tumor by spiral CT, and 12 cases were diagnosed as PSIL. Eleven cases were given Ba-meal examinations, and positive results was found in 4 cases, and only 1 case was considered to be PSIL. All the 15 patients received operation. All the patients were diagnosed as non-Hodgkin lymphoma (NHL) by postoperative pathology (8 patients as diffuse large B-cell lymphoma, 5 as mucosa associated lymphoid tissue type B cell lymphoma and 2 as enteropathy-type intestinal T cell lymphoma). No perioperative death occurred. Ten patients received adjuvant chemotherapy with the regimen of CHOP (cyclophosphamide + epirubicin + vincristine + prednisone) after the operation. Fourteen cases were followed-up for a mean time of 30 months (range, 6 - 52 months). The 1- and 3-years survival rate was 85.7% and 57.1%, respectively. CONCLUSIONS: PSIL has no specific clinical manifestations, the diagnostic rate with barium study is low, spiral CT scan is a promising diagnostic method for PSIL. Operation combined with chemotherapy is important for PSIL.

RESUMO

Dense and good catalytic performance TS-1 zeolite membranes were rapidly prepared on porous mullite support by secondary hydrothermal synthesis. The properties of seed crystals were very important for the preparation of high-catalytic performance TS-1 zeolite membranes. Influences of seed crystals (Ti/Si ratios, size, morphology, and zeolites concentration of the seed suspension) on the growth and catalytic property of TS-1 zeolite membranes were investigated in details. High Ti/Si ratio, medium-size, and morphology of the seed crystals were critical for preparing the high-performance TS-1 zeolite membrane. Compared with the bi-layer TS-1 zeolite membrane (inner and outer of the mullite tube), the mono-layer TS-1 zeolite membrane had a better catalytic performance for Isopropanol IPA oxidation with H2O2. When the Ti/Si ratio, size, and morphology of the TS-1 zeolites were 0.030, 300 nm, ellipsoid, and the zeolites concentration of the seed suspension was 5%, the IPA conversion, and flux through the TS-1 zeolite membrane were 98.23% and 2.58 kg·m-2·h-1, respectively.

RESUMO

So far, at least 12 cases of immunoglobulin G4 related disease (IgG4-RD) coexisting with colorectal cancer have been reported in the literature, but IgG4-RD with rectal cancer is still extremely rare. In addition, recently, the correlation between IgG4-RD and malignancy strongly motivates immunologists and pathologists to conduct further research. In this case study, we present a case of IgG4-RD with rectal cancer and review the pathological characteristics of IgG4-RD and discuss the association between IgG4-RD and malignancy. Indeed, the relationship between IgG4-RD and malignancy is controversial, but we remind physicians that they should be aware of the possible coexistence of malignancy among IgG4-RD patients.

RESUMO

The ENF peptide family, so termed after the consensus sequence in their amino termini (Glu-Asn-Phe-), is assumed to play multiple important roles in defense reactions, growth regulation, and homeostasis of Lepidopteran insects. The paralytic peptide of Bombyx mori (BmPP) is one such peptide that is involved in the paralytic and plasmatocyte-spreading activities in the hemocyte immune reaction. The growth-blocking peptide of Pseudaletia separata (PsGBP), which is also a member of the ENF peptide family, has similar functions that can reportedly be attenuated by the growth-blocking peptide-binding protein (GBP-BP). Using the fluorescent differential display (FDD) technique, the differential expression pattern of genes in highly susceptible silkworm strain 306 were analyzed, following infection with B. mori nuclear polyhedrosis virus (BmNPV), and a differential band (G12(782)) was obtained from the hemolymph RNA pools. Using 5'-RACE with a specially designed primer based on the FDD study, a 1,401 bp cDNA clone was obtained containing a 1,311 bp open reading frame (ORF, GenBank accession number DQ306881). The deduced protein was highly homologous in primary structure to GBP-BP and was termed B. mori paralytic peptide-binding protein (PP-BP). The B. mori PP-BP gene is organized into two exons and only one intron, using bioinformatics searches.Using RT-PCR analysis, it was found that the B. mori PP-BP gene was expressed almost exclusively in the hemolymph. Real-time quantitative PCR analysis indicated that the B. mori PP-BP mRNA level in B. mori strain 306 exposed to BmNPV was much higher than that in B. mori strain without the virus infection. This result implies that the B. mori PP-BP is related to the cellular immune response after BmNPV invades the hemolymph.

RESUMO

The genome of Bombyx mori densonucleosis virus type 3 (China isolate) contains two kinds different single-stranded linear DNA molecules (VD1, VD2). The VD2 was purified and cloned into the pUC119 vector, and the complete nucleotide sequence of VD2 was determined. Sequence analysis showed that the VD2 genome sequence consisted of 6022 nts, including inverted terminal repeats (ITRs) of 524 nts. In the viral genome, two major open reading frames (ORF1 and ORF2) in the plus strand and one minor ORF (ORF3) in the complementary strand were identified. Computer analysis suggested the plus stand ORF1 and the minus strand ORF3 most likely encode the major nonstructural protein, while the plus stand ORF2 most likely encodes the major structural protein. Comparing the complete genome sequence of BmDNV-3 VD2 with that of BmDNV-2 VD2 (Yamanashi isolate) demonstrated that they shared 97.7% genome sequence in the VD2 region, with substitutions of 132 nucleotides, deletions of 11 nucleotides and insertions of 2 nucleotides in the VD2 region of BmDNV-3. The results suggested that BmDNV-3 is closely related to BmDNV-2, but with some differences, giving a better understanding about the variation of the viruses and providing clues to their evolution.

After 28 days of administration, the scar hyperplasia rate and scar elevation index( SEI) of each group were calculated; the scar tissues were taken and stained with Masson for observation of collagen fibers and muscle fibers hyperplasia under light microscope, and the expression level of TGF-β1 in each group was detected. The scar hyperplasia rate of the PAE-HAM-Gels group was 62. 50%; SEI was 2. 17±0. 33 and TGF-β1 was( 815. 4±34. 69) ng·L~(-1),significantly different from those in model group( P<0.
