RESUMO

Drought stress is one of the dominating challenges to the growth and productivity in crop plants. Elucidating the molecular mechanisms of plants responses to drought stress is fundamental to improve fruit quality. However, such molecular mechanisms are poorly understood in apple (Malus domestica Borkh.). In this study, we explored that the BTB-BACK-TAZ protein, MdBT2, negatively modulates the drought tolerance of apple plantlets. Moreover, we identified a novel Homeodomain-leucine zipper (HD-Zip) transcription factor, MdHDZ27, using a yeast two-hybrid (Y2H) screen with MdBT2 as the bait. Overexpression of MdHDZ27 in apple plantlets, calli, and tomato plantlets enhanced their drought tolerance by promoting the expression of drought tolerance-related genes [responsive to dehydration 29A (MdRD29A) and MdRD29B]. Biochemical analyses demonstrated that MdHDZ27 directly binds to and activates the promoters of MdRD29A and MdRD29B. Furthermore, in vitro and in vivo assays indicate that MdBT2 interacts with and ubiquitinates MdHDZ27, via the ubiquitin/26S proteasome pathway. This ubiquitination results in the degradation of MdHDZ27 and weakens the transcriptional activation of MdHDZ27 on MdRD29A and MdRD29B. Finally, a series of transgenic analyses in apple plantlets further clarified the role of the relationship between MdBT2 and MdHDZ27, as well as the effect of their interaction on drought resistance in apple plantlets. Collectively, our findings reveal a novel mechanism by which the MdBT2-MdHDZ27 regulatory module controls drought tolerance, which is of great significance for enhancing the drought resistance of apple and other plants.

Microbial reduction of perchlorate (ClO4-) is emerging as a cost-effective strategy for groundwater remediation. However, the effectiveness of perchlorate reduction can be suppressed by the common co-contamination of nitrate (NO3-). We propose a means to overcome the limitation of ClO4- reduction: depositing palladium nanoparticles (Pd0NPs) within the matrix of a hydrogenotrophic biofilm. Two H2-based membrane biofilm reactors (MBfRs) were operated in parallel in long-term continuous and batch modes: one system had only a biofilm (bio-MBfR), while the other incorporated biogenic Pd0NPs in the biofilm matrix (bioPd-MBfR). For long-term co-reduction, bioPd-MBfR had a distinct advantage of oxyanion reduction fluxes, and it particularly alleviated the competitive advantage of NO3- reduction over ClO4- reduction. Batch tests also demonstrated that bioPd-MBfR gave more rapid reduction rates for ClO4- and ClO3- compared to those of bio-MBfR. Both biofilm communities were dominated by bacteria known to be perchlorate and nitrate reducers. Functional-gene abundances reflecting the intracellular electron flow from H2 to NADH to the reductases were supplanted by extracellular electron flow with the addition of Pd0NPs.

Selenium-containing polymers are a group of fascinating functional polymers with unique structures, properties, and applications, which have been developed recently but only with limited examples. The challenges of developing selenium-containing polymers with structural and functional diversity include the lack of economic and safe monomers, lack of efficient and convenient synthetic approaches, and poor stability of selenium-involving covalent bonds. In this work, room-temperature metal-free multicomponent polymerizations (MCPs) of elemental selenium, diisocyanides, and dipropargyl alcohols were developed, and polymers with a selenium-containing aliphatic heterocycle, 1,3-oxaselenolane, were synthesized through these MCPs directly from elemental selenium. The alicyclic poly(oxaselenolane)s enjoyed high yields (up to 93%), high molecular weights (up to 15 600 g/mol), high thermal and chemical stability, good solubility and processability. With the structural design of the poly(oxaselenolane)s and their high selenium contents of up to 33.7 wt %, the refractive indices of their spin-coated thin films could reach 1.8026 at 633 nm and maintain 1.7770 at 1700 nm. It is anticipated that these efficient, convenient, mild, and economic multicomponent polymerizations of elemental selenium can promote the selenium-related polymer chemistry and accelerate the exploration of diversified selenium-containing functional polymer materials.

Here, a novel pseudorotaxanes-type crosslinker of a supramolecular polymer network (WP5-PN) has been constructed from a host water-soluble pillar[5]arene (WP5) and a guest naphthalene dimethylamine derivative (PN) via a stepwise process involving multiple non-covalent interactions. The obtained supramolecular polymers were able to transform into a supramolecular polymer gel (WP5-PN-G) and show AIE properties in DMSO-H2O binary solution. Interestingly, due to the dynamic and reversible nature of non-covalent interactions, the resultant supramolecular polymer gels exhibited external stimuli-responsiveness to different parameters, such as temperature, acid-base, competitive guest and mechanical stress. Moreover, WP5-PN-G showed fluorescent response for Fe3+ and Cu2+, while its xerogel showed excellent recyclable separation properties for these metal ions with adsorption rates up to 98.07% and 95.38%, respectively. Moreover, by rational introduction of these metal ions into the WP5-PN-G, corresponding metal ion coordinated metallogels, such as WP5-PN-FeG and WP5-PN-CuG were obtained. These metallogels could selectively and sensitively sense F- and CN-, respectively. The detection limits of these metallogels for F- and CN- were about 1 × 10-8 M. The WP5-PN-G has potential applications in multi-analytes detection and separation as well as fluorescent display materials.

The terminal branch orders of plant root systems have been proposed as short-lived 'ephemeral' modules specialized for resource absorption. The occurrence of ephemeral root modules has so far only been reported for a temperate tree species and it is unclear if the concept also applies to other woody (shrub, tree) and herb species. Fine roots of 12 perennial dicotyledonous herb, shrub and tree species were monitored for two growing seasons using a branch-order classification, sequential sampling and rhizotrons in the Taklamakan desert. Two root modules existed in all three plant functional groups. Among the first five branch orders, the first two (perennial herbs, shrubs) or three (trees) root orders were ephemeral and had a primary anatomical structure, high nitrogen (N) concentrations, high respiration rates and very short life spans of 1-4 months, whereas the last two branch orders in all functional groups were perennial, with thicker diameters, no or collapsed cortex, distinct secondary growth, low N concentrations, low respiration rates, but much longer life spans. Ephemeral, short-lived root modules and long-lived, persistent root modules seem to be a general feature across many plant functional groups and could represent a basic root system design.

The effects of enhanced UV-B radiation on abundance, community composition and the total microbial activity of soil bacteria in alpine meadow ecosystem of Qinghai-Tibet Plateau were investigated. Traditional counting and 16S rRNA gene sequencing were used to investigate the culturable bacteria and their composition in soil, meanwhile the total microbial activity was measured by microcalorimetry. The population of soil culturable bacteria was slightly reduced with the enhanced UV-B radiation in both of the two depths, 2.46 × 10(6) CFU/g in upper layer (0-10 cm), 1.44 × 10(6) CFU/g in under layer (10-20 cm), comparing with the control (2.94 × 10(6) CFU/g in upper layer, 1.65 × 10(6) CFU/g in under layer), although the difference was not statistically significant (P > 0.05). However, the bacteria diversity decreased obviously due to enhanced UV-B, the number of species for upper layer was decreased from 20 to 13, and from 16 to 13 for the lower layer. The distribution of species was also quite different between the two layers. Another obvious decrease induced by enhanced UV-B radiation was in the total soil microbial activities, which was represented by the microbial growth rate constant (k) in this study. The results indicated that the culturable bacteria community composition and the total activity of soil microbes have been considerably changed by the enhanced UV-B radiation.

The Yangtze River Economic Belt (YREB) is a major national strategic development area in China, and the development of the YREB will greatly promote the development of the entirety China, so research on its agricultural production efficiency is also of great significance. This paper is committed to studying the agricultural production efficiency of 11 provinces in the YREB and adopts a combination of the Data Envelopment Analysis (DEA) model and the Malmquist index to make a dynamic and static analysis on the YREB's agricultural production efficiency from 2010 to 2019. Then, a three-stage DEA Malmquist model that eliminates the factors of random interference and management inefficiency is compared to a model without elimination. The results show that the adjusted technological efficiency changes, technological progress, and total factor productivity increased by -0.1%, 0.24%, and 0.22%, respectively. When comparing these values to the pre-adjustment values, the results indicate that the effect of environmental variables cannot be ignored when studying the agricultural production efficiency of the YREB. At the same time, the differences in the agricultural production efficiency in the YREB are reasonably explained, and feasible suggestions are put forward.

This paper investigates the impact of two types of environmental regulations (ERs), command-and-control environmental regulation (CACER) and market-incentive environmental regulation (MIER), on green total factor productivity (GTFP) through outward foreign direct investment (OFDI) in 30 provinces in China for the period of 2006-2019. The Global Malmquist-Luenberger (GML) Index based on non-radial directional distance function (NDDF) considering undesired outputs is used to measure GTFP growth at the provincial level. To explore the mediating effect of OFDI, the two-step econometric model and the non-linear mediating effect model are employed. The empirical results show that CACER has an inverted U-shaped impact on OFDI and a U-shaped impact on GTFP, while MIER has a linearly positive effect on OFDI and GTFP. The current intensity of CACER lies on the left side of the inflection point of the U-shaped curve. OFDI significantly positively influences the increase in GTFP and is a significant mediating variable in the relationship between ERs and GTFP. Moreover, the introduction of OFDI delays the appearance of the inflection point. Further analysis, taking into account the regional heterogeneity, indicates that the inverted U-shaped and U-shaped curve is still valid in the eastern and western area and that the mediating effect of OFDI on ERs in the western area is stronger than that in the eastern area. Based on these conclusions, policy implications are provided to improve GTFP in China.

With the acceleration of industrialization and urbanization, the Yangtze River Economic Belt (YREB) is facing many environmental problems that need to be solved in the process of development. This paper aims to analyze the environmental governance effects of nine provinces and two municipalities in the Yangtze River Economic Belt from 2009 to 2018. Firstly, based on the input-output index, the slacks-based measure (SBM) undesirable model and Malmquist (ML) index were used to measure the green total factor productivity (GTFP) of the YREB from 2009 to 2018. The results showed that the technological progress index contributed the most to the GTFP of the YREB, followed by the pure technical efficiency index and the scale efficiency index. Environmental regulation has no significant impact on the GTFP of the YREB. Secondly, by analyzing the effect of environmental governance in the YREB, the results show that the main reasons for the ineffective environmental governance in the YREB are the redundant input of environmental resources, excessive unwanted output, and low harmless treatment rate of municipal solid waste, rather than the low level of urban environmental management. Finally, this paper provides recommendations for the ineffective provinces and municipalities of the YREB to further optimize the input-output factors of environmental governance.

A novel supramolecular organic framework gel (PQPA) has been constructed by a novel bilateral 8-hydroxyquinoline modified pillar[5]arene host (P5Q) and a bilateral bromohexyl functionalized pillar[5]arene guest (P5C6). Interestingly, the gel PQPA shows special selective and ultrasensitive sensing property for Fe3+. The limit of lowest detection (LOD) of gel PQPA for Fe3+ is 0.102 nM. Meanwhile, by adding Fe3+ into the gel PQPA, a no fluorescence Fe3+ coordinated metallogel (PQPA-Fe) could be constructed. Due to the competitive coordination of F-, fluorescent "turn-on" ultrasensitive response of the Fe3+ coordinated metallogel (PQPA-Fe) for F- is achieved. The limit of lowest detection of the Fe3+ coordinated metallogel (PQPA-Fe) for F- is 9.79 nM. In addition, the xerogel of the supramolecular organic framework gel (PQPA) is capable for removing Fe3+ in a lower concentration aqueous solution at an adsorption rate of 99.9%. More importantly, the supramolecular organic framework gel (PQPA) could be used to detect Fe3+ in the Yellow River Water samples. The supramolecular organic framework gel (PQPA) and Fe3+ coordinated metallogel (PQPA-Fe) could act as ultrasensitive reversible fluorescent chemosensor, test kits and fluorescent display materials for Fe3+ and F-.

A novel approach for in situ generation of AgI quantum dots by the confinement of a pillar[5]arene-based supramolecular polymer network has been successfully developed. The supramolecular polymer network (SPN-QP) was constructed by using a bis-8-hydroxyquinoline-modified pillar[5]arene derivative as a host (H-QP) and a bis-pyridinium-modified decane as guest (G-PD). The SPN-QP shows ultrasensitive response for Ag+ . The limit of detection is about 7.44×10-9  M.. Interestingly, when I- was added to the SPN-QP+Ag+ system, an unexpected strong warm-white fluorescence emission was observed. After carefu investigation, we found that the strong warm-white fluorescence emission could be attributed to the in situ formation of AgI quantum dots under the confinement of the supramolecular polymer network (SPN-QP). Based on this approach, ultrasensitive detection of I- was realized. The limit of detection for I- is 4.40×10-9  M. This study provides a new way for the preparation of quantum dots under the confinement of supramolecular polymer network as well as ultrasensitive detection of ions by in situ formation of quantum dots.

A novel palladium-catalyzed domino cyclization/alkylation of terminal alkynes was achieved for the synthesis of alkynyl-functionalized 3,3-disubstituted azaindoline derivatives under air atmosphere conditions. Various types of terminal alkynes, including aromatic alkynes, aliphatic alkynes, and ferrocene acetylene, can undergo the process successfully. The protocol provides a range of alkynyl-functionalized azaindoline scaffolds bearing a quarternary carbon center.

RESUMO

In order to gain the optimal extract condition of extraction polysaccharides in Loquate leaf by ultrasound technique, the single factor and the orthogonal experiments were carried. The results showed that the optimal condition turned out to be ultrasound extraction time 35 min, ultrasound power 120W, extraction temperature 60 degrees C and the liquid-to-solid ratio 12: 1. In contrast to conventional extraction method, the ultrasound technique could be a better method for extracting the polysaccharides from Loquat leaf with a higher yield and shorter time.

Diversity of arbuscular mycorrhizal fungi (AMF) is well studied in many ecosystems, but little is known about AMF in cold-dominated regions with very high altitude. Here, we examined AMF communities associated with two plant species in the Tibet Plateau. Roots and rhizosphere soils of Dracocephalum heterophyllum (pioneer species) and Astragalus polycladus (late-successional species) were sampled at five sites with altitude from 4500 to 4800 m a.s.l. A total of 21 AMF phylotypes were identified from roots and spores following cloning and sequencing of 18S rRNA gene, including eight new phylotypes and one new family-like clade. More AMF phylotypes colonized root samples of D. heterophyllum (5.4±0.49) than of A. polycladus (1.93±0.25). Vegetation coverage was the most important factor influencing AMF community composition in roots. Globally infrequent phylotype Glo-B2 in Glomus group B was the most dominant in roots, followed by globally frequent phylotype Glo-A2 related to Glomus fasciculatum/intraradices group. Our findings suggest that a diverse AMF flora is present in the Tibet Plateau, comprising both potentially habitat-selective and generalist fungi.
