RESUMO

Proteasome-mediated degradation of chromatin-bound NF-κB is critical in terminating the transcription of pro-inflammatory genes and can be triggered by Set9-mediated lysine methylation of the RelA subunit. However, the E3 ligase targeting methylated RelA remains unknown. Here, we find that two structurally similar substrate-recognizing components of Cullin-RING E3 ligases, WSB1 and WSB2, can recognize chromatin-bound methylated RelA for polyubiquitination and proteasomal degradation. We showed that WSB1/2 negatively regulated a subset of NF-κB target genes via associating with chromatin where they targeted methylated RelA for ubiquitination, facilitating the termination of NF-κB-dependent transcription. WSB1/2 specifically interacted with methylated lysines (K) 314 and 315 of RelA via their N-terminal WD-40 repeat (WDR) domains, thereby promoting ubiquitination of RelA. Computational modeling further revealed that a conserved aspartic acid (D) at position 158 within the WDR domain of WSB2 coordinates K314/K315 of RelA, with a higher affinity when either of the lysines is methylated. Mutation of D158 abolished WSB2's ability to bind to and promote ubiquitination of methylated RelA. Together, our study identifies a novel function and the underlying mechanism for WSB1/2 in degrading chromatin-bound methylated RelA and preventing sustained NF-κB activation, providing potential new targets for therapeutic intervention of NF-κB-mediated inflammatory diseases.

The evolution of insect vector-pathogen relationships has long been of interest in the field of molecular ecology. One system of special relevance, due to its economic impacts, is that between Diaphorina citri and 'Candidatus Liberibacter asiaticus' (CLas), the cause of the severe Asian form of huanglongbing. CLas-positive D. citri are more fecund than their CLas-negative counterparts, boosting opportunities for pathogens to acquire new vector hosts. The molecular mechanism behind this life-history shift remains unclear. Here, we found that CLas promoted ovarian development and increased the expression of the vitellogenin receptor (DcVgR) in ovaries. DcVgR RNAi significantly decreased fecundity and CLas titer in ovaries, extended the preoviposition period, shortened the oviposition period and blocked ovarian development. Given their importance in gene regulation, we explored the role of miRNAs in shaping these phenotypes and their molecular triggers. Our results showed that one miRNA, miR-275, suppressed DcVgR expression by binding to its 3' UTR. Overexpression of miR-275 knocked down DcVgR expression and CLas titer in ovaries, causing reproductive defects that mimicked DcVgR knockdown phenotypes. We focused, further, on roles of the Juvenile Hormone (JH) pathway in shaping the observed fecundity phenotype, given its known impacts on ovarian development. After CLas infection, this pathway was upregulated, thereby increasing DcVgR expression. From these combined results, we conclude that CLas hijacks the JH signalling pathway and miR-275, thereby targeting DcVgR to increase D. citri fecundity. These changes simultaneously increase CLas replication, suggesting a pathogen-vector host mutualism, or a seemingly helpful, but cryptically costly life-history manipulation.

Huanglongbing, a globally devastating citrus disease, is associated with Candidatus Liberibacter asiaticus (CLas) and is mainly transmitted by Diaphorina citri. Verification of the distribution and dynamics of CLas in D. citri is critical to understanding CLas transmitted by vectors in nature. Here, the distribution and titers of CLas in different sexes and tissues of D. citri adults were investigated by fluorescence in-situ hybridization (FISH) and quantitative real-time PCR (qRT-PCR). Results showed that CLas had widespread distribution in the brain, salivary glands, digestive system, and reproductive system of both females and males, indicating a systemic infection of CLas in D. citri. Moreover, CLas fluorescence intensity and titers were significantly increased in both the digestive system and the female reproductive system with development and there was a marked decreased in both the salivary glands and the male brain, but there was no significant change in the female brain or the male reproductive system. Furthermore, the distribution and dynamics of CLas in embryos and nymphs were investigated. CLas was observed in all laid eggs and subsequent first-second-instar nymphs, indicating that a high percentage of embryos and nymphs resulting from infected D. citri mothers were infected with CLas.

The preparation of methanol chemicals through CO2 and H2 gas is a positive measure to achieve carbon neutrality. However, developing catalysts with high selectivity remains a challenge due to the irreversible side reaction of reverse water gas shift (RWGS), and the low-temperature characteristics of CO2 hydrogenation to methanol. In-plane sulfur vacancies of MoS2 can be the catalytic active sites for CH3OH formation, but the edge vacancies are more inclined to the occurrence of methane. Therefore, MoS2 and a series of MoS2/Nix and MoS2/Cox catalysts doped with different amounts are prepared by a hydrothermal method. A variety of microscopic characterizations indicate that Ni and Co doping can form NiS2 and CoS2, the existence of these substances can prevent CO2 and H2 from contacting the edge S vacancies of MoS2, and the selectivity of the main product is improved. DFT calculation illustrates that the larger range of orbital hybridization between Ni and MoS2 leads to CO2 activation and the active hydrogen is more prone to surface migration. Under optimized preparation conditions, MoS2/Ni0.2 exhibits relatively good methanol selectivity. Therefore, this strategy of improving methanol selectivity through metal doping has reference significance for the subsequent research and development of such catalysts.

Catalytic ammoxidation of alcohols into nitriles is an essential reaction in organic synthesis. While highly desirable, conducting the synthesis at room temperature is challenging, using NH3 as the nitrogen source, O2 as the oxidant, and a catalyst without noble metals. Herein, we report robust photocatalysts consisting of Fe(III)-modified titanium dioxide (Fe/TiO2) for ammoxidation reactions at room temperature utilizing oxygen at atmospheric pressure, NH3 as the nitrogen source, and NH4Br as an additive. To the best of our knowledge, this is the first example of catalytic ammoxidation of alcohols over a photocatalyst using such cheap and benign materials. Various (hetero) aromatic nitriles were synthesized at high yields, and aliphatic alcohols could also be transformed into corresponding nitriles at considerable yields. The modification of TiO2 with Fe(III) facilitates the formation of active •O2- radicals and increases the adsorption of NH3 and amino intermediates on the catalyst, accelerating the ammoxidation to yield nitriles. The additive NH4Br impressively improves the catalytic efficiency via the formation of bromine radicals (Br•) from Br-, which works synergistically with •O2- to capture H• from Cα-H, which is present in benzyl alcohol and the intermediate aldimine (RCH═NH), to generate the active carbon-centered radicals. Further, the generation of Br• from the Br- additive consumes the photogenerated holes and OH• radicals to prevent over-oxidation, significantly improving the selectivity toward nitriles. This amalgamation of function and synergy of the Fe(III)-doped TiO2 and NH4Br reveals new opportunities for developing semiconductor-based photocatalytic systems for fine chemical synthesis.

In the operation of proton exchange membrane fuel cells (PEMFCs), the ionomer-perfluorosulfonic acid (PSFA) membrane side chains are easily attacked by free radicals, resulting in membrane degradation. In this work, the chemical degradation effect of side chains in the PSFA membrane on proton dissociation and transfer behaviors is investigated by means of the quantum chemistry calculation. The rotation of the H atom in the acid group after the degradation is evaluated. The impact of the electrostatic potential (ESP) and electronegativity of the side chains is analyzed. The results demonstrate that the membrane degradation decreases the positive potential of the proton in the acid group, leading to the proton being less active so that more water molecules are required for the spontaneous proton dissociation. The rotation of the H atom in the acid group affects the proton dissociation mode owing to the change of the hydrogen bond network. It is found that the ESP of the acid group in two side chain fragments influences each other and the water molecules between two side chains can be shared to reduce the number of water molecules for the proton dissociation.

In proton exchange membrane fuel cells (PEMFCs), free radicals easily attack ionomers, resulting in membrane degradation. In this work, the multistate empirical valence bond (MS-EVB) method is employed to investigate the chemical degradation impact of Nafion membranes on proton transport and the solvation effect. The hydrophilic domain and its connectivity are analyzed. The numbers of surrounding water and hydronium ions as well as the location of the solvation shell are evaluated. The results demonstrate that the membrane degradation does not change the interaction with the hydronium ions and water molecules but alters the coordination number, leading to a larger hydrophilic domain and a poorer connectivity. The acid groups and protons are distributed more at the edge of the hydrophilic region to accelerate the proton motion, which is not conducive to the proton transport along the membrane side chain.

The explosive growth in citizen science combined with a recalcitrance on the part of mainstream science to fully embrace this data collection technique demands a rigorous examination of the factors influencing data quality and project efficacy. Patterns of contributor effort and task performance have been well reviewed in online projects; however, studies of hands-on citizen science are lacking. We used a single hands-on, out-of-doors project-the Coastal Observation and Seabird Survey Team (COASST)-to quantitatively explore the relationships among participant effort, task performance, and social connectedness as a function of the demographic characteristics and interests of participants, placing these results in the context of a meta-analysis of 54 citizen science projects. Although online projects were typified by high (>90%) rates of one-off participation and low retention (<10%) past 1 y, regular COASST participants were highly likely to continue past their first survey (86%), with 54% active 1 y later. Project-wide, task performance was high (88% correct species identifications over the 31,450 carcasses and 163 species found). However, there were distinct demographic differences. Age, birding expertise, and previous citizen science experience had the greatest impact on participant persistence and performance, albeit occasionally in opposite directions. Gender and sociality were relatively inconsequential, although highly gregarious social types, i.e., "nexus people," were extremely influential at recruiting others. Our findings suggest that hands-on citizen science can produce high-quality data especially if participants persist, and that understanding the demographic data of participation could be used to maximize data quality and breadth of participation across the larger societal landscape.

Passive daytime radiative cooling (PDRC) has drawn significant attention recently for electricity-free cooling. Porous polymers are attractive for PDRC since they have excellent performance and scalability. A fundamental question remaining is how PDRC performance depends on pore properties (e.g., radius, porosity), which is critical to guiding future structure designs. In this work, optical simulations are carried out to answer this question, and effects of pore size, porosity, and thickness are studied. We find that mixed nanopores (e.g., radii of 100 and 200 nm) have a much higher solar reflectance R̅solar (0.951) than the single-sized pores (0.811) at a thickness of 300 µm. With an Al substrate underneath, R̅solar, thermal emittance ε̅LWIR, and net cooling power Pcool reach 0.980, 0.984, and 72 W/m2, respectively, under a semihumid atmospheric condition. These simulation results provide a guide for designing high-performance porous coating for PDRC applications.

RESUMO

'Candidatus Liberibacter asiaticus' (CLas) is an unculturable phloem-restricted α-proteobacterium associated with huanglongbing (HLB). Here, we provide the genome sequence of CLas strain CoFLP1 from its insect vector Diaphorina citri (Hemiptera: Liviidae) collected in the department of La Guajira, Colombia. The CoFLP1 strain is composed of 1,231,639 bp with G+C 36.5% content. This study reports the first CLas genome sequence from Colombia, which will add to CLas genome resources and help to elucidate our understanding of the introduction pathway of HLB in South America.

Tuning fork gyroscopes (TFGs) are promising for potential high-precision applications. This work proposes and experimentally demonstrates a novel high-Q dual-mass tuning fork microelectromechanical system (MEMS) gyroscope utilizing three-dimensional (3D) packaging techniques. Except for two symmetrically decoupled proof masses (PM) with synchronization structures, a symmetrically decoupled lever structure is designed to force the antiparallel, antiphase drive mode motion and eliminate low frequency spurious modes. Thermoelastic damping (TED) and anchor loss are greatly reduced by the linearly coupled, momentum- and torque-balanced antiphase sense mode. Moreover, a novel 3D packaging technique is used to realize high Q-factors. A composite substrate encapsulation cap, fabricated by through-silicon-via (TSV) and glass-in-silicon (GIS) reflow processes, is anodically bonded to the wafer-scale sensing structures. A self-developed control circuit is adopted to realize loop control and characterize gyroscope performances. It is shown that a high-reliability electrical connection, together with a high air impermeability package, can be fulfilled with this 3D packaging technique. Furthermore, the Q-factors of the drive and sense modes reach up to 51,947 and 49,249, respectively. This TFG realizes a wide measurement range of ±1800 °/s and a high resolution of 0.1°/s with a scale factor nonlinearity of 720 ppm after automatic mode matching. In addition, long-term zero-rate output (ZRO) drift can be effectively suppressed by temperature compensation, inducing a small angle random walk (ARW) of 0.923°/√h and a low bias instability (BI) of 9.270°/h.

The GLABROUS1 enhancer-binding protein (GeBP) gene family encodes a typical transcription factor containing a noncanonical Leucine (Leu-)-zipper motif that plays an essential role in regulating plant growth and development, as well as responding to various stresses. However, limited information on the GeBP gene family is available in the case of the Gramineae crops. Here, 125 GeBP genes from nine Gramineae crops species were phylogenetically classified into four clades using bioinformatics analysis. Evolutionary analyses showed that whole genome duplication (WGD) and segmental duplication play important roles in the expansion of the GeBP gene family. The various gene structures and protein motifs revealed that the GeBP genes play diverse functions in plants. In addition, the expression profile analysis of the GeBP genes showed that 13 genes expressed in all tested organs and stages of development in rice, with especially high levels of expression in the leaf, palea, and lemma. Furthermore, the hormone- and metal-induced expression patterns showed that the expression levels of most genes were affected by various biotic stresses, implying that the GeBP genes had an important function in response to various biotic stresses. Furthermore, we confirmed that OsGeBP11 and OsGeBP12 were localized to the nucleus through transient expression in the rice protoplast, indicating that GeBPs function as transcription factors to regulate the expression of downstream genes. This study provides a comprehensive understanding of the origin and evolutionary history of the GeBP genes family in Gramineae, and will be helpful in a further functional characterization of the GeBP genes.

In catalysis science, the electronic structure of the active site determines the structure-activity relationship of the catalyst to a large extent. Therefore, modulating the electronic structure has become a main route for the rational design of metal-based catalyst materials. In this work, we prepared a LaCoSiHx material that has more electronegativity and a lower workfunction than traditional supported Co-based catalysts. Using CO2 methanation as a model catalytic reaction, the facile dissociation of CO2 and CO (a key reaction intermediate) on the surface of the LaCoSiHx catalyst is observed by various experimental methods (e.g., in situ Raman and FTIR) at room temperature. Moreover, theoretical calculation results further show that LaCoSiHx has a much stronger capacity for carbon-oxygen bond activation than the Co surface. The intrinsic mechanism is attributed to the marked electron transfer from catalysts into the antibonding orbital of CO2 and CO.

The removal of corpses (aka 'necrophoric behavior') is critical to sanitation in ant colonies. However, little is known about differences in the necrophoric responses of Solenopsis invicta workers towards corpses of nestmates and non-nestmates. We introduced corpses of S. invicta workers from either intracolony (i.e., nestmate) or intercolony (i.e., non-nestmate) origin at the entrance of artificial nests, and recorded workers' aggressive responses and necrophoric behaviors for analysis. Solenopsis invicta workers displayed distinct responses towards corpses of different origins. Specifically, resident workers were more likely to remove fresh non-nestmate corpses than nestmate corpses, but there was no difference regarding corpses that had been dead for 15 min or longer. Resident workers reacted more aggressively to, and removed more quickly, fresh non-nestmate corpses than corpses of their nestmates. On the other hand, there was no significant difference in the removal time between nestmate and non-nestmate corpses that had been dead for 15 min or longer. Resident workers always displayed stronger aggressiveness towards non-nestmate corpses than nestmate corpses, excepting to corpses that had been dead for 6 h, which elicited a response. No significant correlation between the removal times and aggressiveness levels were detected in any treatments. It remains to be tested whether this differential response is adaptive in how it influences colony fitness and competition.

A miniature piezoresistive pressure sensor fabricated by temporary bonding technology was reported in this paper. The sensing membrane was formed on the device layer of an SOI (Silicon-On-Insulator) wafer, which was bonded to borosilicate glass (Borofloat 33, BF33) wafer for supporting before releasing with Cu-Cu bonding after boron doping and electrode patterning. The handle layer was bonded to another BF33 wafer after thinning and etching. Finally, the substrate BF33 wafer was thinned by chemical mechanical polishing (CMP) to reduce the total device thickness. The copper temporary bonding layer was removed by acid solution after dicing to release the sensing membrane. The chip area of the fabricated pressure sensor was of 1600 µm × 650 µm × 104 µm, and the size of a sensing membrane was of 100 µm × 100 µm × 2 µm. A higher sensitivity of 36 µV/(V∙kPa) in the range of 0-180 kPa was obtained. By further reducing the width, the fabricated miniature pressure sensor could be easily mounted in a medical catheter for the blood pressure measurement.

To understand the chemical origin of platinum promotion effects on iron based Fischer-Tropsch synthesis catalysts, the effects of Pt on CO adsorption and dissociation as well as surface carbon hydrogenation on the Fe5C2(100) facet with different surface C* contents have been studied using the spin-polarized density functional theory method. CO dissociation initiating from diverse sites was calculated through both direct and H-assisted pathways via the CHO intermediate. On the perfect (100) surface, CO can hardly dissociate, and the surface carbon can be facially hydrogenated to CH4. On the C*-defect and C*-free (100) surfaces, CO can strongly adsorb on the C* vacant sites and direct dissociation is favored to occur. The activity is higher with the decrease of the surface carbon content. When platinum atoms are added on the surfaces, the C*-vacancies have a higher activity for CO dissociation than the new sites generated by Pt adsorption. However, both the CO dissociation and the surface carbon consumption through CH4 formation are hindered. The evolution of surface carbon is predicted to be suppressed by the addition of Pt on the Fe5C2(100) surface.

A novel three-dimensional (3D) hermetic packaging technique suitable for capacitive microelectromechanical systems (MEMS) sensors is studied. The composite substrate with through silicon via (TSV) is used as the encapsulation cap fabricated by a glass-in-silicon (GIS) reflow process. In particular, the low-resistivity silicon pillars embedded in the glass cap are designed to serve as the electrical feedthrough and the fixed capacitance plate at the same time to simplify the fabrication process and improve the reliability. The fabrication process and the properties of the encapsulation cap were studied systematically. The resistance of the silicon vertical feedthrough was measured to be as low as 263.5 mΩ, indicating a good electrical interconnection property. Furthermore, the surface root-mean-square (RMS) roughnesses of glass and silicon were measured to be 1.12 nm and 0.814 nm, respectively, which were small enough for the final wafer bonding process. Anodic bonding between the encapsulation cap and the silicon wafer with sensing structures was conducted in a vacuum to complete the hermetic encapsulation. The proposed packaging scheme was successfully applied to a capacitive gyroscope. The quality factor of the packaged gyroscope achieved above 220,000, which was at least one order of magnitude larger than that of the unpackaged. The validity of the proposed packaging scheme could be verified. Furthermore, the packaging failure was less than 1%, which demonstrated the feasibility and reliability of the technique for high-performance MEMS vacuum packaging.

Twenty-one volatile terpenes and terpenoids were found in Monomorium chinense Santschi (Hymenoptera: Formicidae), a native Chinese ant, by using headspace solid-phase microextraction (HS-SPME) coupled with gas-phase chromatography and mass spectrometry (GC-MS), which makes this ant one of the most prolific terpene producers in insect. A sesquiterpene with unknown structure (terpene 1) was the main terpene in workers and neocembrene in queens. Terpenes and terpenoids were detected in poison, Dufour's and mandibular glands of both workers and queens. Worker ants raised on a terpene-free diet showed the same terpene profile as ants collected in the field, indicating that de novo terpene and terpenoid synthesis occurs in M. chinense.

The fire ant Solenopsis invicta Buren is an important invasive pest. Among S. invicta workers behavioral changes depend on age where younger ants are nurses and older ants foragers. To identify potential genes associated with this division of labor, we compared gene expression between foragers and nurses by high-throughput sequencing. In total, we identified 1,618 genes significantly differently expressed between nurses and foragers, of which 542 were upregulated in foragers and 1,076 were upregulated in nurses. Several pathways related to metabolism were significantly enriched, such as lipid storage and fatty acid biosynthesis, which might contribute to the division of labor in S. invicta. Several genes involved in DNA methylation, transcription, and olfactory responses as well as resistance to stress were differentially expressed between nurses and foragers workers. Finally, a comparison between previously published microarray data and our RNA-seq data in S. invicta shows 116 genes overlap, and the GO term myofibril assembly (GO: 0030239) were simultaneously significantly enriched. These results advance knowledge of potentially important genes and molecular pathways associated with worker division of labor in S. invicta. We hope our dataset will provide . candidate target genes to disrupt organization in S. invicta as a control strategy against this invasive pest.

In social insects, social behavior may be changed in a way that preventing the spread of pathogens. We infected workers of the ant Solenopsis invicta with an entomopathogenic fungus Metarhizium anisopliae and then videotaped and/or measured worker feeding and trophallactic behavior. Results showed that fungal infected S. invicta enhanced their preference for bitter alkaloid chemical quinine on 3days after inoculation, which might be self-medication of S. invicta by ingesting more alkaloid substances in response to pathogenic infection. Furthermore, infected ants devoted more time to trophallactic behavior with their nestmates on 3days post inoculation, in return receiving more food. Increased interactions between exposed ants and their naive nestmates suggest the existence of social immunity in S. invicta. Overall, our study indicates that S. invicta may use behavioral defenses such as self-medication and social immunity in response to a M. anisopliae infection.
