RESUMO

The application of binary gas mixtures consisting of heptafluorobutyronitrile (C4F7N) and carbon dioxide (CO2) in AC GIS is currently attracting much attention. Therefore, the evaluation of the gas-solid interface charge distribution characteristics of epoxy resin is indispensable. Additionally, the phase-dependency of the charging behavior remains not fully understood. We simulated coaxial electrode structure in GIS and investigated the surface charge distribution on a down-scaled epoxy insulator. The influence of the truncated phase angle and duration of AC voltage on charge behavior were analyzed, and the charge transport mechanism under AC voltage was theoretically analyzed. The results showed that there was a noticeable negative charge speckle with the presence of the metal particle and the accumulated negative charge on the insulator surface far exceeded that of the positive charge. The maximum surface charge density and the amount of surface charge accumulated first increased and then decreased over time. It was found that the phase angle has a negligible influence on the surface charge distribution at the cut-off moment.

OBJECTIVES: To disclose the link between the composition of urolithiasis, especially that of uric acid calculi, and obesity, prediabetes, type 2 diabetes mellitus, and hypertension. MATERIALS AND METHODS: Patients who had urinary calculi and underwent surgical treatment were registered in the study. The composition of urinary calculi was analyzed and correlated with clinical features and biomedical profiles of the patients before surgical intervention. RESULTS: A total of 666 patients with urolithiasis who underwent surgical management were registered and analyzed. In those who had uric acid calculi, there was a significant association with prediabetic (OR: 20.11, 95% CI: 7.40-54.63, P < 0.001) and diabetic states (OR: 11.55, 95% CI: 4.41-29.97, P < 0.001). It also seemed that uric acid calculi were related to obesity but there was no statistical significance (OR: 2.45, 95% CI: 0.91-6.62, P = 0.078). There was no association of uric acid calculi with hypertension (OR: 1.08, 95% CI: 0.54-2.17, P = 0.822) and concurrent urinary tract infection (OR: 0.93, 95% CI: 0.44-1.96, P = 0.841). CONCLUSION: There was a remarkable association of uric acid calculi with prediabetic and diabetic states. The uric acid calculi were also seemingly associated with obesity in patients with urolithiasis undergoing surgical management.

Oxidative stress induces miR-200c, the predominant microRNA (miRNA) in lung tissues; however, the antioxidant role and biochemistry of such induction have not been clearly defined. Therefore, a lung adenocarcinoma cell line (A549) and a normal lung fibroblast (MRC-5) were used as models to determine the effects of miR-200c expression on lung antioxidant response. Hydrogen peroxide (H2O2) upregulated miR-200c, whose overexpression exacerbated the decrease in cell proliferation, retarded the progression of cells in the G2/M-phase, and increased oxidative stress upon H2O2 stimulation. The expression of three antioxidant proteins, superoxide dismutase (SOD)-2, haem oxygenase (HO)-1, and sirtuin (SIRT) 1, was reduced upon H2O2 stimulation in miR-200c-overexpressed A549 cells. This phenomenon of increased oxidative stress and antioxidant protein downregulation also occurs simultaneously in miR-200c overexpressed MRC-5 cells. Molecular analysis revealed that miR-200c inhibited the gene expression of HO-1 by directly targeting its 3'-untranslated region. The downregulation of SOD2 and SIRT1 by miR-200c was mediated through zinc finger E-box-binding homeobox 2 (ZEB2) and extracellular signal-regulated kinase 5 (ERK5) pathways, respectively, where knockdown of ZEB2 or ERK5 decreased the expression of SOD2 or SIRT1 in A549 cells. LNA anti-miR-200c transfection in A549 cells inhibited the endogenous miR-200c expression, resulting in increased expressions of antioxidant proteins, reduced oxidative stress and recovered cell proliferation upon H2O2 stimulation. These findings indicate that miR-200c fine-tuned the antioxidant response of the lung cells to oxidative stress through several pathways, and thus this study provides novel information concerning the role of miR-200c in modulating redox homeostasis of lung.
