RESUMO

Response surface methodology was employed to optimize the conditions for alkaline extraction of polysaccharides from Ganoderma lucidum. The results indicated that the optimum conditions were an extraction temperature of 60.1 degrees C, an extraction time of 77.3 min, a sodium hydroxide (NaOH) concentration of 5.1% and a substrate/liquid ratio of 1:21.4. Immunological assays results have shown that the alkaline soluble polysaccharides have no noticeable effects on monocyte phagocytosis and immune organ (spleen, thymus) weight of immunocompromised mice at the tested dosages. However, they could restore delayed type hypersensitivity reaction to dinitrofluorobenzene (DFNB), hemolysis antibody levels at the three doses applied, and improve the natural killer cell activity at the high-dose and medium dose.

Five polysaccharides (IOP1b, IOP2a, IOP2c, IOP3a and IOP4) were isolated and purified from Inonotus obliquus by DEAE-Sepharose fast flow and SepharoseCL-6B column chromatography. Their chemical and physical characteristics were determined and antioxidant activities were investigated on the basis of hydroxyl radical assay, superoxide radical assay and ferric-reducing antioxidant power assay. The results showed that five polysaccharides exhibited antioxidant activities, and the higher content of uronic acid and proteinous substances, the stronger antioxidant activities of polysaccharides. Besides, molecular weights of polysaccharides also influence their antioxidant activities. IOP3a and IOP4 showed higher antioxidant properties than IOP1b, IOP2a and IOP2c.

A water-soluble neutral polysaccharide (GLP-F1-1) was isolated from the fruiting bodies of Ganoderma lucidum by DEAE Sepharose Fast Flow and Sephacryl S-500 High Resolution Chromatography. The neutral polysaccharide had an average molecular weight (Mw) of approximately 2.5×10(6) kDa. GC analysis showed that this polysaccharide was mainly composed of glucose and galactose in the molar ratio of 34:1. 1H and 13C NMR spectroscopy in combination with GC-MS technique indicated that the new polysaccharide had a backbone chain of 1,4-disubstituted-ß-glucoseopyranose and 1,4,6-trisubstituted-ß-glucoseopyranosyl, while the branched chains were mainly composed of 1,6-disubstituted-ß-glucopyranosyl and 1,4-disubstituted-ß-galactoseopyranosyl residues.

In the present study, in order to maximize the yield of polysaccharides from Ganoderma lucidum, response surface methodology was employed to optimize the ultrasonic/microwave-assisted extraction (UMAE) conditions. The results indicated that the optimal extraction conditions were ultrasonic power of 50W, microwave power of 284W, extraction time of 701s and water/solid ratio of 11.6:1, respectively. Using UMAE, the yield of polysaccharides was 115.56% above that of classical hot water extraction (HWE) and increased by 27.7% as against ultrasound-assisted extraction (UAE), which confirmed the great potential application of UMAE technology in the extraction of polysaccharides. The immunological assays results demonstrated that polysaccharides of G. lucidum extracted by ultrasonic/microwave (UMP) could improve the weight of immune organ of immunocompromised mice, restore delayed-type hypersensitivity (DTH) reaction to DFNB, improve hemolysis antibody level and natural killer cell activity at high-dose. However, UMP had no noticeable effects on phagocytosis of monocyte at the tested dosage range.

Recently, the use of ultrasonic and microwave has attracted considerable interest as an alternative approach to the traditional extraction methods. In this paper, in order to maximize the yield and purity of polysaccharides from Inonotus obliquus, response surface methodology (RSM) was employed to optimize the ultrasonic/microwave assisted extraction (UMAE) conditions. The results indicated that the optimal conditions for UMAE were 90W microwave power, 50W ultrasonic power together with 40kHz ultrasonic frequency, solid/water ratio was 1:20 (W/V) and the extracting time was 19min, respectively. Under the optimal conditions, the yield and purity of polysaccharides were 3.25% and 73.16%, respectively, which are above that of traditional hot water extraction and close to the predicted value (3.07% and 72.54%, respectively). These results confirmed that ultrasonic/microwave assisted extraction (UMAE) of polysaccharides had great potential and efficiency compared with traditional hot water extraction. At the same time, the anti-tumor activities of the polysaccharides from I. obliquus with UMAE were evaluated. The results suggested that polysaccharides from I. obliquus exhibited obvious anti-tumor activities.
