RESUMO

A vertical confocal observation system capable of high-resolution observation of intracellular structure is demonstrated. The system consists of magnet-active microplates to rotate, incline, and translate single adherent cells in the applied magnetic field. Appended to conventional confocal microscopes, this system enables high-resolution cross-sectional imaging with single-molecule sensitivity in single scanning.

RESUMO

An approach for manipulating single adherent cells is developed that is integrated with an enzymatic batch release. This strategy uses an array of releasable microfabricated mobile substrates, termed microplates, formed from a biocompatible polymer, parylene. A parylene microplate array of 10-70 µm in diameter can be formed on an alginate hydrogel sacrificial layer by using a standard photolithographic process. The parylene surfaces are modified with fibronectin to enhance cell attachment, growth, and stretching. To load single cells onto these microplates, cells are initially placed in suspension at an optimized seeding density and are allowed to settle, stretch, and grow on individual microplates. The sacrificial layer underneath the microplate array can be dissolved on a time-scale of several seconds without cytotoxicity. This system allows the inspection of selected single adherent cells. The ability to assess single cells while maintaining their adhesive properties will broaden the examination of a variety of attributes, such as cell shape and cytoskeletal properties.

RESUMO

This paper describes a simple reusable device that hydrodynamically traps a large number of beads in an array. Guiding pillars allow us to release the trapped beads by simply reversing the flow direction. The trap and reset operations are extremely simple, robust and highly efficient. We analyzed the path of the beads in a microchannel with pillars to optimize the design of the device. We succeeded in arraying hundreds of 100 µm microbeads, subsequently released them in a few minutes, and demonstrated multiple experiments with a single device.

In this study, we have developed a meander-shaped dynamic microfluidic technology that allows us to pair two different types of microbeads in a trapping site. The dynamic microfluidic technology comprises implemented modifications of a conventional dynamic microarray design such as: (i) the combination of a meander-shaped by-pass channel and a trapping channel with a hydrodynamic trapping site and (ii) line-symmetrical formation of the by-pass and trapping channels. Using these modifications, we have successfully trapped different types of sample in one trapping site, and constructed an array of paired beads of different type such as polystyrene beads or hydrogel beads made of agarose, collagen or alginate. We found that this meander-shaped dynamic microfluidic technology is applicable for the observation of interactions between the paired beads such as molecular diffusion.

BACKGROUND: Familial hypobetalipoproteinemia (FHBL) and abetalipoproteinemia (ABL) are rare inherited forms of hypolipidemia. Their differential diagnosis is important for predicting of the prognosis and selecting appropriate therapy. MATERIALS AND METHODS: Genetic analysis was performed in two patients with primary hypocholesterolemia born from consanguineous parents. The oral fat tolerance test (OFTT) was performed in one patient with FHBL (apoB-87.77) and one with ABL as well as in four normal control subjects. After overnight fasting, blood samples were drawn. Serum lipoprotein and remnant-like particle (RLP) fractions were determined by HPLC analysis. RESULTS: Both patients with homozygous FHBL were asymptomatic probably because of preserved levels of fat-soluble vitamins, especially vitamin E. The patients with FHBL were homozygous because of novel apoB-83.52 and apoB-87.77 mutations, and although one of them (apoB-87.77) had fatty liver disease, microscopic findings suggesting nonalcoholic steatohepatitis were absent. Fasting apoB-48 and RLP-triglyceride levels in the patient with homozygous FHBL, which were similar to those in normal control subjects, increased after OFTT both in normal control subjects and the patient with FHBL but not in the patient with ABL, suggesting that the fat load administered was absorbed only in the patient with FHBL. CONCLUSION: Although lipid levels in the patients with homozygous FHBL and ABL were comparable, fasting, postoral fat loading of apoB-48, as well as RLP-triglyceride levels, may help in the differential diagnosis of FHBL and ABL and provide a prompt diagnosis using genetic analysis in the future.

This paper describes a precision-guided surgical navigation system for minimally invasive surgery. The system combines a laser guidance technique with a three-dimensional (3D) autostereoscopic image overlay technique. Images of surgical anatomic structures superimposed onto the patient are created by employing an animated imaging method called integral videography (IV), which can display geometrically accurate 3D autostereoscopic images and reproduce motion parallax without the need for special viewing or tracking devices. To improve the placement accuracy of surgical instruments, we integrated an image overlay system with a laser guidance system for alignment of the surgical instrument and better visualization of patient's internal structure. We fabricated a laser guidance device and mounted it on an IV image overlay device. Experimental evaluations showed that the system could guide a linear surgical instrument toward a target with an average error of 2.48 mm and standard deviation of 1.76 mm. Further improvement to the design of the laser guidance device and the patient-image registration procedure of the IV image overlay will make this system practical; its use would increase surgical accuracy and reduce invasiveness.
