RESUMO

Growth rate (GR), esterase activity (EA), membrane potential (MP), and DNA content were measured by flow cytometry to test if this powerful tool could be included in risk assessment and monitoring programs. This study tests a battery of endpoints that were measured on Scenedesmus (Acutodesmus) obliquus as model species, under high levels of injected CO2 to be proposed as biomarkers of effect. New technologies such as carbon capture and storage (CCS) bring positive consequences on CO2 mitigation strategies but also could have negative consequences if a CO2 leakage occurs during injection. Under this scenario, pH might drop down to 4 pH units around the pipe where the leakage happens. The study focus on to ascertain if the endpoints fulfill the requirements such as sensitiveness, be timely and cost-effective, be easy to measure and interpret, and be nondestructive, valuable attributes in effective biomarkers. CO2 injected at high levels significantly affects the EA, the membrane polarization, as well as GRs. However the DNA content did not shown a clear response under this condition. In conclusion, the GR, the EA, and the MP analyzed by flow cytometry fulfilled the attributes mentioned above and are proposed as biomarkers of effect in CCS monitoring programs. © 2020 International Society for Advancement of Cytometry.

It has been suggested that spinach methanolic extract (SME) inhibits the formation of advanced glycation end products (AGEs), which are increased during diabetes progression, so it is important to know if SME has beneficial effects in the diabetic retina. In this study, in vitro assays showed that SME inhibits glycation, carbonyl groups formation, and reduced-thiol groups depletion in bovine serum albumin incubated either reducing sugars or methylglyoxal. The SME effect in retinas of streptozotocin-induced diabetic rats (STZ) was also studied (n = 10) in the normoglycemic group, STZ, STZ rats treated with SME, and STZ rats treated with aminoguanidine (anti-AGEs reference group) during 12 weeks. The retina was sectioned and immunostained for Nε-carboxymethyl lysine (CML), receptor RAGE, NADPH-Nox4, inducible nitric oxide synthase (iNOS), 3-nitrotyrosine (NT), nuclear NF-κB, vascular endothelial growth factor (VEGF), glial fibrillary acidic protein (GFAP), S100B protein, and TUNEL assay. Lipid peroxidation was determined in the whole retina by malondialdehyde (MDA) levels. The results showed that in the diabetic retina, SME reduced the CML-RAGE co-localization, oxidative stress (NOX4, iNOS, NT, MDA), inflammation (NF-κB, VEGF, S100B, GFAP), and apoptosis (p < 0.05). Therefore, SME could attenuate the retinal degeneration by inhibition of CML-RAGE interaction.

This study aims at elucidating the environmental factors controlling benthic diatom diversity and uniqueness in Mediterranean mountain ponds. Samples of periphytic diatoms were collected in 45 ponds in Andalusia, south of Spain, and analysed by standard methods. Data analysis reveals that diatom diversity is mainly controlled by elevation and hydroperiod. Contrary to the usual findings in the literature, the highest scores on Shannon's diversity index were found in high-elevation temporary ponds, but this effect is hidden by lake clustering in the analysed dataset. Significant distance-decay similarity (DDS) trends were detected in the analysis of floristic composition among the samples, stressing the importance of spatial factors that may override the effect of other abiotic factors. These findings highlight the role of isolation and dispersal limitation in the configuration of the biogeographical patterns of benthic diatoms.

Severe corneal inflammation produces opacity or even perforation, scarring, and angiogenesis, resulting in blindness. In this study, we used the cornea to examine the effect of new anti-angiogenic chemopreventive agents. We researched the anti-angiogenic effect of two extracts, methanol (Met) and hexane (Hex), from the seed of Cucurbita argyrosperma, on inflamed corneas. The corneas of Wistar rats were alkali-injured and treated intragastrically for seven successive days. We evaluated: opacity score, corneal neovascularization (CNV) area, re-epithelialization percentage, and histological changes. Also, we assessed the inflammatory (cyclooxigenase-2, nuclear factor-kappaB, and interleukin-1ß) and angiogenic (vascular endothelial growth factor A, VEGF-A; -receptor 1, VEGFR1; and -receptor 2, VEGFR2) markers. Levels of Cox-2, Il-1ß, and Vegf-a mRNA were also determined. After treatment, we observed a reduction in corneal edema, with lower opacity scores and cell infiltration compared to untreated rats. Treatment also accelerated wound healing and decreased the CNV area. The staining of inflammatory and angiogenic factors was significantly decreased and related to a down-expression of Cox-2, Il-1ß, and Vegf. These results suggest that intake of C. argyrosperma seed has the potential to attenuate the angiogenesis secondary to inflammation in corneal chemical damage.

This observational, open, multicenter clinical trial with a single treatment group aimed to evaluate the effectiveness of a dietary supplement whose main ingredients are hydrolyzed gelatin, chondroitin sulfate, glucosamine sulfate, and devil's claw and bamboo extracts for pain reduction and improvement of functional capacities in patients with osteoarthritis (OA) of the knee and/or hip (REDART study). In all, 130 patients with OA recruited from Spanish hospitals received the dietary supplement for 6 months. The primary outcome was the patients' global assessment of pain in the affected joint as measured with a visual analogue scale (VAS). Other outcome measurements included the Lequesne Functional Index (subindexes for pain/discomfort, distance walked, and daily living) and the Western Ontario and McMaster Universities Arthritis Index (WOMAC; subindexes for pain, stiffness, and physical function). Scores were taken at months 3 and 6 of the treatment. Patients (N = 78) showed a reduction of pain of 3.77 ± 1.77 points after 6 months (p < .0001) in the VAS. The total reduction in the Lequesne Functional Index was 6.30 ± 4.08 points after 6 months (p < .0001), with significant reductions in all subindexes of the scale. A similar pattern was found for the WOMAC index, with an overall reduction of 22.49 ± 14.03 points after 6 months (p < .0001) and significant reductions in all subindexes. No major adverse events were noted during the treatment. This exploratory study shows that treatment with the dietary supplement significantly reduces pain and improves locomotor function in patients with OA of the knee and/or hip.

The carbon capture and storage (CCS) technologies that were proposed to mitigate environmental problems arising from anthropogenic CO2 emissions, also have potential environmental risks. An eventual CCS leak might induce very low pH values in the aquatic system. Due to the lack of knowledge of long-term CO2 exposures with very low pH values, this study aims to know the effects and consequences of such a situation for zooplankton, using the Daphnia magna experimental model. A CO2 injection system was used to provide the experimental condition. A twenty-one days experiment with control and low pH treatment (pH = 7) replicates was carried out under light and temperature-controlled conditions. Survival, individual growth, RNA:DNA ratio, and neonates production were analysed during the aforementioned period. No differences on survival (except last day), individual growth and RNA:DNA ratio were observed between both control and low pH treatments. However, clear differences were detected in neonates production and, consequently, in population growth rates and secondary production. The observed differences could be related with an energy allocation strategy to ensure individual survival but would have ecological consequences affecting higher trophic levels.

To test if capsaicin could improve the bioavailability of ciprofloxacin (1-cyclopropyl-6-fluoro-1,4-dihydro-4-oxo-7-(1-piperazinyl)-3-quinolinecarboxylic acid, CAS 85721-33-1, Bay q 3939) in rats, 0.01, 0.1, 0.5 and 1% capsaicin ((E)-N-[(4-hydroxy-3-methoxyphenyl)methyl]-8-methyl-6-nonenamide, trans-8-methyl-N-vanillyl-6-nonenamide, CAS 404-86-4) dissolved in ethanol and mixed with 20 mg/kg of ciprofloxacin were orally administered to groups of 10 rats each. Control groups were dosed with capsaicin-free, ethanol-containing or ethanol-free ciprofloxacin. Reference intravenous pharmacokinetics of ciprofloxacin was also established. The results revealed that capsaicin increased ciprofloxacin bioavailability by approximately 70% in groups receiving preparations containing capsaicin at a rate of 0.01, 0.1 and 0.5%. Higher concentrations failed to further increase bioavailability. However, capsaicin appears to have little or no impact on the rate of absorption or clearance of ciprofloxacin. Considering that 0.01% or 0.1% capsaicin are unlikely to upset the gastrointestinal tract, it may be worth attempting to study if a similar effect occurs in man, and to evaluate if the addition of capsaicin can be used as a method to increase the area under the curve/minimum inhibitory concentration rate, a key variable to improve clinical efficacy of ciprofloxacin.
