RESUMO

Male reproductive competition can select for condition-dependent, conspicuous traits that signal some aspect of fighting ability and facilitate assessment of potential rivals. However, the underlying mechanisms that link the signal to a male's current condition are difficult to investigate in wild populations, often requiring invasive experimental manipulation. Here, we use digital photographs and chest skin samples to investigate the mechanisms of a visual signal used in male competition in a wild primate, the red chest patch in geladas (Theropithecus gelada). We analysed photographs collected during natural (n = 144) and anaesthetized conditions (n = 38) to understand variability in male and female chest redness, and we used chest skin biopsies (n = 38) to explore sex differences in gene expression. Male and female geladas showed similar average redness, but males exhibited a wider within-individual range in redness under natural conditions. These sex differences were also reflected at the molecular level, with 10.5% of genes exhibiting significant sex differences in expression. Subadult males exhibited intermediate gene expression patterns between adult males and females, pointing to mechanisms underlying the development of the red chest patch. We found that genes more highly expressed in males were associated with blood vessel development and maintenance but not with androgen or oestrogen activity. Together, our results suggest male gelada redness variability is driven by increased blood vessel branching in the chest skin, providing a potential link between male chest redness and current condition as increased blood circulation to exposed skin could lead to heat loss in the cold, high-altitude environment of geladas.

We present a novel statovirus in geladas (Theropithecus gelada), graminivorous primates endemic to the Ethiopian highlands. Using a high-throughput sequencing approach, we identified contiguous sequences in feces from two adult female geladas in the Simien Mountains National Park, Ethiopia, that share similarities to statoviruses. Our phylogenetic analysis of the whole genome, as well as the RNA-dependent RNA polymerase (RdRp) and capsid protein (CP) amino acid sequences, revealed that the gelada statoviruses cluster with those from other primates (laboratory populations of Macaca nemestrina and Macaca mulatta). As the first report of statovirus in wild primates, this finding contributes to our understanding of the phylogenetic and geographic distribution of statoviruses and their hosts.

The objective of the study was to evaluate the effect of follicle ablation, exogenous FSH application, and different coasting time prior to ovum pick-up (OPU) on the number of follicles suitable for aspiration, oocyte quality, and cleavage rate in Ethiopian Boran cows. The experiment was carried out in three parts, (I) cows were synchronized using 500 µg PGF2α given 11 days apart. Cows were then subjected to a biweekly ovum pickup session before ovulation (n = 5) or starting day 7 after ovulation (n = 4) for 3 weeks. (II) Cows were synchronized, and all visible follicles were ablated on the first days of overt estrus, and cows were grouped into those that received a divided dose of 350 IU FSH (n = 5) or 175 IU FSH (n = 5) over 3 days. In both groups, OPU was carried out weekly starting 48 h after the last FSH for 6 weeks. (III) Protocol was similar to part II, but in group with 350 IU FSH (n = 5), coasting period was increased to 72 h. The covariates of follicles and oocyte were not affected (P > 0.05) by corpus luteum presence at OPU. The mean number of medium (7.36 ± 0.57) and large (8.28 ± 0.96) follicles were significantly higher (P < 0.05) in the group that received divided 350 IU FSH. Similarly, the mean number of grade-1 (4.19 ± 0.24) and grade-2 (4.32 ± .27) oocytes, maturation rate (70.41%), and cleavage rate (47.5%) were significantly higher (P < 0.05) in the group that received 350 IU FSH. COC quality was significantly (P < 0.05) influenced by coasting period. However, both maturation and cleavage rates were not affected by the coasting period. This study demonstrated that follicular ablation and treatment with FSH improves follicular population and oocyte recovery rate in Boran cows.

Equine reproduction is unique by having long behavioral estrus and differences in time of breeding between breeds and individuals of mares. An experimental study was conducted at the Balderas Sport Horses and Recreational Center, Addis Ababa, Ethiopia, from January to June, 2018, to evaluate conception rate to frozen semen in local and exotic crossbreed mares. Mares were teased to characterize estrus behavior and examined by ultrasound in determining imminent ovulation. Inseminations were done post ovulation within an average of 6-9 h using frozen-thawed semen. The overall conception rate to frozen semen was 15/21 (71.43%) with 8/11 (72.73%) in crossbreed and 7/10 (70%) in local breed mares. Age and body condition score (BCS) of animals had no significant effect on conception rate to AI with frozen semen. A slightly higher conception rate was obtained when ovulation was from the right ovary than when ovulated from the left ovary. A higher conception rate was obtained when the diameter of the preovulatory follicle was ≤ 45 mm than above diameter. The conception rate increased significantly with increased number of services/conception with an overall mean ± (SEM) of 2.2 ± 0.2 services/conception. A more number of services/conception were required for local breed (2.7 ± 0.2) than crossbreed mares (1.8 ± 0.3) and again for lower body condition scores than higher condition scores of mares. In conclusion, the increased number of services improved the conception rate with significant difference between breed of mares, whereas good management of mares for improved body conditions could be required to decrease the number of services per conception.

Morbidity and mortality of young stock is a challenge for livestock producers globally. In Ethiopia, where camels and small ruminants (sheep and goats) are essential smallholder and pastoral livestock, young stock losses can cause severe consequences to livelihoods. This pilot study, part of a Government-led Young Stock Mortality Reduction Consortium project, was undertaken to identify and evaluate interventions to reduce young stock mortality in mixed crop-livestock and pastoral production systems in Ethiopia. Pastoralists and mixed crop-livestock farmers were enrolled by convenience sampling across four regions. Households were sampled with questionnaire surveys to establish baseline mortality risk and prevalence of diarrhoea and respiratory disease in animals younger than one year, and followed longitudinally over a one-year period, with final evaluations conducted from March to July 2020. Mortality risk and prevalence of diarrhoea and respiratory disease before and after implementation were compared using Poisson regression models including household as random effect. Prior to intervention, median camel mortality, prevalence of diarrhoea, and respiratory disease across production systems in the different households was 0.4, 0.44 and 0.2, respectively. This compared to median pastoralist small ruminant mortality risk and prevalence of diarrhoea and respiratory disease of 0.45, 0.32 and 0.18, respectively. Post-intervention, median camel mortality, prevalence of diarrhoea and respiratory disease dropped to 0.1, 0.08 and 0. Similarly, more than half of the small ruminant households reported no mortality, and no cases of diarrhoea or respiratory disease. In camels, rate ratios of mortality risk, prevalence of diarrhoea, and respiratory disease post-intervention compared to the baseline were 0.41, 0.41 and 0.37. In small ruminants, rate ratios were 0.33, 0.35 and 0.46. All reductions were statistically significant (p < 0.01). Generally, pastoralists experienced higher mortality and disease prevalence compared to mixed crop-livestock smallholders, and the effect of intervention was slightly higher in pastoralist households. The pilot study findings demonstrated highly significant reductions in mortality and risk of diarrhoea and respiratory disease post-interventions. However, not all households benefitted from the interventions, with a few households reporting increased mortality and morbidity. Many households had very few animals which made it challenging to measure impact and the study was conducted over a single year, without a control group, so between year effects could not be accounted for in the reductions observed. These findings should contribute to improved livestock productivity in Ethiopia.

A serial ultrasonographic study was conducted on nine jennies aged 5-15 years from January to April 2008 with the objective of studying ovarian follicular dynamics and estrus manifestations under controlled management. Ovarian follicular activity was determined from the number and size distribution of follicles, length of interovulatory interval (IOI), growth rate of preovulatory follicles, diameter of follicles at the onset of estrus, and incidence of ovulation. Estrus manifestations were characterized using length of estrus and estrous cycle. The mean (± SD) number of follicle detected per ovary was 5.45 ± 2.3 (range, 1-16) with sizes ranging from 2.9 to 44 mm. The mean (± SD) size of follicle encountered at the onset of estrus was 25.9 ± 3.7 mm (range, 20.9-34.4) while that of the preovulatory follicles at -1 day before ovulation was 36.81 ± 3.78 mm. The mean (± SD) IOI, estrus, and estrous cycle length were 25.4 ± 33.6, 7.9 ± 32.9, and 24.2 ± 37.4 days, respectively. The mean (± SD) growth rate of the preovulatory follicle after the day of divergence was 1.9 ± 30.3 mm/day. Serum progesterone profile followed the same patterns of ovarian dynamics with maximum values being detected during midluteal phase. Serum progesterone assay revealed blood progesterone profiles of <1.0 ng/ml during estrus and up to 11 ng/ml during midluteal phase with a pattern following follicular dynamics. Body condition of the study jennies steadily increased and was positively correlated (r = 0.52, p < 0.001) with the diameter of the preovulatory follicle. In conclusion, the ultrasonic evaluation has revealed that follicular dynamics of jennies were generally related with body condition which might have been influenced by the type of management.

Accurate pregnancy diagnosis is an important criterion and management tool for successful dairying. Early identification of non-pregnant dairy heifers and cows after breeding can improve pregnancy rate and life time production. Determination of progesterone hormone levels is more accurate to diagnose failed pregnancies in dairy animals. This method is not always available in developing countries. Some of the kits available are developed for humans and might be used for cattle because in principle, progesterone is not species-specific and detection methods are the same in both animals and human beings. The study aimed at validating a human progesterone ELISA kit for use in cattle as a pregnancy diagnosis tool. Forty Boran and crossbred cattle (22 pregnant and 18 non-pregnant) were selected for the study. Ten milliliter of blood sample was collected from each animal using jugular venipuncture. Serum I and plasma was harvested within 2 hours after venipuncture and serum II after 12 hours, and all samples were analyzed for progesterone concentration using the ELISA procedure provided with the kit. The result showed that 88.9% (n = 16) of non-pregnant cows had progesterone concentration below 1 ng/ml with mean (±SE) of 0.48 ± 0.75 ng/ml while all pregnant cows had mean (±SE) concentration of 19.3 ± 0.68 ng/ml with individual values ranging from 5.2-38 ng/ml. Progesterone concentration between breeds and sample type did not show statistically significant difference for pregnant and non-pregnant cows. Nonetheless, the results of the experiments are very promising as far as pregnancy diagnosis is concerned in dairy cows from an economic perspective and accuracy; the experiments have to be performed on larger scale to proof repeatability and sensitivity.

Morbidity and mortality of young stock present economic and production challenges to livestock producers globally. In Ethiopia, calf morbidity and mortality rates, particularly due to diarrhea and respiratory disease, are high, limiting production, incomes, and the ability of farmers to improve their livelihoods. In this paper, we present findings from the combined experience of the Young Stock Mortality Reduction Consortium, which conducted epidemiological and intervention testing in calves across three production systems. This innovative alliance identified Cryptosporidium parvum and E. Coli K99 as the most common causes of diarrhea in pastoral and peri-urban calves; Strongyloides spp. as the most common fecal parasite in mixed crop-livestock and peri-urban calves; and bovine adenovirus, parainfluenza virus-3, and bovine respiratory syncytial virus as the most common respiratory pathogens in peri-urban calves. Furthermore, by improving producer knowledge with respect to fundamental livestock husbandry, feeding, housing, and neonatal care practices, calf mortality risk across production systems was reduced by 31.4 to 71.4% compared to baseline (between 10.5 and 32.1%), whereas risk of diarrhea was reduced by 52.6-75.3% (baseline between 11.4 and 30.4%) and risk of respiratory disease was reduced by 23.6-80.8% (baseline between 3.3 and 16.3%). These findings have informed scaling strategies and can potentially contribute to improved livestock productivity and human livelihoods in Ethiopia.

Primates have adapted to numerous environments and lifestyles but very few species are native to high elevations. Here we investigated high-altitude adaptations in the gelada (Theropithecus gelada), a monkey endemic to the Ethiopian Plateau. We examined genome-wide variation in conjunction with measurements of haematological and morphological traits. Our new gelada reference genome is highly intact and assembled at chromosome-length levels. Unexpectedly, we identified a chromosomal polymorphism in geladas that could potentially contribute to reproductive barriers between populations. Compared with baboons at low altitude, we found that high-altitude geladas exhibit significantly expanded chest circumferences, potentially allowing for greater lung surface area for increased oxygen diffusion. We identified gelada-specific amino acid substitutions in the alpha-chain subunit of adult haemoglobin but found that gelada haemoglobin does not exhibit markedly altered oxygenation properties compared with lowland primates. We also found that geladas at high altitude do not exhibit elevated blood haemoglobin concentrations, in contrast to the normal acclimatization response to hypoxia in lowland primates. The absence of altitude-related polycythaemia suggests that geladas are able to sustain adequate tissue-oxygen delivery despite environmental hypoxia. Finally, we identified numerous genes and genomic regions exhibiting accelerated rates of evolution, as well as gene families exhibiting expansions in the gelada lineage, potentially reflecting altitude-related selection. Our findings lend insight into putative mechanisms of high-altitude adaptation while suggesting promising avenues for functional hypoxia research.

Lamb and calf preweaning mortality and morbidity account for serious losses in sheep and cattle production and are, thus, a major factor in reducing profitability and adversely affecting the sheep and cattle farming. Thus, a prospective cohort study was conducted in Jamma district, Amhara Regional State, to determine the major cause of calf and lamb morbidity and mortality and associated risk factors. A semi-structured questionnaire and clinical assessment of the animals were conducted from 150 households to assess the potential risk factors. Besides, a total of 102 (81 fecal samples and 21 skin scrapings) were collected from 150 clinically ill and suspected animals to identify the cause of morbidity and mortality. The test of difference and correlation between variables were computed using chi-square and generalized linear model analysis. The total morbidity and mortality in calves were 33.3% and 2%, respectively, whereas for lamb, they were 27.3% and 32.5%, respectively. In calf, septicemia (100%) was a major cause of mortality, and diarrhea (54.6%) was the leading cause of calf morbidity followed by skin disease (30.1%), respiratory problems (12%), and septicemia (3.3%). Malnutrition was the most common problem in lambs causing up to 31.3% mortality followed by diarrhea 24% and respiratory problems 21.3%. The presence of a disease in adult cattle was significantly correlated to the presence of disease in calves (p < 0.001; r = 0.35). There was also a significantly higher positive correlation between sickness in adult sheep and lambs (p < 0.001, r = 0.45). Gastrointestinal parasitosis was identified in 82.7% of the samples collected from diarrheic and suspected calves (87.1% positive) and lambs (80% positive). Monezia species in lamb (33.3%) and Coccidia species in the calf (35.9%) had the largest morbidity rate. Ctenocephalides canis (16.7%) and Linognatus species (41.7%) were common ectoparasites identified in calves, while Melophagus ovinus was the only ectoparasite of lambs recovered (62.5%). In conclusion, the high morbidity found in calves and morbidity and mortality in lambs are known to seriously reduce the profitability of the smallholder cattle and sheep production in the area by affecting the availability of replacement animals and causing a detrimental effect on herd expansion and productivity. In further studies, establishing the specific causative agents, control of diseases in the adult, and improvement in feed resources should be the major areas that need to be considered to mitigate calf and lamb morbidity and mortality currently affecting the area.

: We captured 14 individual African wolves ( Canis lupaster) a total of 16 times in the Ethiopian Highlands in April 2015 and March 2016 by using rubber-lined foothold traps and immobilized them with dexmedetomidine-ketamine. Traps were baited with sheep meat and surveyed every 2 h. Capture efficiency (number of captures per number of visits) was 0.94, and capture rate (number of captures per number of trap nights) was 0.24. Trapped wolves were immobilized with 0.025 mg/kg dexmedetomidine and 8-10 mg/kg ketamine on the basis of respective estimated body mass. Mean (SD) induction times were 3.4 (0.5) min for subadults ( n=4), 3.5 (0.3) min for adult males ( n=4), and 4.7 (1.0) min for adult females ( n=6). Inductions were calm, muscle relaxation was good, and all animals were completely immobilized. Apart from increased rectal temperatures, no major negative side effects were observed. Atipamezole at 10 mg intramuscularly per milligram of dexmedetomidine administered was used for reversal at a mean of 43.5 (7.7) min after administration of dexmedetomidine-ketamine. Recoveries were relatively smooth, and animals were on feet, leaving the site within a mean of 13.6 (3.9) min, after atipamezole administration. Our results indicate that African wolves can be safely captured and immobilized by using rubber-lined foothold traps and dexmedetomidine and ketamine.

In asses, semen collection, cryopreservation, and artificial insemination (AI) with frozen-thawed semen have been scarcely described and success rate, particularly following AI, is reportedly low. In the absence of reliable protocols, assisted reproductive technologies cannot support the conservation efforts aimed at endangered wild ass species and domestic donkey breeds. Two experiments were conducted in this study. In experiment 1 we evaluated freezing Abyssinian donkey (N = 5, 4 ejaculates each) spermatozoa using three freezing extenders (Berliner Cryomedium + glycerol, BC+G; BotuCrio, BOTU; INRAFreeze, INRA) and two cryopreservation techniques (liquid nitrogen vapour, LNV; directional freezing, DF). Post-thaw evaluation indicated that BOTU and INRA were similar and both superior to BC+G (P ≤ 0.004 for all motility tests), and that DF was superior to LNV (P < 0.002 for all evaluation parameters). In experiment 2, relying on these results, we used Abyssinian donkey sperm frozen in BOTU and INRA by DF for AI (N = 20). Prior to AI, thawed samples were diluted in corresponding centrifugation media or autologous seminal fluids at 1:1 ratio. No difference was found between BOTU and INRA or between the addition of seminal fluids or media, all resulting in ~50% pregnancy, and no differences were noted between males (N = 4). The size of pre-ovulatory follicle was a significant (P = 0.001) predictor for AI success with 9/10 pregnancies occurring when follicular size ranged between 33.1-37.4 mm, no pregnancy when it was smaller, and only one when larger. A number of ass species face the risk of extinction. Knowledge gained in this study on the Abyssinian donkey can be customised and transferred to its closely related endangered species and breeds.

The naked mole-rat (Heterocephalus glaber) is a subterranean mouse-sized African mammal that shows astonishingly few age-related degenerative changes and seems to not be affected by cancer. These features make this wild rodent an excellent model to study the biology of healthy aging and longevity. Here we characterize for the first time the intestinal microbial ecosystem of the naked mole-rat in comparison to humans and other mammals, highlighting peculiarities related to the specific living environment, such as the enrichment in bacteria able to utilize soil sulfate as a terminal electron acceptor to sustain an anaerobic oxidative metabolism. Interestingly, some compositional gut microbiota peculiarities were also shared with human gut microbial ecosystems of centenarians and Hadza hunter-gatherers, considered as models of a healthy gut microbiome and of a homeostatic and highly adaptive gut microbiota-host relationship, respectively. In addition, we found an enrichment of short-chain fatty acids and carbohydrate degradation products in naked mole-rat compared to human samples. These data confirm the importance of the gut microbial ecosystem as an adaptive partner for the mammalian biology and health, independently of the host phylogeny.

BACKGROUND: A variety of microbial communities exist throughout the human and animal body. Genetics, environmental factors and long-term dietary habit contribute to shaping the composition of the gut microbiota. For this reason the study of the gut microbiota of a mammal exhibiting an extraordinary life span is of great importance. The naked mole-rat (Heterocephalus glaber) is a eusocial mammal known for its longevity and cancer resistance. METHODS: Here we analyzed its gut microbiota by cultivating the bacteria under aerobic and anaerobic conditions and identifying their species by mass spectrometry. RESULTS: Altogether, 29 species of microbes were identified, predominantly belonging to Firmicutes, and Bacteroidetes. The most frequent species were Bacillus megaterium (45.2 %), followed by Bacteroides thetaiotaomicron (19.4 %), Bacteroides ovatus, Staphylococcus sciuri and Paenibacillus spp., each with a frequency of 16.1 %. CONCLUSION: Overall, the gut of the naked mole-rat is colonized by diverse, but low numbers of cultivable microbes compared with humans and mice. The primary food plants of the rodents are rich in polyphenols and related compounds, possessing anti-microbial, anti-inflammatory, anti-oxidative as well as anti-cancer activity which may contribute to their exceptionally healthy life.

Manual and ultrasonic evaluation of the the reproductive organs of 62 dairy cows and heifers belonging to smallholder farms was conducted. The objective was to assess the efficiency of visual estrus detection method in the dairy animals that were presented for artificial insemination. Data were collected on reproductive status, body condition, estrus signs, and findings of rectal examination. Thirty-three animals were inseminated after ultrasonic evaluation while 29 animals were inseminated after manual evaluation through rectal palpation. Pregnancy diagnosis was performed using ultrasound 30 days post insemination. The most frequently observed estrus signs by the dairy owners were mucus discharge (83.9%) and mounting (67.7%) while the most frequently confirmed physical signs of estrus by rectal examination were cervical opening (88.7%), and uterine turgidity (82.3%). The overall mean (±SD) diameter of the largest follicle was 12.9 ± 3.4 mm with an average number of 3.5 follicles per ovary. The overall pregnancy rate was 29%. Evaluation of post-insemination records showed a significant difference (P < 0.05) in the diameter of largest follicle between the pregnant (14.7 mm) and non-pregnant (12.1 mm) animals. The mean (±SD) time interval between the first observed estrus sign to service were 10.5 ± 7.0 hrs and 14.5 ± 10.0 hrs for pregnant and non-pregnant, respectively. The low pregnancy rate, delayed time of insemination, and the difference in the size of dominant follicle indicate the incompatibility between visual estrus detection and optimal time of insemination. This confirms the significance of improving estrus detection and the need for combining estrus signs with pre-service evaluation of the reproductive organs either manually or using ultrasound.

BACKGROUND: In Ethiopia most childbirth occurs at home and is not assisted by skilled birth attendants. On the other hand having a birth attendant with midwifery skills during child birth is one of the most important interventions in reducing maternal morbidity and mortality. The objective of this study was to make an in-depth assessment of reasons why mothers do not use health facilities for child delivery. METHODS: Focus Group Discussions were used to gather information on use of health facilities for delivery in Butajira districts of South Central Ethiopia. The study was conducted from January to February 2012. Information was collected from four groups of women who had delivered in the past two years and four groups of men whose wives/partners have delivered in the same period. Data was coded and categorized using open code, qualitative data management software and analyzed based on thematic analysis. RESULTS: A total of eight FGD sessions, four with women and four with men groups were conducted involving 81 residents of the Butajira district. FGD participants answered that a large majority of women in the district gave birth at home. Two major themes, client related factors and facility/staff factors, emerged. Factors that emerged within major themes of client factors were decision making on place of delivery, reliance on Traditional Birth Attendants (TBAs), misconception about services provided at health facility, inability of family members to be present at time of labor and delivery, lack of privacy, traditional and/or spiritual factors, economic factors and accessibility to health care facilities. Within major themes of facility/staff factors subthemes that emerged were poor reception, refusal of admission, lack of privacy, information gap, poor competence and shortage of staff and materials at health facilities. CONCLUSION: Women in the study areas do not deliver in health facilities because of reasons that can be attributed to health care system and client related factors. These need to be addressed by considering the specific factors related to the health system and community perspectives.

BACKGROUND: The benefits of Health and Demographic Surveillance sites for local populations have been the topic of discussion as countries such as Ethiopia take efforts to achieve their Millennium Development Goal targets, on which they lag behind. Ethiopia's maternal mortality ratio is very high, and in the 2011 Ethiopia Demographic and Health Survey (2011 EDHS) it was estimated to be 676/100,000 live births. Recent Global Burden of Disease (GBD) and estimates based on the United Nations model reported better, but still unacceptably high, figures of 497/100,000 and 420/100,000 live births for 2013. In the 2011 EDHS, antenatal care (ANC) utilization was estimated at 34%, and delivery in health facilities was only 10%. OBJECTIVES: To compare maternal health service utilization among populations in a Health and Demographic Surveillance System (HDSS) to non-HDSS populations in Butajira district, south central Ethiopia. DESIGN: A community-based comparative cross-sectional study was conducted in January and February 2012 among women who had delivered in the 2 years before the survey. RESULTS: A total of 2,296 women were included in the study. One thousand eight hundred and sixty two (81.1%) had attended ANC at least once, and 37% of the women had attended ANC at least four times. A quarter of the women delivered their last child in a health facility. Of the women living outside the HDSS areas, 715 (75.3%) attended ANC at least once compared to 85.1% of women living in the HDSS areas [adjusted odds ratio (AOR) 0.59; 95% CI 0.46, 0.74]. Of the women living outside the HDSS areas, only 170 (17.9%) delivered in health facilities and were assisted by skilled attendants during delivery, whereas 30.0% of those living in HDSS areas delivered in health facilities (AOR 0.66; 95% CI 0.48, 0.91). CONCLUSION: This paper provides possible evidence that living in an HDSS site has a positive influence on maternal health. In addition, there may be a positive influence on those living nearby or in the same district where an HDSS is located even when not included in the surveillance system.
