RESUMO

Phosphorus (P)-hyperaccumulators for phytoextraction from P-polluted areas generally show rapid growth and accumulate large amounts of P without any toxicity symptom, which depends on a range of physiological processes and gene expression patterns that have never been explored. We investigated growth, leaf element concentrations, P fractions, photosynthetic traits, and leaf metabolome and transcriptome response in amphibious P-hyperaccumulators, Polygonum hydropiper and P. lapathifolium, to high-P exposure (5 mmol L-1), with 0.05 mmol L-1 as the control. Under high-P exposure, both species demonstrated good growth, allocating more P to metabolite P and inorganic P (Pi) accompanied by high potassium and calcium. The expression of a cluster of unigenes associated with photosynthesis was maintained or increased in P. lapathifolium, explaining the increase in net photosynthetic rate and the rapid growth under high-P exposure. Metabolites of trehalose metabolism, including trehalose 6-phosphate and trehalose, were sharply increased in both species by the high-P exposure, in line with the enhanced expression of associated unigenes, indicating that trehalose metabolic pathway was closely related to high-P tolerance. These findings elucidated the physiological and molecular responses involved in the photosynthesis and trehalose metabolism in P-hyperaccumulators to high-P exposure, and provides potential regulatory pathways to improve the P-phytoextraction capability.

In situ immobilization is a widely used measure for passivating Cd-contaminated soils. Amendments need to be continuously applied to achieve stable remediation effects. However, few studies have evaluated the impact of consecutive application of amendments on soil health and the microecological environment. A field experiment was conducted in a Cd-contaminated paddy (available Cd concentration 0.40 mg kg-1) on the Chengdu Plain to investigate the changes in soil Cd availability and response characteristics of soil bacterial communities after consecutive application of rice straw biochar (SW), fly ash (FM) and marble powder (YH) amendments from 2018 to 2020. Compared with control treatment without amendments (CK), soil pH increased by 0.6, 0.5 and 1.5 under SW, FM and YH amendments, respectively, and the soil available Cd concentration decreased by 10.71%, 21.42% and 25.00%, respectively. The Cd concentration in rice grain was less than 0.2 mg kg-1 under YH amendment, which was within the Chinese Contaminant Limit in Food of National Food Safety Standards (GB2762-2022) in the second and third years. The three amendments had different effects on the transformation of Cd fractions in soil, which may be relevant to the specific bacterial communities shaped under different treatments. The proportion of Fe-Mn oxide-bound fraction Cd (OX-Cd) increased by 11% under YH treatment, which may be due to the promotion of Fe(III) and Cd binding by some enriched iron-oxidizing bacteria, such as Lysobacter, uncultured_Pelobacter sp. and Sulfurifusis. Candidatus_Tenderia and Sideroxydans were enriched under SW and FM amendments, respectively, and were likely beneficial for reducing Cd availability in soil through Cd immobilization. These results revealed the significance of the bacterial community in soil Cd immobilization after consecutive application of amendments and highlighted the potential of applying YH amendment to ensure the safe production of rice in Cd-contaminated soil.

The root-associated microbiome assembly substantially promotes (hyper)accumulator plant growth and metal accumulation and is influenced by multiple factors, especially host species and environmental stress. Athyrium wardii (Hook.) is a phytostabilizer that grows in lead (Pb)-zinc (Zn) mine tailings and shows high root Pb accumulation. However, there remains little information on the assembly of the root-associated microbiome of A. wardii and its role in phytostabilization. A field study investigated the structural and functional variation in the root-associated bacterial microbiome of Athyrium wardii (Hook.) exposed to different levels of contamination in Pb-Zn mine tailings. The root compartment dominated the variation in the root-associated bacterial microbiome but the levels of contaminants showed less impact. Bacterial co-occurrence was enhanced in the rhizosphere soil and rhizoplane but tended to be much simpler in the endosphere in terms of network complexity and connectivity. This indicates that the microbial community assembly of A. wardii was non-random and shaped by root selective effects. Proteobacteria, Chloroflexi, Actinobacteria, Cyanobacteria, and Acidobacteriota were generally the dominant bacterial phyla. The genera Crossiella and Bradyrhizobium were enriched in the rhizosphere and cyanobacterial genera were enriched in the endosphere, demonstrating substantial advantages to plant survival and adaptation in the harsh mine environment. Functional categories involved in amino acid and carbohydrate metabolism were abundant in the rhizosphere soil, thus contributing to metal solubility and bioavailability in the rhizosphere. Membrane transporters, especially ATP-binding cassette transporters, were enriched in the endosphere, indicating a potential role in metal tolerance and transportation in A. wardii. The study shows substantial variation in the structure and function of microbiomes colonizing different compartments, with the rhizosphere and endophytic microbiota potentially involved in plant metal tolerance and accumulation during phytostabilization.

BACKGROUND: Studies are needed to assess risk factors pertinent to the incidence of secondary malignancies among childhood and adolescent lymphoma survivors. We aimed to identify risk factors pertinent to the incidence of secondary malignancies and subsequently establish a clinically practical predictive nomogram. METHODS: A total of 5561 patients who were diagnosed with primary lymphoma below the age of 20 years between 1975 and 2013 and survived for at least 5 years were identified. Standardized incidence ratio (SIR) and excess risk (ER) analysis were performed by sex, age, and year when primary lymphoma was diagnosed, sites and types of primary lymphoma, and therapy strategies. Univariable and multivariable logistic regression were used to identify independent risk factors for adolescent and childhood lymphoma-related secondary malignancies. Based on 5 factors (age, time from lymphoma diagnosis, gender, lymphoma type, and therapy), a nomogram for predicting the risk of a secondary malignancy for patients with childhood and adolescent primary lymphoma was established. RESULTS: Among 5561 lymphoma survivors, 424 developed a secondary malignancy. Females (SIR = 5.34, 95% CI, 4.73-5.99; ER = 50.58) exhibited a higher SIR and ER than males (SIR = 3.28, 95% CI, 2.76-3.87; ER = 15.53). Blacks were at a higher risk than Caucasians or others. Nodular lymphocyte-predominant Hodgkin lymphoma survivors exhibited typically high SIR (13.13, 95% CI, 6-24.92) and ER (54.79) among all lymphoma classifications. Lymphoma survivors who underwent radiotherapy, whether they received chemotherapy or not, had typically higher SIR and ER. Among all types of secondary malignancies, "bone and joint neoplasms" (SIR = 11.07, 95% CI, 5.52-19.81) and "soft tissue neoplasms" (SIR = 12.27, 95% CI, 7.59-18.76) presented significantly high SIR whereas "breast cancer" and "endocrine cancer" associated with higher ER. The median diagnosis age of secondary malignancies was 36 years old, and the median time interval between the diagnosis of two malignancies was 23 years. A nomogram was constructed to predict the risk of secondary malignancies in patients diagnosed with primary lymphoma before 20 years of age. After internal validation, the AUC and C-index of the nomogram are 0.804 and 0.804, respectively. CONCLUSION AND RELEVANCE: The established nomogram provides a convenient and reliable tool for predicting the risk of a secondary malignancy among childhood and adolescent lymphoma survivors, concluding significant concern for lymphoma survivors with high-risk estimates.

Four photo-catalysts of the general formula [Ir(CO6/ppy)2 (L)]Cl where CO6=coumarin 6 (Ir1-Ir3), ppy=2-phenylpyridine (Ir4), L=4'-(3,5-di-tert-butylphenyl)-2,2' : 6',2''-terpyridine (Ir1), 4'-(3,5-bis(trifluoromethyl)phenyl)-2,2' : 6',2''-terpyridine (Ir2 and Ir4), and 4-([2,2' : 6',2''-terpyridin]-4'-yl)-N,N-dimethylaniline (Ir3) were synthesized and characterized. These photostable photo-catalysts (Ir1-Ir3) showed strong visible light absorption between 400-550 nm. Upon light irradiation (465 and 525 nm), Ir1-Ir3 generated singlet oxygen and induced rapidly photo-catalytic oxidation of cellular coenzymes NAD(P)H. Ir1-Ir3 showed time-dependent cellular uptake with excellent intracellular retention efficiency. Upon green light irradiation (525 nm), Ir2 provided a much higher photo-index (PI=793) than the clinically used photosensitizer, 5-aminolevulinicacid (5-ALA, PI>30) against HeLa cancer cells. The observed necro-apoptotic anticancer activity of Ir2 was due to the Ir2 triggered photo-induced intracellular redox imbalance (by NAD(P)H oxidation and ROS generation) and change in the mitochondrial membrane potential. Remarkably, Ir2 showed in vivo photo-induced catalytic anticancer activity in mouse models.

Cell wall pectin is essential for cadmium (Cd) accumulation in rice roots and hydrogen peroxide (H2O2) plays an important role as a signaling molecule in cell wall modification. The role of H2O2 in Cd binding in cell wall pectin is unclear. D62B, a Cd-safe rice line, was found to show a greater Cd binding capacity in the root cell wall than a high Cd-accumulating rice line of Wujin4B. In this study, we further investigated the mechanism of the role of H2O2 in Cd binding in root cell wall pectin of D62B compared with Wujin4B. Cd treatment significantly increased the H2O2 concentration and pectin methyl esterase (PME) activity in the roots of D62B and Wujin4B by 22.45-42.44% and 12.15-15.07%, respectively. The H2O2 concentration and PME activity significantly decreased in the roots of both rice lines when H2O2 was scavenged by 4-hydroxy-Tempo. The PME activity of D62B was higher than that of Wujin4B. The concentrations of high and low methyl-esterified pectin in the roots of D62B significantly increased when exposed to Cd alone but significantly decreased when exposed to Cd and exogenous 4-hydroxy-Tempo. No significant difference was detected in Wujin4B. Exogenous 4-hydroxy-Tempo significantly decreased the Cd concentration in the cell wall pectin in both rice lines. The modification of H2O2 in Cd binding was further explored by adding H2O2. The maximum Cd adsorption amounts on the root cell walls of both rice lines were improved by exogenous H2O2·H2O2 treatment significantly influenced the relative peak area of the main functional groups (hydroxyl, carboxyl), and the groups intensely shifted after Cd adsorption in the root cell wall of D62B, while there was no significant difference in Wujin4B. In conclusion, Cd stress stimulated the production of H2O2, thus promoting pectin biosynthesis and demethylation and releasing relative functional groups involved in Cd binding on cell wall pectin, which is beneficial for Cd retention in the roots of Cd-safe rice line.

Nitric oxide (NO) is tightly associated with plant response against cadmium (Cd) stress in rice since NO impacts Cd accumulation via modulating cell wall components. In the present study, we investigated that whether and how NO regulates Cd accumulation in root in two rice lines with different Cd accumulation ability. The variation of polysaccharides in root cell wall (RCW) of a high Cd-accumulating rice line Lu527-8 and a normal rice line Lu527-4 in response to Cd stress when exogenous NO supplied by sodium nitroprusside (SNP, a NO donor) was studied. Appreciable amounts of Cd distributed in RCW, in which most Cd ions were bound to pectin for the two rice lines when exposed to Cd. Exogenous NO upregulated the expression of OsPME11 and OsPME12 that were involved in pectin demethylesterification, resulting in more low methyl-esterified pectin and therefore stronger pectin-Cd binding. Exogenous NO also enhanced the concentration of hemicellulose and the amount of Cd ions in it. These results demonstrate that NO-induced more Cd binding in RCW in the two rice lines through promoting pectin demethylesterification and increasing hemicellulose accumulation. Higher OsPMEs expression and more hemicellulose synthesis contributed to more Cd immobilization in RCW of the high Cd-accumulating rice line Lu527-8. The main findings of this study reveal the regulation of NO on cell wall polysaccharides modification under Cd stress and help to elucidate the physiological and molecular mechanism of NO participating in Cd responses of rice.

Exploring the mechanism of cadmium (Cd) accumulation in Cd-safe rice lines is beneficial for ensuring rice safety. D62B, a Cd-safe rice line, accumulates less than 0.2 mg Cd kg-1 in the brown rice due to strong capacity of Cd retention in the roots, and the root cell wall (RCW) polysaccharides play important roles. However, specific underlying mechanism of Cd binding on the polysaccharides is little known. In this study, the role of polysaccharides, especially pectin and hemicellulose 1 (HC1), in RCW of D62B was investigated by adsorption experiments and Fourier Transform Infrared Spectroscopy (FTIR) analysis compared with a common rice line (Luhui17). Cadmium was adsorbed on RCW of two rice lines by a multilayer and inhomogeneous chemisorption way with the force of ion transfer or exchange. Cadmium was adsorbed on RCW rapidly at first stage with the limit of internal and external diffusion, and gradually reached saturation. With the removal of pectin, the Cd adsorption rate, maximum Cd adsorption amount and the shift degree of carboxyl groups in the RCW of D62B sharply decreased, which showed advantages compared with Luhui17. Sequential removal of HC1 further decreased the maximum Cd adsorption amount and the shift degree of hydroxyl groups. The results showed that more available functional groups, especially carboxyl groups in pectin and hydroxyl groups in HC1, contributed to Cd immobilization within the RCW of Cd-safe rice line, thus limiting Cd translocation to the shoot and reducing Cd accumulation in the brown rice.

Photocatalytic anticancer profile of a IrIII photocatalyst (Ir3) with strong light absorption, high turnover frequency, and excellent biocompatibility is reported. Ir3 showed selective photo-cytotoxicity against cisplatin- and sorafenib-resistant cell lines while remaining dormant to normal cell lines in the dark. Ir3 exhibited excellent photo-catalytic oxidation of cellular co-enzyme, the reduced nicotinamide adenine dinucleotide phosphate (NADPH), and amino acids via a single electron transfer mechanism. The photo-induced intracellular redox imbalance and change in mitochondrial membrane potential resulted in necrosis and apoptosis of cancer cells. Importantly, Ir3 exhibited high biocompatibility and photo-catalytic anticancer efficiency as evident from in vivo zebrafish and mouse cancer models. To the best of our knowledge, Ir3 is the first IrIII based photocatalyst with such a high biocompatibility and photocatalytic anticancer therapeutic effect.

Cell wall (CW) plays an important role in Cd accumulation in roots of metal-tolerant plants, including rice. The role of CW polysaccharides, especially pectin, in binding Cd in roots of a high Cd accumulating (HA) rice line of Lu527-8 and a non-high Cd accumulating (NHA) rice line of Lu527-4 was investigated in this study. About 59%-63% of Cd in roots of the two rice lines was bound to CWs, indicating that CW was the main site for Cd accumulation in roots of the two rice lines. Cd adsorbed on the root CWs of the HA was 1.1-1.2 times more than that of the NHA, demonstrating the root CWs of the HA showed greater Cd binding ability. Cd exposure induced more Cd accumulation in pectin and hemicellulose in the HA. In particular, up to 65% of Cd accumulation in root CWs of the HA was observed in pectin. The removal of pectin lead to a 50% decrease for the amounts of Cd adsorption on root CWs of the HA, indicating that pectin was the major binding site for Cd in root CWs of the HA. The HA showed greater pectin methylesterase activities, resulting in lower degree of pectin methylesterification along with more low-methylesterified pectins in root CWs than the NHA. The more accumulation of low-methylesterified pectins in CWs induced by Cd contributed greatly to the high Cd accumulation in roots of the HA rice line of Lu527-8.

Using an ideal parental line to breed hybrid rice with low cadmium (Cd) accumulation in grain is an environmental-friendly approach to reduce the risk of Cd contamination in field. A grain low-Cd rice line YaHui2816 has stably low Cd in grain but strong Cd accumulation in straw, revealing specific pattern of its straw-grain Cd remobilization is beneficial to effectively breed hybrid rice for safe production as well as phytoremediation. In this study, a pot experiment was conducted to investigate Cd partitioning of YaHui2816 at different stages by comparison with a common rice C268A. The Cd from lower nodes and leaves was restricted in nodeⅡ, this Cd retention led to weak Cd transport from straw to ear in YaHui2816. Cd concentration in nodeⅡ of YaHui2816 was 1.56-fold and 7.36-fold higher than C268A at filling and mature stages. Thus, glutathione (GSH) and phytochelatin (PC) concentration, PC synthase (OsPCS1), GSH S-Transferase (OsGST) and Cd-remobilization associated genes were measured at filling stage. OsHMA2 and OsCCX2 were up-regulated in nodeⅡ of YaHui2816, relative expression of them were significantly higher than C268A. PCs participated in Cd remobilization process, remarkable PC increase in nodeⅡ of YaHui2816 was found in response to Cd treatment under regulation of OsPCS1 and OsGST of which PC2 was dominant form. Taken as a whole, the Cd retention in nodeⅡ of YaHui2816 acts as a 'firewall' to restrain Cd transport to grain. This work provides more insight to understand possible function of alleles for reducing Cd concentration in grain as well as strengthening Cd accumulation in straw of YaHui2816.

Physiological properties involved in cadmium (Cd) transport were investigated in the high Cd accumulating rice line (Lu527-8) in comparison with the normal rice line (Lu527-4) through a soil culture experiment. The results showed that Cd contents in xylem saps of Lu527-8 were 1.68-2.55 times higher than those of Lu527-4 under Cd stress. A high-positive correlation between Cd contents in xylem saps and Cd contents in shoots was observed. Lu527-8 owned a more rapid and effective transport of Cd to above-ground part. By analyzing the relationship between inorganic anions, organic components and Cd contents in xylem saps, the lower HPO42- and oxalate contents were considered to be related to the higher Cd transport in xylem sap of Lu527-8. As for citrate, tartaric and histidine content, significant increases were observed with the increasing Cd contents in xylem saps of two rice lines, and their contents of Lu527-8 were significantly higher than those of Lu527-4. Citrate, tartaric and histidine could take part in root-to-shoot Cd transport in xylem.

Breeding the rice cultivar with high cadmium (Cd) accumulation in straw but with low Cd in brown rice using marker-assisted selection (MAS) based on quantitative trait loci (QTL) is meaningful for phytoremediation as well as safety in production. A restorer rice line, YaHui2816, steadily showed low Cd translocation from straw to brown rice and carried alleles for reducing Cd concentration in brown rice (BRCdC). In this study, one F2 population (C268A/YaHui2816) was used to identify the QTLs for BRCdC in 2016, and other two different F2 populations (Lu98A/YaHui2816 and 5406A/YaHui2816) were used to furtherly validate the QTLs in 2017. Furthermore, a pot experiment was conducted to investigate the relative expression of predicted genes in the regions of these QTLs for BRCdC. Here 4 QTLs for BRCdC were identified, among which, 2 novel QTLs (qBRCdC-9 and qBRCdC-12) were identified on chromosomes 9 and 12 in rice. The YaHui2816 alleles in the QTLs qBRCdC-9 and qBRCdC-12 could effectively reduce BRCdC under different genetic backgrounds. Importantly, the QTL qBRCdC-12 was simultaneously associated with the Cd translocation from shoot to brown rice (T-s-b), genetically explaining that the low T-s-b of the YaHui2816 resulted in its low BRCdC. The interval length of the QTL qBRCdC-12 was only narrowed to 0.28 cM, making it possible to develop molecular markers and excavate genes for reducing BRCdC. It is worth noting that genes existed in these QTL regions have not been reported for regulating the Cd translocation in rice. 6 candidate genes (OS05G0198400, OS05G0178300, OS09G0544400, OS12G0161100, OS12G0162100 and OS12G0165200) up-regulated expressed in nodeⅡof the YaHui2816 in response to Cd treatment, and encoded ZRT/IRT-like protein (ZIP) 4, the protein similar to glutathione transferase (GSTs) 16, heat shock protein Hsp20 domain containing protein, MAP kinase-like protein and Cd tolerant protein 5, respectively.

A hydroponic experiment with two different cadmium (Cd) accumulating rice lines of Lu527-8 (the high Cd accumulating rice line) and Lu527-4 (the normal rice line) was carried out to explore the links among Cd stress, root exudates and Cd accumulation. The results showed that (1) Cd stress increased quantities of organic acids, but had no effect on composition in root exudates of the two rice lines. In Cd treatments, the contents of every detected organic acid in root exudates of Lu527-8 were 1.76-2.43 times higher than those of Lu527-4. Significant positive correlations between organic acids contents and Cd contents in plants were observed in both rice lines, except that malic acid was only highly relevant to Lu527-8, but not to Lu527-4. (2) Both composition and quantities of amino acids in root exudates changed a lot under Cd stress and this change differed in two rice lines. In control, four amino acids (glutamic acid, glycine, tyrosine and histidine) were detected in two rice lines. Under Cd stress, eight amino acids in Lu527-8 and seven amino acids in Lu527-4 could be detected, among which phenylalanine was only secreted by Lu527-8 and alanine, methionine and lysine were secreted by both rice lines. The contents of those four newly secreted amino acids from Lu527-8 increased significantly with the increase of Cd dose and each had a high-positive correlation with Cd contents, but the same change did not appear in Lu527-4. The difference between two rice lines in secretion of organic acids and amino acids may be related to their different Cd uptake properties.

Understanding plant phosphorus (P) assimilation and its root morphological responses is important to acquire an ideal material for remediation of P-enriched environments. Pot experiments were conducted to explore P accumulation and root morphological traits in a mining ecotype (ME) and non-mining ecotype (NME) of Polygonum hydropiper under different organic P (Po) sources (G1P, AMP, ATP, IHP) and inorganic P (Pi) source (KH2PO4), and also their responses to a high level of IHP for different growth periods. Both ecotypes showed higher biomass in Pi and IHP treatments than other Po sources. P accumulation in seedlings were in the order of Pi > IHP > other Po media. Extending the growth period increased biomass and P accumulation in both ecotypes. The ME demonstrated 1.11-1.46 times higher P accumulation than the NME. Seedlings fed with IHP demonstrated significantly greater morphological parameters of fine, medium, and thick roots compared to other Po sources. Total root length, surface area, and volume of both ecotypes significantly increased with the prolonged growth period. The ME has a higher ability to develop root system and exhibits better distribution of fine roots to enhance P accumulation from high P media, and thus it is a worthy material for P-phytoextraction.

Chelate-assisted phytoextraction with biodegradable chelants has been demonstrated as an efficient method to enhance heavy metal remediation efficiency by plants, while there is little available information on phytostabilization. A pot experiment was conducted to investigate the effects of biodegradable [S,S]-ethylenediaminedisuccinic acid (EDDS) and nitrilotriacetic acid (NTA) on plant growth and Pb accumulation of Pb phytostabilizer Athyrium wardii (Hook.) grown in Pb contaminated soils and to explore the feasibility of chelate-assisted phytostabilization. Greater adverse effects on plant biomass under high EDDS treatments were observed than NTA treatments. Significant increase of shoot Pb concentrations of A. wardii was noticed with increasing NTA and EDDS dosages, while EDDS induced higher shoot Pb concentrations than NTA. Moreover, root Pb concentrations of A. wardii under NTA treatments were 1.18-1.28-time higher than EDDS treatments, and a peak value of root Pb concentrations was observed at 2 mmol kg(-1) of NTA. Shoot Pb accumulations significantly increased with increasing dosages, and EDDS treatments caused a 1.44-1.6-time increase of shoot Pb accumulation than NTA. Root Pb accumulations under NTA treatments were 1.18-1.28-time higher than EDDS treatments. Maximum root Pb accumulation (155.5 mg plant(-1)) was found at 2 mmol kg(-1) of NTA on the 14th day. Higher BCF values and lower TF values were found under NTA treatments as compared to EDDS treatments. Available Pb concentrations in soil significantly increased on the 7th day with increasing NTA and EDDS dosages, then gradually decreased on the 14th day. Soil pH slightly decreased with increasing NTA and EDDS dosages. Therefore, chelate-assisted phytostabilization could be a feasible way to enhance the efficiency of Pb phytostabilization by A. wardii.

Introduction: Fingered citron slices possess significant nutritional value and economic advantages as herbal products that are experiencing increasing demand. The grading of fingered citron slices plays a crucial role in the marketing strategy to maximize profits. However, due to the limited adoption of standardization practices and the decentralized structure of producers and distributors, the grading process of fingered citron slices requires substantial manpower and lead to a reduction in profitability. In order to provide authoritative, rapid and accurate grading standards for the market of fingered citron slices, this paper proposes a grading detection model for fingered citron slices based on improved YOLOv8n. Methods: Firstly, we obtained the raw materials of fingered citron slices from a dealer of Sichuan fingered citron origin in Shimian County, Ya'an City, Sichuan Province, China. Subsequently, high-resolution fingered citron slices images were taken using an experimental bench, and the dataset for grading detection of fingered citron slices was formed after manual screening and labelling. Based on this dataset, we chose YOLOv8n as the base model, and then replaced the YOLOv8n backbone structure with the Fasternet main module to improve the computational efficiency in the feature extraction process. Then we redesigned the PAN-FPN structure used in the original model with BiFPN structure to make full use of the high-resolution features to extend the sensory field of the model while balancing the computation amount and model volume, and finally we get the improved target detection algorithm YOLOv8-FCS. Results: The findings from the experiments indicated that this approach surpassed the conventional RT-DETR, Faster R-CNN, SSD300 and YOLOv8n models in most evaluation indicators. The experimental results show that the grading accuracy of the YOLOv8-FCS model reaches 98.1%, and the model size is only 6.4 M, and the FPS is 130.3. Discussion: The results suggest that our model offers both rapid and precise grading for fingered citron slices, holding significant practical value for promoting the advancement of automated grading systems tailored to fingered citron slices.

Chemotherapy is currently a widely used treatment for cancer in clinical settings. Some chemotherapeutic drugs such as oxaliplatin (OXA) can cause tumor immunogenic cell death (ICD), activate immunity, and realize chemoimmunotherapy for tumors. However, the low degree of accumulation and immunosuppressive microenvironment in tumors limit the immunotherapeutic efficacy of these drugs. T cell immunoreceptor with Ig and ITIM domains (TIGIT)/poliovirus receptor (PVR) is an inhibitory immune checkpoint pathway involved in mediating natural killer (NK) cell and T cell exhaustion in tumors. TIGIT expression is up-regulated in NK cells and CD8+ T cells during tumor development. Moreover, we first found that tumors upregulated PVR expression after OXA treatment in previous work. Here, we systematically analyzed the effects of OXA on the TIGIT/PVR pathway, further proving the effectiveness of the combination of OXA and TIGIT/PVR blocking combination. We developed engineered TIGIT-expressing cell membrane nanovesicles loaded with OXA (OXA@TIGIT MVs) for synergistic cancer therapy. OXA@TIGIT showed good efficacy in several cancer models, leading to tumor regression, effectively inhibiting tumor growth and prolonging mouse survival. Furthermore, the OXA@TIGIT MVs activate a strong tumor-specific immune response in the body, providing long-term (more than 2 months) protection from tumor reactivation in the B16F10 melanoma rechallenge mouse model.

With only limited clinical patient benefit, focusing on new immune checkpoint pathways could be an important complement to current immune checkpoint drugs. In addition, not only does T cell-mediated adaptive immunity play an important role, but also macrophage-mediated innate immunity, due to its abundant presence in solid tumors. Here, we developed an engineered M1-like macrophage exosome, OX40L M1-exos. OX40L M1-exos can activate the adaptive immunity by activating the OX40/OX40L pathway and can reprogram M2-like tumor-associated macrophages into M1-like macrophages, thereby restoring and enhancing macrophage-mediated innate immunity. Our OX40L M1-exos achieved an effective synergistic effect of innate and adaptive immunity and achieved a potent therapeutic effect in a mouse breast cancer model, effectively inhibiting tumor growth and metastasis. These results suggest that OX40L M1-exos are an attractive therapeutic strategy and may be an important complement to current cancer immunotherapies.
