RESUMO

Although lithium-sulfur (Li-S) batteries with a high theoretical energy density and low cost have attracted extensive research attention, their commercialization is still unsuccessful due to the poor cycle life caused by the dissolution of polysulfides. It is the key challenge to overcome polysulfide shuttling for achieving long-term cycling stability in Li-S batteries. Here we report a novel strategy for the synthesis of N, S-doped graphene with high nitrogen and sulfur contents via in situ self-assembly of graphene oxide and thiourea-formaldehyde resin and calcination. The N, S-doped graphene serves as a conductive agent and a chemosorbent for suppressing polysulfide shuttling and preventing the Li-metal from corrosion, leading to a high reversible capacity and superior cycling stability. The Li-S batteries with the N, S-doped graphene can achieve an excellent cycling life (622 mA h g-1 after 500 cycles at 1C) and a slow capacity decay rate (0.049% per cycle over 500 cycles at 1C). The proposed strategy has the potential to enhance the high electrochemical properties of Li-S batteries.

RESUMO

PURPOSE: Proper taste-masking formulation design is a critical issue for instant-dissolving tablets (IDTs). The purpose of this study is to use the electronic tongue to design the additives of the 3D printed IDTs to improve palatability. METHODS: A binder jet 3D printer was used to prepare IDTs of levetiracetam. A texture analyzer and dissolution apparatus were used to predict the oral dispersion time and in vitro drug release of IDTs, respectively. The palatability of different formulations was investigated using the ASTREE electronic tongue in combination with the design of experiment and a model for masking bitter taste. Human gustatory sensation tests were conducted to further evaluate the credibility of the results. RESULTS: The 3D printed tablets exhibited rapid dispersion (<30 s) and drug release (2.5 min > 90%). The electronic tongue had an excellent ability of taste discrimination, and levetiracetam had a good linear sensing performance based on a partial least square regression analysis. The principal component analysis was used to analyze the signal intensities of different formulations and showed that 2% sucralose and 0.5% spearmint flavoring masked the bitterness well and resembled the taste of corresponding placebo. The results of human gustatory sensation test were consistent with the trend of the electronic tongue evaluation. CONCLUSIONS: Owing to its objectivity and reproducibility, this technique is suitable for the design and evaluation of palatability in 3D printed IDT development.

RESUMO

α-Conotoxin GeXIVA[1,2] is a highly potent and selective antagonist of the α9α10 nicotinic acetylcholine receptor (nAChR) subtype. It has the advantages of strong efficacy, no tolerance, and no effect on motor function, which has been expected help patients with neuropathic pain. However, drug development for clinical use is severely limited owing to its instability. Lyophilization is applied as the most preferred method to solve this problem. The prepared lyophilized powder is characterized by differential scanning calorimetry (DSC), powder X-ray diffractometry (PXRD), and Fourier transform infrared spectroscopy (FTIR). Molecular simulation is also used to explore the internal distribution and forces formed in the system. The analgesic effect on paclitaxel-induced neuropathic pain following single and 14-day repeated administrations are evaluated by the von Frey test and the tail-flick test. Trehalose combined with mannitol in a ratio of 1:1 is employed as the excipients in the determined formulation, where trehalose acts as the stabilizer and mannitol acts as the bulking agent, according to the results of DSC, PXRD, and FTIR. Both GeXIVA[1,2] (API) and GeXIVA[1,2] lyophilized powder (formulation) could produce stable analgesic effect. These results indicated that GeXIVA[1,2] lyophilized powder could improve the stability and provide an effective strategy to push it into clinical use as a new analgesic drug.

RESUMO

OBJECTIVE: To evaluate the effects of Xihuang Pills (XHP) and its main components on PI3K, AKT and mTOR signaling pathways and cell apoptosis of castration-resistant human PCa PC-3 cell subcutaneously transplanted tumors in nude mice. METHODS: We assigned 36 PC-3 tumor-bearing model mice to six groups of equal numbers to be treated with XHP, musk, calculus bovis (CB), musk + CB and docetaxel, respectively. After 14 days of intervention, we calculated the tumor-inhibition rate in different groups, observed the morphology of the tumor cells by HE staining, determined the levels of PI3K, Akt and mTOR mRNA by RT-qPCR, and determined the expressions of PI3K, Akt and mTOR signaling pathways and caspase-3 and caspase-9 proteins by Western blot. RESULTS: After 14 days of medication, the tumor-inhibition rates in the XHP, musk, CB, musk + CB and docetaxel groups were 29.67%, 5.52%, 7.26%, 12.88% and 6.26%, respectively. HE staining showed the formation of apoptotic bodies in the tumor tissues after intervention, especially in the XHP and musk + CB groups. The mRNA and phosphorylated protein expressions of PI3K, Akt and mTOR were significantly down-regulated (P < 0.01), and so were the expressions of caspase-3 and caspase-9 proteins in the XHP and musk + CB groups in comparison with the control (P < 0.01). CONCLUSIONS: Xihuang Pills, musk and calculus bovis can inhibit the growth of castration-resistant human PCa PC-3 cell subcutaneously transplanted tumors, which is associated with their effects of suppressing the abnormally activated PI3K, Akt and mTOR signaling pathways and promoting the apoptosis of PCa PC3 cells.

RESUMO

Supercapacitors (SCs) have received widespread attention as excellent energy storage devices, and the design of multicomponent electrode materials and the construction of ingenious structures are the keys to enhancing the performance of SCs. In this paper, MoS2 nanorods were used as the carrier structure to induce the anchoring of CoAl-LDH nanosheets and grow on the surface of nickel foam (NF) in situ, thus obtaining a uniformly distributed MoS2 nanorod@CoAl-LDH nanosheet core-shell nanoarray material (MoS2@CoAl-LDH/NF). On the one hand, the nanorod-structured MoS2 as the core provides high conductivity and support, accelerates electron transfer, and avoids agglomeration of CoAl-LDH nanosheets. On the other hand, CoAl-LDH nanosheet arrays have abundant interfacially active sites, which accelerate the electrolyte penetration and enhance the electrochemical activity. The synergistic effect of the two components and the unique core-shell nanostructure give MoS2@CoAl-LDH/NF a high capacity (14,888.8 mF cm-2 at 2 mA cm-2) and long-term cycling performance (104.4% retention after 5000 charge/discharge cycles). The integrated MoS2@CoAl-LDH/NF//AC device boasts a voltage range spanning from 0 to 1.5 V, achieving a peak energy density of 0.19 mW h cm-2 at 1.5 mW cm-2. Impressively, it maintains a capacitance retention rate of 84.6% after enduring 10,000 cycles, demonstrating exceptional durability and stability.

RESUMO

Cincticostellajianchuan sp. nov. from Dali Bai Autonomous Prefecture, Yunnan Province, China, is described based on chorionic structure, nymph, and winged stages. The new species is closely related to C.fusca (Kang & Yang, 1995), but it can be distinguished in the male imago stage by its mesonotum and penes morphology, coloration, and the forking point of the stem of MA+Rs on the forewing; in the nymph stage, it can be distinguished by the length of the posterolateral projections of abdominal segment IX and the setation of the abdominal terga. Compared to other congeners, nymphs and male imagoes of the new species and C.fusca share several morphological characteristics, such as a larger body, mesothorax with medially notched anterolateral projections, forefemur without a subapical band of transverse spines of the nymphs, the area between C, Sc and R1 of the forewings distinctly pigmented, and an apical sclerite on the ventral face of the penes of the male imagoes, supporting the proposition of a new species complex, the jianchuan complex. The systematics of Cincticostella and related genera are discussed briefly.

RESUMO

This study explores the impact of anthropogenic land use changes on the macroinvertebrate community structure in the streams of the Cangshan Mountains. Through field collections of macroinvertebrates, measurement of water environments, and delineation of riparian zone land use in eight streams, we analyzed the relationship between land use types, stream water environments, and macroinvertebrate diversities. The results demonstrate urban land use type and water temperature are the key environmental factors driving the differences in macroinvertebrate communities up-, mid-, and downstream. The disturbed streams had lower aquatic biodiversity than those in their natural state, showing a decrease in disturbance-sensitive aquatic insect taxa and a more similar community structure. In the natural woodland area, species distributions may be constrained by watershed segmentation and present more complex community characteristics.

RESUMO

Siphlonurusdongxi Li & Tong, sp. nov. from Shangri-La City, Yunnan Province, China, is described based on egg, nymph, and winged stages. The new species is closely related to S.davidi (Navás, 1932), and can be distinguished by the colour of the imago, the forking point of MP, the penis, posterolateral spines of tergum IX of imagoes, and first abdominal terga nymph, as well as the structure of the egg. The new species and S.davidi have the same morphological and structural characteristics, such as the long cubital area with many intercalaries, cross veins between C, Sc, RA, and RSa1 surrounded with distinct pigments, the strong curvature of vein CuP in the forewing, the broad expansion of the hindwing, the membranous penis lobes fused without teeth, supporting the proposition of a new species complex, the Siphlonurusdavidi group. The structures of the penis and the egg of the new species could help understand the origin and evolution of the genus Siphlonurus.

RESUMO

Transition metal selenides (TMSs) are viewed as a prospective high-capacity electrode material for asymmetric supercapacitors (ASCs). However, the inability to expose sufficient active sites due to the limitation of the area involved in the electrochemical reaction severely limits their inherent supercapacitive properties. Herein, a self-sacrificing template strategy is developed to prepare self-supported CuCoSe (CuCoSe@rGO-NF) nanosheet arrays by in situ construction of copper-cobalt bimetallic organic framework (CuCo-MOF) on rGO-modified nickel foam (rGO-NF) and rational design of Se2- exchange process. Nanosheet arrays with high specific surface area are considered to be ideal platforms for accelerating electrolyte penetration and exposing rich electrochemical active sites. As a result, the CuCoSe@rGO-NF electrode delivers a high specific capacitance of 1521.6 F/g at 1 A/g, good rate performance and an excellent capacitance retention of 99.5% after 6000 cycles. The assembled ASC device has a high energy density of 19.8 Wh kg-1 at 750 W kg-1 and an ideal capacitance retention of 86.2% after 6000 cycles. This proposed strategy offers a viable strategy for designing and constructing electrode materials with superior energy storage performance.

RESUMO

Primary malignant melanoma of the parotid gland (PGMM) is extremely rare, with a poor prognosis. Surgery is the main treatment option followed by adjuvant treatments such as radiotherapy, but which adjuvant treatment to be optimal is still controversial. In this case, a 63-year-old male PGMM patient was first misdiagnosed as a "myoepithelial tumor" and then treated with surgery, postoperative immunotherapy (sintilimab), chemotherapy, and radiotherapy successfully. The progression free survival was more than 19 months without signs of metastasis or recurrence to date. To our best knowledge, this is the first report of postoperative immunotherapy combined with chemotherapy and radiotherapy for PGMM. Our case indicated that combination therapy including surgery, adjuvant immunotherapy (sintilimab) combined with chemotherapy and radiotherapy may be a potential treatment option for PGMM, which needs further research.

RESUMO

OBJECTIVE: This study aimed to construct an optimal model to predict radiation pneumonia (RP) after radiotherapy for esophageal cancer using unified fractional dosiomics and to investigate the improvements in the prediction efficiency of each model for RP. METHODS: The clinical data, DVH, pre-treatment CT, and dose distribution of 182 patients were retrospectively analyzed. The independent risk factors were screened using univariate and multivariate logistic regression. The mutual information (MI),least absolute shrinkage and selection operator (LASSO), and recursive feature elimination (RFE) methods were used to screen the omics features. The AUC values of ROC, calibration curves, and clinical decision curves were calculated to evaluate the efficacy and trends of each model. RESULTS: The AUC of dosiomics model were 0.783 and 0.760 in the training and test cohorts, higher than 0.585 and 0.579 in the training and test cohorts of the DVH model. The AUC value of the R + D combination was the highest, reaching 0.833. The combined R + D model had a better calibration degree than the other models (mean absolute error = 0.018) and better net benefit in clinical decision-making. CONCLUSIONS: The radiomics combined dosiomics model was the best combined model to predict RP after radiotherapy for esophageal cancer. The dosiomics model could cover the efficiency of the DVH model and significantly improve the efficiency of the combined model. In the future, we will include other centers for further verification. ADVANCES IN KNOWLEDGE: For the first time, this study used CT images combined dose distribution to predict the occurrence of radiation pneumonitis after radiotherapy for esophageal cancer.

RESUMO

OBJECTIVE: We clarified the pathogenic influence of the absence of Streptococcus suis type 2 capsular saliva acid on BLAB/c mice. METHODS: The virulence of the experimental strains were compared; the distribution of strains in vivo was determined by quantitative plating. Histopathological analysis was used to qualitatively compare the different pathogenicity of wild strain and knockout strains. ELISA was used to test the levels of cytokine in whole blood cells for the stimulation of strains. RESULTS: The virulence of mutant strains was significantly reduced, and when the genes were restored, toxicity levels were recovered to that of the wild type strain. The distribution in blood and in the brain between wild strain and knock out strains has significant difference, and Streptococcus suis type 2 strains can cause different degrees of brain damage. During the in vitro test, the mutant strains can stimulate the whole blood cells to secrete higher levels of MCP-1 and IL-6. CONCLUSION: Capsular saliva acid affects bacterial virulence and host cell inflammation response. As an important virulence factor of Streptococcus suis type 2, it can damage the blood brain barrier and cause meningitis.

RESUMO

Notacanthellajinwu Li & Jacobus, sp. nov. is described based on egg, nymph, and winged stages from Dali Bai Autonomous Prefecture, Yunnan Province, China. The nymph of the new species is closely related to N.commodema (Allen, 1971), whose nymphs share a similar tuberculation of head, pronotum, and mesonotum. However, the nymph of our new species can be distinguished based on the structures of male sternum IX and abdominal tergal tubercles. In addition, the new species is distributed in subtropical high-altitude areas. The description of the male imago of the new species is the first certain one for the genus Notacanthella. Data associated with our new species allow for expanded discussion and diagnosis of Notacanthella and closely related genera. An identification key for nymphs of these groups is provided.

RESUMO

Lung adenocarcinoma (LUAD) is the most frequent subtype of lung cancer globally. However, the survival rate of lung adenocarcinoma patients remains low. Immune checkpoints and long noncoding RNAs are emerging as vital tools for predicting the immunotherapeutic response and outcomes of patients with lung adenocarcinoma. It is critical to identify lncRNAs associated with immune checkpoints in lung adenocarcinoma patients. In this study, immune checkpoint-related lncRNAs (IClncRNAs) were analysed and identified by coexpression. Based on the immune checkpoint-related lncRNAs, we divided patients with lung adenocarcinoma into two clusters and constructed a risk model. Kaplan-Meier analysis, Gene Set Enrichment Analysis, and nomogram analysis of the 2 clusters and the risk model were performed. Finally, the potential immunotherapeutic prediction value of this model was discussed. The risk model consisting of 6 immune checkpoint-related lncRNAs was an independent predictor of survival. Through regrouping the patients with this model, we can distinguish between them more effectively in terms of their immunotherapeutic response, tumour microenvironment, and chemotherapy response. This risk model based on immune checkpoint-based lncRNAs may have an excellent clinical value for predicting the immunotherapeutic response and outcomes of patients with LUAD.

RESUMO

BACKGROUND: Non-small cell lung cancer is the most common subtype of lung cancer in the world. However, the survival rate of non-small cell lung cancer patients remains low currently. Immune checkpoint and long non-coding RNAs are emerging as critical roles in prognostic significance and the immunotherapeutic response of non-small cell lung cancer. It is critical to discern LncRNAs related with immune checkpoints in patients with Non-small cell lung cancer. METHODS: In this study, immune checkpoint-linked LncRNAs were determined and achieved by the co-expression analysis. Immune checkpoint-linked LncRNAs with noteworthy prognostic value (P < 0.05) gained were next utilized to separate into two cluster by non-negative matrix factorization (NMF). Univariate and a least absolute shrinkage and selection operator were applied to construct an immune checkpoint-linked LncRNAs model. Kaplan-Meier analysis, Gene Set Enrichment Analysis, and the nomogram were utilized to investigate the LncRNAs model. Lastly, the capability immunotherapy and chemotherapy prediction value of this risk model were also estimated. RESULTS: The model consisting of ten immune checkpoint-related LncRNAs was acknowledged to be a self-determining predictor of prognosis. Through regrouping the NSCLC patients by this model, difference between them more efficiently on immunotherapeutic response, tumor microenvironment and chemotherapy response could be discovered. This risk model related to the immune checkpoint-based LncRNAs may have an excellent clinical prediction for prognosis and the immunotherapeutic response in patients with NSCLC. CONCLUSIONS: We performed an integrative analysis of LncRNAs linked with immune checkpoints and emphasized the significance of NSCLC subtypes classification, immune checkpoints related LncRNAs in estimating the tumor microenvironment score, immune cell infiltration of the tumor, immunotherapy, and chemotherapy response.

RESUMO

A highly active organocatalyst has been shown to affect the asymmetric reductive amination of ketones producing both aromatic and aliphatic amines. At 1 mol% catalyst loading, a series of structurally diverse chiral amines were quickly and economically prepared with good enantioselectivity and generally useful yield. The efficient synthesis of the calcimimetic (+)-NPS R-568 (67%, 89% ee) demonstrated the synthetic applicability of this methodology.

RESUMO

A new species with primitive characteristics, Ameletus daliensis Tong, sp. nov., is described, based on the morphology of imago, larva and egg with molecular data of the mitochondrial COI from Mount Cangshan, Dali, China. The new species is closely related to one of the most primitive mayflies, Ameletus primitivus Traver, 1939, by sharing persistent mouthparts in the alate stage, but it can be distinguished from the latter by the morphological differences of the mouthpart remains, wings and genitals in the imaginal stage. Both morphological and molecular evidence support that A. daliensis Tong, sp. nov. is a new member of the genus Ameletus. The discovery of the new species could help understand the origin and evolution of the genus Ameletus.
