RESUMO

Objective@#: Kinesin family member C1 (KIFC1), a non-essential kinesin-like motor protein, has been found to serve a crucial role in supernumerary centrosome clustering and the progression of several human cancer types. However, the role of KIFC1 in glioma has been rarely reported. Thus, the present study aimed to investigate the role of KIFC1 in glioma progression. @*Methods@#: Online bioinformatics analysis was performed to determine the association between KIFC1 expression and clinical outcomes in glioma. Immunohistochemical staining was conducted to analyze the expression levels of KIFC1 in glioma and normal brain tissues. Furthermore, KIFC1 expression was knocked in the glioma cell lines, U251 and U87MG, and the functional roles of KIFC1 in cell proliferation, invasion and migration were analyzed using cell multiplication, wound healing and Transwell invasion assays, respectively. The autophagic flux and expression levels matrix metalloproteinase-2 (MMP2) were also determined using imaging flow cytometry, western blotting and a gelation zymography assay. @*Results@#: The results revealed that KIFC1 expression levels were significantly upregulated in glioma tissues compared with normal brain tissues, and the expression levels were positively associated with tumor grade. Patients with glioma with low KIFC1 expression levels had a more favorable prognosis compared with patients with high KIFC1 expression levels. In vitro, KIFC1 knockdown not only inhibited the proliferation, migration and invasion of glioma cells, but also increased the autophagic flux and downregulated the expression levels of MMP2. @*Conclusion@#: Upregulation of KIFC1 expression may promote glioma progression and KIFC1 may serve as a potential prognostic biomarker and possible therapeutic target for glioma.

ObjectiveTo explore the association between air pollutants and hospital outpatient visits in a district of Shanghai. MethodsDaily meteorological data, environmental data, data of outpatient visits to two secondary hospitals and two tertiary hospitals in this district from January 1, 2015 to December 31, 2019 were collected. A Poisson regression generalized linear model was used to analyze the exposure-response relationship between the air pollutants and hospital outpatient visits in this area. ResultsDuring the study period, the total number of outpatient visits in the included hospitals was 17 802 634, with an average daily total of (9 750±4 191) outpatient visits,and an average daily of (761±341) respiratory outpatient visits. In the lag effect of single pollutant model, when the concentration of air pollutant increased by 10 μg·m-3, PM2.5, SO2, NO2 had the maximum lag effect on the number of outpatient visits in the department of internal medicine for respiratory diseases on lag day 4, day 5 and day 7, respectively. And the RR values and 95%CI were 1.002 0(1.001 3‒1.002 6), 1.0154(1.012 3‒1.018 5), and 1.006 1(1.005 3‒1.006 9), respectively. ConclusionThere is a exposure-response relationship between air pollutants and the number of outpatient visits in each department of the hospitals, and different pollutants have different degrees of lag effects.

RESUMO

Objective To observe the clinical efficacy on the treatment of diabetic gastroparesis of self-designed Xiaopi Formula. Methods Totally 200 cases of DGP were divided into the observation group and the control group with random number table, with 100 cases in each group. Two groups were given the general treatment scheme, such as the subcutaneous injections of insulin or oral hypoglycemic medicine. The patients in the control group were treated with Domperidone orally, one tablet each time, 3 times a day;while the patients in the observation group were treated with self-designed Xiaopi Formula, twice a day, one time for morning and one time for evening. One course lasted for one month, 3 courses in total. Clinical efficacy and the change of gastric emptying rate, glycosylated hemoglobin and hemorrheology in the two groups were observed. Results Total effective rate of observation group (94.0%) was higher than that of the control group (82.0%), with statistical significance (P0.05); Observation group can obviously improve the indexes of blood rheology, with statistical significance with control group (P<0.05). Conclusion The clinical efficacy of self-designed Xiaopi Formula for the treatment of diabetic gastroparesis is obvious, which can promote gastric emptying rate and decrease HbA1c level, and improve blood rheology.

OBJECTIVE:To analyze the rationality of tamoxifen for ovulation induction,and to provide reference for the stan-dardization of off-label use of tamoxifen. METHODS:Takingtamoxifen,ovulation inductionandpolycystic ovarian syn-dromeas keywords,relevant literatures about off-label use of tamoxifen for ovulation induction were analyzed and summarized by a search of CNKI,Wanfang database,PubMed database up to July 2015. RESULTS:58 literatures were searched,among which clinical trial or systematic review about the effect of tamoxifen on ovulation induction were selected. 11 valid literatures were col-lected to confirm clinical efficacy of tamoxifen for ovulation induction. Referring to Diagnosis and Treatment of Polycystic Ovary Syndrome:An Expert Consensus,pharmaceutical affairs council of our hospital had agreed to off-label use of tamoxifen for induc-tion ovulation,and formulated the standard of off-label use of tamoxifen. CONCLUSIONS:Standard management of off-label use of tamoxifen could be better protect the rights both of patient and doctor.

Objective To analyze the effect ofmethylprednisolone stosstherapy on CD4+CD25+ regulatory T cell (Treg) and apoptosis factor CD95 expressions in the external peripheral blood of myasthenia gravis (MG) patients and explore the influencing factors of different clinical efficacy.Methods Thirty-one patients with MG,admitted to our hospital from January 2013 to September 2015,were included in this study;and 27 normal subjects were selected as control group.Methylprednisolone stosstherapy was given to the patient group.Expressions of Treg and CD95 in peripheral blood were detected by flow cytometry before and after treatment.Results Treg percentage of patients whose clinical absolute scale scores less or equal than 3 points and relative scale scores higher than 0.5 after methylprednisolonestosstherapy (13.39%+2.71%) was significantly higher than that ofhealthy controls (8.35%+1.87%,P<0.05);Treg percentage of patients whose clinical absolute scale scores greater than 3 as well as relative scale scores less than 0.5 after methylprednisolone stosstherapy (8.17%+1.31%) was slightly lower than that of healthy controls (P>0.05).CD95 percentage of patients whose clinical absolute scale scores less or equal than 3 points and relative scale scores higher than 0.5 after methylprednisolone stosstherapy (36.47%±5.32%) was not statistically different as compared with that of healthy controls (35.28%±5.58%,P>0.05),CD95 percentage of patients whose clinical absolute scale scores greater than 3 as well as relative scale scores less than 0.5 after methylprednisolone stosstherapy (34.97%±5.12%) was not statistically different as compared with that of healthy controls (P>0.05).Conclusion Treg ratio increases,CD95 change is not obvious in patients whose clinical curative effect is improved markedly after methylprednisolone stosstherapy.

@#Objective To evaluate the value of diffusion-weighted imaging-Alberta Stroke Program Early Computed Tomography (DWI-ASPECT) score in clinical outcomes in patients with acute anterior circulation occlusion treated by thrombectomy using solitaire AB stent. Methods Between January 2014 to March 2018,a total of 17 patients with acute anterior circulation occlusion were treated by thrombectomy using Solitaire AB stent. All the patients accepted assessment of DWI-ASPECT scores and NIHSS scores before undergoing endovascular treatment. General clinical data of patients,as well as DWI-ASPECTS scores,discharged NIHSS scores,discharged The modified Rankin Scale (mRS) scores,discharged Glasgow Outcome Scale (GOS) scores,intracranial hemorrhage,and presence or absence of radiation crown of each patient,were retrospectively analyzed. 3 months after thrombectomy,mRS scores,GOS scores and the functional activities questionary (FAQ) score of each patient were recorded. According to the DWI-ASPECTS score,the patients were divided into two group,one is the score ≥7 group,and the other is <7 group. The mRS score ≤2 was defined as a good prognosis,and >2 was as a poor prognosis. The correlation between with DWI-ASPECT scores,mRS scores and other factors were also analyzed. Results Mechanical thrombectomy with Solitaire AB stent was performed in 19 patients. DWI-ASPECTS scores ≥7 were in 9 cases,and the score <7 points was in 8 cases. A good prognosis (mRS scores ≤2) was observed in 8 patients in ≥7 group,and a poor prognosis (mRS score≤2) was observed in only 1 patients in <7 group. There was significantly difference in clinical outcomes between the two groups (P<0.001). Moreover,a good prognosis was observed in 9 patients,and there are 8 cases of DWI-ASPECTS score over 7 points among of them. A poor prognosis was observed in 8 patients,and there are only one case over 7 points. Conclusion DWI-ASPECTS score can be used as an indicator for preoperative assessment of patients with acute anterior circulation occlusion. Patients with DWI-ASPECTS score of ≥7 had a good prognosis after mechanical thrombectomy with Solitaire AB stent.

Objective To study the process of hydroxypropyl-β-cyclodextrin (HP-β-CD) inclusion techniques for osthol. Methods The inclusion complex of osthol and HP-β-CD was prepared by unsaturated water solution and freeze-drying technique. Inclusion techniques were selected by screening on quadratic orthogonal rotation combination design method, and the entrapment efficiency was identified by HPLC. Results The optimal technical conditions were as follows:the ratio of HP-β-CD and drug was 4.5∶1;temperature for electric mixer was 35 ℃;the stirring time for thermostatic waterbath was 210 min. Conclusion This method is reasonable and it may have a prosperous future of development and application.

Objective To investigate the effect of moldavica total flavone on pulmonary function and collagen type Ⅲ in lung tissue of ovalbumin-induced bronchial asthmatic rats. Methods A total of 60 SD rats were randomly divided into control group, model group, dexamethasone group, and moldavica total flavone low, medium and high dose group. Rat asthma model was established by ovalbumin challenge methods except the control group. The control and model group were intragastrically given distilled water, medicine groups were intragastrically given moldavica total flavone and dexamethasone for 30 d, once per day. After the last administration, the pulmonary function and airway responsiveness including breathing frequency (F), minute ventilation (MVb), peak inspiratory flow (PIFb), peak expiratory flow (PEFb) and enhanced pause (Penh) were measured by using non-invasive measurement system (Penh system, Buxco, USA). The collagen type Ⅲ in lung tissue were detected by ELISA. Results The moldavica total flavone at 180 and 360 mg/kg decreased F and Penh (P<0.05, P<0.01), increased MVb, PIFb and PEFb (P<0.05), and decreased collagen type Ⅲ in lung tissue (P<0.05, P<0.01). Conclusion Moldavica total flavone can relieve the airway hyperresponsiveness and remodeling, improve respiratory function of ovalbumin-induced bronchial asthmatic rats.

In order to clarify the chemical composition and source of Banxia Xiexin decoction quickly and comprehensively, whole and individual herbs of Banxia Xiexin decoction were analyzed by ultra-performance liquid chromatography with quadrupole-time-of-flight mass spectrometry (UPLC/Q-TOF-MS(E)). Under identical experiment conditions, chromatography results were compared between experiment groups. Based on the Q-TOF-MS(E) analysis, 74 peaks were identified on line. The herbal sources of these peaks were assigned. The results implied that flavonoids, triterpenoid saponins, alkaloids and glycosides were the main components in effective part of Banxia Xiexin decoction. The method established is simple and rapid for elucidation the constituents of Banxia Xiexin decoction and the results could be used for the quality control of Banxia Xiexin decoction.

Objective To investigate the effects of moldavica total flavone on vascular endothelial growth factor (VEGF), intercellular adhesion molecule-1 (ICAM-1), nitric oxide (NO) and endothelin-1 (ET-1) in ovalbumin-induced bronchial asthmatic rats, and explore its mechanism. Methods A total of 60 SD rats was randomly divided into control group, model group, dexamethasone (1 mg/kg) group and moldavica total flavone low (90 mg/kg), medium (180 mg/kg) and high (360 mg/kg) dose group. Rats asthma model was established by ovalbumin challenge methods except the control group. After modeling, rats in control and model groups were intragastrically given distilled water, rats in medicine groups were intragastrically given moldavica total flavone and dexamethasone for 30 d. VEGF in lung tissue and ICAM-1 in bronchoalveolar lavage fluid (BALF) were detected respectively by ELISA. The content of ET-1 and NO in BALF were analyzed with radioimmunity method and nitric acid reductase method. Results The VEGF in lung tissue and ICAM-1, NO and ET-1 in BALF were decreased by moldavica total flavone at 180 and 360 mg/kg (P<0.05, P<0.01) in ovalbumin-induced bronchial asthmatic rats. Conclusion Moldavica total flavone is effective in adjusting abnormal increase of VEGF, ICAM-1, ET-1 and NO of bronchial asthma rats.

BACKGROUND:Studies have shown that the application of electrical stimulation of peripheral nerve fibers can recover partial muscle functions due to the loss of central nervous control. OBJECTIVE:To verify the feasibility of selective nerve excitatory effectively used the symmetric biphasic pulses under the bipolar electrodes stimulation of 1 mm. METHODS:Eight adult Wistar rats were selected to expose the sciatic nerves after anesthesia and then the electrodes were placed on the sciatic nerves careful y to establish the model of selective nerve stimulation. Experimental electrode was homemade Cuff bipolar electrode, and the electrode stimulators were Grass S88 stimulator and AWG2005 arbitrary waveform signal generator. The two-way dual-electrode stimulation was used. The distance between two electrodes was 1 mm, and the stimulation waveform was symmetric biphasic pulse with the width of 0.2 ms. The output pulse amplitude, pulse width and delay could be adjusted. The stimulation intensity was adjusted, and the law of nerve excitability was detected under two-way dual-electrode stimulation, in order to achieve selective nerve excitability, and the feasibility of two-way dual-electrode stimulation to achieve selective nerve excitatory was verified with“col ision”method. RESULTS AND CONCLUSION:The change of nerve action potential was amplified by P511 amplifier and then linked into oscil oscope for displaying, and dual-electrode stimulation waveform was the symmetric biphasic pulse with the width of 0.2 ms. With the increasing of stimulus amplitude, we achieved the selective nerve excitatory. The results indicate the selective nerve excitatory can be achieved with the closed (1 mm) dual-electrode symmetric pulse, and the feasibility and effectiveness of this method can be verified with“col ision”principle.

<p><b>OBJECTIVE</b>To investigate serum APPL1 level in patients with newly diagnosed type 2 diabetes mellitus (T2DM) and analyze its correlation with body mass index (BMI), waist-to-hip ratio (WHR), blood pressure, fasting plasma glucose, fasting blood insulin, HbA1c, and insulin resistance (HOMA-IR).</p><p><b>METHOD</b>Serum APPL1 levels were determined using ELISA in 22 normal control subjects and 63 patients with newly diagnosed T2DM, and HOMA-IR of the subjects was calculated using the HOMA model.</p><p><b>RESULTS</b>The diabetic patients had significantly elevated levels of serum APPL1 compared with the control subjects (85.71∓27.39 vs 64.52∓16.28 pg/ml, P<0.01), with also significantly increased BMI, WHR, SBP, FINS, LgHOMA-IR (P<0.01) and LDL-C (P<0.05) but lowered HDL-C (P<0.01). Fasting serum APPL1 levels were positively correlated with FPG, FINS, and HOMA-IR (r=0.215, 0.297, 0.334, P=0.014, 0.006, 0.002, respectively). In multiple linear regression analysis with APPL1 as the dependent variable, HOMA-IR (β=0.329, P=0.002) was included in the equation.</p><p><b>CONCLUSION</b>Patients with newly diagnosed T2DM have elevated serum APPL1 levels, suggesting the involvement of APPL1 in the development of T2DM.</p>

Objective To explore the possible mechanism of sex hormone-binding globulin (SHBG) in diabetes mellitus by detecting the level of serum SHBG in patients with newly diagnosed type 2 diabetes mellitus (T2DM),and analyze the correlation between serum SHBG and insulin resistance.Methods SHBG levels were determined by enzyme linked immunosorbent assay (ELISA) in 30 normal controls (control group) and 60 newly diagnosed T2DM patients(T2DM group).HOMA-IR was calculated.Results The levels of serum SHBG in T2DM group were significantly lower than those in control group [(22.6 ±9.7)mmol/L vs.43.1 ±11.9) mmol/L,P <0.05]; SHBG level was negatively correlated with triglyceride(TG),low density hpoprotein cholesterol(LDL-C),fasting insulin(FINS),hemoglobintHbA1c) and lgHOMA-IR (r =-0.351,-0.361,-0.426,-0.645,-0.483,P <0.05),and positively correlated with high density lipoprotein cholesterol (HDL-C) (r =0.453,P <0.01).When SHBG was analyzed as a dependent variable by multiple linear regression,HbA1c (β =-0.614,P =0.000),lgHOMA-IR (β =-0.473,P = 0.014) and HDL-C (β = 0.234,P= 0.048) were into the equation.Conclusions Patients who have diagnosed T2DM show low level of serum SHBG.HOMA-IR and HbA1c are key indicators of SHBG,suggesting that SHBG may be closely related with T2DM.

Objective To investigate the correlation between serum level of visfatin and type 2 diabetes mellitus (T2DM). Methods Impaired glucose regulation (IGR) group consisted of 40 patients,T2DM group consisted of 106 patients, while control group consisted of 86 subjects. The serum visfatin levels of all groups were detected by enzyme linked immunosorbent assay (ELISA). Results Fasting serum visfatin levels in IGR group and T2DM group were significantly higher than those in control group [(19.93 ±6.89) μg/L and (29.53 ± 11.33) μg/L vs. (16.12 ± 5.24) μ g/L, P < 0.01]. Fasting serum visfatin levels positively correlated with waist-to-hipratio (WHR) (r = 0.161, P < 0.05); significantly positively correlated with triacylglycerol (TG), low density lipoprotein cholesterol (LDL-C), glycosylated hemoglobin (HbA1c),fasting plasma glucose (FPG), fasting insulin (FINS), lg Homeostasis model assessment-insulin resistance index (HOMA-IR) (r = 0.189,0.266,0.643,0.574,0.285,0.526,P < 0.01), and significantly negatively correlated with high density lipoprotein cholesterol (HDL-C)(r =-0.377,P <0.01). When visfatin was analyzed as a dependent variable in multiple linear regression, HbA1c (β = 0.512, P = 0.000), lg HOMA-IR (β= 0.172, P = 0.026), WHR (β = 0.119, P = 0.036) were into the equation. Conclusion Visfatin may be involved in glucose and lipid metabolism regulation, and closely related with insulin resistance; visfatin may be a risk factor for T2DM.

BACKGROUND: It is indicated in latest research that in cerebral protective measures of cerebral vasospasm induced by subarachnoid hemorrhage due to various factors in acute stage, mild hypothermia has been drawn the attention specially and it has been recommended in clinical practice. But such therapy is generally limited in experimental research and aneurysm hemorrhage, the clinical research on subhypothermia probably provides important influence on cerebral vasospasm in severe craniocerebral injury.OBJECTIVE: Based on cerebral vascular hemodynamical indexes (CVDI),the cerebral protection of subhypothermia was observed on cerebral vasospasm in severe craniocerebral injury.DESIGN: Randomized controlled experiment.SETTING: Department of Neurological Surgery of No.16 Hospital of Chinese PLA and General Military Institute of Neurological Medical Science in Zhujiang Hospital Affiliated to First Military Medical University of Chinese PLA.PARTICIPANTS: Totally 36 cases of severe craniocerebral injury were selected in General Military Institute of Neurological Medical Science in Zhujiang Hospital Affiliated to First Military Medical University of Chinese PLA from July 1997 to August 1999, which were randomized into the control and treatment group, 18 cases in each one. At same period, 24 cases with normal CVDI were screened and taken as normal group. All of receptors participated in the experiment in volunteer.METHODS: In both the control and treatment group, the treatment was applied with anti-inflammation, stopping bleeding, fluid limitation, dehydration, supporting, hyperbaric oxygen, etc. In the control, the normal body temperature was maintained and in treatment group, anus temperature was dropped to about 33 ℃ in 4 hours to 8 hours, which was maintained for 3-test was given on the day of injury (0), on the 1st, 3rd, 7th, 14th and 21st days assay was done on the day of injury (0), on the 1st, 3rd, 7th, 14th and 21st days successively, in which, minimum blood velocity (Vmin) and minimum blood flow (Qmin) reflect blood supply of distal cerebral vessel and blood flow.Cerebral vessel resistance (CVR) reflects smooth degree of cerebral microcirculation. Dynamical resistance (DR) reflects auto-regulation of cerebral vessel. Criteria of evaluation: Recovery state of consciousness was justified according to Glasgow Coma Scale (GCS) in 1 week after injury. The outcomes were evaluated according to Glasgow outcome scale (GOS) in 3 months (5 score: good, 4 score: moderate handicapped, 3 score: severely handicapped, 2 score: vegetative state and 1 score: death). The case over 4 score indicated good outcome.ery of consciousness and outcomes in 1 week after injury in the control and treatment group.RESULTS: Totally 36 cases of severe craniocerebral injury entered result phases after injury in the control, named hypoperfusion phase (0 day), hyperperfusion phase (1-3 days), cerebral vasospasm phase (4-14 days) and improving phase (>15 days). In treatment group, 3 phases were manifested,named hypoperfusion phase (0 day), improving phase (1-3 days) and recovery phase (> 4 days), without hyperperfusion phase. Eight cases and 2 cases of cerebral vasospasmodic changes in CVDI presented in the control in focus: The maximum volume (140.9±22.95) cm3 was on the 14th day after injury in the control and that (95.83-±14.97) cm3 was on the 3rd day in treatment group. On the 14th day after injury, the volume in treatment group in 1 week after injury: It was 22.2% (4/18) in the control and 55.6%(10/18) (P < 0.05) in treatment group. Improving outcome rate: It was 38.9% (7/18) and 66.7% (12/18) in treatment group.CONCLUSION: Subhypothermia reduces incidence of cerebral vasospasm by stabilizing cerebral circulation after severe craniocerebral injury, especially by inhibiting acute hyperperfusion after the injury so that the volume of cerebral edema in focus is lessened remarkably and the prognosis is improved.

RESUMO

AIM: To investigate the protecting mechanisms of adjusting blood pressure and blood fat capsule(Radix angelicae sinesis,Rhizoma alismatis,poria,Rhizoma attactylodis macrocephalae,Rhizoma Chuanxiong,Radix Paeoniae alba,semen arecae,Folium perillae) in diabetic nephropathy rats induced by streptozotocin. METHODS: Forty-eight health male Wister rats were randomly divided into normal group,diabetic nephropathy group,adjusting blood pressure and blood fat capsule group and valstatan group.After experiment urine,blood,kidney samples were taken,and immunohistochemistry were performed. RESULTS: 24 hour urinary microalbumin,creatinine clearance,?_2-microglobulin,creatinine clearance ET1 and NOS in kidneys had distinct difference among three gruops. CONCLUSION: The provention and cure of DN ' function of adjusting blood pressure and blood fat capsule bears relationship with inhibition of ET-1,NOS's expression.
