RESUMO

Homoharringtonine (HHT), was first isolated from the bark of Cephalotaxus harringtonia (Knight ex J. Forbes) K. Koch and Cephalotaxus fortunei Hook trees. The bark extract is used to treat leukemia and in recent years has also been used in traditional Chinese medicine (TCM) to treat solid tumors. However, the inhibitory mechanism of HHT in the progression of hepatocellular carcinoma (HCC) is rarely studied. We aimed to evaluate the antitumor efficacy of HHT on HCC in vitro and in vivo and elucidate the underlying molecular mechanism(s). HCC cell lines, including HCCLM3, HepG2, and Huh7, were used to evaluate the antitumor efficacy of HHT in vitro. Cytotoxicity and proliferative ability were evaluated by MTT and colony formation assays. Cell cycle progression and apoptosis in HHT-treated HCC cells were evaluated by flow cytometry. To determine the migration and invasion abilities of HCC cells, wound-healing and Transwell assays were used. Finally, western blot analysis was used to reveal the proteins involved. We also established a xenograft nude mouse model for in vivo assessments of the preclinical efficacy of HHT, mainly using hematoxylin and eosin staining, immunohistochemistry, ultrasound imaging (USI), and magnetic resonance imaging (MRI). HHT suppressed the proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) of HCC cells, and induced cell cycle arrest at the G2 phase and apoptosis. In the HCC xenograft model, HHT showed an obvious tumor-suppressive effect. Surprisingly, Slug expression was also decreased by HHT via the PI3K/AKT/GSK3ß signaling pathway at least partially suppressed the growth of HCC via the PI3K/AKT/GSK3ß/Slug signaling pathway.

A hexagonal graphene-like boron-carbon-nitrogen (h-BCN) monolayer, a new two-dimensional (2D) material, has been synthesized recently. Herein we investigate for the first time the thermal conductivity of this novel 2D material. Using molecular dynamics simulations based on the optimized Tersoff potential, we found that the h-BCN monolayers are isotropic in the basal plane with close thermal conductivity magnitudes. Though h-BCN has the same hexagonal lattice as graphene and hexagonal boron nitride (h-BN), it exhibits a much lower thermal conductivity than the latter two materials. In addition, the thermal conductivity of h-BCN monolayers is found to be size-dependent but less temperature-dependent. Modulation of the thermal conductivity of h-BCN monolayers can also be realized by strain engineering. Compressive strain leads to a monotonic decrease in the thermal conductivity while the tensile strain induces an up-then-down trend in the thermal conductivity. Surprisingly, the small tensile strain can facilitate the heat transport of the h-BCN monolayers.

In recent years, silver nanoparticles (Ag NPs) have been used as positive electrode material for zinc/silver batteries, and the silver oxides formed during the charging process determine the discharge performance of batteries. Therefore, it is important to study the oxidation behavior of Ag NPs in alkaline solution. Single-nanoparticle collision is an important tool for analyzing oxidation behavior of individual nanoparticles. Based on thermodynamic information from collision events, it is known that oxidation products are potential-dependent and size-dependent. Based on dynamic information, including collisional peak shapes and duration time, it was observed that the Ag NP collision oxidation process changed from stepwise oxidation to direct oxidation as the potential increased or size decreased. This work provides guidance for application of Ag NPs in zinc/silver batteries and proposed a strategy for oxidation behavior of individual NP that could be tracked in situ through an all-encompassing view of thermodynamic and dynamic information.

AIMS: To conduct a systematic review to summarize the definition, measurement tools, prevalence, and contributing factors of impaired awareness of hypoglycemia (IAH) in type 2 diabetes mellitus (T2DM). METHODS: A reproducible search strategy was used to identify factors affecting IAH in T2DM in PubMed, MEDLINE, EMBASE, Cochrane, PsycINFO, and CINAHL from inception until 2022. Literature screening, quality evaluation, and information extraction were performed independently by 2 investigators. A meta-analysis of prevalence was performed using Stata 17.0. RESULTS: The pooled prevalence of IAH in patients with T2DM was 22% (95%CI:14-29%). Measurement tools included the Gold score, Clarke's questionnaire, and the Pedersen-Bjergaard scale. IAH in T2DM was associated with sociodemographic factors (age, BMI, ethnicity, marital status, education level, and type of pharmacy patients visited), clinical disease factors (disease duration, HbAlc, complications, insulin therapy regimen, sulfonylureas use, and the frequency and severity of hypoglycemia), and behavior and lifestyle (smoking and medication adherence). CONCLUSION: The study found a high prevalence of IAH in T2DM, with an increased risk of severe hypoglycemia, suggesting that medical workers should take targeted measures to address sociodemographic factors, clinical disease, and behavior and lifestyle to reduce IAH in T2DM and thus reduce hypoglycemia in patients.

Ischemic stroke (IS) is harmful to human health and social development, and there is no medicine available at present. To find the hit compound for treating ischemic stroke, we screened 28 FDA approved nervous system drugs by using an in vitro OGD-induced stroke model. Notably, our in vitro and in vivo studies demonstrated that low-dose sertraline had good neuroprotective activities, while high-dose sertraline showed significant toxicity. Interestingly, the same high-dose sertraline in the control group did not exhibit any obvious toxic effect. Therefore, it is important to modify the structure of sertraline to improve the activity and reduce the toxicity. Stereoisomers of sertraline were first investigated to analyze the influence of stereochemistry on the neuroprotective activities, which showed no obvious difference. Then we evaluated the activity of our previously reported sertraline analogues and found that introducing amide or alkane groups to the amino moiety might be beneficial to enhance the activity and reduce the toxicity. Thus, 10 new analogues were designed, synthesized, and evaluated. Among them, compound OY-201 showed the best safety and neuroprotective activity in both in vitro and in vivo models. Moreover, it exhibited good blood-brain barrier (BBB) permeability, indicating its potential for the development of anti-ischemic stroke drugs.

Covalent attachment of 2,2'-(ethylenedioxy)-diethylamine to multiwalled carbon nanotubes (MWCNTs) produced amino-functionalized MWCNTs which behaved like liquids at ambient temperature. These liquid-like MWCNTs (l-MWCNTs) could be homogeneously dispersed and chemically embedded in an epoxy matrix by solvent-free processing. In contrast, solid MWCNTs (s-MWCNTs) functionalized by 1,8-diaminooctane were poorly dispersed in epoxy although they possess chemical structures and functionalization comparable to l-MWCNTs. An epoxy composite filled with pristine MWCNTs (p-MWCNTs) was also fabricated in the absence of a solvent at the same loading for comparison. The molecular level coupling of l-MWCNTs and epoxy provided significant improvements in overall mechanical properties relative to those composites containing p-MWCNTs and s-MWCNTs. The Young's modulus, storage modulus, tensile strength, failure strain and toughness of neat epoxy were increased by 28.4, 23.8, 22.9, 24.1 and 66.1%, respectively, by adding 0.5 wt% of l-MWCNTs. Thus, functionalized carbon nanotubes in liquid form contributed to better dispersion and superior interfacial bonding with the epoxy matrix, thereby facilitating greater mechanical reinforcement efficiency.

A simple method was developed to assemble graphite oxide (GO) densely onto electrospun (ES) polyamide 66 (PA66) nanofibrous membranes, used as a guide for the deposition of graphene nanosheet (GNS) conductive networks for preparing transparent conductive thin film (TCF). The main advantage of this technique by comparison with previous methods is that graphene does not form a uniform coating, but a percolated conductive network, when guided by PA66 nanofiber templates. A low surface coverage of the transparent substrate by GNS resulted in high transmittance. Polyvinylpyrrolidone-stabilized GO (PVP-GO) was prepared as a modifier for improving the adsorption to the nanofibers. The resulting PVP-GO material could adsorb well on PA66 nanofibers due to stronger hydrogen bonds. Hence, a lower sufficient concentration of PVP-GO (0.050 wt%) solution was required than that for GO solution (0.100 wt%) to fabricate a complete conductive path through a possible enriched adsorption process. For TCF applications, a reduction step is essential because as-deposited GO is non-conductive. In this work, we reduced GO to GNS by a combination of chemical reduction and thermal annealing. The TCF optical transmittance also could be improved after thermal annealing at 350 °C above the PA66 melting point. Light scattering by PA66 nanofibers was found as the main cause of reduced transmittance. A fused film, obtained after electrospinning PA66 solution for 120 s, and immersing in 0.050 wt% PVP-GO solution, exhibits a surface resistance of 8.6 × 10³ Ω/square, while maintaining 88% light transmittance.

OBJECTIVE: To study on the optimal medium ingredients for PLB induction and proliferation of Dendrobium officinale. METHODS: Seed embryos of Dendrobium officinale were cultivated in MS medium as the basic medium, along with different plant hormones like 6-BA, NAA,2,4-D and KT or their combinations added with organic additives like PE, BE, AE and CM. RESULTS: BA and NAA combination was not conductive to germination and the germination ratio was even lower than that of MS medium; 10% PE and CM was beneficial to PLB induction; 2,4-D was not conductive to growth and proliferation; A certain concentration of BA, KT and NAA was beneficial to PLB proliferation; KT at 1.0 mg/L recorded the highest 40 d PLB proliferation times at 9.0; PE, CM and AE could promote the PLB proliferation at different levels, among which 10% CM was the most effective. CONCLUSION: The optimized medium ingredients suitable for PLB induction are MS +10% CM +1.0g/L AC; The optimized medium ingredients suitable for PLB proliferation are MS + 1.0 mg/L KT + 0.2 mg/L NAA + 10% CM.

Professor CHEN Gui-zhen's experience of acupuncture for perimenopausal panic disorder was summarized. Professor CHEN believes that the etiology and pathogenesis of perimenopausal panic disorder are the deficiency of congenital kidney essence and the injury of acquired spleen and stomach, leading to mental dysfunction. The clinical treatment should be based on the principle of "nourishing the congenital, tonifying the acquired, and regulating thoroughfare vessel and conception vessel". As for the acupoint selection, the combination of back-shu point and the front-mu point is valued; as for the acupuncture technique, quick needling and skillful application of catgut embedding are recommended. The combination of acupuncture and medicine shows synergistic effect. In addition, emotional therapy is also applied for this condition. In conclusion, it is advocated to treat perimenopausal panic disorder with the combination of acupuncture, catgut embedding, herbs and emotional therapy.

Antibiotics and antibiotic resistance genes (ARGs) are highly concerning emerging polltants. Wastewater treatment plants (WWTPs) are considered to be one of the most important anthropogenic sources of antibiotics and ARGs in the environment. Six WWTPs of a city were selected to investigate the antibiotics and ARGs in the influents and effluents, respectively. In total, 73 kinds antibiotics belonging to 7 classes were detected in WWTPs. The total concentration of those antibiotics ranged between 490.2-2288.6 ng·L-1, and ofloxacin, maprofloxacin, and roxithromycin were the most frequently detected antibiotics in the influents. A dramtic difference was observed in the antibiotic removal efficiency among the WWTPs, and the total antibiotic concentration in the effluents ranged between 260.2-1464.3 ng·L-1. Macrolides and quinolones are the main classes in the effluents, and the antibiotic concentration in the effluents was higher than that in the influents. Ten kinds of ARGs were detected in the WWTPs. Among these, sul Ⅰ was the most widely prevalent with an absolute abundance of 2.4×105-5.4×106 copies·mL-1. No significant difference in ARG abundance was observed between industrial wastewater and domesic wastewater. Furthermore, the correlation between ARG abundance and antibiotic concentrations weakened after treatment in the WWTPs.

Incorporation of the Monte Carlo (MC) algorithm in optimizing CyberKnife (CK) plans is cumbersome, and early models unconfgured MC calculations, therefore, this study investigated algorithm-based dose calculation discrepancies by selecting different prescription isodose lines (PIDLs) in head and lung CK plans. CK plans were based on anthropomorphic phantoms. Four shells were set at 2-60 mm from the target, and the constraint doses were adjusted according to the design strategy. After optimization, 30%-90% PIDL plans were generated by ray tracing (RT). In the evaluation module, CK plans were recalculated using the MC algorithm. Therefore, the dosimetric parameters of different PIDL plans based on the RT and MC algorithms were obtained and analyzed. The discrepancies (mean±SD) were 3.72%±0.31%, 3.40%±0.11%, 3.47%±0.32%, 0.17%±0.11%, 0.64%±3.60%, 7.73%±1.60%, 14.62%±3.21% and 10.10%±1.57% for D1%, D(mean), D98% and coverage of the PTV, DGI, V5, V3 and V1 in the head plans and -6.32%±1.15%, -13.46%±0.98%, -20.63%±2.25%, -34.78%±25.03%, 122.48%±175.60%, -12.92%±5.41%, 3.19%±4.67% and 7.13%±1.56% in the lung plans, respectively. The following parameters were signifcantly correlated with PIDL: dD98% at the 0.05 level and dDGI, dV5 and dV3 at the 0.01 level for the head plans; dD98% at the 0.05 level and dD1%, dD(mean), dCoverage, dDGI, dV5 and dV3 at the 0.01 level for the lung plans. RT may be used to calculate the dose in CK head plans, but when the dose of organs at risk is close to the limit, it is necessary to refer to the MC results or to further optimize the CK plan to reduce the dose. For lung plans, the MC algorithm is recommended. For early models without the MC algorithm, a lower PIDL plan is recommended; otherwise, a large PIDL plan risks serious underdosage in the target area.

PURPOSE: We investigated the dosimetric effect of intrafraction tumor motion in lung stereotactic body radiotherapy using the CyberKnife static tracking system. METHODS: Four-dimensional computed tomography scans of a dynamic thorax phantom were acquired. Two motion ranges, 3 collimator sizes, and 4 treatment starting phases were investigated. Monte Carlo dose distributions were calculated on internal target volume with a treatment-specific setup margin for 6 Gy/1 fraction. Dosimetric effects of intrafractional tumor motion were assessed with Gafchromic films. γ (5%/3 mm), dose differences, and distance to agreement were analyzed. RESULTS: With 30 mm collimator plans, the measured dose passed the criteria γ (5%/3 mm) in all tumor motion ranges. The γ passing rates of the plans using 20 mm or 20+35 mm collimators were much lower than that with 30 mm collimator, especially with the 30 mm tumor motion range. The measured dose of 10 mm tumor motion ranges all passed the 90% criteria of γ (5%/3 mm), the results being much better than those of 30 mm tumor motion ranges, which were below 80%. The results of same delivered plan but treated with different starting phases varies greatly. CONCLUSION: Xsight Lung Tracking technique should be used with caution in lung cancer stereotactic body radiation therapy because the temporal dose variations can be significant.

An analytical model was developed to study the interlaminar fracture behaviour of polymer composite reinforced by carbon fibres grafted with carbon nanotubes. Delamination properties, such as load with displacement or crack (R-curve) and toughness with crack (GR-curve), can be obtained from this model. The bridging laws presented, based on the CNT pullout mechanism (CNT pullout from polymer matrix) and the CNT sword-in-sheath mechanism (CNT breakage), were incorporated into the proposed analytical model to investigate the influence of the structure of CNT growth onto CFs (CNT@CFs) on delamination properties. The numerical results showed that different toughening mechanisms led to different features of GR-curves, R-curves, and load with displacement curves. Parametric study demonstrated that strengthening the CNT@CF interface resulted in significant improvement in toughness. Further, it was found that elastic deformation of CNTs played an important role in the toughness improvement in the CNT sword-in-sheath mechanism, but no such role was evident in the CNT pullout mechanism.

The objective of this study is to quantify the clinical accuracy of the Cyberknife Xsight Lung Tracking System (XLTS) in our center and calculate the PTV margin of XLTS treated lung tumors. Data from the treatment log files of 22 lung cancer patients treated with the CyberKnife XLTS were analyzed and the PTV margin was calculated. Segmentation, deformation, correlation, prediction and targeting errors were calculated from the log files of XLTS treatments. Two different methods were used to calculate anisotropic treatment margin. The relationships between tumor motion ranges and the correlation and prediction errors were also analyzed. Based on our estimation of a 4 mm global margin, 95% coverage in the S-I direction and 100% coverage in the L-R and A-P directions were obtained. Strong correlations between tumor motion range and the standard deviation (SD) of correlation and prediction errors were also found. Tumor position motion caused by respiration can be compensated using the Xsight Lung Tracking System. We found total tracking errors to be less than 4 mm in all three directions. This result could provide a reference for the selection of PTV margin for treatment with the CyberKnife XLTS.

A hard template method to prepare N-doped graphene foams (NGF) with superfast template removal was developed through a pyrolyzing commercial polyurethane (PU) sponge coated with graphene oxide (GO) sheets in an ethanol flame. The removal of the template was fast and facile, and could be completed in less than 60 s in an open environment. The synthesized graphene foams consisted of a unique structure of 3D interconnected hollow struts with highly wrinkled surfaces, and the morphology of the hollow struts could be tuned by controlling the GO dispersion concentration. The foams showed high hydrophobicity and were used as absorbents for a variety of organic solvents and oils. The unique NGF structure afforded a high absorption rate and capacity, and a remarkable 98.7% pore volume of the foam could be utilized for absorption of hexane, exhibiting one of the highest capacity values among existing absorptive counterparts. The N-doping brought higher capacitive performance than conventional graphene foams prepared by chemical vapor deposition on nickel foam templates. The NGFs also displayed high elasticity and could recover completely after 50% compressive strain. Owing to easy availability and reduction environment of the flame, complete thermal decomposition of the PU sponge and highly porous open-cell structure, and flame resistance of the graphene foam, the present flame method was demonstrated to be a simple, effective, and ultrafast approach to fabricate ultra-low-density NGFs with good electromechanical response, excellent organic liquid absorption, and high-energy dissipation capabilities.

The reduction of nitrate contaminant in groundwater has gained renewed and intensive attention due to the environmental problems and health risks. Catalytic denetrification presents one of the most promising approaches for the removal of nitrate from water. Catalytic nitrate reduction from water by powder catalysts and catalytic membrane in a batch reactor was studied. And the effects of the initial concentration, the amounts of catalyst, and the flux H2 on the nitrate reduction were also discussed. The results demonstrated that nitrate reduction activity and the selectivity to nitrogen gas were mainly controlled by diffusion limitations and the mass transfer of the reactants. The selectivity can improved while retaining a high catalytic activity under controlled diffusion condition or the intensification of the mass transfer, and a good reaction condition. The total nitrogen removal efficiency reached above 80%. Moreover, catalytic membrane can create a high effective gas/liquid/solid interface, and show a good selectivity to nitrogen in comparative with the powder catalyst, the selectivity to nitrogen was improved from 73.4% to 89.4%.

RESUMO

A facile approach to functionalize carbon nanofibers [CNFs] with nanostructured polyaniline was developed via in situ mechanochemical polymerization of polyaniline in the presence of chemically treated CNFs. The nanostructured polyaniline grafting on the CNF was mainly in a form of branched nanofibers as well as rough nanolayers. The good dispersibility and processability of the hybrid nanocomposite could be attributed to its overall nanostructure which enhanced its accessibility to the electrolyte. The mechanochemical oxidation polymerization was believed to be related to the strong Lewis acid characteristic of FeCl3 and the Lewis base characteristic of aniline. The growth mechanism of the hierarchical structured nanofibers was also discussed. After functionalization with the nanostructured polyaniline, the hybrid polyaniline/CNF composite showed an enhanced specific capacitance, which might be related to its hierarchical nanostructure and the interaction between the aromatic polyaniline molecules and the CNFs.

OBJECTIVE: To study the relationship between obstructive sleep apnea and nocturnal gastroesophageal reflux (GER). METHODS: Seventy-six patients with obstructive sleep apnea hypopnea syndrome (OSAHS) underwent polysomnography (PSG) and nocturnal distal esophageal pH monitoring, to assess the prevalence of GER in OSAHS patients. Among these patients, thirty-two OSAHS patients with GER disease had been operated on. Surgical treatment included uvulopalatopharyngoplasty (UPPP), inferior turbinate reduction, nasal septoplasty et al. After 6 months of surgical treatment, the PSG and pH probe testing were rechecked. The pre and post operative apnea-hypopnea index (AHI), micro-arousal index (MAI), lowest artery oxygen saturation (LSaO2), acidified index of distal esophagus (pHI) and percentage of acid contact time of esophagus were compared. The correlation analysis was also employed. RESULTS: The AHI of 76 OSAHS patients was (38.6 +/- 29.5)/h (average +/- s), GER was present in 48 patients (63.2 %). The pre and post operative AHI, MAI, LSaO2, pHI and percentage of ACT in 32 OSAHS patients associated with GER were (51.2 +/- 23.1)/h and (17.3 +/- 10.3)/h, (38.3 +/- 21.4 )/h and (14.5 +/- 10.0)/h, 0.698 +/- 0.107 and 0.858 +/- 0.076, (12.5 +/- 6.6)/h and (6.3 +/- 4.2)/h, (10.3 +/- 5.2)% and (4.5 +/- 2.9)%. The differences were statistically significant by matching t test (t were 10.95, 7.82, 15.97, 6.12, 7.62 respectively, P < 0.001). There were positive relationships between the reductions of AHI, MAI and the reductions of pHI (r were 0.775, 0.764, P < 0.001). The improved levels of AHI and MAI were correlated with the reduction of the percentage of ACT (r were 0.607, 0.730, P < 0.001). CONCLUSIONS: GER is prevalent in OSAHS patients. Surgical treatment of OSAHS has significant reduction in AHI, MAI as well as marked improvement in the GER, which suggests that nocturnal gastroesophageal reflux is correlated with the disorder of sleep respiration and the sleep disturbances.

The reactions of diselenophosphates, [dsep, (RO)2PSe2-; R = Et, (n)Pr and (i)Pr] with cadmium(II) and mercury(II) perchlorates in a 2 : 1 molar ratio formed compounds of stoichiometry M[Se2P(OR)2]2{M = Cd, R = Et (1), (n)Pr (2), (i)Pr (3); Hg, Et(4), (n)Pr (5), (i)Pr (6)}, and with zinc(II) perchlorates, chalcogen centered tetranuclear clusters, [Zn4(micro4-E){Se2P(OR)2}6]{E = Se, R = Et (7), (n)Pr (8), (i)Pr (9); E = O, R = Et (10), (n)Pr (11), (i)Pr (12)} were formed. All these complexes have been characterized with the help of analytical data, X-ray crystallography (1, 3, 6, 10, 11 and 12), and FAB-mass spectrometry (7-12). Compound 1 is a linear double-chain polymer, in which each pair of Cd atoms is bridged by two dsep ligands; the mercury 6 polymer has a helical chain structure, in which two Hg atoms are bridged by one dsep ligand, and the other ligand chelates the Hg atom. The chelating dsep ligands lie on either side of the helical chain. Compound 3 exists as a dimer in which two cadmium atoms are connected by two bridging dsep ligands, and each cadmium atom is further chelated by a dsep ligand. The metal atoms in 1, 3 and 6 are each coordinated by four selenium atoms in a distorted tetrahedral geometry. Clusters 10-12 have tetrahedral array of zinc atoms with an oxygen atom in the center with edge-bridging dsep ligands. Positive FAB-mass spectra support the formation of selenium-centered clusters,7-9, of which the cluster 8 was structurally confirmed earlier. The solution state behavior of compounds 1-12 has been studied by using multinuclear NMR spectroscopy. Dimer 3 in CD2Cl2 showed monomer-dimer exchange equilibrium in the temperature range 20 to -90 degrees C and the free energy of activation is calculated from the coalescence temperature as DeltaG++(223 K)= 38.5 kJ mol(-1). Polymer undergoes depolymerization in CDCl3 and exhibits monomer-dimer exchange equilibrium in the temperature range 20 to -60 degrees C.
