BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) used to be defined as germinal center B-like and non-germinal center B-like subtypes, associated with different prognoses, but the conventional classification does not meet the needs of clinical practice because of DLBCL heterogeneity, a problem that might be improved by selection of miRNAs as biomarkers. METHODS: Twelve patients with DLBCLs were used to screen out the aberrant miRNA profile using miRNA microarray technology in two patient subtypes (six germinal center B-like and six non-germinal center B-like patients). The potential biomarkers were further analyzed using the quantitative reverse transcription-polymerase chain reaction method in 95 DLBCL patients to investigate relationships among expression levels of potent miRNA, clinicopathological features and survival rates of patients. RESULTS: miR-208a-5p, miR-296-5p and miR-1304-5p were screened as potential biomarkers. miR-208a-5p and miR-296-5p were shown to be associated with better survival of patients after Kaplan-Meier analysis, whereas miR-1304-5p overexpression indicated a poor survival prognosis independent of the DLBCL subtype. In addition, changes of miR-296-5p and miR-1304-5p expression, the International Prognostic Index (IPI) status and the age of patients were all independent indicators for DLBCL prognosis. We also found that high miR-208a-5p expression led to better outcomes in DLBCL patients with similar IPI scores; however high miR-1304-5p expression tended to indicate the opposite. CONCLUSIONS: MiR-208a-5p, miR-296-5p and miR-1304-5p levels might be potential biomarkers for the prediction of the prognosis of DLBCL patients.

OBJECTIVE: To detect the expression of miRNA-296-5p in diffuse large B cell lymphoma (DLBCL) cells and study the proliferation, migration and apoptosis of DLBCL cells after interfering the expression of miRNA-296-5p. METHODS: The expression of miRNA-296-5p in DLBCL cells line DB cells was detected by real-time quantitative reverse-transcription polymerase chain reaction (qRT-PCR). The possible roles of miRNA-296-5p in the biological and behavioral properties of DLBCL-DB cells were explored by transfection of miRNA-296-5p inhibitor for miRNA-296-5p knockdown, and detected by using CCK-8 method, Transwell method and Annexin V-FITC/PI. RESULTS: miRNA-296-5p was highly expressed in DLBCL-DB cells, and the inhibition of miRNA-296-5p could induce suppression of cell proliferation and migration, but the cell apoptosis was not changed significantly. CONCLUSION: The high expression of miRNA-296-5p may relate with the occurence and development of DLBCL, which may be a new therapeutic target for DLBCL.

RESUMO

Soil seed bank consists of all living seeds existed in soil and its surface litter, especially in topsoil, and can reflect the characteristics of regional biodiversity. As the base of vegetation restoration and potential greening material, topsoil and its seed bank are the limited and non-renewable resources in mining areas. The study of soil seed bank has become one of the hotspots in the research field of vegetation restoration and land reclamation in China mining areas. Owing to the special characteristics of mining industry, the soil seed bank study of mining areas should not only concern with the seed species, quantities, and their relations with ground surface vegetation, but also make use of the research results on the soil seed bank of other fragile habitats. Besides, a breakthrough should be sought in the thinking ways and research approach. This paper analyzed the particularity of mining area's soil seek bank research, summarized the research progress in the soil seed bank of mining areas and other fragile habitats, and put forward the challenges we are facing with. It was expected that this paper could help to reinforce the soil seed bank research of China mining areas, and provide scientific guidelines for taking great advantage of the significant roles of soil seed bank in land reclamation and vegetation restoration in the future.
