RESUMO

The authors present epidemiologic characteristics of COLD in workers of major atomic machinery enterprise. The article covers high prevalence of this disease on the enterprise, evaluation of connection between occupational and non-occupational risk factors and COLD development. Constant broncholytic therapy in the occupational health office caused significant improvement of clinical and functional parameters and decrease in transitory disablement duration.

RESUMO

The authors studied frequency, periods of development and morbidity structure of malignances among chronic uranium intoxication patients. tumors were diagnosed in 19.1% of the cases. Neoplasms developed in the presence of uranium intoxication symptoms in 57.8% of the patients, occurred after the occupational disease was ruled out in 42.2% of the cases. The oncologic morbidity structure was presented mostly by tumors of gastro-intestinal tract (33.3%) and respiratory system (20%).

Unidirectional fluxes from ATP to phosphocreatine (PCr) catalyzed by MM-isoenzyme of creatine kinase (CK) were measured by using 31P-NMR saturation transfer technique and by means of radioactively labeled [gamma-32P]ATP. At 30-37 degrees C and pH 7.4 in a wide range of [PCr]/[creatine] ([PCr]/[Cr]) ratios (0.2 to 3.0) both of these methods gave similar results, thus showing that magnetization (saturation) transfer allows to determine fluxes close to real ones under "physiological" conditions. However, at [PCr]/[Cr] ratio higher than 5 ([ADP] less than 30 microM) or at decreased temperatures (7-15 degrees C, [PCr]/[Cr] approximately 1) fluxes determined by saturation transfer substantially exceeded those measured with the radioactive label. These data imply that under "physiological" conditions phosphoryl group transfer is actually rate-determining step of the CK reaction. On the contrary, at high [PCr]/[Cr] values or at low temperature the control step could be shifted from the phosphoryl group transfer or distributed among other steps of the reaction.

It has been shown that the contents of mitochondria and mitochondrial isoenzyme of creatine phosphokinase are almost identical in skeletal and heart muscles. In mitochondria from both types of muscle creatine phosphokinase is functionally coupled to ATP-ADP translocase. This kind of coupling ensures complete conversion of mitochondrial ATP energy into the energy of creatine phosphate and effective control of oxidative phosphorylation by the creatine phosphokinase reaction. It has also been shown that all isoenzymes of creatine phosphokinase from heart and skeletal muscle have very similar kinetic properties. Significant differences have been found to exist between isoenzyme patterns of these muscles and also in distribution of different isoenzymes in the cells. In skeletal muscle cells creatine phosphokinase is present mainly as cytosolic MM isoenzyme; about 6% of total cellular activity is localised also in mitochondria. Due to high activity of cytosolic isoenzyme the total activity of creatine phosphokinase is about three times higher in skeletal muscle than in cardiac muscle. It has been also shown that phosphoenolpyruvate and glucose-6-phosphate do not have any inhibitory effect on creatine phosphokinase.

The kinetics of the creatine phosphokinase reaction catalyzed by different creatine phosphokinase (CPK) isoenzymes from the heart and the mechanism of the regulatory action of calcium ions on this reaction have been studied. It has been shown that the kinetic parameters of the reaction in the presence of calcium are similar to those in the presence of magnesium with the exception of the maximal rate of the reverse reaction, which is about three times lower in the presence of calcium. Calcium ions are able to exert a significant regulatory action on the CPK reaction. Simultaneous regulation of the CPK reaction by calcium and magnesium ions can be quantitatively described by a kinetic model, which takes into account the formation of complexes of adenine nucleotides with calcium and magnesium. The mechanism of the CPK reaction regulation by metal ions involves changes in concentrations of the metal -- adenine nucleotide compexes. The regulatory action of calcium on the creatine kinase reaction is the same for all CPK isoenzymes studied.
